특정 계통의 생쥐 초기 배아의 체외 배양때에 나타나는 "In Vitro 2-Cell Block" 현상을 규명할 것을 목적으로 하고 본 실험이 행해졌다. 먼저 이 현상이 발생하는 ICR 계통의 생쥐의 수정란 또는 2세포기의 배아를 일정시간 대 (배란을 유도하기 위한 hCG주사시간을 기준)를 두고 수란관으로부터 회수한 뒤 이를 3-4일간 배양하면서 배낭으로까지의 발생능력을 알아보았다. 그 결과 hCG주사 후 약 30시간이 지난 뒤 수란관에서 회수한 수정란이나 2세포기의 배아의 일부가 배낭으로까지 발생하였으며 만일 48시간이 지나면 수란관 내에서 회수된 배아는 대부분이 2세포기 배아이며 이것들은 거의 배낭으로 발생하였다. HCG 주사 후 27시간이 지난 수란관으로부터 회수한 수정란을 2시간에서 24시간을 배양한 뒤 이들을 다시 수란관에 이식하여 기관배양법에 의해 72시간 배양하고 다시 수란관 밖에서 배아를 24시간 배양해 본 결과, 배아들이 수란관 밖의 환경에서 배양된 시간이 길수록 이것들을 다시 수란관내에 되돌려 준다 하더라도 배낭으로까지 발생할 능력을 크게 상실하였다. 이같은 실험 결과로 보아 생쥐의 수정란이 "2-Cell Block" 현상을 극복하기 위해서는 수정후 일정시간 이상을 수란관이라는 환경내에 머물러 있어야 한다는 것을 알게 되었다. 즉, 배아는 수란관에 오래 머물러 있을수록, 그리고 수란관으로부터 축출되더라도 다시 수란관으로 돌려보내질 때까지 밖에 머물러 있는 시간이 짧을수록, 수정란 혹은 2세포기 배아의 배낭으로의 발생능력은 정상에 가깝게 유지되는 것이다. "2-Cell Block"에 걸려있는 2세포기의 배아는 배양 후 24시간까지에는 광학현미경적인 관찰 결과 정상적인 형태를 보여주고 있었으나 48시간이 되면 핵의 이상응축이 나타나며 72시간이 경과하면 세포질 내에 비정상적인 공포들이 나타나고 있었다.

The cyutotoxic effects of hot water soluble polysaccharides extract(PS) from the mushroom, Lentinus edodes, and in combinations with vitamin A or vitamin E on life span of ICR mice bearing P388 cancer cells and in vitro against P388 cancer cells were examined. The chemical components of PS and fractions were analyzed and survival time and cell number of P388 treated with extract fractions with and without vatamin A or E supplementation were also measured. The results obtained were summarized as follows; The extract of fraction B was shown to have the highest antitumor activity against P388 implanted in ICR mice. The antitumor fraction B was consisted of 82.0% of polysaccharide and 4.2% of protein. All three fractions seemed to have in vivo antitumor activity against P388, and fraction B showed the highest activity, In vitro P388 cell growht was inhibited 76%, 89%, 54% by the addition of fraction A, B and C respectively. Vitamin A or E did not appear to have any accelerating effects on either in vivo or in vitro cell cytotoxicity when each of them was combined with the PS and fractions. All three fractions contained more than 68% of polysaccharides. The fraction B showed the highest value of 88% in polysaccharides. Monsaccharides of the fraction B were identified as galactose(59.1%), glucose(29.2%), fructose(2.8%) and uronic acid(4.2%). Hydrolysis of protein from the fraction B was didentifed to have 17 kinds of defined and 5 undifined amino acids. The inhibitory effects of the hot water extracts from mushroom against cancer cell growth of P388 were stronger than the control group. And the survival time of ICR mice was shown to be 161% between the control group and the experimental groups.

본 연구에서는 탄소성형 분리판의 물리화학적, 전기화학적 특성을 분석하여 수소 및 개미산연료 전지의 성능에 미치는 영향을 알아보았다. 기존 기계가공 탄소분리판과 탄소복합소재 탄소성형 분리판의 접촉저항, 부식특성, 소수성을 비교 평가하였다. 특히, 현재 연구개발 중인 탄소복합소재 성형분리판의 경우 계면접촉저항이 기계가공 분리판보다 1.5배 높게 나타났으며, 내식성 실험에서는 산에 취약하여 분리판 표면이 거칠어지고 결정성이 감소하였다. 연료전지의 성능은 분리판의 계면접촉저항에 크게 영향을 받았으며, 계면저항이 적은 기계가공 분리판이 수소 및 개미산 연료 전지에서 높은 성능을 나타냈다.

The study was carried out to obtain the basic data for types, incidence and histopathological features of the spontaneous tumors of the specific pathogen free (SPF) Ktc : ICR mice bred in a barrier system in Technology Research Center, Korea Research Institute of Chemical Technology. One hundred of the mice consisted of 50 males and 50 females were examined for 18 months. 1. The overall incidence rate of spontaneous tumors was 51(51%) of 100 heads tested. The male mice showed slightly higher incidence(28%) than the female(23%), and the incidence rate and the number of affected organs were increased with the increasing age of mice. 2. The incidence rate of primary tumor was 59(59%) of 100 heads tested, consisted of 30 cases (50.8%) of benign tumors and 29 cases(49.2%) of malignant tumors. Among the malignant tumors twenty cases were metastasized to various organs. 3. In tumor incidence rates by systems and organs, the male mice showed the high incidence rate in the liver (18%), hematopoietic system (16% ) and lung (14%), while the female mice, in the hematopoietic system(18%), lung(12%), liver(8%) and uterus(8%). 4. The tumors showing the particularly low incidence rates (<1.0%) were rhabdomyosarcoma in the skeletal muscle, malignant schwannoma in the peripheral nerve, cortical adenoma in the adrenal gland, transitional cell carcinoma in the urinary bladder, tubular cell adenoma in the kidney and adenoma in the pituitary gland and harderian gland.

Flavin mononucleotide (1,4-butanediamine) Pt(II) complex (7FMN) was synthesized and screened anticancer activity [J. Pharm. Soc. Korea 43(6),762-770 (1999)]. 7FMN have good water solubility and moderate anticancer activiy In this paper cell-cycle specificity and nephrotoxicity were studied. Interaction of DNA with cisplatin and synthesized 7FMN was analyzed by flow cytometry and showed G2 arrest in L1210 cell line. It means that cell-cycle on L1210 was inhibit in S phase by cisplatin and 7FMN. In order to biochemically analyze nephrotoxicity of cisplatin and 7FMN, after injecting each agent intraperitoneally, blood was exsanguinated after 6 hours, 1 day, 3 days and 7 days, respectively. Then, serum was separated from the blood. The serum level of BUN, creatinine and uric acid in cisplatin and 7FMN administated mice (25~35 g, ICR strain, a dose each 8,12 and 16 times of the $IC_{50}$/ value, cisplatin; 7 times) were determined by autochemistry analyzer. In cisplatinadministered mice group, BUN level was elevated than normal control group at 3rd day and repaired at 7th day. In 7FMN administrated group was not elevated. Creatinine and uric acid level were no difference with the normal control group. Therefore synthesized 7FMN is less toxic than cisplatin in nephrotoxiciaty.

The effects of Chelidonii Herba on the cell mediate and humoral immune responses were investigated in ICR mice. ICR mice received intraperitoneal injection of methanol extracts (ether fraction, buthanol fraction, water fraction) for 5 days before sensitization. The change of body weight (%), the organ weight ratio (%), the numbers of RBC and WBC, the ear swelling by dinitrofluorobenzene, the titers of hemagglutination (HA) and hemolysin (HY) to SRBC were determined. The results were as follows; 1. The change of body weight (%) showed a tendency of increasing generally. 2. The weight of spleen (%) revealed decreasing but that of thymus revealed increasing. 3. Ear swelling was maxium at 48 hours after challenge and was significantly decreased in the groups treated with Ether, Buthanol and Water. 4. The numbers of RBC and WBC were decreased generally. 5. HA and HY titers were decreased significantly.

The present study was carried out to investigate the viability of frozen-thawed aggregated mouse embryos and to produce the chimeras. Different phenotypic embryos were obtained by mating ICR female mice with ICR or CBA male mice. The aggregated morulae were produced following aggregation of embryos at 4-, 8- and 12- to 16-cell stage. The desirable stage for the aggregation of the mouse embryos was 8- to 16-cell stage. The post-thawed in vitro survival rates of aggregated embryos in glycerol, DMSO and ethylene glycerol were 51.5, 78.6 and 69.4%, respectively. Although the higher survival was obtained in DMSO, there were no significant differences in the survival rates among the three cryoprotectants. A total of 155 frozen-thawed aggregated embryos were transferred to 11 recipient mice, 3 out of 7 offsprings were born to overt chimera. These experiments have proven that mouse chimeras can be obtained from frozen-thawed aggregated embryos.

When a dielectric layer, in-cell retarder (ICR) is formed between electrode and LC layer to get a single gap transflective fringe-field switching (FFS) display, the operating voltage ($V_{op}$) is highly increased due to the thickness of dielectric material. But, we also knew the phenomenon that the increasing rate of Vop is different whether the 1st common electrode was composed of plate type or slit type. In this paper, the common electrode in transmissive part was composed of slit type which had less steepness effect of the Vop and in reflective part was composed of plate type. The rubbing angle of reflective part can be adjusted properly to match the voltage dependent transmittance and reflectance.
