시클로덱스트린과의 포접에 의한 케토코나졸의 용해성 및 용출 증가 (Improvement of Solubility and Dissolution of Ketoconazole by Inclusion with Cyclodextrin)

  • Inclusion complexes of ketoconazole (KT) with ${\alpha}-$, ${\beta}-cyclodextrin$ (CD) and dimethyl-${\beta}-cyclodextrin$ $(DM{\beta}CD)$ in a molar ratio of 1:2 were prepared by freeze-drying and solvent evaporation methods. The interactions of KT with ${\alpha}-CD$, ${\beta}-CD$ and $DM{\beta}CD$ in aqueous solution and in solid state were investigated by solubility study, infrared (lR) spectroscopy and differential scanning calorimetry (DSC). The stability constant of $KT-DM{\beta}CD$ inclusion complex (lC) was found to be the largest among three inclusion complexes. Clear differences in IR spectra and DSC curves were observed between inclusion complexes and physical mixtures (PM) of KT-CDs. It was also shown by IR spectra and DSC curves that solvent evaporation method might be. superior to the freeze-drying method in preparing the inclusion complexes of KT-CDs. The dissolution rate of KT was markedly increased by inclusion complex formation with CDs in the buffer solution at pH 4.0 and pH 6.8. The mean dissolution time (MDT,min), which represents the rapidity of dissolution, was in the order of $KT-DM{\beta}CD$ IC (3.20) < $KT-{\beta}-CD$ IC (4.36) < $KT-{\alpha}-CD$ IC (6.99) < $KT-{\alpha}-CD$ PM (17.46)< $KT-{\beta}-CD$ PM (19.36) < $KT-{\beta}-CD$ PM (28.53). The dissolution rates of KT-CD ICsprepared by solvent evaporation method were faster than those of KT-CD ICs prepared by freeze-drying method.

Gender Differences in Paediatric Patients of the Swiss Inflammatory Bowel Disease Cohort Study

  • Purpose: Gender differences in paediatric patients with inflammatory bowel disease (IBD) are frequently reported as a secondary outcome and the results are divergent. To assess gender differences by analysing data collected within the Swiss IBD cohort study database since 2008, related to children with IBD, using the Montreal classification for a systematic approach. Methods: Data on gender, age, anthropometrics, disease location at diagnosis, disease behaviour, and therapy of 196 patients, 105 with Crohn's disease (CD) and 91 with ulcerative or indeterminate colitis (UC/IC) were retrieved and analysed. Results: The crude gender ratio (male : female) of patients with CD diagnosed at <10 years of age was 2.57, the adjusted ratio was 2.42, and in patients with UC/IC it was 0.68 and 0.64 respectively. The non-adjusted gender ratio of patients diagnosed at ${\geq}10$ years was 1.58 for CD and 0.88 for UC/IC. Boys with UC/IC diagnosed <10 years of age had a longer diagnostic delay, and in girls diagnosed with UC/IC >10 years a more important use of azathioprine was observed. No other gender difference was found after analysis of age, disease location and behaviour at diagnosis, duration of disease, familial occurrence of IBD, prevalence of extra-intestinal manifestations, complications, and requirement for surgery. Conclusion: CD in children <10 years affects predominantly boys with a sex ratio of 2.57; the impact of sex-hormones on the development of CD in pre-pubertal male patients should be investigated.

  • Nonclassical aminobenz[cd]indole antifolates 4, 5 and 6, in which the glutamic acid moiety of the classical antifolates is substituted by 2-phenylglycinamide or 3-aminobenzamide, were synthesized and their in vitro antitumor activity was evaluated. The purpose of this substitution is that the lipophilicity is enhanced due to the aromatic ring of the target compounds for the passive transport through lipid membrane of cells while the hydrogen bonding of the amide is retained in the active site of the enzyme, thymidylate synthase, where the glutamate is originally present. The target compounds were highly cytotoxic against tumor cell lines of murine and human origin with micromolar to nanomolar $IC_{50}$ values. Most effective was compound 4 ($N^6-methyl-N^6$-[4-[(${\alpha}$(S)-aminocarbonylbenzyl) aminocarbonyl]benzyl]-2,6-diaminobenz[cd]indole)with $IC_{50}$ of 2 nM against SW480, human colon adenocarcinoma cell line, which is 650-fold more potent than the reference compound 3.

연속회분식반응조 공정에서 슬러지 체류시간과 중금속 독성의 관계 (Relationship between Toxicity of Heavy Metals and Sludge Retention Time in Sequencing Batch Reactor Process)

  • 연속회분식반응조(SBR) 공정에서 슬러지 체류시간(SRT)에 따른 중금속의 독성도 변화를 측정하였다. 중금속은 구리(Cu), 카드뮴(Cd) 및 아연(Zn)을 사용하였고, SRT는 $2\sim30$일로 변화시켰으며, 독성도는 INT-dehydrogenase 활성도의 변화로 측정하였다. 중금속의 농도가 증가함에 따라 독성도가 증가하였으며, Cu가 Zn 및 Cd 보다 독성도가 높았다. SRT를 변화시켰을 때 $IC_{50}$ 값이 Cu의 경우 $0.37\sim1.96$ mg/L의 범위를 나타내었으며, Cd의 경우는 $15.4\sim16.9$ mg/L를 나타내었다. 또한 Zn의 경우는 $9.70\sim23.4$ mg/L의 범위를 나타내었다. Cu와 Zn의 경우, SRT가 증가함에 따라 독성이 감소하였으며, 이는 긴 SRT에서 세포외 중합체의 농도가 증가하기 때문인 것으로 판단된다. 따라서 중금속을 포함한 산업폐수를 처리하는 SBR 공정에서 SRT를 길게 운영하는 것이 바람직할 것으로 판단된다.

A VLSI DESIGN OF CD SIGNAL PROCESSOR for High-Speed CD-ROM

  • We implemented a CD signal processor operated on a CAV 48-speed CD-ROM drive into a VLSI. The CD signal processor is a mixed mode monolithic IC including servo-processor, data recovery, data-processor, and I-bit DAC. For servo signal processing, we included a DSP core, while, for CAV mode playback, we adopted a PLL with a wide recovery range. Data processor (DP) was designed to meet the yellow book specification.[2]So, the DP block consists of EFM demodulator, C1/C2 ECC block, audio processor and a block transferring data to an ATAPI chip. A modified Euclid's algorithm was used as a key equation solver for the ECC block To achieve the high-speed decoding, the RS decoder is operated by a pipelined method. Audio playability is increased by playing a CD-DA disc at the speed of 12X or 16X. For this, subcode sync and data are processed in the same way as main data processing. The overall performance of IC is verified by measuring a transfer rate from the innermost area of disc to the outermost area. At 48-speed, the operating frequency is 210 ㎒, and this chip is fabricated by 0.35 um STD90 cell library of Samsung Electronics.

Toxic Effects of Heavy Metals on the Growth and Phosphorus Removal Efficiency of Phosphorus Accumulating Microorganisms (PAOs)

  • Phosphorus accumulating microorganisms (PAOs) are influenced by various environmental factors and heavy metals. This study was performed to evaluate the effects of the selected heavy metals on the growth and phosphorus removal capacity of Bacillus sp. 3434 BRRJ, Pseudomonas aerunogisa, and Bacillus Subtilis, well known as PAOs. The heavy metals used in this study included Cu, Cd, As, and Zn. The $IC_{50}$ (median inhibition concentration) values of Bacillus sp. 3434 BRRJ for the Cu, Cd, As, and Zn were 8.07 mg $L^{-1}$, 0.18 mg $L^{-1}$, 73.62 mg $L^{-1}$ and 0.25 mg $L^{-1}$, respectively. The $IC_{50}$ values of Pseudomonas aerunogisa for the Cu, Cd, As, and Zn were 4.45 mg $L^{-1}$, 0.16 mg $L^{-1}$, 18.51 mg $L^{-1}$ and 2.34 mg $L^{-1}$, respectively. The $IC_{50}$ values of Bacillus Subtilis for the Cu, Cd, As, and Zn were 3.81 mg $L^{-1}$, 0.18 mg $L^{-1}$, 11.31 mg $L^{-1}$ and 0.47 mg $L^{-1}$, respectively. The phosphorus removal efficiencies of the three bacteria, Bacillus sp. 3434 BRRJ, Pseudomonas aerunogisa, and Bacillus subtilis were 93.12%, 71.81%, and 65.31%, respectively. Based on the results of the three PAOs obtained from the study, it appears that Bacillus sp. 3434BRRJ may have the best results in terms of their growth rate and P removal efficiencies.

Alcaligenes sp.의 생장과 인 제거에 미치는 이종 중금속 혼합의 독성 효과 (Toxic Effects of Binary Mixtures of Heavy Metals on the Growth and P Removal Efficiencies of Alcaligenes sp.)

  • 본 연구는 인 축적능을 갖는 것으로 알려진 미생물인 Alcaligenes sp.의 생장 및 인 축적에 중금속이 미치는 영향을 정량적으로 평가하고자 실시되었다. 미생물의 생장 및 인 제거에 미치는 저해효과를 정량적 평가를 위해 생장저해 정도와 인 제거반응저해 정도를 IC50와 EC50를 이용하여 표현하였고, 중금속 간 교호작용의 평가를 위해 Additive Index Value method (A.I.V) 를 이용하여 중금속 간 혼합효과를 판단하였다. 실험 결과를 통해 Alcaligenes sp.의 생장 및 인 제거에 미치는 저해효과가 가장 큰 중금속은 카드뮴(Cd)이었다. 반면, 상대적으로 저해효과가 약한 중금속은 니켈(Ni)이었다. 모든 중금속은 처리 농도가 증가할수록 미생물의 생장과 인 제거율을 감소시키는 것을 확인할 수 있었다. 단일 중금속 처리시 IC50에 해당하는 농도는 Cd 0.38, Cu 16.43, Zn 12.17, Pb 11.68, Ni 15.35 mg/L 이다. 연구에 사용된 균주 Alcaligenes sp. 의 인 제거율은 88.1%였고, EC50에 해당하는 농도는 Cd 0.31, Cu 10.37, Zn 5.57, Pb 6.74, Ni 12.04 mg/L 이다. 두 종의 중금속을 혼합 처리하였을 때 IC50에 해당하는 농도가 가장 낮은 중금속 처리구는 Cd + Cu 처리구였으며, 가장 높은 처리구는 Zn + Pb 처리구였다. EC50의 농도는 Cd + Cu 처리구에서 가장 낮은 농도를, Cu + Ni 처리구에서 가장 높은 농도를 보였다. 중금속 간 교호작용은 대부분 길항효과(antagonistic effect)를 보였으나, IC50의 경우, Cu + Zn, Cu + Pb, Cu + Ni, Pb + Ni 처리구에서 상승효과(synergistic effect)를 보이는 것을 확인할 수 있었고, EC50에서는 Cu + Zn, Pb + Ni 처리구에서 상가효과(additive effect)를, Cu + Ni 처리구에서 상승효과(synergistic effect)를 보였다. 연구 결과는 중금속을 단일 및 2종 혼합 처리하였을 때 중금속이 인 제거 미생물의 생장 및 기능에 미치는 독성효과에 대한 기초자료를 제공함으로써 추후 진행될 미생물 단백질 동정, 중금속 독성평가에 관한 연구에 활용될 수 있을 것이라 사료된다.

Characterization of Dye Decolorization in Cell-Free Culture Broth of Trametes versicolor CBR43

  • The dye decolorization rate in a cell-free culture broth of the white-rot fungus Trametes versicolor CBR43 was studied, including the effects of inhibitors of NaCl, Zn(II), and Cd(II) on dye decolorization activity. The maximum rates of dye decolorization in cell-free culture broth were 1,410, 44.7, 41.2, and $0.19{\mu}mol{\cdot}l^{-1}{\cdot}min^{-1}$ for Acid Blue 62, Acid Black 175, Reactive Blue 4, and Acid Red 114, respectively. The inhibition effects of NaCl, Zn(II), and Cd(II) on dye decolorization were quantitatively compared using the half maximal inhibition concentration ($IC_{50}$), which indicates the concentration of an inhibitor required for 50% inhibition. Based on $IC_{50}$ values, dye decolorization in the cell-free culture broth of CBR43 was most potently inhibited by Cd(II), whereas the inhibitory effect of NaCl was relatively low. The dye decolorization rates and $IC_{50}$ data can be used in the design and development of a dye-wastewater treatment process using T. versicolor CBR43 and its operating factors.

IC-MS/MS를 이용한 수중의 과염소산이온 분석방법 최적화 및 한강수계내 분포조사 (Determination of Trace-Level Perchlorate by IC-MS/MS and Distribution in the Han River)

  • 과염소산이온의 분석을 위해 전기전도도 검출기(CD; conductivity detector)를 연결한 이온크로마토그래피(IC; ion chromatography) 방법이 지금까지 가장 널리 사용되어 왔다. 그러나 이 방법은 시료의 조성에 영향을 받을 수 있고, 정량한계를 낮추는데 한계가 있다. 따라서, 본 연구에서는 미량의 과염소산이온 분석을 위해 이온크로마토그래피 질량분석법(IC-MS/MS; ion chromatography mass spectrometry)을 제시하였다. IC-MS/MS를 이용한 과염소산이온의 분석방법을 적용한 결과 평균회수율 104.4 ${\pm}$ 5.7%, 상대표준편차 1.9 ${\pm}$ 1.3%를 얻었으며, 검출한계(MDL; method detection limit)는 0.0207 ${\pm}$ 0.0099 ${\mu}g/L$를 얻을 수 있었다. IC-MS/MS를 한강수계 지류천과 본류에 적용시킨 결과, 최소 <0.1 ${\mu}g/L$에서 최고 18.30 ${\mu}g/L$ 범위로 나타났다. 서울시 6개 아리수정수센터의 취수원수와 정수에서는 과염소산이온이 0.18~0.34 ${\mu}g/L$의 농도로 존재하는 것으로 나타났다.

Effect of Cadmium on $C_6$ Glioma Cells in Culture

  • It is demonstrated that cadmium has cytotoxic effect on glial cells, oxygen radicals are involved in cadmium-induced cytotoxicity. However, the toxic mechanism of cadmium is left unknown so far. The purpose of this study was to examine the cytotoxicity of $CdCl_2$ on $C_6$ glioma cells. The cytotoxicy was measured by cell viability via XTT assay in $C_6$ glioma cells. Colorimetric assay such as XTT assay is regarded as a very sensitive screening method for the determination of the cell viability on a lots of chemicals. In this study, $CdCl_2$ decreased cell viability according to the dose- and time dependent manners after $C_6$ glioma cells were treated with various concentrations of $CdCl_2$ for 48 hours. $IC_{90}\;and\;IC_{50}$ values for XTT assay was determined at $5{\mu}M\;and\;55{\mu}M$ of $CdCl_2$, respectively. These results suggest that $CdCl_2$ has highly cytotoxic effect on $C_6$glioma cells by the decrease of cell viability.
