Construction of a Fusion-Stoffel Fragment to Improve 3′-5′Exonuclease Activity

  • Taq DNA polymerase exhibits a sizable drawback compared to the other thermophilic DNA polymerases in that it demonstrates lower proof-reading activity due to the deficiency of 3'-5'exonuclease activity. A study was undertaken to improve the 3'-5' exonuclease activity in the PCR of Taq DNA polymerase. The three-dimensional structural alignment of the polymerase and 3'-5' exonuclease domains from the pol I family DNA polymerases explains why Taq DNA polymerase has just a background level of 3'-5'exonuclease activity. A comparison indicated that the two polymerase domains are very similar in primary and tertiary conformations, even though Taq DNA polymerase carries a much shorter 3'-5'exonuclease domain than that of E. coli DNA polymerase I. Those two polymerase domains were interchanged between Taq DNA polymerase and E. coli DNA polymerase I. The 3'-5' exonuclease domain from E. coli DNA polymerase I was separated and pasted into the polymerase domain of Taq DNA polymerase I, which resulted in a functional fusion-Stoffel fragment. The 3'-5'exonuclease activity of the fusion-Stoffel fragment increased up to 48% of the value of the Klenow fragment, while that of Taq DNA polymerase remained at 6.0% of the Klenow fragment.

A Study on the I-Ching Perspective in Shanghansuyuanji (『상한소원집(傷寒溯源集)』에 나타난 역학적(易學的) 관점 고찰)

  • Objectives : This paper aims to study the I-Ching perspective in Shanghansuyuanji. Methods : Key words associated with I-Ching was searched in Shanghansuyuanji and analyzed its meaning and categorized thematically. Results & Conclusions : 1. Qianhuang symbolized Shaoyang, The Power of the Great hexagrams, Jueyin Peace hexagrams, Taiyin Gen Earth, Yangming Kun Earth, Shaoyin Return hexagrams, but didn't symbolize Taiyang, making it not uniform. 2. Qianhuang explained physiology focusing the change of YangQi with I-Ching perspective. This is possible because Shanghanlun thought that much of YangQi and I-Ching set a high value on Yang. 3. Qianhuang explained pathology such as insomnia, epigastric fullness, splenic constipation syndrome, Taiyinbing syndrome, somnolence, thirst, YinYang exchange with I-Ching perspective, it is meaningful because it enriches medical YinYanglun. 4. Qianhuang explained prescriptions such as Daqinglongtang, Zhenwutang, Shizaotang, Fuzixiexintang, Dachaihutang, Baihutang, a comparison between Daqinglongtang and Xiaoqinglongtang with I-Ching perspective. This is helpful to grasp the image of prescriptions. 5. Qianhuang explained nature of drugs such as Guizhi, Fuling, Fuzi, Qiandan, Rougui with I-Ching perspective, and it combines well with the features of I-Ching with drugs efficacy. 6. Qianhuang explained diseases of the six Meridians curing time with I-Ching perspective. This shows diseases of the six Meridians recover when Corresponding Qi is vigorous or Conflict Qi appears, and it is persuasive. For reasons mentioned above; Shanghansuyuanji is meaningful as Shanghanlun commentary.

Characterization of the Restriction Endonuclease Bki I from Brevibacterium divaricatum (Brevibacterium divaricatum의 제한효소 Bdi I의 특성)

  • A new type II restriction endonuclease, Bdi I, has been isolated from Brenibacterium divaricatum FERM 5948 by procedures of ammonium sulfate fractionation, DEAE-cellulose chromatography and heparin agarose chromatography. The purified Bdi I restriction endonudlease had the same cleavage patterns of Cla I whose recognition sequence is 5' ATCGAT 3'. From the result that ${\lambda}-Cla$ I DNA frahment could be cloned in pBR 322 digested with Bdi I, it has been proven that Bdi I cuts between T and C(5' AT/CGAT3') within the recognition sequence and produces 5'pCG cohesive end. The optimal temperature for the Bdi I restriction endonuclease activity was $37^{\circ}C$, and optimal salt (NaCl) concentration was 50-100 mM.

  • Based on the elegant properties of the spectral norm and Thompson metric, we firstly give two perturbation estimates for the positive definite solution of the nonlinear matrix equation $$X-\sum^m_{i=1}A^{\ast}_iX^{\delta_i}A_i=Q(0<|{\delta}_i|<1)$$ which arises in an optimal interpolation problem.

Benzo(a)pyrene-Triglyceride Adduct: a Potential Molecular Biomarker for Carcinogen Exposure

  • In this study, we demonstrated the in vitro and in vivo formation of carcinogen-lipid adduct and its correlation with DNA or protein adducts. The lipids from serum or hepatocyte membranes of Spragu-Dawley rats. human serum, and standard major lipids were in vitro reacted with benzo[a]pyrene(BP) and BP metabolites. 7,8-Dihydroxy-9,10-epoxy-7,8,9,10-tetrahydrobenzo[a]-pyrene(BPDE-I), an ultimate carcinogenic form of BP, was covalently bound to triglyceride(TG). BPDE-I-TG adducts isolated by thin-layer chromatography (TLC) were further detected by high performance liquid chromatography(HPLC). TGs, including triolein, tripalmitin and tristearin, showed positive reactions with BPDE-I. However, cholesterol, phospholipids(Phosphatidylcholine, phosphatidyl-ethanolamine, phosphatidyl-inositol and sphingomyelin) and nonesterified fatty acids(palmitic acid, oleic acid, linoleic acid and stearic acid) did not react with BPDE-I. In addition, other BP metabolites (BP-phenols and -diols) did not react with TG, which TG appeared to be the most reactive lipid yet studied with respect to its ability to form an adduct with BPDE-I. There was a clear-cut dose-respect to its ability to form an adduct with BPDE-I-lipid adduct in vitro between TG and [1,3-3H]BPDE-I. In an animal study, BPDE-I-TG was also formed in the serum of rats orally treated with BP(25 mg/rat). Also, obvious correlations between [3H]BP related-biomolecule adducts (DNA, protein) or lipid damage and the BPDE-I-TG adduct were obtained in various tissues of mice i.p. treated with [3H]BP. These data suggest that TG can form an adduct with BPDE-I, as do other macromolecules (DNA, RNA, and protein). Therefore, a carcinogen-lipid adduct would be a useful biomarker for chemical carcinogenesis research and cancer risk assessment.

Comparison of the Quality of Various Polychromatic and Monochromatic Dual-Energy CT Images with or without a Metal Artifact Reduction Algorithm to Evaluate Total Knee Arthroplasty

  • Objective: To compare the quality of various polychromatic and monochromatic images with or without using an iterative metal artifact reduction algorithm (iMAR) obtained from a dual-energy computed tomography (CT) to evaluate total knee arthroplasty. Materials and Methods: We included 58 patients (28 male and 30 female; mean age [range], 71.4 [61-83] years) who underwent 74 knee examinations after total knee arthroplasty using dual-energy CT. CT image sets consisted of polychromatic image sets that linearly blended 80 kVp and tin-filtered 140 kVp using weighting factors of 0.4, 0, and -0.3, and monochromatic images at 130, 150, 170, and 190 keV. These image sets were obtained with and without applying iMAR, creating a total of 14 image sets. Two readers qualitatively ranked the image quality (1 [lowest quality] through 14 [highest quality]). Volumes of high- and low-density artifacts and contrast-to-noise ratios (CNRs) between the bone and fat tissue were quantitatively measured in a subset of 25 knees unaffected by metal artifacts. Results: iMAR-applied, polychromatic images using weighting factors of -0.3 and 0.0 (P-0.3i and P0.0i, respectively) showed the highest image-quality rank scores (median of 14 for both by one reader and 13 and 14, respectively, by the other reader; p < 0.001). All iMAR-applied image series showed higher rank scores than the iMAR-unapplied ones. The smallest volumes of low-density artifacts were found in P-0.3i, P0.0i, and iMAR-applied monochromatic images at 130 keV. The smallest volumes of high-density artifacts were noted in P-0.3i. The CNRs were best in polychromatic images using a weighting factor of 0.4 with or without iMAR application, followed by polychromatic images using a weighting factor of 0.0 with or without iMAR application. Conclusion: Polychromatic images combined with iMAR application, P-0.3i and P0.0i, provided better image qualities and substantial metal artifact reduction compared with other image sets.

  • The Fe(II)-isothiocyanato complex $trans-[FeH(NCS)(dppe)_2]$ (1) eactedwith iodomethane(Mel) to give methyl isothiocyanide-Fe(n) complex, $trans-FeH(NCS(Me)-S)(dppe)_2]I(2)$. Compound 2 was oxidized to $trans-[Fe(NCS)_2(Ph_2P(O)CH_2CH_2P(O)Ph_2)_2][I_3]$ (3), which was structurally characterized by X-ray diffraction. The molecular structure of 3 showed a bent Fe-NCS group, Crystallographic data for 3: triclinic space group P1,a=11.071(2) A,b=12.054(2)A,c=12.121(1)A, $\alpha=101.02(1){\circ}C{\beta}=95.887(9){\circ}Cr=110.34(1){\circ}C$, $Z=1R(wR_2)=0.0567(0.1294)$.

  • A sensitive spectrophotofluorometric method was developed for the analysis of 2[[3-(trifluoromethyl)phenyl]amino]-3-phridine carboxylic acid (I) in urine. The method is based on the fluoroscence behavior of the I-aluminum complex in absolute ethanol. This fluorophore has activation and emission wavelengths of 355 and 450 nm, respectively. Optimum conditions for the reaction were investigated. The fluorescence was linear in the range of 0.25 3.0 ug of I/ml. Replicate studies of spiked urine samples, each containing 2.5 ug of I/ml showed good precision with a relative standard deviation of 0.019. Overall recovery percent from five spiked urine samples was 99.4 $\pm$1.32%.

Let ${\mu}_{[k]}$ be the largest of all ${\mu}$'s and the population ${\pi}_{i}$ is defined to be 'good' if ${\mu}_{i}\;{\geq}\;{\mu}_{[k]}\;-\;{\delta}_1$, where ${\delta}_1\;>\;0$, i = 1,...,$textsc{k}$. A selection procedure based on sample median is proposed to select a subset of $textsc{k}$ logistic populations which includes all the good populations with probability at least $P^{*}$(a preassigned value). Simultaneous confidence intervals for the differences of location parameters, which can be derived with the help of proposed procedures, are discussed. If a population with location parameter ${\mu}_{i}\;<\;{\mu}_{[k]}\;-\;{\delta}_2({\delta}_2\;>{\delta}_1)$, i = 1,...,$textsc{k}$ is considered 'bad', a selection procedure is proposed so that the probability of either selecting a bad population or omitting a good population is at most 1­ $P^{*}$.
