수지상세포에 있어서 베로독소 수용체의 발현조절 (Regulation of Gb3 Expression on Dendritic Cells)

  • Shiga-like toxin(SLT)을 생산하는 Esherichia coli에 의한 감염은 설사, 출혈성 대장염(hemorrhagic colitis) 및 용혈성 요독 증후군(hemolytic uremic syndrome)을 특징으로 하며, 특히 5세 이하의 소아에게서 심각한 결과를 초래한다. SLT-I의 병인으로는 다양한 숙주 매개 인자들이 알려져 있다. 본 연구에서는 E. coli 0157:H7(ATCC 43890)로 부터 정제한 SLT-I이 포유동물 세포들에 대한 세포독성과 종양괴사인자(tumor necrosis $factor-{\alpha},\;TNF-{\alpha}$)의 생산에 미치는 효과를 측정하였으며, SLT-I의 수용체인 glycolipid globotriaosylceramide(Gb3)의 발현과 SLT-I의 세포독성의 관계를 규명하고자 하였다. SLT-I 과 SLT-I B를 순수분리 정제하고 SLT-I B-FLTC 접합체를 제조하여 vero 세포, 대식세포 및 수지상세포를 대상으로 세포독성능을 측정하고 세포독성능의 차이가 SLT-I의 수용체인 Gb3의 발현과 상관관계가 있는지를 Flow cyotmetry로 분석하였다. 또한 대식세포의 종양괴사인자 생산능은 ELISA법으로 시행하였다. SLT-I은 대식세포(Raw264.7)로부터 $TNF-{\alpha}$의 생산을 증가시켰다. 연구 대상 세포 중 SLT-I에 감수성을 나타낸 Vero 세포와 수지상세포(dendritic cells)는 Gb3 발현이 각각 83%와 68%로 높았으며, 29%의 낮은 Gb3 발현을 보인 Raw264.7 세포는 감수성을 보이지 않았다. 따라서 위의 결과로부터 SLT-I에 감수성을 보이지 않은 Raw264.7세포를 대상으로 Gb3 발현 정도와 SLT-I의 세포독성의 관계를 규명하고자 Gb3의 발현을 증가시킨 후 SLT-I의 세포독성을 재차 평가하였다. 이 결과 $TNF-{\alpha}$의 처리에 의하여 6 hrs에 Gb3의 발현이 정점(43.5%)에 이르렀으며 36 hrs에 정상 수준(25.0%)으로 환원되었다. 그러나, Gb3의 발현이 증가함에도 불구하고 SLT-I의 세포독성에는 변화가 관찰되지 않았다. 따라서, SLT-I에 의한 세포독성은 세포의 종류에 따라서 다르며 또한, Gb3의 발현정도에만 의존적이지는 않을 것으로 생각된다. 이와 같은 결과는 E. coli 0157의 감염증 병인 연구에 있어 SLT-I과 Gb3의 발현의 상관관계에 대한 보다 심도 있는 연구가 필요함을 시사한다.

THE ARTINIAN COMPLETE INTERSECTION QUOTIENT AND THE STRONG LEFSCHETZ PROPERTY

  • It has been little known when an Artinian (point) quotient has the strong Lefschetz property. In this paper, we find the Artinian complete intersection quotient having the SLP. More precisely, we prove that if ${\mathbb{X}}$ is a complete intersection in ${\mathbb{P}}^2$ of type (2, 2) and ${\mathbb{Y}}$ is a finite set of points in ${\mathbb{P}}^2$ such that ${\mathbb{X}}{\cup}{\mathbb{Y}}$ is a basic configuration of type (2, a) with $a{\geq}3$ or (3, a) with a = 3, 4, 5, 6, then $R/(I_{\mathbb{X}}+I_{\mathbb{Y}})$ has the SLP. We also show that if ${\mathbb{X}}$ is a complete intersection in ${\mathbb{P}}^2$ of type (3, 2) and ${\mathbb{Y}}$ is a finite set of points in ${\mathbb{P}}^2$ such that ${\mathbb{X}}{\cup}{\mathbb{Y}}$ is a basic configuration of type (3, 3) or (3, 4), then $R/(I_{\mathbb{X}}+I_{\mathbb{Y}})$ has the SLP.

  • Let R be a ring. It is well-known that R is NI if and only if ${\sum}^n_{i=0}Ra_i$ is a nil ideal of R whenever a polynomial ${\sum}^n_{i=0}a_ix^i$ is nilpotent, where x is an indeterminate over R. We consider a condition which is similar to the preceding one: ${\sum}^n_{i=0}Ra_iR$ contains a nonzero nil ideal of R whenever ${\sum}^n_{i=0}a_ix^i$ over R is nilpotent. A ring will be said to be quasi-NI if it satises this condition. The structure of quasi-NI rings is observed, and various examples are given to situations which raised naturally in the process.

  • Ten, heretofore unreported, $ 5^I-methyl-5^I-[2-(5-substituted uracil-1-yl)ethyl)]-2^I-oxo-3^I$-methylenetetrahydrofurans (H, F, Cl, Br, I, $ CH_3$,$CF_3$,$CH_2CH_3$,$ CH=CH2$, SePh) (7a-j) were synthesized and evaluated against four cell lines (K-562, FM-3A, P-388 and U-937). For the preparation of ${\alpha}$-methylene-${\gamma}$-butyrolactone-linked to 5-substituted uracils (7a-j), the convenient Reformasky type reaction was employed which involves the treatment of ethyl ${\alpha}$-(bromomethyl)acrylate and zinc with the respective 1-(5-substituted uracil-1-yl)-3-butanone (6a-j). The 5-substituted uracil ketones (6a-j) were directly obtained by the respective Michael type reaction of vinyl methyl ketone with the $K_2CO_3$(or NaH)-treated 5-substituted uracils (5a-j) in the presence of acetic acid in the DMF solvent. The .alpha.-methylene-.gamma.-butyrolactone compounds showing the most significant antitumor activity are 7e, 7f, 7h and 7j (inhibitory concentration $(IC_50)$ ranging from 0.69 to $2.9 {\mu}g/ml$), while 7b, 7g and 7i have shown moderate to significant activity. The compounds 7a, 7c and 7d were found to be inactive. The synthetic intermediate compounds 6a-j were also screened and found marginal to moderate activity where compounds 6b and 6g showed significant activity $(IC_50:0.4~2.8 {\mu}g/ml)$.

고온연소로를 이용한 방사성 폐기물 내 I-129 정량 분석법 최적화 연구 (Optimization for I-129 analytical method of radioactive waste sample using a high-temperature combustion tube furnace)

  • 원자력 이용시설에서 발생하는 방사성 폐기물의 처분을 위해서 129I 와 같이 긴 반감기를 지니는 핵종의 농도를 결정하는 것은 매우 중요하다. 특히, 토양과 콘크리트와 같은 고체 시료내의 방사성 핵종들을 분석하기 위해서는 시료 중의 관심 핵종만을 효과적으로 분리하고 정제하는 과정이 필수적이다. 본 연구에서는 고온 연소로를 이용하여 고체 시료 중 129I를 분석하는 절차를 확립하였다. 시료에서 휘발된 129I은 환원제(NaHSO3)를 첨가한 0.01 M HNO3으로 포집 되어 ICP-MS로 신속하게 측정할 수 있었다. 이때, 시료에 첨가한 129I의 회수율을 높이고자 연소온도, 이동상 가스의 종류, 촉매 그리고 포집용액과 같은 연소로 분석 조건들을 최적화하였다. 또한, 본 연구에서 확립된 129I의 분석조건을 다른 휘발성 핵종(3H, 14C)의 동시분석에 적용할 수 있도록 최적화하였다. 최종적으로 고온 연소로를 사용하여 휘발성 핵종들을 분리한 후, 이들을 LSC (3H, 14C)와 ICP-MS (129I)로 각각 측정하는 분석 절차의 유효성을 평가하였다.

방사선조사가 MC3T3-E1 조골세포주의 type I collagen과 alkaline phosphatase mRNA 발현에 미치는 영향 (The effects of irradiation on the mRNA expression of type I collagen and alkaline phosphatase in the MC3T3-E1 osteoblastic cell line)

  • Purpose: To investigate the effects of irradiation on the phenotypic expression of the MC3T3-El osteoblastic cell line, particularly an the expression of type I collagen and alkaline phosphatase mRNA. Materials and Methods: Cells were irradiated with a single dose of 0.5, 1, 2, 4, and 8 Gy at a dose rate of 5.38 Gy/min using a cesium 137 irradiator. The specimens were then harvested and RNA extraction was carried out at 1 and 3 days after irradiation. The extracted RNA strands were reverse-transcribed and the resulting cDNA fragments were amplified by PCR. Results: The irradiated cells demonstrated a dose-dependent increase in type I collagen mRNA expression relative to the control group, with a maximum level of type I collagen mRNA expression occurring at 8 Gy. The degree of type I collagen mRNA expression increased significantly at 1 day after irradiation, but little differences were found between the control group and at the 3rd day. The amount of alkaline phosphatase mRNA expression increased significantly at land 3 days after irradiation in the 1 Gy exposed group compared with the control group. Conclusion: The amount of type I collagen and alkaline phosphatase mRNA expression increased significantly 1 day after irradiation when compared with the control group.

마우스 폐암 세포에서 Insulin-Like Growth Factor-I (IGF-I) 및 IGF Binding Protein (IGFBP)의 역할 (The Role of Insulin-like Growth Factor I(IGF-I), and IGF Binding Protein (IGFBP) in Mouse Lung Cancer Cells)

  • 연구배경 : 세포성장에 관여하는 여러 growth factor중 IGF-I은 IGF-IR와 결합하여 세포증식을 유발하는 mitogen으로 알려져 있다. 대부분의 정상세포 및 암세포는 IGFBPs을 분비하는데 이들은 IGF-I과 결합하여 IGF-I의 세포증식 효과를 증가 혹은 억제시킨다. 마우스 폐암세포주 (3LL)에서 IGF-I이 세포성장에 미치는 효과를 보고, 3LL 세포에서 분비되는 IGFBP을 추출 확인하고, IGFBPs이 세포 성정에 미치는 영향을 관찰하였다. 방 법 : 마우스 폐암세포주 (3LL)를 이용하여, IGF-I을 투여하여 세포성장을 MTT assay로 측정하였고, 3LL에서 분비되는 IGFBP을 추출하여 Western ligand blot 및 western immunoblot으로 확인하였다. 또한 분비된 IGFBP이 세포성장에 미치는 영향을 보기위해, anti-IGFBP antibody을 첨가하여 이의 기능을 억제하여 세포성장에 관한 기능을 관찰하였다. 결 과 : IGF-I은 serum free media에서 3LL 세포성장을 증가시켰다. 3LL 세포는 IGFBP-4를 생성하는 것을 확인하였고, anti-IGFBP-4 antibody를 첨가시 세포성장이 증가된 소견이 관찰되어 IGFBP-4는 세포증식을 억제하는 기능을 가지고 있음을 간접적으로 알 수 있었다. 결 론 : 이상의 실험 결과로 3LL 마우스 폐암세포에서 IGF-I은 세포성장을 증가시키며, 3LL에서 생성된 IGFBP-4는 세포증식을 억제하는 기능을 가지고 있다는 것을 관찰하였다. 향후 IGF-I과 IGFBPs이 암 성장에 미치는 기전과 임상적 적용에 대한 연구가 필요하리라 생각된다.

간호사의 수분 섭취배설량 측정에 대한 인식도와 수행도 (Nurses' Perception and Practice of Fluid Intake and Output Measurement)

  • Purpose: This research aimed to identify levels of nurses' perception and practice of fluid intake and output (I & O) measurement and to explore the relationship between perception and practice of it. Methods: Using a cross-sectional survey design, 195 nurses who practiced fluid I & O measurement were recruited from a general hospital. Nurses who agreed to participate in this study completed a structured study questionnaire to assess their levels of perception and practice of fluid I & O measurement. Results: A level of perception of I & O measurement was high (3.46 points out of 5), and scores for 3 subdomains of I & O (importance, accuracy, and efficacy) were evenly high. The level of practice of I & O was fairly high (3.76 points out of 5). Perception and practice of I & O were highly correlated (r=.73, p<.001). Conclusion: Nurses seem to have higher levels of perception and do practice fluid I & O measurement correctly. In order to have reliable and valid I & O measures, nurses need to have continuous education on I & O measurement based on clinical guideline to utilize it as an invaluable clinical instrument.

Insulin-Like Growth Factor-I-Induced Androgen Receptor Activation Is Mediated by the PI3K/Akt Pathway in C2C12 Skeletal Muscle Cells

  • Although insulin-like growth factor-I (IGF-I) and androgen receptor (AR) are well known effectors of skeletal muscle, the molecular mechanism by which signaling pathways integrating AR and IGF-I in skeletal muscle cells has not been previously examined. In this study, the role of PI3K/Akt on IGF-I-induced gene expression and activation of AR in skeletal muscle cells was investigated. C2C12 cells were treated with IGF-I in the absence or presence of inhibitors of PI3K/Akt pathway (LY294002 and Wortmannin). Inhibition of the PI3K/Akt pathway with LY294002 or Wortmannin led to a significant decrease in IGF-I-induced AR phosphorylation and total AR protein expression. Furthermore, IGF-I-induced AR mRNA and skeletal ${\alpha}-actin$ mRNA were blocked by LY294002 or Wortmannin. Confocal images showed that IGF-I-induced AR translocation from cytosol to nucleus was inhibited significantly in response to treatment with LY294002 or Wortmannin. The present results suggest that modulating effect of IGF-I on AR gene expression and activation in C2C12 mouse skeletal muscle cells is mediated at least in part by the PI3K/Akt pathway.

Ginsenoside Rg3 protects against iE-DAP-induced endothelial-to-mesenchymal transition by regulating the miR-139-5p-NF-κB axis

  • Background: Emerging evidence suggests that endothelial-to-mesenchymal transition (EndMT) in endothelial dysfunction due to persistent inflammation is a key component and emerging concept in the pathogenesis of vascular diseases. Ginsenoside Rg3 (Rg3), an active compound from red ginseng, has been known to be important for vascular homeostasis. However, the effect of Rg3 on inflammation-induced EndMT has never been reported. Here, we hypothesize that Rg3 might reverse the inflammation-induced EndMT and serve as a novel therapeutic strategy for vascular diseases. Methods: EndMT was examined under an inflammatory condition mediated by the NOD1 agonist, γ-d-glutamyl-meso-diaminopimelic acid (iE-DAP), treatment in human umbilical vein endothelial cells. The expression of EndMT markers was determined by Western blot analysis, real-time polymerase chain reaction, and immunocytochemistry. The underlying mechanisms of Rg3-mediated EndMT regulation were investigated by modulating the microRNA expression. Results: The NOD1 agonist, iE-DAP, led to a fibroblast-like morphology change with a decrease in the expression of endothelial markers and an increase in the expression of the mesenchymal marker, namely EndMT. On the other hand, Rg3 markedly attenuated the iE-DAP-induced EndMT and preserved the endothelial phenotype. Mechanically, miR-139 was downregulated in cells with iE-DAP-induced EndMT and partly reversed in response to Rg3 via the regulation of NF-κB signaling, suggesting that the Rg3-miR-139-5p-NF-κB axis is a key mediator in iE-DAP-induced EndMT. Conclusion: These results suggest, for the first time, that Rg3 can be used to inhibit inflammation-induced EndMT and may be a novel therapeutic option against EndMT-associated vascular diseases.
