Tripterygium regelii has been used as an oriental medicine, especially antiparasitic, anti-inflammatory and detoxifying agents in East asia. During our research to develop new antitumor agents from natural products, MeOH ext. and CH2Cl2 ext. of Tripterygium regelii showed the potent antitumor activity. In order to purify active compounds from Tripterygium regelii, activity-guided fractionation was carried out. Silica gel and RP-18 column chromatography for the active fraction led to the isolation of two compounds and their antitumor activities were studied. Those two compounds didn't show potent antitumor activity against human tumor cell lines. The structure of two compounds were determined by $^1H-NMR$, $^{13}C-NMR$, DEPT, $^1H-^{13}C$ COSY and IR spectrum. Compound I and Compound II were turned out to be Celastrol, and ${\beta}-sitosteryl-3-o-{\beta}-D-glucopyranoside$ respectively.
The evaluation of the anti-inflammatory analgesic and antipyretic activities of loxoprofen sodium given in intramuscular route was investigated as compared to oral application in rats and mice. The intramuscular $ED_{50}$ values of loxoprofen sodium in carrageenan edema and vascular permeability tests are 1.15 and 7.8 mg/kg, respectively, which represent more potent than in case of oral application. Its therapeutic effects in adjuvant arthritis were shown at 6 mg/kg l.m. and 3mg/kg p.o. Analgesic effect was shown to be more potent as given intramuscularly. Similar potency of antipyretic effects was shown in both administration routes. Considerably weak gastric damages were observed in intramuscular application.
4-(4-Methoxyphenyl)-2-aminothiazole and 3-(4-methoxyphenyl)-5-aminothiadiazole derivatives have been synthesized and evaluated as selective antagonists for human adenosine A$_3$ receptors. A methoxy group in the 4-position of the phenyl ring and N-acetyl or propionyl substitutions of the aminothiazole and aminothiadiazole templates displayed great increases of binding affinity and selectivity for human adenosine A$_3$ receptors. The most potent A$_3$ antagonist of the present series, N-[3-(4-methoxy-phenyl)-[1,2,4]thiadiazol-5-yl]-acetamide exhibiting a K$\_$i/ value of 0.79 nM at human adenosine A$_3$ receptors, showed antagonistic property in functional assay of cAMP biosynthesis involved in one of the signal transduction pathways of adenosine A$_3$ receptors. Polypeptide growth factor belong to a class of potent biologic mediator which regulate cell differentiation, proliferation, migration and metabolism. 1,25-dihydroxyvitamin $D_3$ decrease cell proliferation, and stimulate alkaline phosphatase activity which express in osteoblast during cell differentiation period. IGF-I is known to stimulate cell proliferation and differentiation too. 1,25-dihydroxyvitamin $D_3$ is known to increase IGF-I binding sites and IGF binding protein which inhibite the effect of IGF. The purpose of this study is to evaluate potential role of IGF-I as mediator that control the action of 1,25-dihydroxyvitamin $D_3$. MC3T3-E1 cell were seeded $5{\times}10^5/ml$ at 100mm culture plate in ${\alpha}-MEM$ containing 10% fetal bovine serum. After 48 hour incubation period, medium were changed ${\alpha}-MEM$ containing 5% fetal bovine serum. After 24 hours, $10^{-9}M$ 1,25-dihydroxyvitamin $D_3$ added. Total mRNA was extracted at 0, 6, 24, 48, 72 hour. PRPCR method was programed for the detection of IGF-I mRNA. In the both groups of 1,25-dihydroxy vitamin $D_3$ treated and control, alternative splicing form of IGF-I, IGF-IA and IGF-IB were expressed. In the 1,25-dihydroxyvitamin $D_3$ treated group, IGF-I mRNA expression was matained until 24 hour, there after expression was decresed. MC3T3-E1 cell were seeded $2.5{\times}10^4/ml$ at 24well plate in ${\alpha}-MEM$ containing 10% fetal bovine serum. After 48 hour incubation period, medium were changed ${\alpha}-MEM$ containing 3% fetal bovine serum. After 24 hours, $10^{-9}M$ 1,25-dihydroxyvitamin $D_3$ and 10 ng/ml IGF-I were added separately or together. Cell were cultured for 1 and 3 days, $2{\mu}Ci/ml\;[^3H]$ -thymidine was added for the last 24h of culture of each days. ${[^3H]}$-thymidine incorporation in to DNA was measured and expressed counter per minute(CPM). DNA synthetic activity was significantly decreased by 1,25-dihydroxyvitamin $D_3$ both at 1 day and 3 day, and in the combination group of 1,25-dihydroxyvitamin $D_3$ and IGF-I, DNA synthetic activity was also decreased both at 1 day and 3 days. IGF-I did not affect the DNA synthetic activity compared to control group both at 1 day and 3 day. From the above results, 1,25-dihydroxyvitamin $D_3$ was potent inhibitor of cell proliferaton in MC3T3-E1 cells. It assumed that the effect of 1,25-dihydroxyvitamin $D_3$ on osteoblast proliferation may be mediated in part by decreased level of IGF-I.
Porpilyromonas endodontalis is specifically involved in endodontic infections. The bacterium can be isolated almost exclusively only from infected rool canals. P. gingivalis also has been implicated in endodontic infection. Pathogemcity of P. gingival is is attributed to a variety of virulence factors, especially proteases, produced by the bacterium. Importance of P. endodontalis in endodontic infection has been revealed. However, the pathogenic property of P. endodontalis has not been extensively studied. The present study was undertaken to characterize the proteolytic activity of P. endodontalis and compare the activity with that of P. gingivalis which has the most potent and diverse proteases among oral bacteria. For this purpose, culture supematants(SUP) and cell extracts(CE) were obtained from these two bacteria and were subjected to zymography using 15% polyacrylamide gel copolymerized with gelatin, type I, IV collagens or albumin. Hydrolysis of the collagens was further investigated by the cleavage assay using native type I and IV collagens in solution-phase. The results were as follows: 1. P. endodontalis apparently has a proteolytic activity that is comparable with that of P. gingivalis. 2. SUP and CE obtained from P. endodontalis and P. gingival is showed the strongest activity for gelatin, followed by type I and IV collagens, and albumin. 3. In the zymography, no noticeable difference in proteolytic activity for gelatin and albumin between the SUP and CE was observed, but in the cleavage assay using native collagens, the SUP showed a stronger collagenolytic activity than the CE. 4. The gelatinolytic activity of both the SUP and CE from these two bacteria was diminished in the presence of $CaCl_2$ or reducing agents such as ${\beta}$-mercaptoethanol and dithiothreitol(DTT). 5. Type I(calf skin and human placenta) collagenolytic activity of P. endodontalis and P. gingivalis was reduced by DTT but not affected by $CaCl_2$. The inhibitory effect of DTT, however, was reduced to some extent by $CaCl_2$. 6. Type IV collagenolytic activity of these two bacteria was not affected by $CaCl_2$ but increased to some extent in association with the reducing agents. 7. Hydrolysis of albumin by P. endodontalis and P. gingivalis was demonstrated only in the presence of the reducing agents. The overall results indicate that with respect to proteolytic activity, P. endodontalis appears to be as potent as P. gingivalis, or maybe more, and its proteolytic characteristic is similar to that of P. gingivalis. This suggests that P. endodontalis has so potent proteolytic activity that can participate by itself in endodontic infections and apical periodontitis, causing tissue destruction.
The nephrotoxicity of dichromate was reduced upon the administration of red ginseng $S_I-fraction$. Rabbits treated with dichromate showed acute renal failure and the majority of animals died within ten days. Mortality and physioligical status were significantly improved by the administration of ginseng. The effect of red ginseng $S_I-fraction$ was studied by hematological analysis. urinalysis. and histophathological examination. It was found that red ginseng $S_I-fraction$ has a potent effect on glomerulotubular imbalance induced by dichromate. In addition, $S_I-fraction$ normalized the rate of glycogenolysis. glycolysis. and the rate of lactate production. which had been accelerated by intravenous dichromate injection. Red ginseng $S_I-fraction$ was superior to ascorbic acid. CaEDT A and furocemide in the protection and recorvery from dichromate-induced acute renal failure.
The intersection of all two-sided ideals of an ordered semigroup, if it is non-empty, is called the kernel of the ordered semigroup. A left ideal L of an ordered semigroup ($S,{\cdot},{\leq}$) having a kernel I is said to be simple if I is properly contained in L and for any left ideal L' of ($S,{\cdot},{\leq}$), I is properly contained in L' and L' is contained in L imply L' = L. The notions of simple right and two-sided ideals are defined similarly. In this paper, the author characterize when an ordered semigroup having a kernel is the class sum of its simple left, right and two-sided ideals. Further, the structure of simple two-sided ideals will be discussed.
Drosophila Sog and vertebrate Noggin play important roles during development. They function as antagonists against BMP4 signaling and induce neural ectoderm during embryogenesis. They are also engaged in appendage formation by inhibiting BMP4 signaling during late development. To understand further functions of Sog, Supersog, which is a more potent form of Sog, and Noggin BMP4 antagonists during development, I performed the molecular genetic analysis using Drosophila embryogenesis and wing formation as assay systems. In cellular blastoderm embryos, Sog inhibited Dpp signaling, Drosophila BMP4 signaling, whereas Supersog or Noggin did not block Dpp signaling. During wing formation, Sog inhibited Sax type I receptor of Dpp signaling whereas Noggin inhibited Tkv type I receptor of Dpp signaling. However, Supersog inhibited both Sax and Tkv type I receptors. These results suggest that functions of BMP4 antagonists are developmental stage dependent and indicate that each BMP4 antagonist inhibits BMP4 signaling by blocking different BMP4 receptors.
Analogues of $2(S)-5, 2^{l}, 5^{l}$-trihydroxy-7, 8-dimethoxyflavanone, a naturally-occurring compound, which had been reported to have potent antitumor activity, were synthesized and examined for the cytotoxicity against three cancer cell lines. Among the intermediate chalcones and synthetic 5-hydroxy-7, 8-dimethoxyflavanone analogues, $({\pm})2^{l}, 5^{l}-dibenzyloxy-5, 7, 8-trimethoxyflavanone$ exhibited about 2-8 times stronger activity than $2(S)-5, 2^{l}, 5^{l}$-trihydroxy-7, 8-dimethoxyflavanone against L1210, K562 and A549 cancer cell lines. In the structure-activity relationship, it is suggested that among analogues of 5-hydroxy-7, 8-dimethoxyflavanone, the existence of two oxygenated groups of para-relation at $C-2^{I} and C-5^{I}$ positions on flavanone B-ring, may be necessary to exhibit effective cytotoxic activity.
This study was carried out to identify the skin anti-aging effect of cycloheterophyllin on dermal fibroblasts. To elucidate anti-aging effects of cycloheterophyllin on dermal fibroblasts, I measured cell viability, mRNA expressions, and Collagen, type I/matrix metallopeptidase 1(MMP1)-ELISA assay. In this study, I investigated the effects of cycloheterophyllin on Collagen, type I, alpha 1(COL1A1)/Collagen, type III, alpha 1(COL3A1)/MMP1/Superoxide dismutases/Catalase(CAT) mRNA expressions and Collagen, type I/MMP1 protein production. Quantitative Real-time RT-PCR showed that cycloheterophyllin increased mRNA level of COL1A1/COL3A1/CAT genes and collagen, type I protein by ELISA assay compared to UVA-treated dermal fibroblasts. Furthermore MMP1 mRNA and protein expressions were decreased by cycloheterophyllin treatment. These observations revealed that cycloheterophyllin increased anti-aging effects in dermal fibroblasts. Therefore, I identified the anti-aging effects of cycloheterophyllin, and these results showed that the cycloheterophyllin can be a considerable potent ingredient for skin anti-aging. Based on this, I anticipated further researches about cycloheterophyllin for mechanism to develop not only cosmetics but for healthcare food or medicine.
