The Stem Bark of Kalopanax pictus Exhibits Anti-inflammatory Effect through Heme Oxygenase-1 Induction and NF-${\kappa}B$ Suppression

  • Backgroud: The stem bark of Kalopanax pictus (KP) has been used in traditional medicine to treat rheumatoidal arthritis, neurotic pain and diabetes mellitus in China and Korea. In this study, the mechanism responsible for anti-inflammatory effects of KP was investigated. Methods: We examined the effects of KP on NO production, nitric oxide synthase (iNOS) and HO-1 expression, NF-${\kappa}B$, Nrf2 and MAPK activation in mouse peritoneal macrophages. Results: The aqueous extract of KP inhibited LPS-induced NO secretion as well as inducible iNOS expression, without affecting cell viability. KP suppressed LPS-induced NF-${\kappa}B$ activation, phosphorylation and degradation of $I{\kappa}B-{\alpha}$, phosphorylation of extracellular signal-regulated kinase 1/2 (ERK1/2) and c-Jun N-terminal kinase (JNK). Furthermore, KP induced HO-1 expression and Nrf2 nuclear translocation. Conclusion: These results suggest that KP has the inhibitory effects on LPS-induced NO production in macrophages through NF-${\kappa}B$ suppression and HO-1 induction.

들깨 새싹 추출물의 췌장 RINm5F 세포에서 NF-κB 경로를 통한 사이토카인에 의한 손상 예방 효과 (Perilla frutescens Sprout Extracts Protected Against Cytokine-induced Cell Damage of Pancreatic RINm5F Cells via NF-κB Pathway)

  • 들깨(Perilla frutescents (L.) Britton var.) 새싹은 꿀풀과에 속하는 1년생 초본이다. 본 연구의 목적은 들깨 새싹 에탄올 추출물이 사이토카인으로 유도된 췌장 베타 세포 손상에 대한 예방 효과를 평가하기 위함이다. 췌장 소도 주위에 염증 세포 침습으로 의해 분비되는 사이토카인은 1형 당뇨병의 발병원인에 해당된다. 인터루킨-$1{\beta}$ (IL-$1{\beta}$), 인터페론-${\gamma}$ (IFN-${\gamma}$), 종양괴사인자-${\alpha}$ (TNF-${\alpha}$) 등의 사이토카인은 활성산소 형성을 유도한다. 세포 내 활성산소 축적은 췌장 베타 세포 기능장애와 세포사멸을 이끈다. 들깨 새싹 추출물은 항산화 효과를 증가 시켰으며 활성산소 생성을 억제하였다. 사이토카인은 세포생존율을 감소시켰고, iNOS와 COX-2의 발현을 증가시키고 산화질소 생성을 유도하였다. 들깨 새싹 추출물은 사이토카인으로 유도된 세포생존을 농도 의존적으로 예방하였다. 또한, 사이토카인에 의한 산화질소 생성과 iNOS와 COX-2의 단백질 발현 증가를 억제하였다. 더 나아가 들깨 새싹 추출물은 췌장 베타 세포주(RIN-m5F)에서 $I{\kappa}B{\alpha}$ 인산화 억제를 통해서 NF-${\kappa}B$의 활성화를 상당히 감소시켰다. 요약하자면, 본 연구 결과는 들깨 새싹 추출물이 사이토카인으로 유도된 췌장 베타 세포 손상에 대한 보호 효과를 가지고 있다는 것이 확인되었다. 결과적으로 들깨 새싹은 혈당 증가에 의한 산화 스트레스와 염증성 사이토카인에 의한 베타 세포 손상을 완화하여 당뇨에 유익할 것으로 사료된다.

아위느타리 신품종 GW10-45 클로로포름 추출물의 항염증 효과 (Anti-inflammatory properties of chloroform extracts from GW10-45, a new cultivar derived from Pleurotus ferulae, in RAW264.7 murine macrophages.)

  • 만성적 염증 반응은 지속적인 항원에 대한 노출에 의해 발생할 수 있으며 관절염이나 2형 당뇨와 같은 여러 염증성 질환의 가장 큰 발병 원인이 된다. 본 연구에서는 아위 느타리 버섯 (Pleurotus ferulae)의 새로운 육종 균주인 GW10-45의 추출물(헥산, 클로로포름, 메탄올, 메탄올/물, 물)이 가지는 RAW264.7 대식세포에 대한 항염증 활성을 연구하였다. 먼저, 각각의 추출물은 RAW264.7 세포주에 대하여 세포독성을 가지지 않았으며, 헥산 및 클로로포름, 물 추출물이 산화질소의 생성을 억제하였다. 그 중, 클로로포름 추출물(CG45)을 처리한 실험군에서 가장 높은 산화질소 생성 억제효과가 나타남을 확인하였다. 또한 주요한 염증성 인자들의 단백질 발현 및 인산화에 미치는 영향을 확인하기 위하여 Western blot 분석을 시행하였다. 그 결과, LPS로 유도된 염증 모델에 CG45를 처리하였을 때 iNOS 및 COX-2 단백질의 발현이 급격히 감소함을 확인하였다. 또한 $NF-{\kappa}B$ 신호전달에서 $NF-{\kappa}B$와 이의 억제 단백질인 $I{\kappa}B{\alpha}$의 인산화가 LPS만을 처리한 군과 비교하여 CG45를 함께 처리한 실험군에서 크게 감소함을 확인하였다. 실험을 통하여 아위느타리 버섯의 새로운 육종균주 GW10-45의 추출물 CG45는 $NF-{\kappa}B$ 신호전달 과정에서 인산화 정도를 억제함으로써 iNOS 및 COX-2 단백질의 발현을 억제하여 LPS로 유도된 RAW264.7 세포주의 염증 모델에서 그 염증 반응을 억제할 수 있다는 것을 확인하였다.

WIN-34B May Have Analgesic and Anti-Inflammatory Effects by Reducing the Production of Pro-Inflammatory Mediators in Cells via Inhibition of IκB Signaling Pathways

  • WIN-34B showed analgesic and anti-inflammatory effects in various animal models of pain and osteoarthritis. However, the molecular mechanism by which WIN-34B inhibits pain and inflammation in vivo remains to be elucidated. We investigated the molecular mechanisms of the actions of WIN-34B using various in vitro models using fibroblast-like synoviocytes from patients with rheumatoid arthritis (RA FLSs), RAW264.7 cells and peritoneal macrophages. WIN-34B inhibited the level of IL-6, $PGE_2$, and MMP-13 in IL-$1{\beta}$-stimulated RA FLSs in a dose-dependent manner. The mRNA levels were also inhibited by WIN-34B. The level of $PGE_2$, NO, IL-$1{\beta}$, and TNF-${\alpha}$ were inhibited by WIN-34B at different concentrations in LPS-stimulated RAW264.7 cells. The production of NO and $PGE_2$ was inhibited by WIN-34B in a dose-dependent manner in LPS-stimulated peritoneal macrophages. All of these effects were comparable to the positive control, celecoxib or indomethacin. I${\kappa}B$B signaling pathways were inhibited by WIN-34B, and the migration of NF-${\kappa}B$ into the nucleus was inhibited, which is consistent with the degradation of $I{\kappa}B-{\alpha}$. Taken together, the results suggest that WIN-34B has potential as a therapeutic drug to reduce pain and inflammation by inhibiting the production of pro-inflammatory mediators.

홍삼 비사포닌 분획의 단핵세포 분화와 염증반응에 대한 억제효과 (Non-saponin fraction of red ginseng inhibits monocyte-to-macrophage differentiation and inflammatory responses in vitro)

  • 본 연구에서는 홍삼 비사포닌 분획(NSF)의 항 염증 효과를 마우스 대식세포와 인간유래 단핵세포에서 확인하였다. NSF는 마우스 대식세포에서 LPS로 유도된 NO, iNOS 그리고 COX-2의 양 뿐만 아니라 IL-6, $TNF-{\alpha}$, MCP-1과 같은 염증성 싸이토카인의 생성량을 유의적으로 감소시켰다. 인간 유래 단핵세포에서는 PMA에 의해 유도되는 대식세포로의 분화를 효과적으로 억제하면서 분화인자인 $CD11{\beta}$와 CD36의 발현을 유의적으로 감소시켰다. 마우스 대식세포에서와 마찬가지로 염증성 싸이토카인들의 생성량 또한 감소하였는데, 이러한 NSF의 항 염증 효과는 두 전사인자의 조절작용에 의한 것으로 사료된다. 즉 NSF는 $NF-{\kappa}B$의 핵으로 이동을 감소시킴으로써 전사활성을 억제하여 염증성 싸이토카인들의 발현을 저해하고 이와 반대로 Nrf2의 발현과 핵으로의 이동을 증가시켜 항산화 효소이면서 항 염증 작용을 나타내는 HO-1의 발현을 촉진하는 것으로 관찰되었다. 따라서 NSF는 $NF-{\kappa}B$와 Nrf2의 두 가지 신호전달체계를 조절함으로써 항 염증 작용을 나타냈으며 이를 홍삼 NSF의 항 염증 기작으로 보고하는 바이다.

Anti-inflammatory effect of aged black garlic on 12-O-tetradecanoylphorbol-13-acetate-induced dermatitis in mice

  • BACKGROUND/OBJECTIVES: Although aged black garlic has various biological activities such as anti-allergy, anti-inflammation and neuroprotection, effect of aged black garlic on chemically contact dermatitis is unclarified. MATERIALS/METHODS: To evaluate anti-dermatitic activity of aged black garlic extract, we investigated effects of a fraction of aged black garlic extract (BG10) on both in vivo and in vitro. RESULTS: BG10 almost inhibited formation of nitric monoxide and interleukin-6 (IL-6; $IC_{50}$, $7.07{\mu}g/mL$) at $25{\mu}g/mL$, and dose-dependently reduced production of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$; $IC_{50}$, $52.07{\mu}g/mL$) and prostaglandin $E_2$ ($IC_{50}$, $38.46{\mu}g/mL$) in lipopolysaccharide-stimulated RAW264.7 cells. In addition, BG10 significantly inhibited the expression of inducible nitric oxide synthase, cyclooxygenase-2 and nuclear $NF-{\kappa}B$, and improved that of cytosolic levels of $NF-{\kappa}B$ and $I{\kappa}B{\alpha}$ in the cells. Consistent with in vitro studies, BG10 (0.5 mg/mL) not only reduced ear edema but also suppressed the formation of IL-6 and $TNF-{\alpha}$ induced by 12-O-tetradecanoylphorbol-13-acetate in ear tissues of mice. CONCLUSIONS: These findings suggest BG10 has anti-dermatitic activity through inhibiting activation of macrophages. Therefore, such effects of BG10 may provide information for the application of aged black garlic for prevention and therapy of contact dermatitis.

Oral Concentrated Grape Juice Suppresses Expression of NF-kappa B, TNF-α and iNOS in Experimentally Induced Colorectal Carcinogenesis in Wistar Rats

  • The aim of this study was to evaluate the effects of grape juice on colon carcinogenesis induced by azoxymethane (AOM) and expression of NF-kB, iNOS and TNF-${\alpha}$. Methods: Forty male Wistar rats were divided into 7 groups: G1, control; G2, 15 mg/kg AOM; G3, 1% grape juice 2 weeks before AOM; G4, 2% grape juice 2 weeks before AOM; G5, 1% grape juice 4 weeks after AOM; G6, 2% grape juice 4 weeks after AOM; G7, 2% grape juice without AOM. Histological changes and aberrant crypt foci (ACF) were studied, while RNA expression of NF-kB, TNF- and iNOS was evaluated by qPCR. Results: The number of ACF was higher in G2, and G4 presented a smaller number of crypts per focus than G5 (p=0.009) and G6. Small ACF (1-3) were more frequent in G4 compared to G2, G5 and G6 (p=0.009, p=0.009 and p=0.041, respectively). RNA expression of NF-kB was lower in G3 and G4 compared to G2 (p=0.004 and p=0.002, respectively). A positive correlation was observed between TNF-${\alpha}$ and NF-kB gene expression (p=0.002). In conclusion, the administration of 2% grape juice before AOM reduced the crypt multiplicity, attenuating carcinogenesis. Lower expression of NF-kB was observed in animals exposed to grape juice for a longer period of time, regardless of concentration.

Anti-inflammatory Effect of MeOH Extracts of the Stem of Polygonum multiflorum in LPS-stimulated Mouse Peritoneal Macrophages

  • Polygoni multiflori Ramulus (PM), the stem of Polygonum multiflorum Thunb. has been widely used as a traditional medicine for the treatment of lots of diseases. In macrophages, nitric oxide is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions in inflammation. In the present study, it was investigated that the inhibitory effects on NO and proinflammatory cytokines such as tumor necrosis factor alpha (TNF-${\alpha}$), interleukin-6 (IL-6) and the mechanism of down-regulation of immune response by 85% methanol extracts of PM in mouse (C57BL/6) peritoneal macrophages. Extracts of PM (0.1, 1 mg/ml) suppressed NO production and showed inhibition of proinflammatory cytokines like TNF-${\alpha}$, IL-6 and it attenuated iNOS and COX-2 expression via down-regulation of NF-${\kappa}$B activation. The present results indicate that the 85% methanol extracts of PM has an inhibitory effect on the production of NO through down-regulation of iNOS expression in LPS stimulated mouse peritoneal macrophages and therefore may be beneficial in diseases which related to macrophage-mediated inflammatory disorders.

Immunostimulating activity of maysin isolated from corn silk in murine RAW 264.7 macrophages

  • Corn silk (CS) has long been consumed as a traditional herb in Korea. Maysin is a major flavonoid of CS. The effects of maysin on macrophage activation were evaluated, using the murine macrophage RAW 264.7 cells. Maysin was isolated from CS by methanol extraction, and preparative $C_{18}$ reverse phase column chromatography. Maysin was nontoxic up to $100{\mu}g/ml$, and dose-dependently increased TNF-${\alpha}$ secretion and iNOS production by 11.2- and 4.2-fold, respectively, compared to untreated control. The activation and subsequent nuclear translocation of NF-${\kappa}B$ was substantially enhanced upon treatment with maysin ($1-100{\mu}g/ml$). Maysin also stimulated the phosphorylation of Akt and MAPKs (ERK, JNK). These results indicated that maysin activates macrophages to secrete TNF-${\alpha}$ and induce iNOS expression, via the activation of the Akt, NF-${\kappa}B$ and MAPKs signaling pathways. These results suggest for the first time that maysin can be a new immunomodulator, enhancing the early innate immunity.

Can herbal drug(s) meet the challenges of genomewide screen results on rheumatoid arthritis

  • Rheumatoid arthritis (RA) is an autoimmune/inflammatory disorder with a complex genetic component. RA is characterized by chronic inflammation of the synovial membrane in the joint, which leads to the progressive destruction of articular cartilage, ligament and bone. Several cytokines such as tumor necrosis $factor-{\alpha}\;TNF-{\alpha}\;and\;interleukin-1{\beta}\;(IL-1{\beta})$ and interleukin-6 (IL-6) have been implicated in the pathological mechanisms of synovial tissue proliferation, joint destruction and programmed cell death in rheumatoid joint. Genome wide screening of subjects suffering from autoimmune diseases especially arthritis revealed linkage to inflammatory molecules like $TNF-{\alpha},\;IL-1{\beta}$ and IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), nuclear factor-kappaB $(NF-{\kappa}B)$ and human leucocyte antigen/major histocompatibility complex (HLA/MHC) locus. The status of the pharmacological mechanism of herbal drugs in the light of genome wide screening results has been discussed to reinforce the therapeutic potential and the pharmacological basis of the herbal drugs.
