Antioxidant and Anti-inflammatory Activities of Butanol Extract of Melaleuca leucadendron L.

  • Melaleuca leucadendron L. has been used as a tranquilizing, sedating, evil-dispelling and pain-relieving agent. We examined the effects of M. leucadendron L. extracts on oxidative stress and inflammation. M. leucadendron L. was extracted with methanol (MeOH) and then fractionated with chloroform ($CHCl_3$) and butanol (BuOH). Antioxidant activity of the MeOH extract and BuOH fraction were higher than that of both ${\alpha}$-tocopherol and butyrated hydroxytoluene (BHT). Total phenol content in the extracts of M. leucadendron L., especially the BuOH fraction, well correlated with the antioxidant activity. The anti-inflammatory activity of BuOH extracts were investigated by lipopolysaccharide (LPS)-induced nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production, and cyclooxygenase-2 (COX-2) expression in RAW 264.7 macrophages. The BuOH fraction significantly inhibited LPS-induced NO and $PGE_2$ production. Furthermore, BuOH extract of M. leucadendron L. inhibited the expression of COX-2 and iNOS protein without an appreciable cytotoxic effect on RAW264.7 cells. The extract of M. leucadendron L. also suppressed the phosphorylation of inhibitor ${\kappa}B{\alpha}$ ($I{\kappa}B{\alpha}$) and its degradation associated with nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activation. Furthermore, BuOH fraction inhibited LPS-induced NF-${\kappa}B$ transcriptional activity in a dose-dependent manner. These results suggested that M. leucadendron L. could be useful as a natural antioxidant and anti-inflammatory resource.

후박에서 추출한 Obovatol의 염증매개 생성의 억제와 세포자멸사를 통한 Human Prostate Carcinoma PC-3 및 LNCap 세포의 세포증식에 대한 영향 (Obovatol Extracted from Magnolia Obovata Inhibits Inflammation Mediator Generation and Prostate Carcinoma PC-3, LNCap Cell Growth Through Induction of Apoptotic Cell Death via Inactivation of $NF-{\kappa}B$)

  • 목적 : 후박(厚朴)(Magnolia Obovata)에서 추출한 낮은 농도의 Obovatol 약침액의 RAW264.7 세포에서 LPS로 유발된 염증,$TNF-{\alpha}$로 유발된 human Prostate carcinoma PC-3 및 LNCap 세포의 세포증식에 대한 영향과 그 기전을 살펴보고자 하였다. 방법 : RAW264.7 세포에서 LPS로 염증을 유발하고 낮은 농도의 Obovatol 약침액을 처리한 후 cell viability, NO 생성량,iNOS와 COX-2의 발현, $NF-{\kappa}B$활성,전사능력을 관찰하기 위해 MTT assay, NO determination assay, western blot analysis, EMSA, luciferase activity assay를 시행하였고,LNCap, PC-3 세포에 $TNF-{\alpha}$로 증식을 유도하고 낮은 농도의 Obovatol 약침액을 처리한 후 cell growth, apoptosis 및 apoptosis와 연관된 $NF-{\kappa}B$의 활성 변화를 관찰하기 위해 WST-1, Cell morphogy test, DAPI staining and TUNEL assay, EMSA, luciferase activity assay를 시행하였다. 결과 : 1. RAW264.7 세포에서 낮은 농도의 Obovatol 약침액 처리는 $NF-{\kappa}B$의 활성 및 전사능력을 낮추고 iNOS와 COX-2의 발현과 NO 생성을 감소시켜 LPS로 유발된 염증을 억제하였다. 2. LNCap, PC-3 세포에서 낮은 농도의 Obovatol 약침액 처리는 $NF-{\kappa}B$의 활성을 낮추어 세포자별사를 촉진함으로써 $TNF-{\alpha}$로 유발된 암세포의 성장을 억제하였다. 결론 : 이상의 결과는 낮은 농도의 Obovatol 약침액이 항염 및 인간 전립선암세포주인 PC-3, LNCap에 대한 증식억제 효과가 있음을 입증한 것이며,향후 이를 바탕으로한 생체 연구에서의 긍정적인 결과는 Obovatol 약침액이 만성염증성 질환 및 전립선암의 예방과 치료에 대한 효과적인 치료제 개발에 초석이 될 것으로 기대된다.

Mouse의 신장상피세포에서 패장(敗醬)추출물이 산화 스트레스 및 NF-${\kappa}B$ signaling에 미치는 영향 (The Effect of Patriniae Radix on the Oxidative Stress and the NF-${\kappa}B$ Signaling in Mouse LLC-$PK_1$ Cell)

  • Objectives : The aims of this study were to investigate the cytoprotective, antioxidative and inflammation genes inhibitory effects of Patriniae Radix on the mouse LLC-$PK_1$ cells (renal epithelial cells). Methods : The cytoprotective effect of Patriniae Radix was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The antioxidative effect was measured in terms of generation amount of superoxide anion radical (${\cdot}{O_2}^-$) by 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), nitric oxide (NO) by 4,5-diaminofluorescein (DAF-2), peroxynitrite ($ONOO^-$) by dihyldrorhodamine 123 (DHR 123) and prostaglandin $E_2$ ($PGE_2$) by $PGE_2$ immunoassay on $H_2O_2$-treated LLC-$PK_1$ cells. For measuring of inflammation genes inhibitory effects, western blot was performed to detect IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, NF-${\kappa}B$ (p50, p65), COX-2, iNOS, IL-$1{\beta}$ and VCAM-1 protein level in cytosol fractions from LLC-$PK_1$ cells. Results : Patriniae Radix extract reduced the $H_2O_2$-induced cell death and inhibited the amount of $H_2O_2$-induced ${\cdot}{O_2}^-$, NO, $ONOO^-$, $PGE_2$ generation dose-dependently on the mouse LLC-$PK_1$ cells in vitro. Also Patriniae Radix extract inhibited the expression of IKK-$\alpha$, phospho-$I{\kappa}B-\alpha$, COX-2, iNOS, IL-$1\beta$ and VCAM-1 genes dose-dependently by means of decreasing activation of NF-${\kappa}B$. Conclusions : According to above results, it was identified that Patriniae Radix had the cytoprotective, antioxidative and inflammation genes inhibitory effects. So it was suggested that Patriniae Radix would be effective to the treatment for the inflammatory process and inflammation-related diseases.

Rhizopus oryzae으로 발효한 울금의 항산화 및 항염효과 (Anti-oxidative and Anti-inflammatory Activities of Fermented Turmeric (Curcuma longa L.) by Rhizopus oryzae)

  • 본 연구의 목적은 울금의 발효를 통해 쓴맛의 관능적 기호도를 향상시키면서, 발효 울금의 항산화 및 항염효능 검증을 위한 것으로, 발효 울금의 관능적 기호도 확인을 위해 맛센서 분석과 항산화 평가를 위해 총 폴리페놀 및 플라보노이드 함량 및 DPPH radical 소거능, 지표성분 검출을 위해 HPLC분석을 진행하였고, 항염증평가는 RAW 264.7 세포에서 LPS에 의해 유도되는 염증인자인 NO, $PGE_2$, iNOS, COX-2, $NF-{\kappa}B$, IL-6와 $TNF-{\alpha}$에 대한 감소효과를 측정하였다. 실험결과 Rhizopus oryzae로 울금의 발효가 진행 될수록 쓴맛이 감소함을 보였다. 총 폴리페놀, 플라보노이드 함량과 DPPH radical 소거능은 비 발효 울금보다 울금 발효 1, 3일차에 증가함을 보였다. 발효 기간별 발효 울금은 10, 50, $100{\mu}g/ml$ 모든 농도에서 세포독성이 나타내지 않았다. RAW 264.7 세포에 LPS로 유도된 염증인자인 $NF-{\kappa}B$, IL-6와 $TNF-{\alpha}$ 생성량이 발효 기간별 발효 울금의 염증인자 생성억제를 평가한 결과 $NF-{\kappa}B$와 IL-6에서 발효 울금 $100{\mu}g/ml$에서 현저히 생성량이 억제되었다. LPS로 유도된 cytokines 억제효과를 보인 발효 기간별 발효 울금($100{\mu}g/ml$)의 NO, $PGE_2$ 생성억제를 평가한 결과, 대조군과 비교해 LPS를 처리한 군에서 NO, $PGE_2$의 생성이 현저히 증가되었고, 울금 에탄올 추출물에 의해 유의성 있는 생성억제효과를 보였고 비 발효에 비해 발효 울금의 생성억제 효과를 보였다. 또한 COX-2와 iNOS의 단백질 발현 억제효과를 확인한 결과, 대조군과 비교해 LPS에 의해 증가 된 COX-2와 iNOS의 단백질발현이 발효 울금에 의해 감소됨을 확인하였다. 위 결과 발효 울금은 RAW 264.7 세포에서 LPS에 의해 유도되는 NO와 $PGE_2$의 생성억제, iNOS와 COX-2의 발현을 억제시켰으며, $NF-{\kappa}B$, IL-6와 $TNF-{\alpha}$의 분비량도 억제시켰다. 이는 발효 울금의 항산화와 항염증성을 입증하기 위한 기초자료로 활용이 가능하다고 사료된다.

LPS로 유도된 RAW 264.7 대식세포에 대한 미역(Undaria pinnatifida) Ethyl Acetate 분획물의 항염증 효과 (Anti-Inflammatory Effect of Ethyl Acetate Fraction Isolated from Undaria pinnatifida on Lipopolysaccharides-Stimulated RAW 264.7 Cells)

  • An ethanolic extract of Undaria pinnatifida was fractionated using several solvents. Of the fractions, the ethyl acetate fraction had the greatest inhibitory effect on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 macrophage cells. Using this fraction (U. pinnatifida ethyl acetate extract, UPE), we investigated the molecular mechanism underlying its inhibitory effect on LPS-stimulated RAW 264.7 cells. Pretreatment of the cells with up to $100{\mu}g/mL$ UPE significantly inhibited NO production and inducible nitric oxide synthase (iNOS) expression, in a dose-dependent manner. Similarly, UPE treatment markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-1, IL-6 and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), while it strongly suppressed the nuclear translocation of nuclear factor-kappa B (NF-${\kappa}B$) by preventing proteolytic degradation of inhibitor of nuclear factor ${\kappa}B$ $(I{\kappa}B)-{\alpha}$. Moreover, UPE treatment significantly reduced the phosphorylation of phosphatidylinositol 3-kinase (PI3K)/Akt and mitogen-activated protein kinase (MAPK) in LPS-stimulated cells. These results indicate that UPE contains anti-inflammatory compounds and suggest that it might be used as a functional food material that assists in prevention of inflammatory diseases.

목과(木瓜)의 항염(抗炎) 및 면역반응(免疫反應)에 대한 실험적(實驗的) 연구(硏究) (Anti-inflammatory Effect of Fructus Chaenomelis(FC))

  • Purpose: The purpose of this study is to investigate anti-inflammatory effect and immune responses of aqueous extract from Fructus Chaenomelis (FC). Methods: We studied anti-inflammatory effect by means of examining the production of NO(nitric oxide) and expressions of pro-inflammatory cytokine (TNF-$\alpha$(tumor necrosis factor-alpha), IL(Interleukin)-6, IL-12) in the LPS-induced peritoneal macrophages of mice. Also, The western blot analysis has been done to look into the mechanism of anti-inflammatory effect. Results: 1. The FC extract did not have any cytotoxicity in the peritoneal macrophages. 2. The FC extract inhibits the productions of NO, IL-6. IL-12 in the LPS-stimulated peritoneal macrophages of mice, but not of TNF-$\alpha$. 3. The FC extract inhibits the activation of NF-${\kappa}B$(nuclear factor-kappa B) by keeping $I{\kappa}B-\alpha$(inhibitory kappa B-alpha) from degradating, but not of MAPKs(mitogen-activated protein kinases) such as ERK(extracelluar signa 1-regulated kinase), JNK(c-Jun N-terminal kinase), p38. Conclusion: These results show that FC extract inhibits the production of pro-inflammatory cytokines such as IL-6. IL-12. NO by inhibiting NF-${\kappa}B$ activation in the peritoneal macrophages of mice. In conclusion, this experiment suggests that FC extract may be effective for the treatment of acute and chronic inflammation including genitourinary infection.

창출(蒼朮) 에탄올 추출물이 비만세포 매개 염증반응에 미치는 영향 (The Effects of Ethanol Extract from Atractylodes Chinensis Rhizome on the Mast Cell-Mediated Inflammatory Responses)

  • Objective : Atractyloides Chinensis Rhizome (ACR) is widely used in oriental medicine as a remedy for an inflammation and an allergic disease. However, as yet there is no clear explanation of how ACR affects the production of inflammatory cytokine. This study was to determine the effects of ACR on the mast cell-mediated inflammatory responses. Method : The amount of inflammatory cytokine production induced by the phorbol myristate acetate (PMA) plus calcium ionophore(A23187) in the human mast cell line (HMC-1) incubated with various concentrations of ACR was measured. The TNF-${\alpha}$ protein levels were analysised by Western blots. The TNF-${\alpha}$, IL-6 and IL-8 secreted protein levels were measured by the ELISA assay. The TNF-${\alpha}$, IL-6 and IL-8 mRNA levels were measured by the RT-PCR analysis. NF-${\kappa}$B, phospho-I${\kappa}$B and MAPKs were examined by Western blot analysis. The NF-${\kappa}$B promoter activity was examined by a luciferase assay. Results : 1. The expressions of TNF-${\alpha}$ and TNF-${\alpha}$ mRNA were decreased dose-dependently at 0.05-0.2mg/$m\ell$ of ACR and significantly decreased at 0.2mg/$m\ell$. 2. The expressions of IL-6 and IL-6 mRNA were decreased dose-dependently at 0.05-0.2mg/$m\ell$ of ACR and significantly decreased at 0.2mg/$m\ell$. 3. The expressions of IL-8 and IL-8 mRNA were decreased dose-dependently at 0.05-0.2mg/$m\ell$ of ACR and significantly decreased at 0.2mg/$m\ell$ specially. 4. The expressions of Phosphorylated-JNK were decreased, not p38, ERK 5. The expressions of NF-${\kappa}$B were decreased dose-dependently at 0.1-0.2mg/$m\ell$ of ACR. The expressions of Phosphorylated I${\kappa}$B were significantly decreased at 0.2mg/$m\ell$. In addition, ACR suppressed PMA plus A23187-induced NF-${\kappa}$B promoting activity. Conclusion : It is suggested that ACR should suppress through inhibition of NF-${\kappa}$B activity and cytokine production.

Rosmarinic acid와 luteolin의 항염증에 대한 상승효과 (Synergistic Anti-inflammatory Effect of Rosmarinic Acid and Luteolin in Lipopolysaccharide-Stimulated RAW264.7 Macrophage Cells)

  • 본 연구에서는 들깨 유래 기능성 물질인 rosmarinic acid (RA)와 luteolin이 RAW264.7 세포에서 항염증작용에 대한 상승 효과가 있는지 알아보고자 하였다. 그 결과 RAW264.7 세포에 RA($50{\mu}M$)와 luteolin ($1{\mu}M$)을 동시에 처리하였을 경우 염증 매개인자인 NO, iNOS, $PGE_2$, COX-2의 생성을 RA ($100{\mu}M$)와 luteolin ($2{\mu}M$)을 각각 처리하였을 때 보다 더 뛰어나게 억제하였다. 또한 RA ($50{\mu}M$)와 luteolin ($1{\mu}M$)을 동시에 처리하였을 경우 TNF-${\alpha}$, IL-6, IL-$1{\beta}$ 같은 염증성 사이토카인의 생성량을 RA ($100{\mu}M$)와 luteolin ($2{\mu}M$)을 각각 처리하였을 때 보다 더 뛰어나게 억제하는 것을 확인하였다. 그리고 RA ($50{\mu}M$)와 luteolin ($1{\mu}M$)을 동시에 처리하였을 경우 RA ($100{\mu}M$)와 luteolin ($2{\mu}M$)을 각각 처리하였을 때 보다 NF-${\kappa}B$의 subunit인 p65의 translocation과 $I{\kappa}B$-${\alpha}$의 degradation을 더 뛰어나게 억제하는 것을 볼 수 있어 두 화합물 간의 상승작용이 뚜렷함을 확인 할 수 있었고, RA와 luteolin 두 화합물을 동시에 처리할 경우 염증관련 질환 치료에 유용하게 활용될 수 있을 것으로 판단된다.

꽃송이버섯 추출물의 항염활성 효과 (Anti-inflammatory effects of Sparassis crispa extracts)

  • 꽃송이버섯 추출물은 염증반응 시 유도되는 NO 생성을 저해하는 활성이 있는 것으로 나타났으며, 200 ${\mu}g/ml$의 꽃송이버섯 추출물을 처리하였을 때 NO 저해능이 최대 효과를 보였고 RAW264.7 cell에서는 대조군과 유사한 수준으로 NO 생성을 억제하였다. 이러한 NO 생성의 저해는 iNOS의 발현이 감소한 것에 의한 결과임을 단백질과 mRNA의 발현량 변화를 통하여 확인하였으며, mRNA 발현 변화는 iNOS 유전자의 전사를 담당하는 전사인자인 $NF-{\kappa}B$와 STAT-1의 활성감소에 의한 결과임을 western blot을 통하여 확인하였다. 특히, $NF-{\kappa}B$의 활성감소는 $I{\kappa}B{\alpha}$의 활성증가에 의한 $NF-{\kappa}B$의 억제능 향상에 의한 것임을 확인하였고, 활성억제된 $NF-{\kappa}B$가 핵 내부로의 이동이 저해되면서 iNOS 유전자 발현에 영향을 미친 것임을 확인하였다. 그러므로, 꽃송이버섯 추출물은 NO 저해능을 이용한 항염증소재로서 염증성질환의 완하에 도움이 될 것으로 사료된다.

Emodin Isolated from Polygoni cuspidati Radix Inhibits TNF-α and IL-6 Release by Blockading NF-κB and MAP Kinase Pathways in Mast Cells Stimulated with PMA Plus A23187

  • Emodin, a naturally occurring anthraquinone derivative isolated from Polygoni cuspidati radix, has several beneficial pharmacologic effects, which include anti-cancer, anti-diabetic, and anti-inflammatory activities. In this study, the authors examined the effect of emodin on the production of proinflammatory cytokines, such as, tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-6, in mouse bone marrow-derived mast cells (BMMCs) stimulated with phorbol 12-myristate 13-acetate (PMA) plus the calcium ionophore A23187. To investigate the mechanism responsible for the regulation of pro-inflammatory cytokine production by emodin, the authors assessed its effects on the activations of transcriptional factor nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and mitogen-activated protein kinases (MAPKs). Emodin attenuated the nuclear translocation of (NF)-${\kappa}B$ p65 and its DNA-binding activity by reducing the phosphorylation and degradation of $I{\kappa}B{\alpha}$ and the phosphorylation of $I{\kappa}B$ kinase B (IKK). Furthermore, emodin dose-dependently attenuated the phosphorylations of MAPKs, such as, extracellular signal-regulated kinase 1/2 (ERK1/2), p38 MAP kinase, and the stress-activated protein kinases (SAPK)/c-Jun-N-terminal kinase (JNK). Taken together, the findings of this study suggest that the anti-inflammatory effects of emodin on PMA plus A23187-stimulated BMMCs are mediated via the inhibition of NF-${\kappa}B$ activation and of the MAPK pathway.
