Peanut sprouts extract (Arachis hypogaea L.) has anti-obesity effects by controlling the protein expressions of $PPAR{\gamma}$ and adiponectin of adipose tissue in rats fed high-fat diet

  • BACKGROUD/OBEJECTIVES: This study aims to find out the effects of peanut sprout extracts on weight controls and protein expressions of transcription factors related to adipocyte differentiation and adipocytokine in rats under high-fat diets. MATERIALS/METHODS: Four week-old Sparague-Dawley (SD) were assigned to 4 groups; normal-fat (NF) diets (7% fat diet), high-fat (HF) diets (20% fat diet), high fat diets with low peanut sprout extract (HF + PSEL) diet (20% fat and 0.025% peanut sprout extract), and high fat diets with high peanut sprout extract (HF + PSEH) diet (20% fat and 0.05% peanut sprout extract). Body weight changes, lipid profiles in adipose tissue, and the mRNA protein expressions, such as peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), CCAAT element binding protein ${\alpha}$ (C/EBP ${\alpha}$), leptin, and adiponectin, were determined. RESULTS: After 9 weeks of feeding, the HF + PSEH group had significantly less weight gains than the HF group (P < 0.05). However, the total dietary intakes or food efficiency ratios among groups were not significantly different. The weight of epididymal fat in HF + PSEH group, $3.61{\pm}0.5g$, or HF + PSEL group, $3.80{\pm}0.7g$, was significantly lower than the HF group, $4.39{\pm}0.4g$, (P < 0.05). Total lipids and total cholesterol in adipose tissue were significantly decreased in HF + PSEH group compared to those in the HF group, respectively (P < 0.05). PSEH supplementation caused AST and ALT levels to decrease when it compared to HF group, but it was not statistically significant. The protein expression of $PPAR{\gamma}$ in HF + PSEH group was significantly lower than the HF group (P < 0.05). Comparing with the HF group, the protein expression of adiponectin in HF + PSEH group was significantly increased (P < 0.05). The protein expressions of C/EBP ${\alpha}$ and leptin in HF + PSEH group were lower than the HF group, but it was not statistical significant. CONCLUSIONS: In conclusion, peanut sprout extract has anti-obesity effect by lowering the expressions of $PPAR{\gamma}$ which regulates the expression of adiponectin.

  • Transparent vinyl-mulching cultivation plots heavy-fertilized (6-14-20 kg $10a^{-1}\; of\; N-P_2O_5-K_2O)$ in growing peanuts produced 315-344Kg $10a^{-1}$ of quality kernels through improvement of the low temperature conditions of underground environment during the early growing stage and kernel-filling stage. The yields of which were 82-99 percent higher than that of conventional cultivation plot. The vinyl-mulching cultivation techniques accelerated emergence, young seedling growth, flowering, pod bearing and kernel filling of peanuts in Korea.

  • We investigated the effect of peanut sprout extract (PSE) on andropause symptoms. PSE was evaluated for impotency and benign prostatic hyperplasia via phosphodiesterase (PDE) or 5-alpha reductase II inhibition assay. Inhibition of PDE and 5-alpha reductase II activities in PSE was significantly higher than that of the non-treated control. To investigate the effects of testosterone levels by PSE, we performed cell media test using TM3 cells. Production of testosterone in TM3 cells was elevated by PSE. These results indicate that PSE was able to alleviate andropause symptoms.

  • In order to determine the optimum sowing date of peanut in the middle part of Korea, peanut's 100-grain-weight and physico-chemical compositions for peanut cultivars such as runner (Yeoju landrace), semi-spanish (Suweon No. 15) and spanish types (Wasedairu) to different seeding date which planted at interval of ten days from May 5 to July 4, in 1981, was carried out. The 100-grain-weight and crude fat of peanut were increased from May 5 to May 25, but after that decreased. And the 100-grain-weight of peanut was great in order of Wasedairu, Suweon No. 15 and Yeoju landrace. The highest amount of crude fat was seen in peanuts which was sown at May 25 and May 15. Unsaturated fatty acids are composed of $80{\sim}85%$ of oleic and linoleic acid, but saturated fatty acids are constituted of $10{\sim}15%$ of palmitic acid and $1{\sim}5%$ of stearic and arachidic acid. Composition ratio of fatty acid in peanut was changed greatly according to sowing date. According to the late sowing date linoleic and palmitic acid were increased but oleic acid was decreased, respectively.

Effects of Dry Roasting on the Vitamin E Content and Microstructure of Peanut (Arachis hypogaea)

  • Effects of roasting on vitamin E content, color, microstructure and moisture of peanuts, and vitamin E content in peanut oils prepared from the roasted peanuts were investigated. Runner-type peanuts were roasted at 140, 150, and $160^{\circ}C$ for 10-20 min. As roasting temperature and time increased, the CIELAB $L^*$ value of peanuts decreased while $a^*$ and $b^*$ values increased, resulting in formation of the golden brown color of roasted peanuts. Moisture ratio (M/Mo) and color $b^*$ value of peanuts roasted at 140 to $160^{\circ}C$ showed a correlation of $b^*=21.61\;(M/Mo)^2-40.62\;(M/Mo)+34.12$ ($R^2=0.9123$). Overall changes in the tocopherol contents of peanuts and peanut oils were significantly affected by roasting temperature and time (p<0.05). Roasting at $140^{\circ}C$ caused a slight increase in the levels of tocopherols of peanuts over roasting time up to 20 min (p<0.05). There was no significant change in the tocopherol levels of peanuts during roasting at $150^{\circ}C$ for 20 min (p>0.05). At $160^{\circ}C$, the levels of tocopherols significantly decreased during the initial 10 min of roasting (p<0.05) while there was no extended loss after 10 min, resulting in about 5, 12, 20, and 10% losses of ${\alpha}$-, ${\beta}$-, ${\gamma}$- and ${\delta}$-T, respectively. After 20 min, total tocopherols decreased by 18%. However, tocopherol contents of pressed peanut oils significantly decreased at all roasting temperatures (p<0.05). After roasting peanuts at $160^{\circ}C$ for 20 min, about 84% of initial ${\alpha}$-T in peanut oils was retained. ${\alpha}$-T was the most stable to roasting while ${\gamma}$-T was the least. Swollen epidermal cells on the inner surface and broken cell walls of parenchyma tissue of peanut cotyledon were observed in peanuts after roasting at $160^{\circ}C$ for 15 min. Severe changes in microstructure of peanut by roasting would contribute to vitamin E stability because of exposure of oil droplets in peanuts to oxygen.

Enzyme Inhibitory and Anti-Proliferation Effects of Peanut Skin Extracts Depending on Cultivar (품종별 땅콩 종피 추출물의 효소저해활성 및 암세포주 증식억제 효과)

  • The purpose of this study was to investigate the inhibitory effect of enzyme activity and anti-proliferation of human cancer cell lines (HCT 116, NCI-H460 and MCF-7) of peanut skin depending on cultivars (Arachis hypogaea L. cv. K-Ol, cv. Sinpalkwang, cv. Daan, cv. Heuksaeng) and extraction solvent. Peanut skin was extracted with 80% ethanol, 80% methanol, 80% acetone, and distilled water, followed by analysis of the enzyme inhibitory activity and anticancer activity. Methanol extract of Daan cultivar most effectively inhibited ${\alpha}$-gluosidase (65.08%, 0.025 mg/mL), tyrosinase (82.49%, 2 mg/mL) and ACE (73.61%, 10 mg/mL). The inhibitory effect of peanut skin extracts on colon cancer cell (HCT-116), lung cancer cell (NCI-H460) and breast cancer cell (MCF-7) growth were investigate using MTT assay. The highest anti-proliferation of cancer cell line of peanut skin extracts was observed in the methanol extract of Daan cultivar. The cell viability on HCT 116, NCI-H460 and MCF-7 cell lines of methanol extracts from peanut skin of Daan cultivar was 48.13%, 41.03%, and 36.02% at $200{\mu}g/mL$, respectively. These results suggest that peanut skin extracts may mediate physiological activity, and provide valuable information for the use of peanut byproduct as a functional food material.

Analysis of Seed Oil Fatty Acids and Their Effect on Lipid Accumulation and Leptin Secretion in 3T3-L1 Adipocytes (헥산 추출 종실유의 지방산 분석 및 3T3-L1 지방세포의 지방 축적과 랩틴 분비에 미치는 영향 연구)

  • In this study, we evaluated the fatty acid composition and physiological activities of oils extracted from eight types of seeds, pepper (Capsicum annuum L.), green tea (Camellia sinensis L.), perilla (Perilla frutescens var. japonica Hara), peanut (Arachis hypogaea L.), cotton (Gossypium indicum LAM.), sesame (Sesamum indicum L.), walnut (Juglans regia L.), and safflower (Carthamus tinctorius L.). The composition and quality analysis showed that the oils were potentially suitable for foo-grade applications. The composition analysis showed that the oils were mostly composed of unsaturated fatty acids including linoleic acid and oleic acid. In 3T3-L1 adipocytes, green pepper, perilla, and peanut seed oils inhibited lipid accumulation, and green pepper, perilla, peanut, sesame, walnut, and safflower seed oils induced leptin secretion. These results show that the inhibitory effect of edible seed oils on lipid accumulation, and induction of leptin secretion may be useful for obesity management.

Phytotoxic Effect of Herbicides on Upland Crops and Weeds (밭작물(作物) 및 잡초(雜草)에 대한 제초제(除草劑)의 약해(藥害) 약효(藥效))

  • This study was conducted to select herbicides effective for upland crops and to investigate the cause of crop injury in peanut cultivated with mulching. Crop such as radish (Raphanus acanthiformis Moor.), Chinese cabbage (Brassica raps L.), soybean (Glycine max Merr.), Peanut (Archis hypogaea L.), and marsh mallow (Malva olitoria Nakai) were tolerant to napropamide [2-(${\alpha}$-naphthoxy)-N, N-diethylpropionamide], alachlor [2-chloro-2', 6'-diethyl-N-(methoxymethyl) acetanilide], trifluralin (${\alpha},{\alpha},{\alpha}$-trifluoro-2, 6-dinitro-N, N-dipropylp-toluidine) and nitrofen (2,4-dichlorophenyl-p-nitrophenylether). Napropamide, diphenamide (N, N-dimethyl-2, 2-diphenylacetamide) and alachlor were safe for red pepper (Capsicum annuum L.), eggplant (Solanum melongena L. and tomato (Lycopersicon esculentum Mill.), while trifluralin, nitrofen and chlonitrofen (2,4,6-trichlorophenyl-4-nitrophenyl ether) could be used for water melon (Citrullus battich Forsk.), carrot (Daucus carota L.) and lettuce (Lactuca scariola L.) without crop injury. Out of nine major weed species studied, Capsella bursa-pastoris Medicus was the most resistant species to the herbicides tested. Napropamide and alachlor could not control P. hydropiper, while P. oleracea and C. album were tolerant to diphenamide :and alachlor, respectively. Urea herbicides such as methabenzthiazuron [3-(2-benzothiazolyl)-1,3-dimethylurea], linuron [3-(3, 4-dichlorophenyl~l-methoxy-i-methyl urea], and isoproturon [3-(4-isopropylphenyl) -1, 1-dimethylurea]gave a great injury to the crops studied. The weeding effect was greater for broadleaf weeds than for grasses. Isoproturon and linuron provided good selectivity for marsh mallow and carrot, respectively. In peanut, the crop injury caused by Four herbicides studied was greater when cultivated with mulching than when cultivated without mulching. With dinitroaniline herbicides the crop injury decreased as the gaseous herbicide was removed out of mulching. Alachlor gave little phytotoxicity to peanut grown under mulching condition and nitralin [4-(methylsuphonyl)-2, 6-dinitro-N, N-dipropylaniline] showed less toxicity to the peanut than pendimenthalin (3,4-dimethyl-2, 6-dinitro-N-1-ethyl propylaniline) and trifluralin.

Influence of Soil Temperature on Growth and Nodulation Competition of Bradyrhizobium sp. Strains in the Rhizosphere of Peanut (온도(溫度)가 땅콩근류균(根瘤菌)의 근류형성(根瘤形成) 경합(競合)에 미치는 영향(影響))

  • Greenhouse experiments were conducted to avaluate strain competition, nodulation, patterns of nodule occupancy and population changes of Bradyrhizobium sp. strain HCR-46 $str^{r}cep^{r}$ and CB756 $str^{r}rif^{r}$ in the rhizosphere of peanut(Arachis hypogaea L.) under different root temperatures. Inoculated with two strains using seed coating with peat slurry under different root temperatures, population of each strain in the rhizosphere increased with plant growth and multiplication rate of inoculum in the unit weight of root were showed the highest from 10 to 15days after sowing. The multiplication rate of inoculum in the rhizosphere was $28^{\circ}C$>$34^{\circ}C$>$22^{\circ}C$. The density of HCR-46 $str^{r}cep^{r}$ was more increased than that of CB756 $str^{r}rif^{r}$ under $22^{\circ}C$ and $28^{\circ}C$. While the density of two strains showed no difference under $34^{\circ}C$. Inoculated with HCR-46 $str^{r}cep^{r}$ and CB756 $str^{r}rif^{r}$, respectively at 22, 28 and $34^{\circ}C$, nodulation of each strain was dominated in its inoculation portion. Inoculated with the mixture of HCR-46 $str^{r}cep^{r}$ and CB756 $str^{r}rif^{r}$, occupancy rate of HCR-46 $str^{r}cep^{r}$ was dominated over that of CB756 $str^{r}rif^{r}$ at $22^{\circ}C$ and $28^{\circ}C$, but that was similar between them at $34^{\circ}C$. Dry mass, nodulation, nitrogen content per plant and nitrogenase activity showed higher at $28^{\circ}C$ than at $32^{\circ}C$ and $22^{\circ}C$, while those were higher in HCR-46 $str^{r}cep^{r}$ and mixing HCR-46 $str^{r}cep^{r}$ with CB756 $str^{r}rif^{r}$ than in CB756 $str^{r}rif^{r}$.

Evaluation of Anti-wrinkle Effect of Peanut Shell Extract and Stability of the Extract in Cosmetic Products (땅콩 겉껍질 추출물의 주름개선 효능 및 화장품 제형에서 추출물의 안정성 평가)

  • This study was conducted to investigate the possibility of peanut shell, a by-product of peanut, as a functional cosmetic ingredient. Peanut shell extract showed high antioxidant activity with IC50 values of 75.00, 46.33, and 472.83 ㎍/mL for DPPH and ABTS radical scavenging and SOD-like activity, respectively. Furthermore, peanut shell extract was efficiently decreased the MMP-1 and MMP-3 protein level in the UVB treated-HaCaT cell and maintained procollagen protein level similar to normal control. Similar to anti-wrinkle related protein expression assay, the IC50 value of elastase and collagnease inhibition in peanut shell extract was lower as 0.30 and 0.09 mg/mL, respectively, than that of the positive control. Additionally, eriodictyol and luteolin, which are isolated from peanut shell extract, showed 53.8 and 98.0% elastase inhibition rate, respectively, and 60.1 and 72.5% collagenase inhibition rate, respectively, at a concentration of 0.1 mg/mL. Thus, luteolin was assumed to be the effective ingredient for wrinkle inhibition in peanut shell extract. As a result of stability evaluation of lotion and cream formulations containing peanut shell extract, it was confirmed to be a stable formulation with no significant changes. Therefore, it is considered that peanut shell extract can be applied as a cosmetic ingredient for wrinkle inhibition.
