BACKGROUND/OBJECTIVES: Cadmium is a toxic metal that is an occupational and environmental concern especially because of its human carcinogenicity; it induces serious adverse effects in various organs and tissues. Even low levels of exposure to cadmium could be harmful owing to its extremely long half-life in the body. Cadmium intoxication may be prevented by the consumption of dietary components that potentially reduce its accumulation in the body. Dietary chitosan is a polysaccharide derived from animal sources; it has been known for its ability to bind to divalent cations including cadmium, in addition to other beneficial effects including hypocholesterolemic and anticancer effects. Therefore, we aimed to investigate the role of dietary chitosan in reducing cadmium accumulation using an in vivo system. MATERIALS/METHODS: Cadmium was administered orally at 2 mg (three times per week) to three groups of Sprague-Dawley rats: control, low-dose, and high-dose (0, 3, and 5%, respectively) chitosan diet groups for eight weeks. Cadmium accumulation, as well as tissue functional and histological changes, was determined. RESULTS: Compared to the control group, rats fed the chitosan diet showed significantly lower levels of cadmium in blood and tissues including the kidneys, liver, and femur. Biochemical analysis of liver function including the determination of aspartate aminotransferase and total bilirubin levels showed that dietary chitosan reduced hepatic tissue damage caused by cadmium intoxication and prevented the associated bone disorder. CONCLUSIONS: These results suggest that dietary chitosan has the potential to reduce cadmium accumulation in the body as well as protect liver function and bone health against cadmium intoxication.
Black chokeberry (Aronia melanocarpa), a rich source of polyphenols, exerts hypocholesterolemic effects. However, little is known about its effects on the regulation of the hepatic cholesterol metabolism and the underlying mechanisms. In the present study, the effects of polyphenol-rich black chokeberry extract (CBE) on hepatic cholesterol metabolism were investigated by measuring the expression of genes involved in the absorption, de novo synthesis, and efflux of cholesterol in HepG2 cells. There was a significant reduction in the expression levels of genes involved in cholesterol metabolism, the low-density lipoprotein receptor, 3-hydroxy-3-methylglutaryl coenzyme A reductase, and sterol regulatory element-binding protein 2, in CBE-treated HepG2 cells. Meanwhile, CBE increased the expression levels of genes involved in cholesterol and bile acid efflux. The expression levels of mitochondrial fatty acid oxidation genes increased, whereas those of lipogenic genes decreased following CBE treatment. These data suggest that the consumption of black chokeberry may be beneficial for the prevention of hypercholesterolemia.

Plantain was extracted with ethanol and fractionated systemically with n-hexane, chloroform, ethylacetate, n-butanol and water to study inhibitory effect on cholesterol synthesis in vitro. To screen the effect, inhibitory activities on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase obtained from Saccharomyces cerevisiae were examined using the five fractions of Plantain. The HMG-CoA reductase activity was inhibited most by ehylacetate fraction among the fractions, although the all five fractions had the effect To see the hypocholesterolemic effect of the ethylacetate fraction of Plantanin (PAE) in vivo, male Sprague-Dawley rats were received 5 types of diets for 6 weeks: normal diet group (NOR), high cholesterol diet group($1\%$ cholesterol and $0.25\%$ sodium cholate, CON), normal diet and PAE 70 mg/kg administered group(S1), high cholesterol diet and PAE 140 mg/kg administrated group(S2), and high cholesterol diet and PAE 140 mg/kg administered group(S3). Body weight gains of the CON were significantly increased compared to those of S1, S2 and S3. Activities of serum AST and ALT were tended to be increased in CON compared with NOR and reduced by the PAE administration. Concentrations of serum total cholesterol, free cholesterol, LDL-cholesterol, triglyceride and the atherogenic index were tended to be decreased in the PAE administered groups compared with the CON. HDL-cholesterol and phospholipid concentrations were significantly decreased in the CON and markedly increased by the PAE administered groups. Taten together, it is suggested that the ethylacetate fraction of Plantanin exerts antiatherosclerotic effect by reducing serum cholesterol concentrations in rats fed high cholesterol diets.
Hypocholesterolemic effect of soy protein was examined in comparison with casein and three other dietary protein sources in chicks. In two feeding trials, 40 (Expt.1) or 50 (Expt. 2), three-day-old, male chicks were forced-fed each of five semi-purified-type diets containing isolated soy protein (ISP, cp 82%), casein (cp 92%), rice protein (RP, cp 70%), corn gluten meal (CGM, cp 65%) or fish meal (FM, cp 70%) for two weeks. The diets for Expt. 2 were supplemented with 0.3% cholesterol by replacing glucose. Each protein source was the only source of protein of each diet. Essential amino acids were added to the diets to satisfy their requirements according to NRC. The diets were forced-fed to equalize the intake of all nutrients except the amino acids which were inherently variable in the diets. Chicks fed casein showed lower body weight gain than those fed the other proteins in both experiments. Birds fed ISP and FM gained better body weight than the others. Chicks fed casein showed significantly higher levels of plasma total cholesterol, non-HDL cholesterol and triacylglycerol (TG) than those fed ISP and the other protein sources. Meanwhile, the chicks fed ISP, RP, CGM and FM showed comparable levels of plasma total cholesterol, non-HDL cholesterol and TG. In Expt. 2, the birds fed casein and FM showed markedly elevated plasma total cholesterol and non-HDL cholesterol levels. Liver weight and levels of total lipids and cholesterol of chicks fed casein appeared significantly higher than those of the other protein diets, whereas those of the chicks fed ISP, RP, CGM and FM appeared comparable except cholesterol in FM group. In conclusion, only the chicks fed casein diets in both experiments always showed significantly higher levels of plasma cholesterol and TG compared to those fed ISP and the other protein sources. These results support the views that casein, which has unique lysine-arginine ratio, is inherently hyper-cholesterolemic, and ISP is hypocholesterolemic only when compared to casein.
The purpose of this study was to investigate the effects of medical nutrition therapy(MNT) on plasma lipid levels of hyperlipidemic patients with apo E3 genotype according to Sasang Constitutions. From March to July, 2001, the 33 hyperlipidemic patients admitted to K University Medical Center were studied. The study subjects were classified according to their Sasang constitutions by QSCC II questionnaire which have been used at K University Oriental Medical Center. The anthropometric assessments, blood analysis, and apo E genetic typing were carried out. Nutrient intake was determined by food record method of food taken during two weekdays and one weekend. The MNT including the instruction for hypolipidemic and hypocholesterolemic diet (step l diet) was performed for 12 weeks. The results are as follow; (1) The mean age of hyperlipidemic patients was $49.91{\pm}8.48$ years. (2) The distributions of Sasang Constitution were 60.6% of Tae-eum, 21.2% of So-yang, and 18.2% of So-eum. The distributions of apo E genotype were 6.5% of apo E2/3, 78.8% of apo E 3/3, and 15.2% of apo E 3/4. (3) The nutrient consumption of the apo E3 subjects before the MNT showed lower calorie, iron, calcium, and vitamin B2 intakes than the RDAs for each nutrients with no significant differences among the constitutions. After 12-week of MNT, only the fat consumption was decreased in the Tae-eum group. The MNT did not change the pattern of food intake. (4) The plasma level of triglyceride, total cholesterol, and LDL-C were not changed after MNT in the three constitutional groups. The level of HDL-C was significantly increased significantly in Tae-eum and So-yang group and the level of homocystein was lowered in Tae-eum group after MNT. It could be concluded that the 12-weeks MNT with hypolipidemic and hypocholesterolemic diet did not change the level of total cholesterol, triglyceride, and LDL-C effectively regardless of Sasang constitutions even though the subjects' dietary intake was improved by MNT.
This study was performed to investigate the effects of lotus root ethanol extinct (LRE) on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity in vitro, and lipid metabolism in the serum and liver of rate fed normal or high cholesterol diet in vivo. LRE (200 mg/kg/day and 400 mg/kg/day) was administered only to rats with fed high cholesterol diet for 6 week. We divided into 6 groups: normal diet group (NC), high cholesterol diet group (1% cholesterol and 0.25% sodium cholate)(HC), LRE 200 mg/kg treated group (NC-LREL), LRE 400 mg/kg teated group (NC-LREH), high cholesterol diet and LRE 200 mg/kg treated group (HC-LREL), and high cholesterol diet and LRE 400 mg/kg teated group (HC-LREH). LRE significantly inhibited the HMG-CoA reductase activity in a concentration-dependent manner in vitro. The body weight gain and liver weight of the high cholesterol diet group were higher than the normal diet group whereas the groups administered LRE were gradually decreased. The high cholesterol diet group increased serum triglyceride, total cholesterol, free cholesterol and LDL-cholesterol levels, and decreased atherogenic index, HDL-cholesterol and phospholipid levels as compared with the normal diet group. LRE administrated groups were increased in serum HDL-C/T-C, HDL-cholesterol and phospholipid levels, and decreased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels as compared with the high cholesterol diet group. These effect of LRE within the high cholesterol diet groups were concentration-dependent manners. There were no differences in the levels of serum triglyceride, total cholesterol, phopholipid, HDL-cholesterol and free cholesterol between normal diet groups. The hepatic LRE administrated groups than in the high cholesterol diet group. Teken together, it is suggested that LRE exerts hypocholesterolemic effect by reducing serum cholesterol concentration in rats with high cholesterol diet-induced hypercholesterolemia.

This study was performed to investigate the effect of ethanol extract of Pimpinella brachycarpa (PB) on serum and liver lipid metabolism in rats. Male Sprague Dawley rats were administered 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia. PB ethanol extract (200 mg/kg/day or 400 mg/kg/day) was also administered orally to rats with high cholesterol diet for 6 weeks. We divided 40 rats into five groups; normal diet group (NC), high cholesterol diet group (HC), normal diet and PB ethanol extract (200 mg/kg) administered group (NC-PB), high cholesterol diet and PB ethanol extract (200 mg/kg) administered group (HC-PBL), and high cholesterol diet and PB ethanol extract (400 mg/kg) administered group (HC-PBH). The growth rate and liver weight of the high cholesterol diet group was higher than those of the normal diet group, whereas those of the groups administered PB ethanol extract were gradually decreased. There was a signigicant increase in the activities of serum alanine aminotransferase (ALT), asparate aminotransferase (AST) and alkaline phosphatase (ALP) in the high cholesterol diet group. The administration of PB ethanol extract decreased serum ALT, AST and ALP activities in a dose-dependent manners. The high cholesterol diet group showed increased serum triglyceride, total cholesterol, free cholesterol and LDL-cholesterol levels, and decreased atherogenic index, HDL-cholesterol and phospholipid levels as compared with the normal diet group. PB ethanol extract administrated groups showed increased HDL-C/T-C, HDL-cholesterol and phospholipid levels, and decreased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels as compared with the high cholesterol diet group. There were no differences in the concentrations of serum triglyceride, phopholipid, LDL-cholesterol, HDL-cholesterol and free cholesterol between normal diet groups. The hepatic concentrations of total cholesterol and triglyceride were also lower in PB ethanol extract administrated groups than in the high cholesterol diet group. These results suggest that ethanol extract of PB exerts hypocholesterolemic effect by reducing serum and liver cholesterol contents.
The objective of this study was to determine the effect of slaughter season on the fatty acid profile in four types of fat deposits in crossbred (Polish Holstein Friesian Black-and-White${\times}$Limousine) beef bulls. The percentage share of fatty acids was determined by gas chromatography and were divided into the following categories of fatty acids: saturated (SFAs), unsaturated (UFAs), monounsaturated (MUFAs), polyunsaturated (PUFAs), desirable hypocholesterolemic (DFAs) and undesirable hypercholesterolemic (OFAs), n-3 and n-6. Perinephric fat was characterized by the highest SFA concentrations (59.89%), and subcutaneous fat had the highest MUFA content (50.63%). Intramuscular fat was marked by a high percentage share of PUFAs and the highest PUFA/SFA ratio. The slaughter season had a significant effect on the levels of C18:3, C20:4 ($p{\leq}0.01$) and conjugated linoleic acid ($p{\leq}0.05$). There was an interaction between the slaughter season and fat type for the content of C20:4 ($p{\leq}0.01$) and C20:5 ($p{\leq}0.05$). The results of this study show that beef from cattle slaughtered in the summer season has a higher nutritional value and more health-promoting properties.
The main constituent of green tea, catechins have been reported to have numberous biological anti-vites including antimutagenic, antibacterial, hypocholesterolemic, antioxidant and antitumor properties. In the present study, we examined the protective effect of catechin on UVB-induced skin damage. Catechin (3 mg/mouse) was topically treated to dorsal area of SHK-1 hairless mouse daily for 2 weeks. UVB (100 mJ/$\textrm{cm}^2$) was also treated soon after application of catechin alone or with catechin for 2 weeks. Catechin reduced UVB-induced infiltration of inflammatory cells, fibrosis of cells and collagen-fiber formation. In addition, catechin also prevented UVB-induced DNA fragmentation and apoptosis cell number, but not changed p53 level. Furthermore catechin inhibited UVB-induced cell proliferation. There results showed that catechin have preventive effect aganinst UVB-induced skin damages. and these effects could contribute to the antitumor promoters activity.

This experiment was undertaken to investigate the effect of caluium and magnesium on the lipid and mineral composition of serum and tissues in cholesterol-fed rats. GOT of serum was significantly decreased in normal Ca and high Mg supplemented group. Serum cholesterol level was significantly decreased in high Mg supplemented group, whereas that of low Mg supplemented group was significantly increased. Content of total lipid in liver were significantly decreased by supplementation with high calcium of magnesium. However, the content of cholesterol was not affected by supplementation with high calcim and that of low Mg supplemented group was significantly increased. In fatty acid composition of liver, he the order of fatty acid content is as following; oleic acid (18:1), stearic acid(18:0), and palmitic acid(16:0). Levels of Ca, Mg, K and Cu in liver were significantly affected by dietary calcium and magnesium levels. The Mg level of kidney was decreased but Cu level was elevated in cholesterol supplemented group. Levels of Ca, Mg, and Zn in serum were significantly elevated by supplementation with high calcium or magnesium.
