고혈압 흰쥐에서 Cumambrin A의 혈압 강하 효과 (Effect of Cumambrin A Treatment on Blood Pressure in Spontaneously Hypertensive Rats)

  • We have isolated a sesquiterpene lactone, cumambrin A from the dried flowers of Chrysanthemum boreale Makino and reported its chemical structure. The aim of the present study was to investigate whether the exogenous administration of cumambrin A has a pharmacological effect on normalization of blood pressure in the spontaneously hypertensive rats (SHR). In vitro studies: Relaxative response induced by cumainbrin A was increased with dose-dependent manner and showed maximizing response at a concentration of $5{\times}10^{-4}M$. Further, this relaxative response was significantly increased at a condition of endothelium present than that of endothelium denuded. In vivo studies: The normalizing effect of cumambrin A on blood pressure was also increased with time-dependent manner and then gradually recovered to normal condition at approximately 4 hrs after cumambrin A treatment.

The Relationship between Hypertension and Central Serotonergic Nervous System Activity in Spontaneously Hypertensive Rats

  • Relationship between the maintenance of hypertension and central serotonergic nervous system activity in opontaneously hypertensive rats (SHR) was studied. Serotonin turnover-rates were measured in 5 brain areas as an index of serotonergic neuronal activity and compared at the ages of 14 weeks in two types of animals; (1) spontaneously hypertensive rats (SHR) (2) normotensive wistar kyoto rats (WKY). In 14-week old SHR, central serotonin turnover rate was significantly lower in telencephalon, hypothalamus/thalamus and midbrain than normotensive rat, but significantly higher in cerebellum. There were no significant differences between serotonin turnover rate in pons/medulla of SHR and that of normotensive rat. THese data suggest that the abnormally lower turnover rates of serotonin in telencephalon, hypothalamus/thalamus and midbrain may be one of the underlying neuronal factors for manifestation of hypertension in SHR.

오령산(五苓散)의 본태성 고혈압 동물모델에서 혈압강하 및 혈관 이완 효과 (Effect of Oryeongsan on Spontaneously Hypertensive Rat decrease of Blood Pressure and Vasodilatory)

  • Oryeongsan (ORS), a formula composed of five herbal medicines, has long been used to treat impairments of the regulation of body fluid homeostasis. The purpose of this study was to determine the antihypertensive and renal protective effects of ORS in rats with hypertension. Spontaneously hypertensive rats (SHR) were divided into two groups with similar mean baseline systolic blood pressure (SBP). Then, 1 mL/kg of vehicle (distilled water) or 1.5, 3 g/kg of ORS extract were administered orally once a day for 4 weeks. SBP and diastolic blood pressure (DBP) were measured at weeks 1, 2, 3 and 4. At the end of the experiment, blood was collected, and heart were removed for histology. By the 2 weeks after initiation of treatment, the ORS treated group had significantly lower SBP than SHR rats. The ORS treatment significantly improved blood pressure and echocardiogram parameters compared to hypertensive rats. Additionally, the left ventricular (LV) remodeling and LV dysfunction were significantly improved in ORS treated group hypertensive rats. Furthermore, an increase in fibrotic area has been observed in SHR rats compared with Wistar-Kyoto rats (WKY). Furthermore, administration of ORS significantly attenuated cardiac fibrosis in hypertensive rats. Therefore, these findings suggest that ORS has a protective effect on heart failure by alleviating hypertensive heart disease and cardiovascular dysfunction in SHR.

  • Objective : The aim of this experiments was to investigate the effects of Kyongo Puryu(originated from ${\ll}$Classic on Difficulty${\gg}$), Kyongo Um-gok(originated from ${\ll}$Ling Shu${\gg}$) acupuncture on the Blood Pressure, Cardiomegalic index, and plasma levels of atrial natriuretic peptide in Hypertensive RAT induced by 2K1C. Methods : The effects of Kyongo(L8) Puryu(K7), Kyongo(L8) Um-gok(K10) acupuncture on Blood Pressure in Hypertensive RAT induced by Two Kidney One Clip(2K1C). Results : I. Blood Pressure was decreased significantly after acupuncture of L8 K7. II. Cardiomegalic index decreased significantly after acupuncture of L8 Puryu, L8 K10. IlI. Plasma levels of atrial natriuretic peptide was increased significantly after acupuncture of L8 K7.

Kami-bang-pung-tong-sung-san is Involved in Regulating Physiological Parameters Associated with Hypertension in Spontaneously Hypertensive Rat

  • KBPT is the fortified prescription of Bang-pung-tong-sung-san(BPTS) by adding Spatholobi Clulis and Salviae Miltiorrzae Radix. BPTS prescription has been utilized in oriental medicine for the treatments of vascular diseases including hypertension, stroke, and arteriosclerosis. Yet, the overall mechanism underlying its activity at the cellular levels remains unknown. Using spontaneously hypertensive rat (SHR) model, we investigated whether the KBPTS has an effect on the pathophysiological parameters related to hypertension. Pretreatment of SHR with KBPTS was found to lower blood pressure and heartbeat rate. Levels of aldosterone. dopamine, and epinephrine were found to be significantly reduced in the serum of KBPTS-treated SHR. Histological examination of adrenal cortex and superior aorta showed that tissues from KBPTS-treated SHR rats were more intact and cleaner compared to saline-treated control. Levels of superoxide dismutase (SOD) protein in adrenal gland, aorta, myocardial tissue, and kidneys were higher in KBPTS-treated animals than control group. The present data suggest that KBPTS may play a role in normalizing cardiovascular function in SHR by controlling hypertension-related blood factors and superoxide stressors.

Effect of gemigliptin on cardiac ischemia/reperfusion and spontaneous hypertensive rat models

  • Diabetes is associated with an increased risk of cardiovascular complications. Dipeptidyl peptidase-4 (DPP-IV) inhibitors are used clinically to reduce high blood glucose levels as an antidiabetic agent. However, the effect of the DPP-IV inhibitor gemigliptin on ischemia/reperfusion (I/R)-induced myocardial injury and hypertension is unknown. In this study, we assessed the effects and mechanisms of gemigliptin in rat models of myocardial I/R injury and spontaneous hypertension. Gemigliptin (20 and 100 mg/kg/d) or vehicle was administered intragastrically to Sprague-Dawley rats for 4 weeks before induction of I/R injury. Gemigliptin exerted a preventive effect on I/R injury by improving hemodynamic function and reducing infarct size compared to the vehicle control group. Moreover, administration of gemigliptin (0.03% and 0.15%) powder in food for 4 weeks reversed hypertrophy and improved diastolic function in spontaneously hypertensive rats. We report here a novel effect of the gemigliptin on I/R injury and hypertension.

발현성 고혈압쥐의 관상순환 기능과 심장근의 에너지 대사에 관한 생체외 에너지원의 관류 연구 (Substrate-Perfusion Studies on Coronary Circulation and Myocardial Energy Metabolism in Spontaneously Hypertensive Rat Hearts)

  • The effects of energy-yielding substrates on coronary circulation, myocardial oxygen metabolism, and intramyocytic adenylates of perfused Wistar control rat(WC) and spontaneously hypertensive rat(SHR) hearts were examined under basal and $\beta$-adrenergic stimulation conditions. The perfusion medium (1.0mM Ca2+) contained 5mM glucose (+5U/l insulin) in combination with 5mM pyruvate, 5mM lacate, 5mM acetate, or 5mM octanoate as energy substrates. Hearts were perfused with each substrate buffer for 20min under basal conditions. Coronary functinal hyperemia was induced by infusing for 20min isoproterenol (ISO, 1uM), a $\beta$-receptor agonist. Cardiac adenylates, glycolytic intermediates, and coronary venous lactate were measured by using an enzymatic analysis technique. Under basal conditions, acetate and octanoate significantly increased coronary flow(CF) of WC in parallel with myocardial oxygen consumption. However, CF of SHR was partly attenuated by coronary vasoconstriction despite metabolic acidosis. In addition, pyruvate and lactate depressd ISO-induced coronary functional hyperemia in SHR. It should be noted that octanoate exhibited coronary dysfunction under ISO conditions. On the other hand, fat substrates depleted myocardial high energy phosphate pool and accumulated breakdown intermediates. In SHR with coronary vasoconstriction under basal conditions, and with depressed coronary functional hyperemia, high energy phosphates were greatly depleted. These results suggest that energy substrates in the myocardium and coronary smooth muscle alter remarkably coronary circulation, and that coronary circulatory function is associated with a reserve of high energy phosphates and a balance between breakdown and nono synthesis of energy phosphates. These findings could be explained by alterations in the cytosolic redox state manipulated by LDH and hence in the cytosolic phosphorylation potential, which might be involved in hypertension of SHR.

  • There have been reports on the aberration of the control mechanisms of the blood pressure, hormone secretion, and renal functions in spontaneously hypertensive rats (SHR). However, the contribution of the renin-angiotensin system in the maintenance of high blood pressure in SHR is still controversial. Recently, it has been reported that the negative feedback short loop control mechanism of the renin-angiotensin system may be changed in SHR. In the present experiment, it was attempted to explore the possible alterations in the effect of arginine vasopressin (AVP) on the renal function in SHR. Experiments have been done in anesthetized SHR as well as in normotensive Wistar and Sprague-Dawley rats as control groups. Pharmacologic doses of AVP (10-13 mU/rat/10 min) decreased urine volume, excreted amount of creatinine and para-amino-hippuric acid. No differences in these parameters was observed between normotensive and hypertensive rats. AVP increased sodium and potassium excretion, but the responses in SHR were suppressed as compared with normotensive rats. Intravenous infusion of AVP also increased blood pressure in normotensive and hypertensive rats and a vasopressor effect of AVP was attenuated in SHR. There was a positive correlation between the changes in blood pressure and excreted amount of sodium during AVP infusion. These data suggest that the attenuated natriuretic effect of intravenous infusion of AVP may be due to a difference in renal tubular responsiveness to AVP but not due to a difference in vasopressor responsiveness.

Anti-hypertensive Effects of Electroacupuncture at ST36 In Spontaneously Hypertensive Rat

  • The present study investigated whether electroacupuncture (EA) can alleviate hypertension and concomitant disorders such as decreased kidney weight and elevation of serum creatinine in spontaneous hypertensive rats. EA (2 Hz, 3 mA, 10 min) was applied to Joksamni (ST36) once daily for 7 days. Body weight, blood pressure and heart rate were measured on Day 0, 2, 6, 8, 10 and 12, and kidney weight and serum creatinine levels were examined after sacrifice (on Day 12 after last examination). In the ST36 group, the blood pressure were significantly decreased from 6thdays and its effects lasted up to Day12 (up to 5days after cessation of acupuncture), compared to control. And, significant decreases of the heart rates after EA at ST36 were also observed on Day 2, 6, and 8. However, there were no significant differences in daily body weight, kidney weight and serum creatinine between acupuncture and control group. These results showed that EA at ST36 caused anti-hypertension by decreasing blood pressure and heart rates in spontaneous hypertenstive rats, although it failed to alleviate concomitant disorders such as the decreased kidney weight and elevation of serum creatinine shown in hypertensive rats.

Charateristics of Voltage Dependent Calcium Uptake and Norepinephrine Release in Hypothalamus of DOCA-salt Hypertensive Rats

  • Purpose of the present study was to clarify the role of noradrenergic neural activities in hypothalamus for either triggering or maintaining hypertension in deoxycorticosterone (DOCA)-salt hypertensive rats. Two groups of animals were prepared: 1) normotensive Wistar rats and 2) DOCA-salt induced hypertensive rats. Voltage dependent $^{45}Ca^{++}$ uptake, endogenous norepinephrine release, and the catecholamine content in the hypothalamus of DOCA-salt hypertensive and normotensive Wistar rats were compared. Animals at 4, 6 and 16 week-old of two groups were sacrificed by decapitation and hypothalamus was dissected out. Voltage dependent calcium uptake and norepinephrine release were determined from hypothalamic synaptosomes either in low potassium or high potassium stimulatory condition by using $^{45}Ca^{++}$ isotope and HPLC-ECD technique. Degrees of voltage dependent $^{45}Ca^{++}$ uptake and norepinephrine release in hypothalamic synaptosomes of 16-week-old DOCA-salt hypertensive rats were significantly greater than those of age matched normotensive control rats. The norepinephrine and dopamine contents of hypothalamus were about the same in two groups of animals. These results suggest that the alteration of evoked norepinephrine release related to calcium uptake in hypothalamus may play a role in the maintenance of hypertension in DOCA-salt hypertensive rats.
