병독성 콩모자이크바이러스계통에 감염된 콩판별품종의 미세구조의 비교 (Ultrastructural Comparison of Soybean differentials Infected with a Virulent SMV Strain)

  • Two soybean cultivars, Kwanggyo and Hwanggeum (soybean mosaic potyvirus (SMV)-resistant cultivars), that had been inoculated with a virulent strain (G-5H, 4) of soybean mosaic potyvirus produced necrotic lesions on inoculated leaves as well as on upper trifoliate leaves. Cells in the lesion area contained sparse numbers of virus particles and very few characteristic pinwheel inclusions. Although a hypersensitive-like cellular response occurred in the two resistant cultivars, this response did not prevent the virus from spreading systemically in these resistant hosts, indicating a different mechanism from the general hypersensitive reaction in relation to host resistance.

  • 1. The purpose of study To study the effectiveness of Yangkyuksanhawtang(YST) against Allergic Contact Dermatitis(ACD) I observed the change of cutaneous shape, histochemistry, immunohistochemistry, and the distribution of apoptotic cells. 2. The material and method of study I divided 4-month-old rats into three groups of 10, which are a contrastive group of having applied Acetone olive oil only, ACD group to have intentionally activated Allergic Contact Dermatitis by DNCB and YST group to give medication of Yangkyuksanhawtang extract. And I observed each group of mice after 24, 48 and 72 hours. 3. The result and conclusion of study The change of cutaneous shape and hypersensitive reaction of immunity was significantly decreased by YST in ACD rats. I consider that YST suppressed hypersensitive reaction of immunity and inflammation in ACD.

Identification of hrcC, hrpF, and maA Genes of Xanthomonas campestris pv. glycines 8ra: Roles in Pathogenicity and Inducing Hypersensitive Response on Nonhost Plants

  • Nonpathogenic mutants of Xanthomonas campestris pv. glycines were generated with Omegon-Kim to isolate genes essential for pathogenicity and inducing hypersensitive response (HR). Three nonpathogenic multants and two mutants showing slow symptom development were isolated among 1,000 colonies tested. From two nonpathogenic mutants, 8-13 and 26-13, genes homologous to hrcC and hrpF of X. campestris pv. vesicatoria were identified. The nonpathogenic mutant 8-13 had a mutation in a gene homologous to hrpF of X. campestris pv. vesicatoria and failed to cause HR on pepper plants but still induced HR on tomato leaves. The nonpathogenic mutant 26-13 had an insertional mutation in a gene homologous to hrcC of X. campestris pv. vesicatoria and lost the ability to induce HR on pepper leaves but still caused HR on tomato plants. Unlike other phytopathogenic bacteria, the parent strain and these two mutants of X. campestris pv. glycines did not cause HR on tobacco plants. a cosmid clone, pBL1, that complemented the phenotypes of 8-13 was isolated. From the analysis of restriction enzyme mapping and deletion analyses of pBL1, a 9.0-kb Eco RI fragment restored the phenotypes of 8-13. pBL1 failed to complement the phenotypes of 26-13, indicating that the hrcC gene resides outside of the insert DNA of pBL1. One nonpathogenic mutant, 13-33, had a mutation in a gene homologous to a miaA gene encoding tRNA delta (2)-isopentenylpyrophosphate transferase of Escherichia coli. This indicated that tRNA modifications in X. campestris pv. glycines may be required for expression of genes necessary for pathogenicity. The mutant 13-33 multiplied as well as the parent strain did in the culture medium and in planta, indicating that loss of pathogenicity is not due to the inability of multiplication in vivo.

Identification and Expression Profiles of Six Transcripts Encoding Carboxylesterase Protein in Vitis flexuosa Infected with Pathogens

  • Plants protect themselves from pathogen attacks via several mechanisms, including hypersensitive cell death. Recognition of pathogen attack by the plant resistance gene triggers expression of carboxylesterase genes associated with hypersensitive response. We identified six transcripts of carboxylesterase genes, Vitis flexuosa carboxylesterase 5585 (VfCXE5585), Vf-CXE12827, VfCXE13132, VfCXE17159, VfCXE18231, and VfCXE47674, which showed different expression patterns upon transcriptome analysis of V. flexuosa inoculated with Elsinoe ampelina. The lengths of genes ranged from 1,098 to 1,629 bp, and their encoded proteins consisted of 309 to 335 amino acids. The predicted amino acid sequences showed hydrolase like domains in all six transcripts and contained two conserved motifs, GXSXG of serine hydrolase characteristics and HGGGF related to the carboxylesterase family. The deduced amino acid sequence also contained a potential catalytic triad consisted of serine, aspartic acid and histidine. Of the six transcripts, Vf-CXE12827 showed upregulated expression against E. ampelina at all time points. Three genes (VfCXE5585, VfCXE12827, and VfCXE13132) showed upregulation, while others (VfCXE17159, VfCXE18231, and VfCXE47674) were down regulated in grapevines infected with Botrytis cinerea. All transcripts showed upregulated expression against Rhizobium vitis at early and later time points except VfCXE12827, and were downregulated for up to 48 hours post inoculation (hpi) after upregulation at 1 hpi in response to R. vitis infection. All tested genes showed high and differential expression in response to pathogens, indicating that they all may play a role in defense pathways during pathogen infection in grapevines.

Rpi-blb2-Mediated Hypersensitive Cell Death Caused by Phytophthora infestans AVRblb2 Requires SGT1, but not EDS1, NDR1, Salicylic Acid-, Jasmonic Acid-, or Ethylene-Mediated Signaling

  • Potato Rpi-blb2 encodes a protein with a coiled-coil-nucleotide binding site and leucine-rich repeat (CC-NBSLRR) motif that recognizes the Phytophthora infestans AVRblb2 effector and triggers hypersensitive cell death (HCD). To better understand the components required for Rpi-blb2-mediated HCD in plants, we used virus-induced gene silencing to repress candidate genes in Rpi-blb2-transgenic Nicotiana benthamiana plants and assayed the plants for AVRblb2 effector. Rpi-blb2 triggers HCD through NbSGT1-mediated pathways, but not NbEDS1- or NbNDR1-mediated pathways. In addition, the role of salicylic acid (SA), jasmonic acid (JA), and ethylene (ET) in Rpi-blb2-mediated HCD were analyzed by monitoring of the responses of NbICS1-, NbCOI1-, or NbEIN2-silenced or Rpi-blb2::NahG-transgenic plants. Rpi-blb2-mediated HCD in response to AVRblb2 was not associated with SA accumulation. Thus, SA affects Rpi-blb2-mediated resistance against P. infestans, but not Rpi-blb2-mediated HCD in response to AVRblb2. Additionally, JA and ET signaling were not required for Rpi-blb2-mediated HCD in N. benthamiana. Taken together, these findings suggest that NbSGT1 is a unique positive regulator of Rpi-blb2-mediated HCD in response to AVRblb2, but EDS1, NDR1, SA, JA, and ET are not required.

알레르기 비염을 포함하는 과민성 비염 환자 580례에 대한 임상적 고찰sdf (A Clinical Study of 580 Cases on Hypersensitive Rhinitis including Allergic Rhinitis)

  • Objective : The purpose of this study is to investigate outpatients of hypersensitive rhinitis including allergic rhinitis who had visited to Hospital of Oriental Medicine. Method : We analyzed statistics study in 580 patients, who had visited to the Dept. of dermatology, ophthalmology & otorhinolayngology Hospital of Oriental Medicine Dongeui University from March, 1998 to December, 2006. Results : The results were as follows. 1. There is more male(53.28%) than femal(46.72%). The age distributions of patients showed highest in 20-29 years, followed by 10-19 years, 30-39 years in order. 2. The yearly distributions of patients showed highest in 2004, followed by 2001, 2005 in order. The seasonal distributions of patients showed highest in fall, followed by winter, summer, spring in order. The monthly distributions of patients showed highest in September, followed by August, October in order. 3. The distributions of number of times in treatment showed highest in 1-5 times. The distributions of duration of treatment showed highest in 1 week. In the treatment method, a major portion of treatment methods was acupuncture, followed by herbal medicine treatment, aroma therapy in order. 4. The Frequency of use of Prescriptions showed highest in Bojungikgi-Tanggami. The Frequency of use of Extract showed highest in Sochungryong-Tang. 5. The distribution of past history showed highest in Atopic dermatitis. The distribution of Rhinologic Complication showed highest in Paranasal sinusitis.

Role of Trehalose Synthesis in Ralstonia syzygii subsp. indonesiensis PW1001 in Inducing Hypersensitive Response on Eggplant (Solanum melongena cv. Senryo-nigou)

  • Ralstonia syzygii subsp. indonesiensis (Rsi, former name: Ralstonia solanacearum phylotype IV) PW1001, a causal agent of potato wilt disease, induces hypersensitive response (HR) on its non-host eggplant (Solanum melongena cv. Senryo-nigou). The disaccharide trehalose is involved in abiotic and biotic stress tolerance in many organisms. We found that trehalose is required for eliciting HR on eggplant by plant pathogen Rsi PW1001. In R. solanacearum, it is known that the OtsA/OtsB pathway is the dominant trehalose synthesis pathway, and otsA and otsB encode trehalose-6-phosphate (T6P) synthase and T6P phosphatase, respectively. We generated otsA and otsB mutant strains and found that these mutant strains reduced the bacterial trehalose concentration and HR induction on eggplant leaves compared to wild-type. Trehalose functions intracellularly in Rsi PW1001 because addition of exogenous trehalose did not affect the HR level and ion leakage. Requirement of trehalose in HR induction is not common in R. solanacearum species complex because mutation of otsA in Ralstonia pseudosolanacearum (former name: Ralstonia solanacearum phylotype I) RS1002 did not affect HR on the leaves of its non-host tobacco and wild eggplant Solanum torvum. Further, we also found that each otsA and otsB mutant had reduced ability to grow in a medium containing NaCl and sucrose, indicating that trehalose also has an important role in osmotic stress tolerance.
