Detect of Hypericin (HyH) gene in Hypericum erectum in Korea and Comparison of H. perforatum in Europe (한국내 고추나물의 하이퍼리신 유전자(HyH)의 탐색과 유럽의 서양고추나물과 비교)

  • Hypericin (HyH) is a substance which is isolated a medicinal herb, Hypericum perforatum L., commonly known as St. John's Wort. Hypericum erectum is a long-lived herb that is distributed in Korea. Cloned HyH genes H. erectum of were conformed by sequencing. The cDNA Hyp-1 sequence has 732 bp with an open reading frame of 567. Thus coding for a protein of 152 amino acid residues. A BLAST re-search using the deduced nucleotide sequences in HyH gene produced significant alignments with the H. perforatum. Sequences in HyH gene showed significant homology with Rubus idaeus putative allergen Rub-i-1 mRNA, Protein sequence comparisons revealed significant homology between Hyp-1 and the phenolic oxidative coupling protein hyp-1 of H. perforatum (98%). Additionally, Hyp-1 showed sig-nificant homology with various other classes of allergens, including Pru-av-1 (62%) from Prunus avium and allergen Bet-vl-Sc3 from Betula pendula (60%). Thus, the result of this study may offer an important information to establish an assay system for chemicals of the herbal medicines for H. erectum as well as H. perforatum.

Antioxidative Flavonoids from Hypericum erectum (고추나물의 항산화 활성 Flavonoid 성분)

  • Four antioxidative flavonoids were isolated from the aerial parts of Hypericum erectum. Their structures were identified as quercitrin (I), isoquercitrin (Ⅱ), hyperoside (Ⅲ) and orientin (IV) on the basis of spectroscopic means. Antioxidative activities for flavonoids $I{\sim}IV$ were determined by measuring lipid peroxide using 2-thiobarbituric acid (TBA) method and by evaluation the radical scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Compound IV, orientin, was found to have strong antioxidative potency.

Analysis of Genetic Relationships in Hypericum erectum Thunb. by RAPD (RAPD 방법을 이용한 고추나물의 유연관계분석)

  • The genetic relationships of Hypericum erectum, H. ascyron and H. perforatum Thunb. by RAPD using total 46 primers were analysed 30 primers among primers tested showed the amplification band in all. The amplified DNA fragment ranged from 0.25 to 6.6 kb in size. The 411 bands (34.4%) among 1194 bands derived from 30 primers were polymorphic, and 13.7 bands per primer were observed. The phenograms for six analyzed individuals by RAPD markers were not matched well with those of the result by morphological characters. They were clustered monophyletic at the similarity coefficient value ranged from 0.24 to 0.96.

  • The effect of plant growth regulators was investigated on in vitro shoot proliferation from axillary bud explants of Hypericum erectum. To determine the optimal cytokinin for proliferation of axillay buds, we carried out screening four cytokinins (BA, kinetin, 2iP, TDZ). When nodal segments were cultured on MS medium supplemented with $4.5\;{\mu}M$ TDZ (thidiazuron), a number of shoots were induced. Our results indicated that the addition of TDZ to culture medium resulted in the induction of significantly more axillary buds than in the addition of other cytokinins. The optimal concentration of TDZ for proliferation of axillary buds was $10\;{\mu}M$. 92% of shoots spontaneously rooted without any plant growth regulator (PGR) and formed whole plantlets within one month. More than 95% of these regenerants survived and they did not show any detectable variation in morphology or growth characteristics compared to their donor plants.

  • Inter simple sequence repeat (ISSR) markers were performed in order to analyse the phylogenetic relationships of eight Hypericum electum populations in Korea. The six primers were produced 37 reproducible ISSR bands. Analysis of ISSR from individual plants of Korean H. erectum resulted in 22 polymorphic bands with 59.5%. Across populations, the mean number of alleles per locus was 1.348 and Shannon's information index was 0.203.Population Mt. Gyeryong had the highest expected genetic diversity (0.175) among all populations. When species were grouped by eight populations, within group diversity was 0.140 (Hs), while among group diversity was 0.472 (G$_{ST}$) on a per locus basis. The estimated gene flow (Nm) for H. erectum was very low (0.561). It is suggested that reproductive isolation by the isolation of geographical distance among H. electum populations and genetic drift may have played roles in shaping the population structure of this species. In phonetic tree, all populations were well separated from each other. Thus, ISSR markers are very effective in classifying natural population levels of genus Hypericum in Korea.

  • To explore the Korea native plants to substitute for St. John's wort, which produce hypericin that use commercially for the treatment of mild to moderate depression, hypericin contents of Hypericum erectum and H. ascyron collected in two mountain of Korea were examined. From TLC and TLC-densitometer analysis of hypericin contents and biosynthetic pattern, hypericin was detected in flower and leaf of H. erectum, but not in all organs of H. ascyron. The hypericin content of H. erectum grown 200 m high hill in Mt. Byoung-pung was higher than that of Mt. Ji-ri 500 m and 1300 m high hill. When the seasonal variation of hypericin contents in H. erectum leaf collected from two regions was investigated, leaf collected from both regions on July was higher than other seasons.

  • One of micropropagation methods was investigated by using a multiple-shoots protocol. Multiple shoot formation was obtained from excised axillary buds of Hypericum erectum on half-strength or basal MS medium supplemented with TDZ or BA. The optimal combination of shoot multiplication for the production of more shoots with a suitable size was MS medium supplemented with $0.005\;mg{\cdot}L^{-1}$ TDZ (6.5 adventitious shoots per node). In vitro rooting was carried on half-strength MS medium with $1\;mg{\cdot}L^{-1}\;GA_3\;and\;0.5\;mg{\cdot}L^{-1}$ IBA treatment. In addition, the rooted cuttings were showed a better root growth in the greenhouse and survived in more than 90%. The results show that the species can be micropropagated effectively by the application of axillary bud culture systems.

Screening of Natural Plant Resources with Acetylcholinesterase Inhibition and Antioxidant Activity (천연 식물자원으로부터 Acetylcholinesterase 저해 및 항산화 활성 탐색)

  • This study was performed to investigate the effect of natural plant extracts on acetylcholinesterase (AChE) activity and the free radical scavenging activity. The methanolic extracts of plants were tested for AChE inhibitory activity using Ellman's colorimetric method in 96-welled microplates and antioxidant activity as the scavenging effect of 1, 1-diphenyl-2-picryl hydrazyl radical (DPPH). The results showed that AChE activities were inhibited (about 20-30%) in whole plant extract of Daucus carota var. sativa, Hypericum erectum and Fragaria yezoensis. AChE activities were inhibited (about 32-34%) in stems extract of Gingko biloba and leaves extract of Rhododendrondron yedoensa var. poukhanense. Fruit extract of Zanthoxylum schinifolium inhibited (about 18%) AChE activity. And the DPPH scavenging effects as antioxidant activity were similar to L-ascorbic acid in whole plant extract of Fragaria yezoensis and fruits extract of Comus officinalis.

Investigation of DPPH Radical Scavenging and Pyolyl Endopeptidase Inhibitory Activities of Plant Extracts (천연 식물자원에서 DPPH 라디칼 제거능과 Prolyl Endopeptidase 활성 저해능 탐색)

  • The present study was performed to investigate the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging and Proly1 endopeptidase (PEP) inhibitory activities of plant extracts. The whole extract of Fragaria yezoensis inhibited the DPPH radical by $90.4\%$ and the stem of Gingko biloba, Gardenia jasminoides for. grandiflora and Rhododendron yedoensa var. poukhanene, the loaves of Rhododendron mucronulatum var. ciliatum, Gardenia jasminoides for. grandiflora and Corylus sieboldiana var. mandshurice, the fruit of Cornus officinalis, and the root of Gingko biloba showed high DPPH radical scavenging activities. In the case of PEP inhibitory activities, high inhibition was observed in the whole Plant of Fragaria ananassa, Fragaria yezoensis and Hypericum erectum, the stem of Actinidia arguta and Rhododendron yedoensa var. poukhanese, the leaves of Rhododendron yedoensa var. poukhanense and Rosa davurica, the fruit of Cornus officinalis. and the root of Acer okamotoanum. There was significant correlation (P=0.000) between DPPH radical scavenging and prolyl endopeptidase inhibitory activities, thus some of plant extracts such as whole Fragaria yezoensis, fruit of Cornus officinalis had high activities in both DPPH-scavenging and prolyl endopeptidase inhibition. Therefore, it is required to examine the mechanical interaction between DPPH-scavenging and prolyl endopeptidase inhibitory activities and further studying plant extracts with both these activities is desired to develop agents for preventing and treating of Alzheimer's disease.
