Comparison Analysis of MR Images Before and After External Beam Radiotherapy in Brachytherapy (근접치료 시 외부 방사선 치료 전후의 MR 영상 비교분석)

  • Purpose: To analyze availability of MR images before and after external beam radiotherapy in brachytherapy, we will acquire MR images before and after external beam radiotherapy and compare the change of direction of uterine cavity and analyze the accuracy of applicator insertion. Materials and Methods: From January 2009 to December 2010, we compared MR images before and after external beam radiotherapy for uterine cervical cancer only with radical purpose treatment. MR images which was acquired after external beam radiotherapy has done with inserted status of CT/MR applicator. Results: As a consequence, the tumor was markedly reduced after external beam radiotherapy. The change of anteflexion of uterus turned into retroflexion of the uterine cavity was 17.1%. The case of wrong insertion of tandem include direction or length was 14.3%. Conclusion: According to MR images taken after external beam radiotherapy, we recognized not only reduced the tumor volume but the marked change of exact direction or length of the uterine cavity. So the confirmation of accurate insertion based on MR images before brachytherapy could be very helpful for optimal brachytherapy treatment planning with reduced applicator insertion errors.

A Study of Burn Accidents in Children towards Developing a Program for Prevention (어린이 화상사고의 실태와 예방대책 프로그램 개발에 대한 연구)

  • This study is a descriptive study which was done to examine the circumstances surrounding burn accidents in children in order to develop a burn prevention program. The subjects for the study were the mothers of 107 children admitted to two burn specialty hospitals in seoul and the mothers of 172 healthy children attending day care or kindergarten. The tool used in the study was developed by the researchers, pretested and refined. Cronbach's alpha was 0.72. Data collection was done between November 1995 and April 1996. Statistically analysis was done using SAS Version 6.04 and the data was analyzed using test, chi square, regression and logistic analysis. A suumary of the results shows the following : Eighty percent of the children in both groups lived in nuclear families but comparatively more of the children with burn accidents lived in apartments. More of the parents in the group of healthy children had university education and held white collar or professional jobs, while the parents of the children with burn accidents were slightly younger than the parents of the healthy children. but not significantly younger. A significantly greater number of families of children with burn accidents had more than one child and the characteristics of the children with burn accidents that they were more frequently boys, either first or last born and very active. Most of the burn accidents occurred in fall or winter. sometime between noon and midnight and the majority occurred in the home and were caused by hot liquid. In 86% of the cases emergency care was not given at home but the child was brought to the hospital emergency unit. Most of the burns were second degree or greater. involved the arms or legs. required admission, and 41.5% occurred when the parents were preparing or eating meals. In a test of knowledge of preventive measures, the mothers of children who had suffered burn accidents scored slightly higher than the mothers who had no experience with burn accidents, but this was not statistically significant. As to what to do in the case of a fire, the mothers of the healthy children had more knowledge about who to notify. but 70% of the mothers in both groups did not know how to escape from the house if it was of fire. 80 to 96% did not know what do to in case of a fire, such as rolling if clothes were on fire. 80% did not know what emergency care to give for a burn, and 74 to 87% did not know when it is appropriate to try to put out a fire. It can be concluded from this study that. since the results show that the most frequent situation which resulted in burn accidents was when the parents were not aware of the danger of burns or were not paying attention to the child. and when there were no protective devices to prevent burns. and that accidents most frequently occurred at home. the most effective way of preventing accidents is to develop in the parents a "safety mind", Since burn accidents in children are usually more severe than in adults. the physical. emotional. psychological and social damage is greater. In order to develop and awareness of this in the general public. the use of mass media. books and pamphlets and other educational media that provide education on prevention of burn accidents need to be utilized.

Gene Cloning and Characterization of a Cold-Adapted Esterase from Acinetobacter venetianus V28

  • Acinetobacter venetians V28 was isolated from the intestine of righteye flounder, Poecilopsetta plinthus caught in Vietnam seawater, and the esterase gene was cloned using a shotgun method. The amino acid sequence deduced from the nucleotide sequence (1,017 bp) corresponded to a protein of 338 amino acid residues with a molecular weight of 37,186. The esterase had 87% and 72% identities with the lipases of A. junii SH205 and A. calcoaceticus RUH2202, respectively. The esterase contained a putative leader sequence, as well as the conserved catalytic triad (Ser, His, Asp), consensus pentapeptide GXSXG, and oxyanion hole sequence (HG). The protein from the strain V28 was produced in both a soluble and an insoluble form when the Escherichia coli cells harboring the gene were cultured at $18^{\circ}C$. The maximal activity of the purified enzyme was observed at a temperature of $40^{\circ}C$ and pH 9.0 using p-NP-caprylate as substrate; however, relative activity still reached to 70% even at $5^{\circ}C$ with an activation energy of 3.36 kcal/mol, which indicated that it was a cold-adapted enzyme. The enzyme was a nonmetallo-protein and was active against p-nitrophenyl esters of $C_4$, $C_8$, and $C_{14}$. Remarkably, this enzyme retained much of its activity in the presence of commercial detergents and organic solvents. This cold-adapted esterase will be applicable as catalysts for reaction in the presence of organic solvents and detergents.

Characteristics of Gaseous Dissolved Mercury and Total Mercury in Yangsuri Marsh of Korea (양수리 용늪의 용존 수은 및 총수은 농도 특성에 대한 연구)

  • Long-term measurement of total mercury (TM) and dissolved gaseous mercury (DGM) were performed in Yangsuri marsh. Average TM and DGM concentrations were $2.0{\pm}2.0$ ng/L and $15.0{\pm}2.8$ pg/L, respectively, indicating that only 2.6% of TM existed as the form of DGM in Yangsuri marsh. While TM did not show the seasonal variation a statistically high DGM concentration was observed in warm season, indicating that DGM was effectively produced by strong solar radiation and high water temperature. There was no relationship between TM and DGM concentrations in Yangsuri marsh, as observed in other studies. DGM in Yangsuri marsh was supersaturated for most of sampling period; therefore, one can conclude that $Hg^0$ in water surface can readily volatilize to the atmosphere.

Sugar composition and glycemic indices of frequently consumed fruits in Korea (우리나라 다빈도 섭취 과일의 당 함량 및 혈당지수에 관한 연구)

  • Fruits are generally recommended for a balanced meal, as they are good sources of vitamins, minerals, and fiber, which may improve blood glucose control. However, fruits have simple sugars with a wide glycemic index (GI) range. The purpose of this study was to analyze the sugar content and composition and to determine the glycemic indices of the most frequently consumed fruits in Korea, including apple, tangerine, pear, water melon, persimmon, grape, oriental melon, and peach. The sugar content and composition of the fruits were analyzed by high performance anion-exchange chromatography (Dinonex model DX-600). The GI of the fruits was measured in 13 healthy subjects (seven females and six males) after permission was received from the University Hospital institutional review board (KHU-IRB 1114-06). The subjects consumed 50 g of glucose as a reference and carbohydrate portions of eight fruits. Blood samples were collected at 0, 30, 60, 90, and 120 min after consuming the fruits. The GI values for the fruits were calculated by expressing the increase in the area under the blood glucose response curve for each subject. As a result, the total sugar contents of 100 g fruits were: grape (13.9 g), apple (12.3 g), persimmon (11.9 g), oriental melon (11.2 g), watermelon (9.3 g), tangerine (8.9 g), peach (8.6 g), and pear (8.3 g). The GI values of the fruits were as follows: GI value of peach ($56.5{\pm}14.17$), watermelon ($53.5{\pm}18.07$), oriental melon ($51.2{\pm}18.14$), tangerine ($50.4{\pm}15.16$), grape ($48.1{\pm}14.05$), persimmon ($42.9{\pm}18.92$), pear ($35.7{\pm}14.38$), and apple ($33.5{\pm}11.92$). These findings will help individuals choose fruit for controlling blood sugar.

Toxicity Assessment and Establishment Acceptable Daily Intake of Lepimectin (레피멕틴(Lepimectin)의 독성평가와 일일섭취허용량 설정)

  • Lepimectin is a insecticide agent. In order to register this new pesticide, the series of toxicity data on animal testing were reviwed to evaluate its hazards to consumers and to determine its acceptable daily intake. Lepimectin was mostly excreted by feces. It has low acute oral toxicity while it has no dermal, ocular irritation and skin sensitization (As the result of subchronic, chronic toxicity and carcinogenicity showed changes of hematology and clinical biochemistry parameter of serum and blood.). Two-generation reproduction toxicity, genotoxicity, carcinogenicity and prenatal development toxicity were not proven. Therefore, the ADI for Lepimectin is 0.02 mg/kg/ bw/day, based on the NOAEL of 2.02 mg/kg/ bw/day of two-years carcinogenic toxicity study in rats and applying an uncertainty factor of 100.

Effects of Korean Red Ginseng Extract on Cisplatin-Induced Nausea and Vomiting

  • Ginseng, the root of Panax ginseng CA Meyer, is well known as a tonic medicine for restoring and enhancing human health. In traditional medicine, ginseng is utilized for the alleviation of emesis, which includes nausea and vomiting. However, it has not yet been demonstrated whether ginseng exhibits in vivo anti-nausea and anti-vomiting properties. In this study, we examined the anti-emetic effect of Korean red ginseng total extract (KRGE) on cisplatin-induced nausea and vomiting using ferrets. Intraperitoneal administration (i.p.) of cisplatin (7.5 mg/kg) induced both nausea and vomiting with one-hour latency. The episodes of nausea and vomiting reached a peak after 1.5 h and persisted for 3 h. Treatment with KRGE via oral route significantly reduced the cisplatin-induced nausea and vomiting in a dose-dependent manner. The anti-emetic effect was 12.7 $\pm$ 8.6, 31.8 $\pm$ 6.9, and 67.6 $\pm$ 4.0$\%$ with doses of 0.3, 1.0, and 3.0 g/kg of KRGE, respectively. Pretreatment with KRGE via oral route 1 and 2 h before cisplatin administration also significantly attenuated the cisplatin-induced nausea and vomiting. However this did not occur with a pretreatment 4 h before cisplatin administration. These results are supportive of KRGE being utilized as an anti-emetic agent against nausea and vomiting caused by chemotherapy (i.e. cisplatin).

Inhibitory Effects of Dunaliella salina Extracts on Thermally-Induced Skin Aging (두날리엘라 살리나 추출물의 피부 열노화 억제 효과)

  • Just like UV radiation, heat increases collagen degradation and accumulation of abnormal elastin fiber and this is termed thermal skin aging. Dunaliella salina (DS), a green alga, is known for its beta-carotene accumulation, having various applications in the health and nutritional products. However, the effects of DS on heat-induced skin aging remain unexplored. In this study, we performed anti-thermal aging tests of the ethanol extract of DS (DSE). We measured the cellular levels of type I procollagen and MMP-1 using ELISA in human dermal fibroblast cells after heat shock. DSE reduced the expression of MMP-1 protein and increased the expression of type I procollagen. In addition, DSE upregulated the mRNA expression of HSP47 reduced by heat shock, which is involved in collagen synthesis. Also, DSE reduced the expression of inflammation mediator (TGF-${\beta}$, IL-12, etc). We demonstrate that DSE regulates the heat-induced solar elastosis through the regulation of tropoelastin and fibrillin-1, two major proteins of elastic fibers, and MMP-12 expression. These results suggest that DSE may be effective for preventing thermally induced skin aging.

A Study of Stabilization for Insoluble Active Ingredients Using Swollen Micelles (Swollen Micelle을 이용한 난용성 효능물질의 안정화 연구)

  • Micelles, which are called solubilization formulation, have been widely used in skin lotion, mist or various formulations for the purpose of solubilizing the fragrance rather than the role of the active ingredient carrier because the particles are very small. In this study, we developed the swollen micelle having a transparent appearance to deliver ${\beta}$-sitosterol. When preparing the swollen micelle, solubility parameter was considered with active ingredient. This method allowed insoluble active ingredient to be safely entrapped inside the micelles. Stability of micelle was evaluated by dynamic light scattering (DLS). The transparency and shape of the micelles were confirmed by cryo-TEM. In addition, through the thermal analysis using DSC, ${\beta}$-sitosterol was found to be stably present in the swollen micelles. These results indicate that swollen micelles considered solubility parameter could be used as a new carrier for the insoluble active ingredients.
