Determination of Optimum Operating Parameters for Enhanced Alkaline Hydrolysis of Soils Contaminated with TNT (TNT 오염토의 염기성 가수분해 효율 향상을 위한 최적 운전인자 도출)

  • Nitro-aromatic Compounds (NACs) of explosives are structurally non-degradable materials that have an adverse effect to humans and ecosystems in case of emissions in natural due to the strong toxicity. In this study, batch test in the laboratory-scale has been conducted to find some process parameters of alkaline hydrolysis by considering the characteristics of NACs which are unstable in a base status and field application evaluation have been performed on the batch test results. Based on the experimental results of both laboratory and pilot-scale test, the optimum conditions of parameters for the alkaline hydrolysis of soils contaminated with explosives were pH 12.5, above the solid-liquid ratio 1 : 3, above the room temperature and 30 minute reaction time. In these four process parameters, the most important influencing factor was pH, and the condition of above pH 12.0 was necessary for high contaminated soils (more than 60 mg/kg). In the case of above pH 12.5, the efficiency of alkaline hydrolysis was very high regardless of the concentrations of contaminated soils. At pH 11.5, the removal efficiency of TNT was increased from 76.5% to 97.5% when the temperature in reactor was elevated from room temperature to 80℃. This result shows that it is possible to operate the alkaline hydrolysis at even pH 11.5 due to increased reaction rate depending on temperature adjustment. The results found in above experiments will be able to be used in alkaline hydrolysis for process improvement considering the economy.

Lipase Catalyzed Kinetic Resolution of rac-2-(3-Methoxy-4-methylphenyl) propan-1-ol and rac-2-(3-Hydroxy-4-methylphenyl)propyl propanoate for S-(+)-Xanthorrhizol

  • Xanthorrhizol is a bisabolane type of natural sesquiterpene, the major component of essential oils of Curcuma xanthorrhiza. 2-(3-Methoxy-4-methylphenyl)propan-1-ol and 2-(3-hydroxy-4-methyl phenyl)propan-1-ol could be essential building block for enantioselective synthesis of xanthorrhizol. Enantioselective (c = 53%, E = $80{\pm}3$) for R-(+)-2-(3-hydroxy-4-methylphenyl) propan-1-ol and (c = 58%, E = $27{\pm}1$) for R-(+)-2-(3-methoxy-4-methylphenyl) propan-1-ol resolution processes were developed via lipase-catalyzed reaction. We found lipase Aspergillus oryzae (AOL) and Porcine pancreas (PPL) are selective to transesterification and hydrolysis in organic and aqueous phase. Modified demethylated substrate is appropriate for enantioselective hydrolysis reaction without any additives. Enantiopure chiral alcohol was crystallized from ethyl acetate/n-hexane co-solvent system. Gram scale resolved chiral intermediate will facilitate the synthesis of the unnatural S-(+)-xanthorrhizol, the corresponding isomer of the natural one.

  • Acid and halide ion catalyses for the hydrolysis of benzenesulfinamides were kinetically investigated. The rates of hydrolysis increased with increasing concentration of both acid and halide ions and also showed to speed up as the electron donating ability of the benzenesulfinyl moiety and the electron withdrawing ability in the leaving group increased. The reactivity of halide ions was in the order of $Br^-$ > $Cl^-$. The reaction mechanism may be accommodated by including a hypervalent intermediate and sulfonium cation.

Preparation of Pronase Hydrolysate from Alaska-pollack (명태단백 Pronase 가수분해물의 제조)

  • In order to enhance the utility of alaska-pollack, the optimum conditions for the preparation of pronase hydrolysate. The optimum temperature and pH for the hydrolysis of alaska-pollack by pronase were 4$0^{\circ}C$ and pH 7.0. The reaction time and enzyme concentration were 4 hr and 1,000 units per g of substrate. Under the above optimum conditions alaska-pollack was hydrolysed by pronase yielding a hydrolytic degree of about 89eye. The bitterness and hyrophobicity of pronase hydrolysate were decreased with increasing reaction time. Hydrophobic amino acids(Tyr, Met, Ala, flu, Leu, and Phe) were increased for 2 hr, but fur thor hydrolysis was showed decrease of hydrophobic amino acids content. Palatable amino acids (Asp, Glu, Pro, Ser, Thr and Gly) were increased with hydrolysis time.

  • 81.9% of the cellulose delignified by acetosolv process was hydrolyzed in HCl-$AlCl_3$ hydrolysis system when $AlCl_3$ was used as catalyst in breaking down of glycosidic bond of cellulose. It was well compared that the HCl hydrolysis system without $AlCl_3$ as catalyst showed only 60~61% of the hydrolyzed yield. Also monosaccharide yield including glucose clearly increased when $AlCl_3$ was use. When concentration of HCl and $AlCl_3$ was increased, the hydrolyzed monosaccharide was increased within certain range. The monosaccharid yield out of the hydrolyzed reached 55.4% at optimum conditions which were identified as 20% of Hel solution, 0.03 Mol of $AlCl_3$, $120^{\circ}C$ of reaction temperature and 7 hours of reaction time employed in this study.

  • The rate constants of the hydrolysis of phenylvinylsulfone were determined by ultraviolet spectrophotometry at various pH and a rate equation which can be applied over wide pH range was obtained. The reaction mechanism of hydrolysis of phenylvinylsulfone and especially the catalytic contribution of hydroxide ion which did not study carefully before in acidic media, can be fully explained by the rate equation obtained. The rate equation reveals that: below pH 7, the reaction is initiated by the addition of water molecule to phenylvinylsulfone. At above pH 9, the overall rate constant is only dependent upon the concentration of hydroxide ion.

  • Various mechanistic aspects of the A2 hydrolysis of methyl acetimidate were explored using the MNDO method. As in thecorresponding reactions of acetamide and methyl carbamate, a proton transfer pre-equilibrium exists between the N-protonated and the O-protonated tautomers, and the subsequent hydrolysis proceeds from the more stable N-protonated form. Of the two reaction pathways, the $A_{AL}2$ path is favored in the gas phase and in concentrated acid solutions, whereas the $A_{AC}2$ path is favored in less acidic solutions with a stable cationic tetrahedral intermediate formed in the rate determining step. Negative charge development on the alkoxy oxygen in the transition state suggested a rate increase with the increase in the electron withdrawing power of the alkoxy group. Calculations on the reaction processes with AM1 indicated that MNDO is more reliable in this type of work, although AM1 is better than MNDO in reproducing hydrogen bonds.

  • The effect of substrate micellization on the hydrolysis rate in the production of lysopho-sphatidylcholine (LPC) from phosphatidylcholine (PC) using hog pancreas phospholipase A2(PLA2) was studied. The optimal temperature and pH for the reactions in aqueous phase was found 42C and 7.2, respectively. For a given PC concentration, initial reaction rate was progressively increased with the addition of sodium deoxycholate (DOC), which could transform the bilayer of phospholipids into micellar structure.
