Studies on the Optical Resolution of DL-Amino Acids by Aminoacylase Immobilized on Chitosan: Properties and Reactivity of Immobilized Aminoacylase (Chitosan 고정화 Aminoacylase 를 이용한 DL 아미노산의 광학적 분할에 관한 연구 : 고정화 Aminoacylase의 성질 및 반응성)

  • Aminoacylase immobilized on chitosan was applied for optical resolution of DL-amino acids. Optimun pH's for hydrolysis of N-ac DL Met, N-ac DL Try and N-ac DL Phe by immobilized aminoacylase were 8.0, 7.0, and 7.5, respectively. The pH stability of immobilized aminoacylase was less than that of soluble enzyme, while there was no difference in thermostability between immibilized and soluble enzymes. The reaction rate of immobilized enzyme was maximum, when concentrations of N-ac DL Met, N-ac DL Try and N-ac DL Phe were 0.05, 0.03 and 0.05M, respectively. Continuous resolution of M/20 N-ac DL amino acids with immobilized aminoacylase packed in a column resulted in 100% hydrolysis upto space velocity $2.0\;at\;45^{\circ}C$, and the half-life of the column at space velocity 5.0 was about 25 days. The yield of L-Met, L-Try and L-Phe recovered from 2 liter of column effluent were 57%, 52% and 52%, respectively.

Production of Chitosna Oligosaccharides Using Chitin-Immobilized Enzyme (키틴 고정화 효소를 이용한 키토산 올리고당의 생산)

  • Enzymatic hydrolysis using an immobilized enzyme was carried out to produce chitosan oligosaccharides (COSs) from chitosan effectively. Chitosanase was immobilized on eight different carriers by physical adsorption. The enzyme immobilized on chitin had higher activity than those immobilized on the other carriers in spite of its lower adsorption. The activity of chitin-immobilized enzyme was more than 90% of the original activity. Optimal temperature of the immobilized enzyme increased by about $15^{\circ}C$ and its thermostability was excellent in relatively wide range of temperature. But its effects of pH did not improve compared to the free enzyme. The immobilized enzyme produced 153 mg/g chitosan of the reducing sugar for 3hrs of hydrolytic incubation time. The total content of higher oligomers, tetramer to hexamer, among amount of total COSs obtained for 2hrs was more than 90%. In kinetic parameters for both enzymes, immobilized enzyme showed lower affinity for substrate and reaction rate than free enzyme, however, no reduction of the rate for high substrate concentrations. Consequently, chitin-immobilized could effectively hydrolyse chitosan and produce the higher COSs without activity decrease in comparison with the free enzyme.

Synthesis and Application of cPSMA-PSMA Microcapsule Absorbent for Cement Mortars (시멘트 모르타르용 cPSMA-PSMA 마이크로캡슐 흡수제 제조 및 적용)

  • We synthesized microcapsule absorbent with crosslinked poly(styrene-$alt$-maleic anhydride) (PSMA) as a core and PSMA as a shell by a precipitation polymerization method for the delayed absorption of excess water in cement mortar. cPSMA-PSMAs with core-shell structure were synthesized with ratios of 1/1, 1/2 and 1/3 as core monomer mass to shell monomer mass to control shell thickness. We observed the hydrolysis of PSMA in cement-saturated aqueous solution by a FTIR spectrometer. We observed good core-shell structure microcapsules for 1/2(cPSMA #3), but observed incomplete core-shell structure for 1/1(cPSMA #2) and 1/3(cPSMA #4) of core/shell monomer ratios. The swelling ratio of cPSMA #3 in cement-saturated aqueous solution was increased until 20 min. After that it was decreased until 2 hrs swelling time, and they started to increase again. The viscosities of cement paste with cPSMA #3 microcapsules were very slowly increased until 1 hr and increased fast after 1.5 hrs. Cement mortar with 0.5 wt% cPSMA #1 having only core part showed about 5% increase in compressive strength compared to that of plain cement mortar. cPSMA #3 added cement mortar showed the highest compressive strength with 7% increase.

Determination of diclofenac and its metabolites in human urine by GC-MS (GC-MS를 이용한 소변 중 Diclofenac 및 대사체 분석)

  • This study has been described the metabolism and excretion in a healthy male urine collected for 26hrs after oral administration of diclofenac. To detect conjugated metabolites of diclofenac, urine sample was acid-hydrolyzed under the conditions of 6M-HCl at over $110^{\circ}C$ for 1hr. During the acidic hydrolysis process, diclofenac and its metabolites were converted into their corresponding lactam-ring through dehydration reaction. As results of chemical conversion by means of hydrolysis, the structures of diclofenac and its metabolites were also changed acidic to basic forms. However, lactam-ring was degraded by hydroxyl ion at basic condition. Thus, the extraction rate of dehydrated diclofenac and its metabolites was not favored at basic condition. For the determination of trace amounts of diclofenac and its metabolites in urine, trimethylsilylation (TMS) with MSTFA was applied and followed by analysis with gas chromatograph-mass spectrometer. In this study, four metabolites that are formed by the hydroxylation of parent drug were mainly detected. Each metabolite was tentatively identified by both interpretation of mass spectra and comparison with previously reported results. In addition, time profile of urinary excretion rate for parent drugs and metabolites was studied. Finally, the metabolic pathway of diclofenac was suggested on the basis of the elucidation of its metabolites and excretion profiles.

The Quality Characteristics of Soy Cutlets Using Textured Soy Protein Treated with Different Enzymes (효소처리를 달리한 조직대두단백을 이용하여 제조한 콩까스의 품질특성)

  • The development of soy cutlets containing textured soy protein (TSP) as a meat analog was studied. In order to decrease the beany flavor and to increase the texture, TSP was treated with 0.3% Flavourzyme or 0.1% Protamex for 10 or 20 min, respectively. The degree of hydrolysis for TSP treated with Protamex was higher than that treated with Flavourzyme. Hydrolysis was observed to increase as the reaction time was increased for both Flavourzyme and Protamex. The water holding capacity of TSP treated with Protamex for 10 min was the highest, and that treated with Flavourzyme for 20 min was similar to that of Protamex treatment for 20 min. The oil binding capacity of TSP treated with Protamex for 20 min was the highest. The hardness of the soy cutlets using TSP treated with Flavourzyme for 10 min was higher than that treated for 20 min, while that of Protamex treated for 20 min was higher than that treated for 10 min. The cohesiveness of the soy cutlets using TSP treated with Flavourzyme or Protamex for 10 min was higher than those treated for 20 min. The chewiness of the soy cutlets treated with Flavourzyme for 10 min was higher than for those treated for 20 min, while those treated with Protamex for 20 min was higher than those treated for 10 min. The springiness of TSP treated with Flavourzyme for 20 min was higher than those treated for 10 min, and higher than those treated with Protamex for 10 or 20 min. For sensory evaluation, the beany flavor of the soy cutlets treated with Protamex for 20 min was the weakest. The flavor and chewiness of both a pork cutlet and a soy cutlet treated with Protamex for 20 min were the best. In the overall quality, soy cutlets treated with Protamex for 20 min was the most desirable. In conclusion, soy cutlets treated with 0.1% Protamex for 20 min could be a reasonable substitute of pork cutlets.

Improvement of Radiosynthesis Yield of [11C]acetate ([11C]아세트산의 방사화학적 수율 증가를 위한 연구)

  • Purpose $[^{11}C]$acetate has been proved useful in detecting the myocardial oxygen metabolism and various malignancies including prostate cancer, hepatocellular carcinoma, renal cell carcinoma and brain tumors. The purpose of study was to improve the radiosynthesis yield of $[^{11}C]$acetate on a automated radiosynthesis module. Materials and Methods $[^{11}C]$acetate was prepared by carboxylation of grignard reagent, methylmagnesium chloride, with $[^{11}C]$$CO_2$ gas, followed by hydrolysis with 1 mM acetic acid and purification using solid phase extraction cartridges. The effect of the reaction temperature ($0^{\circ}C$, $10^{\circ}C$, $-55^{\circ}C$) and cyclotron beam time (10 min, 15 min, 20 min, 25 min) on the radiosynthesis yield were investigated in the $[^{11}C]$acetate labeling reaction. Results The maximum radiosynthesis yield was obtained at $-10^{\circ}C$ of reaction temperature. The radioactivities of $[^{11}C]$acetate acquired at $-10^{\circ}C$ reaction temperature was 2.4 times higher than those of $[^{11}C]$acetate acquired at $-55^{\circ}C$. Radiosynthesis yield of $[^{11}C]$acetate increased with increasing cyclotron beam time. Conclusion This study shows that radiosynthesis yield of $[^{11}C]$acetate highly dependent on reaction temperature. The best radiosynthesis yield was obtained in reaction of grignard reagent with $[^{11}C]$$CO_2$ at $-10^{\circ}C$. This radiolabeling conditions will be ideal for routine clinical application.

Reusability of Surfactant-coated Candida rugosa Lipase Immobilized in Gelatin Microemulsion-based Organogels for Ethyl Isovalerate Synthesis

  • In our previous study, a surfactant-coated Candida rugosa lipase immobilized in microemulsion-based organogels was exploited for the synthesis of ethyl isovalerate. In the present study, we are focusing on the effective reuse of lipase immobilized in microemulsion-based organogels (MBGs) in terms of retainment of the catalytic activity. As water is one of the co-products in esterification reactions, the removal of water becomes a priority to allow the reaction to work in the forward direction and to prevent back hydrolysis. Taking this fact into consideration, the lipase-containing microemulsion-based organogels were given pretreatment and/or several intermittent treatments with dry reverse micellar solution of AOT in organic solvent during repeated cycles of ester synthesis. The pretreated MBGs with dry reverse micellar solution exhibited lower water content and higher initial rates of esterification in comparison with untreated freshly prepared MBGs. The esterification efficiency of untreated MBGs started decreasing after 5 cycles of reuse and was almost completely lost by the end of the $8^{th}$ cycle. In contrast, pretreated MBGs exhibited a gradual decrease in esterification efficiency after 5 cycles and retained about 80% of the initial activity at the end of the $8^{th}$ cycle. The intermittent treatment of MBGs after every 3 cycles resulted in enhanced reusability of immobilized lipase for up to 9 cycles without significant loss in esterification activity, after which it resulted in a slow decrease in activity with about 27% lower activity at the end of the $12^{th}$ cycle. Furthermore, the treatment conditions such as concentration of AOT in liquid dessicant and time of treatment were optimized with respect to our system. The granulated MBGs proved to be better in terms of initial esterification rates (1.2-fold) as compared with the pelleted MBGs.

Characterization of the Interaction of Sulfiredoxin (Srx1) with a Vacoular Protein $\alpha$-Mannosidase (Ams1) in Saccharomyces cerevisiae (설피리독신과 알파-만노시다제 간의 단백질 결합 특성에 관한 고찰)

  • Most redox-active proteins have thiol-bearing cysteine residues that are sensitive to oxidation. Cysteine thiols oxidized to sulfenic acid are generally unstable, either forming a disulfide with a nearby thiol or being further oxidized to a stable sulfinic acid, which have been viewed as an irreversible protein modification. However, recent studies showed that cysteine residues of certain thiol peroxidases (Prxs) undergo reversible oxidation to sulfinic acid and the reduction reaction is catalyzed by sulfiredoxin (Srx1). Specific Cys residues of various other proteins are also oxidized to sulfinic acid ($Cys-So_2H$). Srxl is considered one of the oxidant proteins with a role in signaling through catalytic reduction of oxidative modification like in the reduction of glutathionylation, a post-translational, oxidative modification that occurs on numerous proteins. In this study, the role of sulfiredoxin in cellular processes, was investigated by studying its interaction with other proteins. Through the yeast two-hybrid system (Y2HS) technique, we have found that Ams1 is a potential and novel interacting protein partner of Srxl. $\alpha$-mannosidase (Ams1) is a resident vacuolar hydrolase which aids in recycling macromolecular components of the cell through hydrolysis of terminal, non-reducing $\alpha$-D-mannose residues. It forms an oligomer in the cytoplasm and under nutrient rich condition and is delivered to the vacuole by the Cytoplasm to Vacuole (Cvt) pathway. Aside from the role of Srxl as a catalyst in the reduction of cysteine sulfenic acid groups, it may play a completely new function in the cellular process as indicated by its interaction with Ams1 of the yeast Saccharomyces cerevisiae.

Optimization of Rod-shaped γ-LiAlO2 Particle Reinforced MCFC Matrices by Aqueous Tape Casting (수계 테이프 케스팅 법에 의한 봉상 γ-LiAlO2 입자 강화 MCFC 매트릭스 제조 공정의 최적화)

  • Rod-shaped particle reinforced $LiAlO_2$ matrices for MCFC were fabricated by an aqueous tape-casting technique. The hydrolysis reaction and agglomeration of $\gamma-LiAlO_2$ particles in aqueous slurries were inhibited by additions of $LiOH{\cdot}H_2O$ and glycerin to the aqueous $\gamma-LiAlO_2$ slurry. The tape-casting, performed using the aqueous slurry containing protein albumin, was fast and led to an effective drying at casting temperature range of $60{\sim}65^{\circ}C$. The strength of the particle reinforced matrix was improved about 4 times compared to that of matrix without reinforcement. Pore size distribution ($0.1{\sim}0.4{\mu}m$) and porosity ($50{\sim}60%$) of the reinforced matrices were determined to be appropriate for the MCFC matrix. The aqueous tape casting process is not only environmental-friendly but also efficient for fabricating MCFC matrices compared to non-aqueous tape casting.

Validation of Analytical Method for Male Sex Hormone Monitoring in Urine due to the Chemical Castration (성충동약물치료 시행에 따른 소변 중 남성호르몬의 분석법 확립)

  • "The Act on Medication Treatment of Sexual Impulse of Sex Offenders" known as chemical castration has been effective since July 2011 in Korea. According to the law, monitoring of male sex hormone in urine is enforced to request National Forensic Service more than once a month after injection of medicine designed to reduce sex impulse. We established a rapid and sensitive method for the monitoring of testosterone (T) and epitestosterone (E) in human urine by liquid chromatography with tandem mass spectrometry (LC-MS/MS). Three mL of urine was pretreated by solid-phase extraction for purification and performed enzymatic hydrolysis. The pretreated samples were extracted twice with 2 ml of ethyl acetate and n-hexane (2 : 3). The separation was applied on Thermo Hypersil GOLD C18 column ($1.9{\mu}m$, $100{\times}2.1mm$). A gradient elution of methanol and water of 0.1% formic acid were used as mobile phase and the retention time was less than 10 min. LC-MS/MS system coupled with an electrospray ionization source was performed in multiple reaction monitoring mode. The transitions of the analytes executed as following: m/z $289{\rightarrow}97$, 109 for T and E, m/z $292{\rightarrow}109$ for $T-d_3$ and $E-d_3$ as internal standards. The validation results of the method were satisfactory. The limits of detection were 0.05 ng/ml and the limits of quantification were 0.1 ng/ml. This method was successfully applied to real human urine sample. The developed method will be useful for monitoring T/E ratio in urine of sex offenders.
