Immobilization of Lipase using Alginate Hydrogel Beads and Enzymatic Evaluation in Hydrolysis of p-Nitrophenol Butyrate

  • The immobilization of enzyme is one of the key issues both in the field of enzymatic research and industrialization. In this work, we reported a facile method to immobilize Candida Antarctica lipase B (CALB) in alginate carrier. In the presence of calcium cation, the enzyme-alginate suspension could be cross-linked to form beads with porous structure at room temperature, and the enzyme CALB was dispersed in the beads. Activity of the enzyme-alginate composite was verified by enzymatic hydrolysis reaction of p-nitrophenol butyrate in aqueous phase. The effects of reaction parameters such as temperature, pH, embedding and lyophilized time on the reactive behavior were discussed. Reuse cycle experiments for the hydrolysis of p-nitrophenol butyrate demonstrated that activity of the enzyme-alginate composite was maintained without marked deactivation up to 6 repeated cycles.

Titanium(IV) isoproxide 유도체의 합성 및 특성연구 (Characterization and Synthesis of Titanium (IV) Isopropoxide Derivatives)

  • Titanium(IV) isoproxide에 유기첨가제로 아세트산을 첨가하여 단일성분계인 치환된 titanium(IV) isoproxide를 합성한다. 합성된 titanium 착화합물은 세가지 형태로 된 5배위 및 6배위의 구조로 존재함을 FT-IR, $^1H$$^{13}C$ NMR 그리고 UV-Vis 스펙트라로 확인하였다. 이 착물에 대한 가수분해 반응을 $^1H$-NMR spectroscopy를 이용해 관찰하였고 FT-IR spectroscopy로 가수분해 및 건조시킨 후의 구조 변화를 관찰하여 보았으며 리간드의 존재 유무도 살펴보았다.

  • Pretreatment of wastepaper using aqueous glycerol under high pressure was studied to enhance the enzymatic hydrolysis. The pretreatment was conducted over a wide range of conditions including temperatures of $150{\sim}170^{\circ}C$, sulfuric acid concentrations of 0.5~1.5%, and reaction times of 30~90 minutes. After the effect of glycerol concentration on the pretreatment performance was investigated, 70% glycerol was selected. As glycerol concentration was increased, higher digestibility was achieved due to higher lignin removal. The optimum condition was found to be around $160^{\circ}C$, 1%, and 60 minutes. At this condition, 60% and 35% of hemicellulose and lignin, respectively, were removed, while only 5% of cellulose was lost. The enzymatic digestibility was 76%, meaning that 73% of the glucan present in the initial substrate was recovered as glucose after enzymatic hydrolysis. Also, it was found that the temperature and acid concentration than the reaction time were more strongly related to the compositional removals and enzymatic digestibility.

Ketoprofen Resolution by Enzymatic Esterification and Hydrolysis of the Ester Product

  • Immobilized Candida antarctica lipase was used to catalyze the separation of ketoprofen into its components by means of esterification followed by the enzymatic hydrolysis of the ester product. In this study, ketoprofen underwent esterification to ethanol in the presence of isooctane. When the reaction was complete, 58.3% of the ketoprofen had been transformed into an ester. The ketoprofen remaining in solution after the reaction was complete consisted primarily of its S-enantiomer (83.0%), while the 59.4% of the ketoprofen component of the ester consisted of its R-enantiomer. We then subjected the ester product to enzymatic hydrolysis in the presence of the same enzyme and produced a ketoprofen product rich in the R-enantiomer; 77% of this product consisted of the R-enantiomer when 50% of the ester had been hydrolyzed, and 90% of it consisted of the R-enantiomer when 30% of the ester had been hydrolyzed. By contrast, the R-enantiomer levels only reached approximately 42 and 65%, respectively, when 50 and 30% of the racemic ester was hydrolyzed under the same conditions.

Thienyl Chalcone 유도체의 가수분해 반응메카니즘과 그 반응속도론적 연구 (The Kinetics and Mechanism of the Hydrolysis to Thienyl Chalcone Derivatives)

  • The hydrolysis reaction kinetics of 2-thienyl chalcone derivatives $[II]{\sim}[V]$ was investigated by ultraviolet spectrophotometery in 20% dioxane-$H_2O$ at $25^{\circ}C$ and the structure of these compounds were ascertained by means of ultraviolet, infrared and NMR spectra. The rate equations which were applied over a wide pH range(pH $1.0{\sim}13.0$) were obtained. The substituent effects on 2-thienyl chalcone derivatives$[II]{\sim}[V]$ were studied, and the hydrolysis were facilitated by electron attracting groups. On the basis of the rate equation, substitutent effect and final product, the plausible hydrolysis reaction mechanism was proposed : At pH $1.0{\sim}9.0$, not relevant to the hydrogen ion concentration, neutral $H_2O$ molecule competitvely attacked on the double bond. By contraries, above pH 9.0, it was proportional to concentration of hydroxide ion.

  • A formation of aluminum hydroxide by hydrolysis reaction in the water has been studied by using nano aluminum powder fabricated by pulsed wire evaporation(PWE) method. The hydroxide type and morphology depending on temperature and pH were examined by structural analysis. The Boehmite($Al_2O_3$.$H_2O$ or AIO(OH)) was predominantly formed in high temperature region over 4$0^{\circ}C$, while the Bayerite($Al_2O_3$.$H_2O$ or $Al(OH)_3$) below $30^{\circ}C$ of hydrolysis temperature. The Boehmite formation was preferred to the Bayerite in acidic solution in the same hydrolysis temperature. The slowly formed Bayerite phase showed facet crystalline structure, while the fast formed Boehmite was fine fiber with a large aspect ratio of several nm in diameter and several hundred nm in length, and with much larger specific surface area(SSA) than that of Bayerite. The highest SSA was about $420m^2$/g.

  • Titania powders were obtained from the various hydrolysis condition the variation of Alkoxide, water/Alcohol, REaction, Temp., reaction time and solvent system were investigated. In this result, spherical monodisperesed titania gel powders(≒0.7${\mu}{\textrm}{m}$) were obtained using EtOH(as solvent), and this method had rapid reaction time compare with iso-PrOH(as solvent).

  • A 2-step process for the safe disposal of chemical warfare agents(agent hydrolysis followed by incineration In the submerged-quench incinerator) was studied to obtain basic data for the design of pilot plant in the future. Sample materials used for the hydrolysis reaction were sarin(GB), sulfur mustard (HD), and methylphosphonic difluoride(DF). The hydrolysates of these materials were thermally destroyed in a submerged-quench incineration system. Experimental conditions for achieving destruction efficiency of 99.99% in both steps were established and phosphoric acid was recovered from the waste water when destroying DF hydrolysate in the incinerator. Treated water could be reused as process water for the agent hydrolysis.

  • The objective of this study is to prepare ketoprofen (KP) - poly(ethylene glycol) (PEG) conjugates and to investigate their degradation kinetics. KP-PEG conjugates were synthesized from KP and PEG methy1ester by esterification in the presence of DCC. The KP-PEG conjugates (KPEG) were characterized by IR and $^{1}H-NMR$ spectroscopy. The hydrolysis of KPEG with time was studied using HPLC by simultaneous quantification of KP and KPEG. The hydrolysis rate constant was high at low and high pHs, and showed minimum at pH 4 and 5. As the size of KPEG increases, hydrolysis rate increased. The slope of degradation rate profile suggests that catalytic reaction seems to occur by specific acid/base catalysis. These results suggest that KPEG could be used as a prodrug for KP, which releases KP slowly in the body.

  • Sulfuric acid hydrolysis is a typical approach for producing cellulose nanocrystals. The method has been widely used, but it has a disadvantage of low yield of cellulose nanocrystals compared to mechanical method. To expand the application of cellulose nanocrystals in practical, we should be able to produce them with higher yield and the controlled properties. In this study, therefore, we intended to investigate the effect of sulfuric acid hydrolysis condition on the characteristics of the prepared cellulose nanocrystals. The concentration of sulfuric acid, temperature and hydrolysis time were varied, and the yield as well as diverse properties including the morphology, size and zeta potential were examined. We could obtain cellulose nanocrystals up to 70% of yield and found that the properties were dependent on the reaction condition. It would be helpful to select an appropriate condition for producing cellulose nanocrystals.
