The ethanol extract of the peony root (Paeonia Lactiflora Pall, Paeoniaceae) as well as its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. In addition, the protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were examined. The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.
Objectives : This study aims at examining the immuno-modulating activity in the fermentative extract of the root of Scutellaria baicalensis Georgi (Scutellariae Radix) on the production of inflammatory mediator in LPS-stimulated RAW264.7 mouse macrophages. Method : Measurements were done for the influences on the cell viability, generation of hydrogen peroxide in cells and nitric oxide (NO) generation using the macrophage of mouse with the specimen SBS as the fermentative extract of Scutellariae Radix (SBS) with Saccharomyces cerevisiae STV89. Result : As a result of carrying out MTT assay to check the cellular toxicity of the fermentative extract of Scutellariae Radix, any excessive toxicity to the macrophage did not occur from treatments by concentration for SBS. SBS increased the generation of hydrogen peroxide in the macrophage. SBS suppressed the NO generated in macrophages and SBS concentration higher than $25{\mu}g/mL$ significantly suppressed the increased NO generated in LPS-stimulated macrophages. SBS concentration higher than $25{\mu}g/mL$ significantly suppressed the generation of IL-6, IL-10, IL-12p40 and MCP-1 in LPS-stimulated macrophages. Conclusion : Our findings indicate that SBS has an immuno-modulating activity in macrophage activation through suppressing the generation of inflammatory substances, NO, IL-6, IL-10, IL-12p40 and MCP-1.

지금까지 연기중의 Nicotine, CO등 몇몇 화합물의 함량 측정과 관능 검사등을 통하여 담배의 품질을 평가해 왔다. 그러나 담배 연기중의 수 많은 화학 성분들을 고려할 때 전연기 성분이 생체에 미치는 영향을 측정함으로써 그 품질을 평가할수 있는 새로운 방범이 요구된다. 담배연기에 의한 생체 손상 쿵 가장 일차적이고 영향이 큰 것으로 알려진 활성 산소종(H2O2, O2-, ·OH)은 그 반응성이 크고 life time이 짧기 때문에 측정이 어렵다. 저자들은 이들을 분해하는 효소를 이용하여 연기중에서 생성되는 이들 산소 자유 라디칼을 측정하였으며 그 결과 담배의 종류에 따라 이들의 생성이 현저한 차이를 보여 일반적으로 고급 담배로 알려진 것들은 낮았고 저급 담배일 수록 높았다. 따라서 이들 산소 자유 라디칼의 측정은 흡연과 건강이라는 측면에서 매우 유용한 담배의 품질 평가 방법으로 이용될수 있을 것으로 생각된다.
Objective : The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide, sodium nitroprusside and hydrogen peroxide induced expression phospholipase $A_2$ and calcium concentration in RAW 264.7 cells, a murine macrophage cell line. Method : The expression of phospholipase $A_2$ was determined by western blotting with corresponding antibodies, and the generation of intracellular calcium concentration was investigated by delta scan system in RAW 264.7 cells. Results : 1. Compared with control, expressions of lipopolysaccharide-induced phospholipase $A_2$ were decreased significantly by $1\;{\mu}g/{\mu}l$ of bee venom and decreased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 2. Compared with control, expressions of sodium nitroprusside-induced phospholipase $A_2$ were decreased significantly by $5\;{\mu}g/{\mu}l$ of bee venom but increased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 3. Compared with control, expressions of hydrogen peroxide-induced phospholipase $A_2$ were decreased significaltly by $1{\mu}g/{\mu}l$ of bee venom and decreased by $0.5\;{\mu}g/{\mu}l$ of bee venom but increased by $5\;{\mu}g/{\mu}l$ of bee venom. 4. The protective role against oxidative stress under cellular system using C6 glioma cells was studied using the ethyl acetate (EtOAc) fraction, luteolin (1), and luteolin-7-glucoside (2) of Taraxacum coreanum. C6 glioma cells showed low cell viability and high generation of reactive oxygen species (ROS) by the treatment with generator of hydrogen peroxide ($H_2O_2$) and amyloid beta ($A{\beta}_{25-35}$). However, the treatment of the EtOAc fraction attenuated the cellular oxidative stress, resulting in significant elevation of cell viability. In addition, the production of ROS formation was also decreased by the treatment of the EtOAc fraction. Compounds 1 and 2 were isolated from the EtOAc fraction, and the protective effect was evaluated. Compounds 1 and 2 led to the increase of cell viability and decrease of production of ROS against oxidative stress by $H_2O_2$ and $A{\beta}_{25-35}$. The present study indicated that the EtOAc fraction, compounds 1 and 2 from T. coreanum demonstrated protective effects against oxidative stress, suggesting the preventive role against neurodegenerative diseases.
최근 들어 새로운 차세대 우주발사체의 추진기관의 개발 방향에서 친환경성과 저비용성이 주요 관심사이며, 과산화수소와 액체메탄은 이러한 추진제로 대표적인 관심을 받고 있다. 과산화수소는 로켓 개발 초기에 널리 사용되었으나, 냉전시대에 성능을 최우선시함에 따라 사용이 현격히 감소해오다가 1990년대 이후 안정성의 향상과 더불어 다시 재조명 받기 시작하였다. 액체메탄은 실제 발사체의 추진기관으로 사용되지는 않았지만, 액화천연가스(LNG)로 사용할 수 있기 때문에 친환경/저비용 추진제로 지속적으로 관심을 받아왔으며 최근에는 달/화성 탐사와 관련하여 지속적인 연구가 수행되고 있다. 이러한 대표적인 친환경 추진제에 대한 활용 역사와 최근 연구 동향을 고찰하여, 향후 국내에서 소요될 각종 임무 조건을 만족하는 엔진의 연구 개발 방향 설정에 도움이 될 것으로 예상된다.

Alpha-lipoic acid (ALA) is a naturally occurring antioxidant and has been previously used to treat diabetes and cardiovascular disease. However, the autophagy effects of ALA against oxidative stress-induced dopaminergic neuronal cell injury remain unclear. The aim of this study was to investigate the role of ALA in autophagy and apoptosis against oxidative stress in the SH-SY5Y human dopaminergic neuronal cell line. We examined SH-SY5Y phenotypes using the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay (cell viability/proliferation), 4′,6-diamidino-2-phenylindole dihydrochloride nuclear staining, Live/Dead cell assay, cellular reactive oxygen species (ROS) assay, immunoblotting, and immunocytochemistry. Our data showed ALA attenuated hydrogen peroxide (H2O2)-induced ROS generation and cell death. ALA effectively suppressed Bax up-regulation and Bcl-2 and Bcl-xL down-regulation. Furthermore, ALA increased the expression of the antioxidant enzyme, heme oxygenase-1. Moreover, the expression of Beclin-1 and LC-3 autophagy biomarkers was decreased by ALA in our cell model. Combined, these data suggest ALA protects human dopaminergic neuronal cells against H2O2-induced cell injury by inhibiting autophagy and apoptosis.
Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.

To evaluate the anti-oxidative activities of GDE, we measured scavenging activities on DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of GDE. To detect the protective effects of GDE against UVB, we irradiated with 40 mJ/㎠`s UVB to HaCaT cells then we measured reactive oxygen species(ROS) generation, apoptotic bodies and cell viability using DCFH-DA assay, Hoechst 33342 staining and MTT assay. GDE showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also GDE showed high reducing values. GDE reduced oxidative stress conditions by inhibition of ROS expression. Also the cell apoptosis by UVB-induced oxidative conditions was decreased by GDE treatment. These results could suggest that GDE had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. GDE would be useful for the development of cosmetics treating UVB-induced skin aging.
