Effect of Allopurinol on Vascular Endothelial Cells Damaged by Hydrogen Peroxide In Vitro (Hydrogen Proxide에 의해 손상된 배양 혈관내피세포에 대한 Allopurinol의 영향)

  • In order to examine the effect of oxygen free radicals on the vascular endothelial cells, cell viability was measured by XTT assay after bovine pulmonary vascular endothelial cell line(BPVEC) was treated only with hydrogen peroxide. In addition, the antioxidant effect of allopurinol on cells treated with hydrogen peroxide was examined by colormetric assay. in this study, the BPVEC treated with hydrogen peroxide showed the significantly decreased cell viability compared with control. Whereas, the viability of cells treated with hydrogen peroxide and allopurinol has significantly increased when compared with that of cells treated only with hydrogen peroxide. These results suggested that hydrogen peroxide, one of the oxygen free radicals showed cytotoxic effect and allopurinol has protective effect on oxygen free radical-induced cytotoxicity.

  • In this paper, analyzes the type of failure and its effect on the hydrogen fueling nozzle used in hydrogen station. Failure of hydrogen fueling nozzle was analyzed using a qualitative risk assessment method, failure mode and effect analysis. The failure data of hydrogen fueling nozzles installed in domestic hydrogen stations are collected, and the failure types are classified, checked the main components causing the failure. Criticality analysis was derived based on frequency and severity depending on the failure mode performed. A quality function is developed by a performance test evaluation item of the hydrogen fueling nozzle, and the priority order of design characteristics is selected. Through the analysis results, the elements to improve the main components for enhancing the quality and maintenance of the hydrogen fueling nozzle were confirmed.

The Function of Hydrogen Chloride on Methane-Air Premixed Flame (메탄-공기 예혼합 화염에서 염화수소의 역할)

  • Numerical simulations were performed at atmospheric pressure in order to understand the effect of additives on flame speed, flame temperature, radical concentrations, $NO_x$ formation, and heat flux in freely propagating $CH_4-Air$ flames. The additives were both carbon dioxide and hydrogen chloride which had a combination of physical and chemical behavior on hydrocarbon flame. In the flame established with the same mole of methane and additive, hydrogen chloride significantly contributed toward the reduction of flame speed, flame temperature, $NO_x$ formation and heat flux by the chemical effect, whereas carbon dioxide mainly did so by the physical effect. The impact of hydrogen chloride on the decrease of the radical concentration was about $1.4\~3.0$ times as large as that of carbon dioxide. Hydrogen chloride had higher effect on the reduction of $EI_{NO}$ than carbon dioxide because of the chemical effect of hydrogen chloride. The reaction, $OH+HCl{\rightarrow}Cl+H_2O$, played an important role in the heat flux from flames added by hydrogen chloride instead of the reaction, $OH+H_2{\rightarrow}H+H_2O$ which was an important reaction in hydrocarbon flames.

  • Hydrogen peroxide has been a bleaching chemical for varied pulp, especially mechanical and deinking pulp. It is catalytically decomposed by some transition metals in pulp slurry. In this paper, some metals which can be contained in pulp such as manganese, copper, iron, magnesium and calcium were used to investigate their effect on the decomposition of hydrogen peroxide. From the result, hydrogen peroxide was more decomposed in the order of Mn, Cu, $Fe^{3+}\;and\;Fe^{2+}$, while Mg and Ca had little effect on the decomposition of hydrogen peroxide. The effect of Mg/Mn ratio on the decomposition of hydrogen peroxide was also investigated. At the specific ratio of them(Mg/Mn=10), hindering effect of peroxide decomposition by Mg was decreased.

  • We investigate the effect of hydrogen on adsorption of hydrocarbon molecules on graphene with density functional theory (DFT) calculations. In this study, we calculate the binding energies of hydrogen molecule, carbon atom and other hydrocarbon fragments such as CHx (x=1, 2, 3, 4) on graphene to find the most stable adsorption site. Then, to study the effect of hydrogen, we investigate the adsorption of hydrocarbon fragments in the presence of hydrogen atoms on graphene.

Effect of Geometrical Shape and Cold Work on Deformation of the Hydrogen Absorption Metal with Hydrogen Absorption-Desorption Cycling (수소저장-방출싸이클링에 의한 수소저장금속의 변형거동에 관한 형상 및 가공의 영향)

  • By using the electrochemical method, the relation between the deformation and the geometrical shape, and the effect of cold work on hydrogen absorption-desorption cycling in palladium were investigated, In order to study this problem, four kinds of the Pd specimens used were plates and bars as cold worked and annealed states. As results, it is found that the deformation of thickness direction in the palladium plates increased whereas other lateral directions decreased. But the palladium bars showed the same deformation ratio in all directions because of uniform distribution of the $\beta$ phase. Grains in the plate specimens were greatly deformed after hydrogenation cycling whereas grains in the bar specimens were pulverized. Also, deterioration of the hydrogen absorption rate of the bar specimen was larger than the plate specimen. And the effect of cold work on hydrogen absorption capacity was relatively small.

  • Numerical simulations of freely propagating flames burning $H_2/HCl/Air$ Air mixtures were performed at atmospheric pressure in order to understand the effect of hydrogen chloride on flame structures. The chemical and physical effects of hydrogen chloride on flame structures were observed. A chemical kinetic mechanism was developed, which involved 26 gas-phase species and 198 forward elementary reactions. Under several equivalence ratios the flame speeds were calculated and compared with those obtained from the experiments, the results of which were in good agreement. As hydrogen chloride as additive was added into $H_2/Air$ flame, the flame speed, radical concentration and NO production rate were decreased. The chemical effect of hydrogen chloride caused the reduction of radical concentration, and then the decrease of the net rate of NO production. It was found that the influence in the reduction of $EI_{NO}$ with the addition of hydrogen chloride was attributed more due to the chemical effect than the physical effect.

  • Kimchi showed protective effect from oxidative damage generated by hydrogen peroxide and paraquat. To investigate the major components of kimchi which reduce the cytotoxicity against reactive oxygen species, keratinocyte(A431, epidermoid carcinoma, human) and fibroblast(CCD-986SK, normal control, human) were cultured under oxidative stress condition provoked by paraquat, a superoxide anion generator, and hydrogen peroxide in the absence or presence of kimchi ingredients. Most keratinocyte and fibroblast cells were killed by hydrogen peroxide and paraquat over 1mM concentration, but kimchi ingredients showed protective effects from oxidative damage generated by hydrogen peroxide and onion, among those, garlic showed the most remarkable preventive effect. Most of kimchi ingredients showed protective effect against paraquat, especially leek notably increased cell survival. For fibroblast cells, ginger had the preventive effect against paraquat, especially leek notably increased cell survival. For fibroblast cells, ginger had the preventive effect from cell killing by high dose of hydrogen peroxide, but most ingredients were not effective against paraquat.

Effect of Emodin on Hydrogen Peroxide Production in Polyinosinic-Polycytidylic acid-induced Human Neuroblastoma SH-SY5Y Cells (Emodin이 polyinosinic-polycytidylic acid로 유발된 인간 신경모세포종 SH-SY5Y의 hydrogen peroxide 생성증가에 미치는 영향)

  • The purpose of this study is to investigate the modulatory effect of emodin on hydrogen peroxide production in human blastoma SH-SY5Y cells induced by the synthetic analog of double-stranded RNA [polyinosinic-polycytidylic acid]. Hydrogen peroxide production was measured by dihydrorhodamine 123 (DHR) assay. Emodin significantly inhibited the polyinosinic-polycytidylic acid (PIC)-induced production of hydrogen peroxide for 0.5, 2, 12, 18, and 24 hr incubation at the concentrations of 5, 10, 25, and 50 uM in SH-SY5Y (P < 0.05) in dose dependent manner. These results suggest that emodin has neuroprotective property related with its inhibition of hydrogen peroxide production in PIC-induced neuronal cells.

  • The purpose of this study is to investigate the modulatory effect of oroxylin A on hydrogen peroxide production in TM4 mouse testis sertoli cells induced by the synthetic analog of double-stranded RNA [polyinosinic-polycytidylic acid]. Hydrogen peroxide production was measured by dihydrorhodamine 123 (DHR) assay. Oroxylin A significantly inhibited the polyinosinic-polycytidylic acid (PIC)-induced production of hydrogen peroxide for 0.5, 2, 12, 18, and 24 hr incubation at the concentrations of 5, 10, 25, and $50{\mu}M$ in TM4 (P < 0.05) in dose dependent manner. These results suggest that oroxylin A has a protective effect against PIC-induced cellular toxicity with its inhibition of hydrogen peroxide production in PIC-induced sertoli cells.
