Feature Analysis of Different In Vitro Antioxidant Capacity Assays and Their Application to Fruit and Vegetable Samples (In Vitro 항산화능 측정법에 대한 특징 분석과 채소.과일 시료에 대한 적용 사례 고찰)

  • Reactive oxygen species (ROS), including singlet oxygen (${O_2}^1$), superoxide anion radical ($O_2{\cdot}^-$), hydroxyl radical ($HO{\cdot}$), peroxyl radical ($ROO{\cdot}$), hydrogen peroxide ($H_2O_2$), and hypochlorous (HOCl), are generated as byproducts of normal cellular metabolism. ROS induce damage to many biological molecules, such as lipids, proteins, carbohydrates, and DNA. It is widely believed that some degenerative diseases caused by ROS can be prevented by the high intake of fruits and vegetables due to their antioxidant activities. Recently, research on natural antioxidants has become increasingly active in various fields. Several assays have been developed to measure the total antioxidant capacity of antioxidants in fruits and vegetables in vitro. These assays include those for DPPH radical scavenging activity, SOD-like activity, total polyphenol content, oxygen radical absorbance capacity, reducing power, trolox equivalent antioxidant capacity (ABTS assay), single-cell gel electrophoresis (comet assay), and a cellular antioxidant activity assay. Because different antioxidant compounds may act through different mechanisms in vitro, no single assay can fully evaluate the total antioxidant capacity of foods. Due to the complexity of the composition of foods, it is important to be able to measure antioxidant activity using biologically relevant assays. In this review, recently used assays were selected for extended discussion, including a comparison of the advantages and disadvantages of each assay and their application to fruits and vegetables.

Steam Gasification of Coal and Petroleum Coke in a Thermobalance and a Fluidized Bed Reactor (열천칭과 유동층반응기에서 석탄과 Petroleum Coke의 수증기 가스화반응)

  • Lignite of low rank coal and petroleum coke of high sulfur content can be high potential energy sources for coal gasification process because of their plentiful supply. The steam gasification of lignite, anthracite, and pet coke has been carried out in both an atmospheric thermobalance reactor and a lab-scale fludized bed reactor (0.02 m i.d. ${\times}$ 0.6 m height). The effects of gasification temperature ($600{\sim}900^{\circ}C$) and partial pressure of steam (0.15~0.95 atm) on the gasification rate and on the heating value of product gas have been investigated. The modified volumetric reaction model was applied to the experimental data to describe the behavior of carbon conversion, and to evaluate kinetic parameters of char gasification. The results shows that higher temperature bring more hydrogen in the product syngas, and thus increased gas heating value. The feed rate of steam is needed to be optimized because an excess steam input would lower the gasification temperature which results in a degradation of fuel quality. The rank of calorific value of the product gas was anthracite > lignite > pet coke. Their obtained calorific value at $900^{\circ}C$ with 95% steam feed were 10.0 > 6.9 > 5.7 $MJ/m^3$. This study indicates that lignite and pet coke has a potential in fuel gas production.

Effects of Radiation on Thermal and Mechanical Properties of Modified Epoxy Resin and Hydrogenated Bisphenol-A Type Epoxy Resin Based Shielding Materials (개질 에폭시수지 및 수소 첨가된 비스페놀-A형 에폭시수지계 차폐재의 열적 및 역학적 성질에 미치는 방사선 영향)

  • ffects of radiation on the thermal and mechanical properties of modified epoxy resin and hydrogenated bisphenol-A type epoxy resin based neutron shielding materials to be used for radioactive material shipping and storage casks have been investigated. The onset temperatures of the shielding materials of KNS(Kaeri Neutron Shield)-201 and KNS-302 increased with the radiation dose, but those of KNS-202 and KNS-301 decreased at radiation dose above 0.5 MGy. In addition, the radiation dose rarely affected the change of weight of shielding materials with the variation in temperature. At radiation dose up to 0.1 MGy, thermal conductivities of shielding materials were not affected. The thermal expansion coefficients of the shielding materials of KNS-301 and 302 were affected to a less extent than those of KNS-201 and 202 by radiation. At radiation dose up to 0.1 MGy, the tensile strength, compressive strength and flexural strength of the shielding materials of KNS-202 and KNS-301 and 302 increased with the radiation dose. In contrast, those of KNS-201 decreased with an increase in the radiation dose. In addition, the amount of radiation dose on the shielding materials did not result in a measurable loss of specific gravity, weight and hydrogen content.

Quality Characteristics of Sweet Persimmon Jams added with Aronia Juice (아로니아즙 첨가에 따른 단감잼의 품질특성)

  • This study investigates the quality characteristics of sweet persimmon jam added with aronia juice. Sweet persimmon jam was prepared with sweet persimmon, sugar, pectin and various amounts (20, 40, 60, 80%) of aronia juice, and it was assessed for pH, total acidity, color, sensory evaluation and hydrogen donating ability on 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging effects and content of the total polyphenols, respectively. Increasing the amount of aronia juice decreased the pH, whereas the total acidity gradually increased. In addition, with an increased amount of aronia juice, the lightness (L) and yellowness (b) significantly decreased, while redness (a) gradually increased. By increasing the amount of aronia juice, the polyphenol contents, DPPH and ABTS radical scavenging activities were increased. The results of the sensory evaluation showed that the sweet persimmon jam added with 80% aronia juice had the highest overall-acceptability scores. The results suggest enhancing the quality, palatability and acceptability of sweet persimmon jam by adding 60~80% of aronia juice.

Geochemistry and Isotope Studies of the Shinchon $CO_2$ -rich Waters in the Gyeongsang Province (경상지역 신촌 탄산약수의 지화학적 및 동위원소 특성)

  • The Geochemica] and isotope studies on the $CO_2$-rich water from the Shinchon area were carried out. The Shinchon $CO_2$-rich water belongs to Ca(Na)-$HCO_3$ type showing very high $P_{CO_{2}}$ ( $10^{-0.35}$ ~ $10^{0.29}$ atm) and TDS (835-3,144 mg/L). The results of geochemical and isotope analysis indicate that $CO_2$ gas is originated from the deep seated source such as mantle or magmatic gases. The $CO_2$-rich water was evolved by interaction with deep-seated granite and major water-rock interaction was dissolution of p]agioclase resulting high Na content of $CO_2$-rich water. Precipitation and dissolution of secondary calcite might be accompanied with the dissolution of plagioclase maintaining Na/Ca ratio. High contents of K and $SO_4$ indicate that the geochemical characteristics of $CO_2$-rich water were partially affected by interaction with upper sedimentary rock during uprising to surface. N03 and tritium contents suggest that the $CO_2$-rich water was mixed with low $CO_2$ groundwater at some locations. The oxygen-hydrogen isotopes show that all water samples were derived from meteoric waters and the $CO_2$-rich water was isotopically re-equilibrated with lighter $CO_2$ gas. Although some carbon isotope data show isotopically heavy values, carbon isotope data indicate that the $CO_2$ gas was possib]y derived by deep source.

Effect of Well Depth, Host Rocks and Mineralization Zone on Hydrochemical Characteristics of Groundwater in the Umsung Area (음성지역 지하수의 수리화학적 특성에 대한 심도, 모암 및 광화대의 영향)

  • The purpose of this study is to investigate the hydrochemical characteristics of groundwater in the Umsung area, and to elucidate the effect of host rock type, well depth and mineralization zone on the groundwater chemistry. The geology of the study area consists of Jurassic granite and Cretaceous sedimentary rocks, which are bounded by a fault. Most of shallow groundwaters exploited in the Jurassic granite area are used for agricultural purpose, whereas the deep groundwaters in the Cretaceous sedimentary rocks are used for a drinking water. The shallow groundwater shows weak acidic pH, the electrical conductivity ranging from $142\;to\;903\;{\mu}S/cm$, and the chemical type of $Ca-HCO_3\;to\;Ca-Cl(SO_4,\;NO_3)$. A few of shallow groundwaters are contaminated by nitrate, and show high concentration of Fe, Mn and Zn, that reflects the effect of a mineralization zone. The deep groundwater shows neutral to weak alkaline pH, higher electrical conductivity than that of shallow groundwater, and the chemical type of $Ca-HCO_3$. The seepage water from the abandoned mines does not have the characteristics such as acidic pH, high concentration of heavy metals and high sulfate content. The hydrogen and oxygen isotopes of groundwater indicates an altitude effect of the recharge area between deep groundwater and shallow groundwater. In conclusion, the chemical composition of groundwater complicately reflects the effects of their host rocks, well depth, agricultural activity and mineralization zone in the study area.

Adsoptive Properties of Cellulose Thermally Treated at Low Temperature and Its Solubility to Water (저온 열처리 셀룰로오스의 염기성가스 흡착과 용해특성)

  • The purpose of this study was to investigate how to modify the physical properties of cellulose after thermal treatment. Cellulose was treated between $225^{\circ}C$ and $325^{\circ}C$ for 3 hrs under air flow, and then the thermally treated cellulose was measured to specific surface area, constitute elements, consumption ofacid and base, as well as the adsorption capacity of ethylamine vapor. The higher was the treating temperature from $225^{\circ}C$ to $325^{\circ}C$, the lower was the total yield of cellulose. Elemental analysis revealed that carbon content in thermally treated cellulose was gradually increased in proportion to temperature increment. The amount of acidic functional groups tended to increase up to $300^{\circ}C$, after then to be lowered slightly. In principle, no alkaline functional groups were found in thermally treated cellulose. In case of treatment with $325^{\circ}C$, only a few amount of alkaline functional groups were detectable. Specific surface area of thermally treated cellulose are determined to $1.9m^2/g$, which value can become higher when the treated temperature rises. The thermally treated cellulose at $275^{\circ}C$ shows the highest adsorption capacity of ethylamine at $40^{\circ}C$ for 4 hrs. Solubility of those two celluloses with WPG (Weight Percent Gain) value of 113% and 108%, respectively, was determined to almost 100%. X-ray diffractogram of thermally treated cellulose suggested that the crystalline structure of cellulose began to be destroyed at the temperature of $275^{\circ}C$. As a conclusion, changes of such a physical properties make it possible to weaken inter and/or intra hydrogen bond in crystal region of cellulose macromolecules. When thermally treated cellulose adsorbs ethylamine, it turns to be well soluble to water.

Methane Production and T-RFLP Patterns of Methanogenic Bacteria Dependent on Agricultural Methods (농법에 따른 메탄생성과 메탄생성 세균의 T-RFLP 패턴)

  • We studied soil components, methane production, the number of methanogens, and T-RFLP patterns dependent on agricultural methods with the change of seasons. There is no regular increase or decrease tendency of the most soil components followed by sampling period. And the water content in soil was higher in October than May. Also a lot of methanogens existed in soil, and acetotrophs were relatively of smaller number than hydogenotrophs and formate utilizing methanogens using MPN (most probable number) enumeration. In the experiment using the formate, it was used from the first week, and only a minute amount was detecte after four weeks. However in the acetate, it was increased until the third week, and after that was consumed. And there was higher methane production for all soil samples which administered with the hydrogen spike. The activity of methanogens was higher in the organic and low-agrichemical agricultural method samples, and the organic agricultural method had high methanogen activity among the other samples. A result of T-RFLP pattern of mcrA gene digested with Sau96I, methanogen community have a little relation with agricultural methods and seasons. This results also agreed to no critical difference the soil components dependent on agricultural methods, but some analytical data have a positive relationship with a agricultural methods. Therefor we could concluded that the comparison study of community for soil bacteria sufficiently could be useful for the microbiological indicator.

Optimal Condition of Torrefaction for the High-density Solid Fuel of Larch (Larix kaempferi) (낙엽송(Larix kaempferi) 고밀도 에너지화를 위한 반탄화 최적조건 탐색)

  • In this study, torrefaction was performed to improve fuel properties of Larch. The optimal condition for torrefaction was investigated by response surface methodology. The torrefaction temperature and time ranged $220{\sim}280^{\circ}C$ and 20~80 min, respectively. As the torrefaction temperature and time increased, the carbon content of torrefied biomass increased from 49.36 to 56.65%, while its hydrogen and oxygen contents decreased from 5.56 to 5.48% and from 37.62 to 31.67%, respectively. The weight loss and calorific value increased with SF, while energy yield decreased. At the severe torrefaction condition (SF 7), the weight loss and calorific value were 26.58% and 22.30 MJ/kg, respectively. The energy contained in torrefied biomass increased to 20.41%, when compared with the untreated biomass. As the torrefaction severity increased, the energy yield decreased due to the relatively high weight loss of biomass. Therefore, the highest energy yield was obtained at high calorific value and low weight loss of biomass (SF 5.72).

Influence of Ni/CeO2-ZrO2 Catalysts on Methane Autothermal Reforming (메탄 자열개질 반응에 대한 Ni/CeO2-ZrO2 촉매의 영향)

  • The catalytic behavior of $Ni/Ce_XZr_{(1-X)}O_2$ loaded on the alumina coated honeycomb monolith was studied for the autothermal reforming reaction of methane. Among the catalysts with the different Ce/Zr ratios, the $Ni/Ce_{0.80}Zr_{0.20}O_2$ Catalyst showed the highest conversion of methane. By investigating the effect of Ni content on the $Ni/Ce_{0.80}Zr_{0.20}O_2$ catalysts, the catalyst loaded with 15wt% Ni had the highest activity. Also, $H_2$ yield was increased as $H_2O/CH_4$ ratio increased. Methane conversion was improved as $O_2/CH_4$ ratio was increased, whereas the yield of $H_2$ was decreased. Among the catalysts tested for 30 hours, $Ni(15wt%)/Ce_{0.80}Zr_{0.20}O_2$ showed the excellent conversion(${\geq}99%$) of methane and the stability at the condition of $GHSV=30,000h^{-1}$, feed ratio S/C/O=2/1/0.5 and reaction temperature $800^{\circ}C$.
