Pharmacokinetics of oxytetracycline in hybrid catfish (Clarias macrocephalus x C. gariepinus) after intravascular and oral administrations

  • Importance: Over the past decade, catfish farming has increased in Southeast Asia. However, there has been no existing for pharmacokinetic data in the hybrid catfish (Clarias macrocephalus x C. gariepinus). Objective: This study was designed to evaluate the pharmacokinetic characteristics of oxytetracycline (OTC) in the hybrid catfish, following single intravascular (IV) or oral (PO) administration at a single dosage of 50 mg/kg body weight (BW). Methods: In total, 140 catfish (each about 100-120 g BW) were divided into two groups (n = 70). Blood samples (0.6-0.8 mL) were collected from ventral caudal vein at pre-assigned times up to 144 h (sparse samples design). OTC plasma concentrations were analyzed using high-performance liquid chromatography-photodiode array detector. Results: The pharmacokinetic parameter of OTC was evaluated using a non-compartment model. OTC plasma concentrations were detectable for up to 144 and 120 h after IV and PO, respectively. The elimination half-life value of OTC was long with slow clearance after IV administration in hybrid catfish. The average maximum concentration value of OTC was 2.72 ㎍/mL with a time at the maximum concentration of 8 h. The absolute PO bioavailability was low (2.47%). Conclusions and Relevance: These results showed that PO administration of OTC at a dosage of 50 mg/kg BW was unlikely to be effective for clinical use in catfish. The pharmacodynamic properties and clinical efficacy of OTC after multiple medicated feed are warranted.

Effects of dietary Boesenbergia rotunda (L.) Mansf. rhizome extract on the growth of hybrid catfish (Clarias macrocephalus × Clarias gariepinus)

  • The objective of this study was to examine the effects of dietary Boesenbergia rotunda (L.) Mansf. rhizome extract on the growth performance, intestinal histology, digestive enzymes, hematological, and biochemical indices of hybrid catfish (Clarias macrocephalus × C. gariepinus). The phytochemicals and antioxidant activity of a methanolic extract from the rhizome of B. rotunda were also evaluated. Hybrid catfish, with an average weight of 5.24 ± 0.26 g, were given diets containing B. rotunda extract at concentrations of 0 (control), 250, and 500 mg/kg for a duration of 8 weeks. Upon completion of the trial, various factors were assessed including growth performance, intestinal histology, digestive enzymes, hematology, and biochemical indices. The presence of phenolics, saponins, alkaloids, flavonoids, steroids, and tannins was identified in the extract of B. rotunda. The inclusion of B. rotunda in the diet resulted in a substantial improvement in final weight, weight gain, specific growth rate, and average daily gain (p < 0.05). The fish that were given diets containing B. rotunda showed a significant increase in the activity of digestive enzymes in their intestines (p < 0.05). Fish that were fed B. rotunda diets exhibited improved histoarchitecture of intestines (p < 0.05). The experimental fish exhibited a considerable increase in white blood cells, red blood cells, hemoglobin, and hematocrit compared to the control group (p < 0.05). Fish that were fed B. rotunda diets showed a significant increase in serum glucose levels, whereas serum cholesterol levels decreased (p < 0.05). The study found that a diet containing 250 mg of B. rotunda/kg had the potential to improve the growth performance and physiological features of hybrid catfish.

  • This study investigated the effects of dietary monobasic potassium phosphate (MKP) on the growth of the far eastern catfish Silurus asotus and four leafy vegetables in a hybrid biofloc technology aquaponic system. To an experimental diet containing 45% protein and 7% lipid, 1, 2, 3 or 4% MKP was added and was designated as MKP1, MKP2, MKP3, and MKP4, respectively. The optimum MKP levels were determined for the growth of fish and four leafy vegetables over 10 weeks. After the 10-week feeding trial, weight gain, feed efficiency, specific growth rate and protein efficiency ratio were higher in the fish groups fed MKP2 and MKP3 than in the other groups (P<0.05). The growth of the four leafy vegetables was also higher in the fish groups fed MKP2 and MKP3. Water quality [dissolved oxygen, pH, water temperature, electrical conductivity, turbidity, total ammonia nitrogen (TAN), $NO_2-N$, $NO_3-N$ and $PO_4-P$] was measured six times a week using a portable water quality meter and reagent measurements. The TAN (4.58-20.40 mg/L), $NO_3-N$ (24.12-52.40 mg/L) and $PO_4-P$ (20.38-48.48 mg/L) levels increased with time, while the $NO_2-N$ level remained below 0.1 mg/L throughout the study.

  • 동자개(Pseudobagrus fulvidraco), 대농갱이(Leiocassis ussuriensis) 및 이들간 잡종의 염색체수는 모두 2n=52로 동일하였으나, 핵형은 서로 상이하여 동자개가 22 metacentrics (M)+22 submetacentric (SM)+8 acrocentric (A) chromosome으로, 대농갱이는 22M+14SM+16A, 그리고 잡종은 22M+18SM+12A로 구성되어 있었다. 잡종은 동자개, 대농갱이 각각의 반수체로 구성되어 있었다. 적혈구 세포 및 핵의 크기, 표면적 및 체적을 측정한 결과 유도된 잡종은 양친의 중간값을 나타내었다. 동자개, 대농갱이 및 잡종의 DNA 함량을 측정한 결과, 동자개는 2.06 pg/cell, 대농갱이는 2.09 pg/cell, 동자개와 대농갱이 잡종은 2.08 pg/cell로 나타나, 잡종의 DNA 함량은 양친의 중간값으로 나타났다.

  • This study compared the productivity of fish and vegetables grown using the hybrid biofloc technology-aquaponics (HBFT-AP), a semi-recirculating aquaculture system (SRAS), and hydroponics (HP). For the study of fish productivity (HBFT-AP vs. SRAS), fish were provided feed containing 3.0% monobasic potassium phosphate (MKP) for 18 weeks. After the 18-week feeding trial, the average weight of the sampled population (n=100) was not significantly different (P>0.05), while hematocrit (PCV, %), hemoglobin (Hb, g/dL), and plasma K (mEq/L) were significantly different (P<0.05) between the two groups (HBFT-AP: 47.83%, 15.48 g/dL, and 1.39 mEq/L; SRAS 34.83%, 11.81 g/dL, and 2.48 mEq/L). Leaf vegetable productivity (HBFT-AP vs. HP) was compared in three experiments (EXP 1-3), and slower growth was observed in both groups in EXP 2, in which pH was maintained at 5.0 or less throughout the experiment. During the 18-week feeding trial, total ammonia nitrogen (TAN), $NO_3-N$, and $PO_4-P$ levels increased with time in the HBFT-AP system, while the concentration of $NO_2-N$ remained below 0.1 mg/L throughout the study.

Evaluation of fish oil-rich in MUFAs for anti-diabetic and anti-inflammation potential in experimental type 2 diabetic rats

  • The advantages of monounsaturated fatty acids (MUFAs) on insulin resistance and type 2 diabetes mellitus (T2DM) have been well established. However, the molecular mechanisms of the anti-diabetic action of MUFAs remain unclear. This study examined the anti-hyperglycemic effect and explored the molecular mechanisms involved in the actions of fish oil- rich in MUFAs that had been acquired from hybrid catfish (Pangasius larnaudii${\times}$Pangasianodon hypophthalmus) among experimental type 2 diabetic rats. Diabetic rats that were fed with fish oil (500 and 1,000 mg/kg BW) for 12 weeks significantly reduced the fasting plasma glucose levels without increasing the plasma insulin levels. The diminishing levels of plasma lipids and the muscle triglyceride accumulation as well as the plasma leptin levels were identified in T2DM rats, which had been administrated with fish oil. Notably, the plasma adiponectin levels increased among these rats. The fish oil supplementation also improved glucose tolerance, insulin sensitivity and pancreatic histological changes. Moreover, the supplementation of fish oil improved insulin signaling ($p-Akt^{Ser473}$ and p-PKC-${\zeta}/{\lambda}^{Thr410/403}$), $p-AMPK^{Thr172}$ and membrane GLUT4 protein expressions, whereas the protein expressions of pro-inflammatory cytokines (TNF-${\alpha}$ and nuclear NF-${\kappa}B$) as well as p-PKC-${\theta}^{Thr538}$ were down regulated in the skeletal muscle. These data indicate that the effects of fish oil-rich in MUFAs in these T2DM rats were partly due to the attenuation of insulin resistance and an improvement in the adipokine imbalance. The mechanisms of the anti-hyperglycemic effect are involved in the improvement of insulin signaling, AMPK activation, GLUT4 translocation and suppression of pro-inflammatory cytokine protein expressions.
