Isolation of Hyaluronidase Inhibitory Component from the Roots of Astraglus membranaceus Bunge (Astragali Radix)

  • In order to isolate hyaluronidase (HAase) inhibitor from Astragali radix (AR), dried roots were extracted with ethanol, prior to sequential fractionations with n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions. The n-butanol soluble fraction was found to exhibit the most pronounced inhibitory effect (68%) on HAase, and the active components were separated using various chromatographic methods, including column chromatography and preparative HPLC. The active component was isolated from the n-butanol soluble fraction of AR and was structurally identified as calycosin-7-O-${\beta}$-D-glucopyranoside by LC-MS, IR, $^1H$ NMR, and $^{13}C$ NMR analysis. The $IC_{50}$ of calycosin-7-O-${\beta}$-D-glucopyranoside's HAase activity was found to be 3.7 mg/mL.

Biochemical characteristics and capsular serotypes of Streptococcus suis isolated from pneumonic lungs of slaughter pigs (도축돈의 폐렴병소로부터 분리한 Streptococcus suis의 생물화학적 특성 및 협막혈청형)

  • The present study was conducted to investigate the incidence of Streptococcus suis infection in the pneumonic lungs of slaughter pigs from December 1992 to April 1994 and to examine the biochemical and cultural properties, the antimicrobial susceptibility and serotype of the isolates. Fifty-six(7.95%) strains of S suis was isolated from 704 pneumonic lungs of slaughter pigs, of which 33 cultures were isolated alone, while remaining 18 were complicated wide Pasteurella multocida. Of the 56 S suis isolates, 18(32.1%) were serotype 2, 5(8.9%) were serotype 1/2, 2 were serotype 1, 1 each for serotype 3 & 5, and the remaining 29 were untypable. The twenty five strains(44.6%) of all isolates produced hyaluronidase while 15 strains(88.3%) of 18 serotype 2 isolates were hyaluronidase producers. All of isolates were highly susceptible to ampicillin, amoxicillin, ceftiofur, cephalothin, ciprofloxacin and penicillin G, although the majority of them were resistant to kanamycin, colistin, erythromycin, oxytetracycline.

Metabolism of Glycyrrhizin in Polyprescriptions Containing Glycyrrhizae Radix by Human Intestinal Bacteria and Their Inhibitory Effects on Some Enzymes (감초 함유 처방의 글리치리진 대사와 몇가지 효소저해효과)

  • To analyze scientifically the prescription principle of polyprescriptions (Gamchotang, Daewhanggamchotang, Jakyakgamchotang, Gamchogungangtang and Gilkyungtang) containing Glycyrrhizae Radix, the transforming rate of glycyrrhizin in these polyprescriptions to 18 ${\beta}-glycyrrhetinic$ acid and their inhibitory effect on ${\beta}-glucuronidase$, hyaluronidase, phosphodiesterase and trypsin were investigated. When Glycyrrhizae Radix containing polyprescriptions were extracted with water, the contents of glycyrrhizin in water extract of Glycyrrhizae Radix with Rhei Rhizoma or with Zingiberis Rhizoma were higher than that of Glycyrrhizae Radix only, but that in water extract of Glycyrrhizae Radix with Platicodi Radix was lower than that of Glycyrrhizae Radix only. By human intestinal bacteria, glycyrrhizin was metabolized to 18 ${\beta}-glycyrrhetinic$ acid. These metabolism of glycyrrhizin in polyprescriptions containing Glycyrrhizae Radix was inhibited by Rhei Rhizoma, Paeoniae Radix and Platicodi Radix, but was not affected by Zingiberis Rhizoma. The inhibitory activity of Glycyrrhizae Radix on hyaluronidase and ${\beta}-glucuronidase$, was synergistic with Rhei Rhizoma or Zingiberis Rhizoma, but was antagonistic by Platicodi Radix.

Anti-allergic Action of Some Medicinal Plants (수종 생약의 항알레르기 작용)

  • Anti-allergic action of each water extracts of some crude drugs was investigated in mice and rats. The activity of hyaluronidase which was used in the screening test of anti-allergic action was inhibited significantly by Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Cinnamomi Ramulus, Glycyrrhizae Radix and Scutellariae Radix. The 48-hour homologous passive cutaneous anaphylaxis(48-hr PCA) in mouse ear was inhibited significantly by intraperitoneal(i.p.) injection of Amomi Semen, Cimicifugae Rhizoma, and ketotifen, a comparative drug of an anti-allergic action. The increase of vascular permeability induced by histamine or serotonin was inhibited significantly by i.p. injection of Amomi Semen, Cimicifugae Rhizoma, Cinnamomi Ramulus and ketotifen. In rat dorsal skin, the increase of vascular permeability which was induced by histamine, serotonin or compound 48/80 was inhibited significantly by i.p. injection of Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Scutellariae Radix and ketotifen. Armeniacae Semen and Liriopis Tuber which had not inhibited hyaluronidase activity did not inhibit 48-hr PCA and the increase of histamine, serotonin or compound 48/80-induced vascular permeability in mice and rats. These results suggest that each water extract of Amomi Semen and Cimicifugae Rhizoma has anti-allergic action.

Efficacy of submucosal injection of hyaluronidase after mandibular third molar surgery: a randomized controlled trial

  • Objectives: This study aimed to investigate the efficacy of postoperative submucosal injection of hyaluronidase (HUD) for reducing sequelae and quality of life (QOL) after mandibular third molar (M3M) surgery. Materials and Methods: Participants with bilateral impacted M3M underwent surgical extraction with a split-mouth randomized controlled study design. M3M were removed by the same surgeon in 2 sessions, one a control and the other experimental. Submucosal injection of HUD was performed in the experimental session and submucosal injection of saline in the control session. Mouth opening, facial swelling, and pain intensity were measured before surgery, and then 2 and 7 days after surgery. The QOL of participants following surgery was evaluated by means of a patient-centered outcome questionnaire (PCOQ). Results: A total of 36 patients was included in the final data analysis. There was a significant reduction in the maximal mouth opening and postoperative pain in the experimental side at the 2 and 7 days after surgery (P<0.05), and a remarkable difference in facial swelling was reported on the experimental side 7 days after surgery (P<0.05). The PCOQ demonstrated that participants reported less pain and swelling on the experimental side. Conclusion: The present study provides clinical evidence that submucosal administration of HUD immediately after M3M surgery reduced postoperative discomfort and improved patients' QOL.

Effects of Allii tuberosi Semen Extract Solution on Reproductive Capacities in Mice (농도별(濃度別) 구자(?子) 투약(投藥)이 수컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

  • Purpose : These studies were undertaken to evaluate the effects of Allii tuberosi Semen (ATS) on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Materials and Methods : We used the 8-week-old mice and administered the 0.2 ml extract solution of ATS in the different concentration (0.1 mg/ml, 1 mg/ml, 10 mg/ml and 100 mg/ml) once a day for 60 days. The control group was administered the distilled water in the same way. After the administration of each extract solution, we examined the number of total, motile and normal sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed the histological changes of isolated testis. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The concentration of total sperm, the motility and normality of spermatozoa were significantly increased in ATS groups, especially in 1 and 10 mg/ml groups, compared to control group. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the ATS groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the ATS groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the ATS groups compared to the control group, especially in 1 mg/ ml group. The activity of testicular catalase was increased in ATS groups. Conclusion : This study shows that ATS has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase and testicular peroxidase. We can suggest that ATS extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Inhibition of Nitric Oxide Production and Hyaluronidase Activities from the Combined Extracts of Platycodon grandiflorum, Astragalus membranaceus, and Schisandra chinensis (길경, 황기와 오미자 혼합추출물의 NO 억제활성과 Hyaluronidase 억제활성 효과)

  • In this study, the optimal extraction conditions for three medicinal herbs as functional sources against inflammatory and arthritic diseases were developed. Traditional medicinal herbs were screened for their inhibition of hyaluronidase (HAse) activity and nitric oxide (NO) synthesis. For the screening of anti-inflammatory properties, ethanolic extracts of 53 species of traditional medicinal herb were examined. We confirmed that Astragalus membranaceus (A.R.), Schisandra chinensis (S.F.), and Platycodon grandiflorum (P.G.) inhibit NO production. For extraction from all three herbs simultaneously, an ethanol concentration of 95%, a 1:2:1 mixture ratio, and at 50 rpm mixing speed, for over 12 h and at $30^{\circ}C$ was the best condition for optimal extract yield and NO inhibition effects. HAse inhibition from the three herb extraction was three fold higher than single samples. The ethanol extracts were fractionated with various solvents (n-hexane, chloroform, ethyl acetate, n-butanol, and water). The ethyl acetate-soluble fraction of the herb mixture showed the highest extract yield (13%) and NO inhibition effects (73%). In conclusion, this study provides experimental evidence that a mixture of P.G., A.R., and S.F. could be used as a source of antioxidant ingredients in the food industry.

Effects of Cynomorii Herba Extract Solution on Reproductive Capacities in Mice (쇄양(鎖陽)의 농도별(濃度別) 투약(投藥)이 수컷 생쥐 생식능력(生殖能力)에 미치는 영향(影響))

The significant differences were observed in the normality of spermatozoa of the CH groups compared to the control group. In the histolocal analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the CH groups compared to the control group. Also, the activity of hyaluronidase was significantly increased in the CH groups compared to the control group. In the antioxidant activity analysis, the activities of testicular peroxidase and testicular catalase were significantly increased in the CH groups compared to the control group, respectively. Conclusion : This study shows that CH has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We can suggest that CH extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Dose Dependent Effect of Morindae officinalis Radix Extract Solution on the Reproductive Capacities in the Mice (농도별(濃度別) 파극(巴戟)투약(投藥)이 숫컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

Methods : We used the 2-month-old mice and administered the extract solution of Morindae officinalis Radix in the different concentration once a day for 60 days. The control group was administered the normal saline in the same way and duration. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed changes of isolated testis before and after administration of Morindae officinalis Radix extract solutions the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The significant dose-dependent differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. Conclusion : This study shows that Morindae officinalis Radix has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase and testicular peroxidase. We can suggest that Morindae officinalis Radix extract solution be useful for the treatment of male sexual dysfunctions and infertility.
