Enhancement of Anti-inflammatory Activities of Fermented Scutellaria baicalensis Extracts using Lactobacillus rhamnosus (유산균 발효를 통한 황금 추출물의 항염증 효과 증진)

  • This study was performed to investigate the anti-inflammatory activities of fermented Scutellaria baicalensis extracts using Lactobacillus rhamnosus. The extracts were WE (water extract at $100^{\circ}C$ for 24 h), EE (70% ethanol extract at $60^{\circ}C$ for 24 h), FWE (fermented and water extract at $60^{\circ}C$ for 24 h), FEE (fermented and 70% ethanol extract at $60^{\circ}C$ for 24 h). The cytotoxicity of the extracts was in the range of 11.2 ~ 15.6 % at 1.0 mg/mL concentratioin. The FEE showed the lowest activity at 1.0 mg/mL concentratioin. Compared to the WE, hyaluronidase inhibitory activity contents in the FEE increased to 9.2% at 1.0 mg/mL concentratioin. Nitric oxide production of WE, EE, FWE and FEE at 1.0 mg/mL concentration was mesured as 7.6, 7.9, 6.9, 6.4 ${\mu}M$, respectively. $PGE_2$ secretion of the human fibroblast of the FEE were lower than 810 pg/mL. Our results suggested that the extracts from fermentation process after 70% ethanol extraction had relatively high anti-inflammatory activities and that the Scutellaria baicalensis could be more extracted in FEE than others.

A Study on Anti-Inflammatory, Antioxidant Activity and Skin Barrier Improvement of Lactobacillus rhamnosus-derived Exosomes (락토바실러스 람노서스 유래 엑소좀의 항염증, 항산화 효능 및 피부 장벽 개선에 관한 연구)

  • In this study, exosomes were isolated by ultrafiltration from Lactobacillus rhamnosus J2K-821 and their various effects for skin were evaluated. Their size and concentration were identified 50 ~ 200 nm and 3.22 × 108 particles/mL, respectively through nanoparticle tracking analysis. In order to verify the inflammatory relief effect of Lactobacillus rhamnosus-derived exosomes (LRDEs), their nitric oxide (NO) production inhibitory ability in RAW 264.7 macrophages induced an inflammatory reaction with lipopolysaccharide (LPS) was confirmed. It was revealed that they inhibited NO production in a concentration-dependent manner. To evaluate the antioxidant activity and skin barrier improvement effect of LRDEs, their 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and hyaluronidase inhibitory activity were confirmed. It was also revealed that their activities were increased by concentration-dependent manner. Through these results, It is believed that LRDEs can be used as a effective natural cosmetic ingredient for anti-inflammation, antioxidation and skin barrier improvement.

Protective Effects of Yinjinchunggan-tang (YJCGT) on Alcohol-induced Oxidative Stress (인진청간탕(茵蔯淸肝湯)의 알코올성 산화스트레스에 대한 보호효과 연구)

  • Objectives : Oxidative stress seems to play a major role in mechanisms by which ethanol causes liver injury. Previous studies have shown that treatment with Yinjinchunggan-tang (Yinchenqinggan-tang, YJCGT) has protective effects on alcoholic liver disease. The aim of this study was to investigate the protective effects of YJCGT on alcohol-induced oxidative stress. Materials and Methods : In vitro, we evaluated the inhibitory activities of YJCHT on DPPH(1,1-diphenyl-2-picryl-hydrazyl), xanthine oxidase, trypsin, and hyaluronidase. In a cell culture model, we measured cell viability and proliferation, and the activities of superoxide dismutase (SOD), and catalase (CAT) after YJCGT treatment in C34 and E47 cell lines, and HepG2 cells transfected with/ without cytochrome P450IIE1 (CYP2E1) gene. In vivo, we estimated serum level of hepatic biochemical markers, and alcohol concentration in the blood. Results : YJCGT showed significant free radical scavenging activity against DPPH and xanthine oxidase and decreased hyaluronidase activity effectively in vitro. YJCGT also increased cell viability, and proliferation in C34 and in E47 cell lines, and increased activities of superoxide dismutase, and catalase in C34 and in E47 cell lines. YJCGT reduced serum AST, LDH, and total cholesterol level in some of the results, and reduced blood alcohol concentration in vivo, as well. Conclusions : This study suggests that YJCGT has protective effects on oxidative stress by inhibiting alcohol-induced suppression of antioxidant enzyme activities.

Anti-oxidant and Anti-inflammation Activities of Prunus persica Flos (도화(桃花, Prunus persica Flos)의 항산화 및 항염증 활성)

  • The solvent extracts of Prunus persica Flos were investigated for the activities of anti-oxidant and anti-inflammation to apply as a functional ingredient for cosmetic products. The electron donating ability of both ethanol (PPE) or acetone (PPA) extracts of P. persica Flos was above 90.0% at the concentration of 500ppm. The superoxide dismutase (SOD)-like activity of P. persica Flos extracts (PPE, PPA) were approximately 40.0% at 1,000 ppm. The xanthine oxidase inhibitory effect of P. persica Flos extracts (PPE, PPA) was approximately 30.0% at 1,000 ppm and equivalent to that of ascorbic acid. Hyaluronidase inhibition activity related to the anti-inflammation effect was 35.0% with the treatment of P. persica Flos extracts (PPW, PPE, PPA) at 1,000 ppm, respectively. In the experiment of anti-inflammation effect, P. persica Flos extracts (PPW, PPE, PPA) inhibited the generation of nitric oxide. In the antimicrobial activity test against the human skin-resident microflora such as Staphylococcus epidermidis and Propionibacterium acnes, a clear zone was identified from 4mg/disc in P. persica Flos (PPE) extract.

Optimization of Hyaluronidase Inhibition Activity from Prunus davidiana (Carriere) Franch Fruit Extract Fermented by its Isolated Bacillus subtilis Strain SPF4211

  • Strain SPF4211, having hyaluronidase (HAase) inhibition activity, was isolated from P. davidiana (Carriere) Franch fruit (PrDF) sugar extract. The phenotypic and biochemical properties based on 16S rDNA sequencing and an API 50 CHB kit suggested that the organism was B. subtilis. To optimize the HAase inhibition activity of PrDF extract by fermentation of strain SPF4211, a central composite design (CCD) was introduced based on three variables: concentration of PrDF extract (X1: 1-5%), amount of starter culture (X2: 1-5%), and fermentation time (X3: 0-7 days). The experimental data were fitted with quadratic regression equations, and the accuracy of the equations was analyzed by ANOVA. The statistical model predicted the highest HAase inhibition activity of 37.936% under the optimal conditions of X1 = 1%, X2 = 2.53%, and X3 = 7 days. The optimized conditions were validated by observation of an actual HAase inhibition activity of 38.367% from extract of PrDF fermented by SPF4211. These results agree well with the predicted model value.

Anti-inflammatory Effect of Jatrorrhizine from Phellodendron amurense in Lipopolysaccharide-stimulated Raw264.7 Cells (Lipopolysaccharide로 유도된 Raw264.7 cell에서 Phellodendron amurense의 Jatrorrhizine에 의한 염증 억제효과)

  • n-Butanol extracts from Phellodendron amurense have about 50% inhibitory activity against hyaluronidase. The anti-inflammatory compound was isolated from P. amurense by Sephadex LH-20 and MCI-gel CHP-20 column chromatography with gradient elution. As a the result, its structure was identified as Jatrorrhizine by the interpretation of spectroscopic analyses including $^1H$- and $^{13}C$-NMR. In anti-inflammatory activity, the expression of nitric oxide (NO) was inhibited as above 60% at 100 ${\mu}g/mL$ concentration of extracts and then purified Jatrorrhizine from P. amurense. The inhibitory activities against the expression of inducible NO syntase (iNOS) and cyclooxygenase-2 (COX-2) were 45% and 29%. It seems that the extracts and purified Jatrorrhizine from P. amurense were expected anti-inflammatory effect in lipopolysaccharide (LPS)-stimulated Raw264.7 cells.

Administration Term-dependent Effects of Allii Tuberosi Semen Extract Solution on the Reproductive Capacities in the Mice (구자 투약기간이 수컷 생쥐의 생식능력에 미치는 영향)

  • 목 적 : 본 연구는 기간별 구자 투약이 생쥐의 생식능력에 미치는 영향을 알아보고자 하였다. 방 법 : 이를 위해 30일, 60일, 90일 및 120일간의 구자 투약이 생쥐의 총 정자수, 활동 정자수, 정상형태 정자수, 고환조직 및 hyaluronidase activity를 측정하여, 다음과 같은 결론을 얻었다. 결 과: 1. 총 정자수는 60일과 90일 투여 후 대조군에 비하여 증가하였다. 2. 활동 정자수는 60일, 90일 및 120일 투여 후 대조군에 비하여 증가하였다. 3. 정상 형태 정자수는 60일, 90일 및 120일 투여 후 대조군에 비하여 증가하였다. 4. 고환 세정관의 조직학적 변화는 60일 투여 후에 가장 현저히 나타났다. 5. Hyaluronidase activity는 모든 투여군에서 대조군에 비하여 증가하였다. 결 론: 이상의 결과를 통해 60일 이상의 구자 투약이 남성 생식능력을 활성화 할 수 있다고 사료된다.

Fourier Transform Ion Cyclotron Resonance (FT-ICR) MASS Spectrophotometric Analysis of Flower Petal from Paeonia lactiflora cv. ‘Red Charm’ and Evaluation of its Functional Activity (작약 레드참 꽃잎의 이온화원-푸리에 변환 질량분석과 기능성 연구)

  • Little attention has been paid to the functional aspect of the flower petal of Paeonia lactiflora, compared to that of its root. To determine the components of flower petal of Paeonia lactiflora, we conducted the Fourier transform ion cyclotron resonance (FT-ICR) MASS spectrophotometric analysis. We detected the 24 different types of ingredients from the 70% ethanol extracts of flower petal of peonia lactiflora cv. ‘Red Charm’. The main compounds were quercetin glucopyranosides, methyl gallate, paonioflolol and kaemperol glucopyranosides. We further tested its functional activity. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the extracts was 87.9-90.4% at 0.1mg/ml. This result showed that these flower extracts have approximately 5-fold stronger antioxidant potential than a previous report with root extracts (Bang et al. 1999). The result of tyrosinase inhibition assay of Paeonia lactflora extract was almost similar to that of arbutin except significantly higher effect in the coral sunset extract at 0.1% concentration. Hyaluronidase inhibition assay showed 76.5% inhibition at 5% concentration of this flower extract, indicating that Peaonia lactiflora flower extracts have the major anti-inflammatory, anti-oxidant and brightening effects. Taken together, these results suggest these three Paeonia lactiflora species extracts might provide the basis to develop a new natural brightening agent.

Antioxidant and Anti-inflammatory Effects of Fractions from Pruni persicae Flos (도화(桃花, Pruni persicae Flos) 분획물의 항산화 및 항염증 효과)

  • Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.
