Increase of ICSI Efficacy with Hyaluronic Acid-Binding Sperm for Low Aneuploidy Frequency in Pig

  • Hyaluronic acid (HA)-binding sites have been shown the diagnostic potential fur assessment of sperm maturity, which is related to male fertility. This study was designed to evaluate chromosomal patterns in porcine embryos produced by in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) with non- or HA-binding sperm (HABS). For binding of sperm with HA, sperm incubated in 10 ${mu}ell$ drop containing HA (0.8 mg/ml)-agarose (0.8%) mixture for 15 min. IVF and ICSI with non- or HA-bound sperm examined with matured oocytes at 44 hr after in vitro maturation. Embryos were cultured in 50 ${mu}ell$ of NCSU 23 containing 0.5% BSA for 5 days and then in 50 ${mu}ell$ of NCSU 23 containing 10% FBS for 2 days. For the evaluation of chromosomal aneuploidies, chromosome 1 sub-metacentric specific probe was used in sperm and embryos by fluorescence in situ hybridization (FISH). The frequency of aneuploidy sperm for chromosome 1 was 6.25%. The significant differences following IVF and ICSI with non- or HA-bound sperm were not observed in blastocyst formation rates (18.6, 23.5, and 23.8%) and cell number (61.8 $\pm$ 12.5, 55.5 $\pm$ 7.3, and 59.3 $\pm$ 9.6). Moreover, the percentage of diploidy in 4-cell stage embryos was 57.1% (IVF), 68.8% (ICSI), and 76.3% (ICSI-HABS). These results suggest that HA-binding sites may be a material for selection of normal sperm for ICSI. Therefore HA selection of normal sperm may be reduce the loss to embryonic mortality prior to embryo transfer in pig.

The Effect of High Molecular Hyaluronic Acid on Bone Formation in Human Fetal Osteoblasts (사람 태아 골모세포에서 고분자 히알루론산의 골형성 유도에 관한 연구)

  • Hyaluronic acid (HA)는 중요한 glycosaminoglycan 중 하나로서 단백질과 화학적 결합을 하지 않기 때문에 분리가 쉽고 결합조직의 세포간 기질의 주요 성분이다. 우리는 점탄성 고분자 hyaluronic acid를 실험실상에서 사람 태아 골모세포의 골 형성 과정에 미치는 영향을 알아보고자 하였다. 우리는 여러 농도의 HA에 대한 사람 태아 골모세포에서의 세포증식, 염기성 인산분해효소 활성, 석회화 결절 형성능, 교원질 합성능 그리고 bone sialoprotein (BSP)의 발현 정도를 검사하였다. 세포증식에서 각 농도의 HA 처리군과 대조군 간에 2일과 4일간의 결과에서 유의한 차이를 보이지 않았다. 염기성 인산분해효소 활성에서는 0.063% HA 처리군에서 음성 대조군에 비해 가장 유의한 염기성 인산분해효소 활성을 보였다 (p<0.05). 0.063% HA 처리군은 교원질 합성능에서도 가장 높은 수준을 보였다 (p<0.05). 석회화 결절 형성능에서는 0.063% HA 처리군에서 대조군에 비해 많은 염색된 석회화 결절을 보였다. BSP의 발현 정도를 분석한 Western blot에서는 대조군에 비해 0.063% HA 처리군에서 증가된 단백질 발현을 나타났다. 본 연구 결과 고분자 HA가 실험실상에서 사람 태아 골모세포의 분화를 통해 새로운 골 형성을 유도할 수 있는 능력이 있음을 시사하였다.

Study of Superparamagnetic Microneedles containing Iron Oxide Nanoparticles (산화철 나노입자를 함유한 초상자성 마이크로니들에 관한 연구)

  • Recently, iron oxide nanoparticles have been used as the subject of many studies on drug delivery system (DDS) due to their excellent magnetic properties and biocompatibility in response to external magnetic fields. In this study, hyaluronic acid-superparamagnetic microneedles (HA-SMNs) and carboxy methyl cellulose-superparamagnetic microneedles (CMC-SMNs) containing superparamagnetic iron oxide nanoparticles (SIONs) were prepared with HA and CMC as a matrix materials of MNs (microneedles). Various properties of SMNs were then investigated with scanning electron microscopy (SEM), superconducting quantum interference device-vibrating sample magnetometer (SQUD-VSM), frequency mixing magnetic detection (FMMD), and polymer/bio membrane. The SQUID-VSM measurements showed superparamagnetism of HA-SMNs and CMC-SMNs containing SIONs. The FMMD results demonstrated that the signal intensity changed significantly as the concentration of SIONs increased. In addition, SMNs exhibited the average skin permeability intensities on the bio membrane for HA-SMNs and CMC-SMNs were 92.5 and 98.5%, respectively. These results suggested that SMNs could be utilized as deliver materials for a TDDS and MR molecular imaging.

Synthesis of Hyaluronic Acid Microsphere Crosslinked with Polyethylene Glycol Diglycidyl Ether Prepared by A Simple Fluidic Device

  • Hyaluronic acid (HA) microspheres (MSs) crosslinked with polyethylene glycol diglycidyl ether (PEGDE) are prepared using a simple fluidic device (SFD) to investigate the optimized parameters. A solution mixture of PEGDE in 2-methyl-1-propanol was prepared as a continuous phase in SFD. HA solutions of 1 wt% concentration were introduced into SFD as a discontinuous phase. The HA solution prepared by stirring for more than 48 h exhibited spherical MSs at the needle tip inside the ring cap. As the flow rate of the continuous phase increased from 0.7 to 1.9 mL/min, the diameter of the MS decreased from 173±36 ㎛ to 129±13 ㎛. Although the PEGDE concentration in the range of 0.2 to 1.8 vol% did not affect the diameter of the MS, the microstructure of MS, consisting of inner hollow void and wall, was changed. The inner void and wall size decreased and increased from 79.5 ㎛ to 57.2 ㎛ and from 10.3 ㎛ to 21.4 ㎛, respectively, with increasing PEGDE concentration from 0.2 vol% to 1.8 vol%. FT-IR peaks located around 2867 cm-1 and 1088 cm-1 indicated that the HA MS prepared at different PEGDE concentrations were chemically crosslinked. The HA MSs containing different PEGDE concentrations exhibited quantitative cell viability of more than 98%. L-929 cells adhered well to the HA MSs and proliferated continuously with increasing culture time to 48 h regardless of PEGDE concentration, implying that the HA MSs are clinically safe and effective.

Anti-aging Effect of Artocarpin in UVA-irradiated Normal Human Epidermal Keratinocytes (자외선 조사에 의해 노화된 인간각질형성세포에서 Artocarpin의 항노화 효능)

  • The aim of this study was to investigate the epidermal moisturizing effects of artocarpin on normal human epidermal keratinocytes (NHEKs). To investigate the effects of artocarpin on NHEKs, cell viability and the expression of mRNAs related to skin hydration were measured. In addition, hyaluronic acid (HA)-ELISA assay was performed. Here, the effects of artocarpin on AQP3, HAS2, KRT1, and KRT10 mRNA expression, and on HA production, following UVA treatment were reported. The Quantitative real-time PCR results demonstrate that artocarpin increased AQP3, HAS2, KRT1, and KRT10 mRNA levels. The HA-ELISA assay revealed that artocarpin also increased HA production in NHEKs. Through these experiments, the epidermal moisturizing effects of artocarpin have been elucidated, providing evidence that artocarpin may be a potent cosmetic ingredient in skin anti-aging and moisturizing products. Based on these results, I anticipate that further research on the mechanisms of action of artocarpin may allow the development of not only cosmetics, but also medicines and healthcare foods.

Non-inferiority study of the efficacy of two hyaluronic acid products in post-extraction sockets of impacted third molars

  • Background: Hyaluronic acid (HA) is well known to exert an anti-inflammatory effect during oral wound healing and is commonly applied after tooth extraction. However, no double-blind randomized controlled study comparing two hyaluronate mouthwash products has been conducted so far. The aim of this study was to comparatively analyze the efficacy of Mucobarrier® and Aloclair® in terms of clinical symptoms. Results: A total of 112 patients were randomly assigned to assess the degree of discomfort, pain reduction, redness, burning sensation, and swelling between two groups on the day of surgery and 7 days later in a double blind test, with a total 56 Aloclair patients and 56 Mucobarrier patients. There was no statistically significant difference in the overall discomfort, degree of pain reduction, redness, burning sensation, and swelling between the Mucobarrier and Aloclair groups. Conclusion: The local application of hyaluronic acid mouth wash after wisdom tooth extraction is beneficial in reducing overall discomfort and pain reduction, and the clinical utility of Mucobarrier® is no different from Aloclair®. APPLICATION OF 0.2% HYALURONIC ACID GEL IN ORAL SURGICAL WOUND : A RANDOMIZED DOUBLE-BLIND STUDY (구강내 외과적 창상 치유에서 0.2% hyaluronic acid gel의 유효성에 대한 이중 맹검 연구)

  • The aim of this study was to evaluate the efficacy of a topical 0.2% hyaluronic acid (HA) preparation in the management of wound after removal of arch bar for facial bone fracture and a suture site after orthognatic, oral cancer or oral surgery. Forty patients participated in a randomized, placebo controlled, double-blind trial to evaluate the efficacy of the topical HA and preparation. HA topically applied to the wound after removal of arch bar or stitch out, 3 times a day for 4 weeks. Evaluation is performed once a week for 4 weeks. For subjective evaluation, relative pain reduction in visual analog scale (VAS) and existence of heat sensation was accessed. For objective evaluation, gross evaluation, papilla index, existence of wound dehiscence, redness and swelling was checked. The same evaluation was performed in each arch bar group and suture group. For whole subject, 0.2% HA group resulted higher reduction than placebo group in pain of site in first week with significancy. Same findings were seen other weeks but there was no significancy. 0.2% HA group had better result than placebo in objective evaluation (papilla index, wound dehiscence, redness and swelling), but in gross evaluation placebo had better result than 0.2% HA group with no significancy. Subject was divided into suture group and arch bar group. Same aspect was seen, but only suture group had significancy not arch bar group in pain reduction score. 0.2% HA group resulted higher reduction than placebo group in pain of site in first week with significancy, especially in suture group. It reveals topical application of HA in wound especially suture site reduced pain in early stage. And 0.2% HA group had better result than placebo in papilla index, redness and swelling with no statistical significancy. In conclusion, HA has effect of pain reduction and healing promotion in the mucosal wound after oral surgery.

CD44 is the principal cell membrane receptor for HA, expressed from the 1-to 8-cell stage in human embryos, during post-implantation mouse and bovine embryogenesis and on the surface of differentiated embryonic stem cells. Characterization of Lactide/Hyaluronic Acid Polymer Cross-Linked by 1,3-Butadiene Diepoxide (1,3-Butadiene Diepoxide에 의해 가교된 락타이드/히아루론산 고분자의 특성)

  • The hyaluronic acid (HA) polymers cross-linked with lactide (LA) using the crosslinking agent, 1,3-butadiene diepoxide (BD), were prepared in order to develop a biomedical material for tissue engineering. The degree of lactide and BD reaction of the crosslinked polymer was determined by the analysis of nuclear magnetic resonance spectroscopy. Both degree of reaction and swelling ratio increased with BD concentration or LA/HA mole ratio. Tensile modulus decreased with increasing BD concentration or decreasing LA/HA mole ratio. Degradation was shown to be progressed at two different stages and became slow with increasing BD concentration. It was shown that the first stage degradation was mainly due to the decomposition of ester linkage in the crosslinked structure. The cell growth inhibition increased with BD concentration. Although cytotoxicity was slightly observed in the high BD concentration, the value was very low (below 6%) enough not to affect the cell growth.
