Dissolved oxygen (DO) has a significant effect on the molecular weight of hyaluronic acid (HA) during the fermentation of Streptococcus zooepidemicus. Therefore, to further investigate the effect of DO on the yield and molecular weight of HA, this study compared the metabolic flux distribution of S. zooepidemicus under aerobic conditions at various DO levels. The metabolic flux analysis demonstrated that the HA synthesis pathway, considered a dependent network, was little affected by the DO level. In contrast, the fluxes of lactate and acetate were greatly influenced, and more ATP was generated concomitant with acetate at a high DO level. Furthermore, the has gene expression and HA synthase activity were both repressed under anaerobic conditions, yet not obviously affected under aerobic conditions at various DO levels. Therefore, it was concluded that the HA molecular weight would seem to depend on the concomitant effect of the generation of ATP and reactive oxygen species. It is expected that this work will contribute to a better understanding of the effect of the DO level on the mechanism of the elongation of HA chains.
Purpose: To evaluate clinical efficacy and safety of hyaluronic acid based autologous dermal fibroblasts (Hyalograft 3D) in the treatment of diabetic foot ulcers. Methods: A total of 28 patients with diabetic ulcers were randomized to either the control group with nonadherent foam dressings(n=14) or the treatment group with autologous tissue - engineered grafts(n=14). Weekly assessment contained vital sign checks, ulcer size measurements, and wound photos. At 12th week, percentages of complete wound healing and mean healing times were compared. Safety was also monitored by adverse events. Results: Complete wound healing was achieved in 84.6% of the treatment group and 23.1% of the control group (p<0.005). The mean times of closures for the treatment versus control groups were 6.1 weeks and 10.9 weeks, respectively. No adverse events related to the study treatment occurred. Conclusion: The use of hyaluronic acid based autologous fibroblast grafts was found to be a safe and effective treatment for diabetic foot ulcers.

Background: Hyaluronic acid (HA) is well known to exert an anti-inflammatory effect during oral wound healing and is commonly applied after tooth extraction. However, no double-blind randomized controlled study comparing two hyaluronate mouthwash products has been conducted so far. The aim of this study was to comparatively analyze the efficacy of Mucobarrier® and Aloclair® in terms of clinical symptoms. Results: A total of 112 patients were randomly assigned to assess the degree of discomfort, pain reduction, redness, burning sensation, and swelling between two groups on the day of surgery and 7 days later in a double blind test, with a total 56 Aloclair patients and 56 Mucobarrier patients. There was no statistically significant difference in the overall discomfort, degree of pain reduction, redness, burning sensation, and swelling between the Mucobarrier and Aloclair groups. Conclusion: The local application of hyaluronic acid mouth wash after wisdom tooth extraction is beneficial in reducing overall discomfort and pain reduction, and the clinical utility of Mucobarrier® is no different from Aloclair®. The hyaluronic acid (HA) conjugate bearing $\alpha$-cyclodextrin ($\alpha$-CD) was synthesized as the potential carrier of poly(ethylene glycol) (PEG)-drug conjugates. The HA conjugate was prepared by the reaction between the carboxylic acid of HA and the primary amine of $\alpha$-CD in the presence of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide and 1-hydroxybenzotriazole. The chemical structure of the conjugate was confirmed using $^1H$ NMR and FT-IR spectroscopy. The conjugate could form nano-sized particles in the presence of PEG by forming the inclusion complexes between $\alpha$-CD at the backbone of HA, which was demonstrated using electrophoretic light scattering and field emission transmission electron microscopy. It is anticipated that this novel kind of nanoparticles can serve as a useful delivery system for PEGylated drugs.
Purpose: Currently, soft tissue filler products based on hyaluronic acid are widely used. They are safe, effective, and convenient to use compared with bovine collagens. However, all commercially available hyaluronic acid based fillers in Korea are imported ones. The purpose of this study is to evaluate efficacy of a new hyaluronic acid filler (HyaFilia; CHA bio&Diostech Co., Seoul), which has been recently developed in Korea. Methods: Three kinds of soft tissue fillers, including Restylane (Q-Med, Uppsala, Sweden), HyaFilia (CHA bio & Diostech Co., Seoul), Juvederm (Allergan, CA, USA), were injected subdermally into the back of hairless mice at six sites. The 2 cephalic sites composed Restylane groups, the 2 middle sites HyaFilia groups, and the 2 caudal sites Juvederm groups. Six hairless mice were included in the study, therefore, a total of 12 injections per group were performed. After 16 weeks after injection, the nodular swellings that resulted from the injections were excised wide enough to include skin beyond the swelling points down to the panniculus carnosus layer using 8mm punches. Volumes and weights were measured using a stereoimage optical topometer system and a weighting machine. Histologic comparisons were also carried out. Results: The mean volumes of the Restylane, HyaFilia and Juvederm groups were $3698{\times}103$, $4820{\times}103$, and $1435{\times}103$ PI, respectively. The mean weights of the Restylane, HyaFilia and Juvederm groups were 36.08, 37.83, and 24.66 mg, respectively. Histologic examination between the 3 groups showed no significant difference in tissue compositions and inflammatory reactions. Conclusion: The results of this study shows that HyaFilia is superior to Restylane and Juvederm in longavity of filling effect. Therefore, HyaFilia may be an effective replacement for the imported materials including Restylane and Juvederm.
Conventional drug delivery methods mainly include subcutaneous injection and oral administration. Subcutaneous injection has the advantages of delivering the correct concentration, but it might cause pain and trauma to patients. Although oral administrations do not accompany pain as the subcutaneous injection, unexpected side effects may occur because they undergo digestion process and it is not easy for many drugs to be exposed to targeted sites with proper concentration. While dissolving microneedles have been extensively studied to overcome the limitations of conventional subcutaneous injection, the effects on microneedle shapes for drug release have not been fully explored. In this study, the characteristics of hyaluronic acid microneedles for cone and quadrangular pyramidal shapes were examined by the size, volume, contact surface area, skin permeability, and dissolution rate. As a result, the cone shapes of hyaluronic acid microneedles had high skin permeability, and the quadrangular pyramid of shapes hyaluronic acid microneedles showed a fast dissolution rate, given by the large contact area on the skin. Based on the results, we could confirm that the shape of a dissolving microneedles can affect skin permeability and the drug delivery rate.
Ambroxol is thought to have antioxidant ability and some antiinflammatory effect. Effect of ambroxol on the oxidative damages of lipid, collagen and hyaluronic acid was examined. F $e_{2+}$(10 $\mu$M) and 100$\mu$Mascorbate-induced lipid peroxidation of liver microsomes was inhibited by 10 and 100$\mu$M ambroxol, 30$\mu$g/ml catalase and 10 mM DABCO but was not affected by 30$\mu$g/ml SOD and 10 mM DMSO. A 10 and 100$\mu$M ambroxol and 10 mM DABCO inhibited the peroxidative action of 10$\mu$M F $e_{3+}$, 160$\mu$M ADP and 100$\mu$M NADPH on microsomal lipids, whereas inhibitory effects of 30$\mu$g/ml SOD,30$\mu$g/ml catalase and 10 mM DMSO were not detected. The degradation of hyaluronic acid caused by 107M Fe2\\`,5007M H2O2 and 100$\mu$M ascorbate was inhibited by 10 and 100$\mu$M ambroxol,30$\mu$g/ml catalase,10 mM DMSO and 10 mM DABCO, while 30$\mu$g/ml SOD did not show any effect. The cartilage collagen degradation caused by 307$\mu$ F $e_{2+}$,500$\mu$M $H_2O$$_2$ and 200$\mu$M ascorbate was prevented by 100$\mu$M ambroxol. $H_2O$$_2$ and OH . were scavenged by ambroxol, whereas $O_2$, was not removed by it. Ambroxol (100$\mu$M) and 1 mM cysteine reduced DPPH to 1,1-diphenyl-2-picrylhydrazine. In conclusion, ambroxol may inhibit the oxidative damages of lipid, hyaluronic acid and collagen by its scavenging action on oxidants, such as OH . and probably iron-oxygen complexes and exert antioxidant ability.
Oral lichen planus (OLP) is a chronic inflammatory immune-mediated condition that has been identified as a potentially malignant oral disorder. Various therapies have been proposed for its management as alternative to corticosteroids. However, no definitive treatment has been identified that can result in complete remission or minimal recurrence. Hyaluronic acid has recently been used as an alternative therapy for the management of OLP. This study aimed to systematically review the effectiveness of Hyaluronic acid in the management of symptomatic OLP. Online electronic databases and manual searches were performed for randomized controlled trials (RCTs) published in English between January 2010 and April 2022. RCTs were identified that compared the efficacy of hyaluronic acid and other interventional therapies at baseline and during follow-up. The Cochrane Risk of Bias tool was used to assess the quality of the included studies. Visual analog scale (VAS) scores, Thongprasom sign scores, lesion size, degree of erythema, clinical severity, and disease severity were assessed both quantitatively and qualitatively. Seven studies were analyzed. Five studies reported a high risk of bias while the remaining two studies reported an unclear risk of bias. The overall quantitative assessment of size, symptoms, degree of erythema, and sign score in OLP lesions treated with HA was not statistically significant compared to that in the control group (P > 0.05). In addition, subgroup analysis comparing HA with placebo or corticosteroids did not yield statistically significant (P > 0.05) results. Qualitatively, both HA and tacrolimus resulted in an effective reduction in signs and symptoms. Clinical/disease severity index/scores were inconsistent. A high degree of heterogeneity was observed among the included studies. None of the included studies reported the side effects of HA. These findings suggest that corticosteroids, tacrolimus, placebo, and HA could be equally effective in OLP management. The clinical/disease severity index or score reduction cannot be determined with certainty. Thus, OLP can be treated with HA as an alternative therapy. Owing to limited clinical trials on HA, high heterogeneity, and high risk of bias in the included studies, definitive conclusions cannot be derived.
Materials currently used as an injectable bulking agent in the dermatologic and urologic fields revealed several drawbacks such as particles migration, inflammatory reaction, allergic reaction, rapid volume shrinkage, and necessity of a donor site. In this study, we have developed injectable biomaterial comprising poly (DL- lactide-co-glycolide)(PLGA) and hyaluronic acid gel to overcome these problems. PLGA is a biocompatible synthetic material and hyaluronic acid is a common substance found in living organisms. We examined the feasibility of injection through needle and tested biocompatibility in animal model. After transplantation, injected sites and distant organs were examined histologically to verify a new tissue formation, inflammation, and particles migrations. Injected volume was maintained approximately 80 percent for 2 months. Results demonstrated that the developed material was injectable through various gauges of needles and induced a new bulking tissue formation without serious inflammatory reaction.

Poly(vinyl alcohol)(PVA) and hyaluronic acid(HA) hydrogel membranes were prepared with varying HA contents from 10 to 50 wt% of PVA. The water contents of the resulting PVA-HA hydrogel membranes in various pH conditions were measured. And the permeation coefficient of indomethacin was determined using several PVA-HA hydrogel membranes at various pH conditions and also 37$^{\circ}C$.
