Study on Dasan's apprehension for I Ching (다산(茶山)의 주역(周易) 해석에 대한 연구)

  • The medicine through I-Ching(The Book of Changes, 易經) is a field of the medical science, which studies physiology, pathology and Yin-Yang philosophy. From ancient times so many scholars have studied I Ching and they are divided into two different school. one is the school of Image and Number(象數學派), the other is the school of reason(義理學派). Da-San Jung Yak-Yong(茶山 丁若鏞) is a distinguished scholar in the I-Ching study, and he had a unique opinion in the analysis about sentences of I Ching. He has done his best to make 'Image and Number(象數)' harmonize with reason(義理). I Ching is the book about changes, which includes everything like natural phenomena, human body and mind. So we can understand human physiology and pathology through I Ching. But it's important to understand it was organized by symbols. The main symbols are Ba-Gua(八卦), 12 Bi-Gua, Zai-Ruo-zhi-Gua(再閏之卦), 50 Yan-Gua(50衍卦) and these symbols originated from the imagess of the four seasons. The image of 12 Bi-Gua coincide with 12 jing-lao(經絡), the images of Zai-Ruo-zhi-Gua(再閏之卦) coinside with Ren-mai(任脈), Du-mai(督脈). 12 Bi-Gua and Zai-Ruo-zhi-Gua(再閏之卦) are fundamental stuffs, on the other hand 50 Yan-Gua(50衍卦) is an application of every phenomenon.

  • Learning can be defined as its objects, main question for the objects, and its unique way to organize all the knowledge acquired as the results of the question. From the point of view like this, Traditional Korean Medicine(TKM) can be defined as learning for human body and its functions, health and diseases based on the theory of the Yin and Yang and of the five elements. Nowaday Many papers based on laboratory work publish for the name of scientization of TKM, but from the viewpoint of definition of learning, they have a problem that there is no basic theory. If TKM could be communicated with western natural science, it has to be solved. And oriental physiology has a same object and same questions with western physiology, so oriental physiology can be useful to make a bridge between TKM and western natural science.

  • To evaluate the fields of human anatomy and physiology important in clinical practice of physical therapy, a questionnaire survey was performed for 70 physical therapists who were practicing in general hospitals, clinics, public health centers, and rehabilitation centers in several provinces including Seoul and Chungcheongdo. Fields of anatomy and physiology were divided into 13 chapters and subdivided into 52 items. And the preferred ranks were statistically analysed according to the career of physical therapists, nature of the institute and the post of duty. As results joint system, muscular system, skeletal system and nervous system were ranked in the fields of great importance. The relative importance of practical knowledge in anatomy and physiology should be considered in educating students as well as reeducation of practitioners.

Vinpocetine inhibits the proliferation and induces apoptosis in human colon cancer cells

  • Vinpocetine induces anti-inflammatory effects in various inflammatory diseases via the inhibition of phosphodiesterase type-1-independent nuclear factor-κB signaling pathway and the release of inflammatory cytokines. In this study, we investigated the effect of vinpocetine on the proliferation of colon cancer cells and its underlying molecular mechanisms. Our data showed that vinpocetine inhibits the viability and proliferation of colon cancer cells. Vinpocetine treatment induced cell death in HCT116 cells, which the percentages of sub-G1 phase were significantly increased, and the apoptosis-related genes were regulated after HCT116 cells were treated with vinpocetine. In sum, our findings indicated that vinpocetine could be a therapeutically useful candidate in the treatment of colon cancer.

$H_2$ Receptor-Mediated Relaxation of Circular Smooth Muscle in Human Gastric Corpus: the Role of Nitric Oxide (NO)

  • This study was designed to examine the effects of histamine on gastric motility and its specific receptor in the circular smooth muscle of the human gastric corpus. Histamine mainly produced tonic relaxation in a concentration-dependent and reversible manner, although histamine enhanced contractility in a minor portion of tissues tested. Histamine-induced tonic relaxation was nerve-insensitive because pretreatment with nerve blockers cocktail (NBC) did not inhibit relaxation. Additionally, $K^+$ channel blockers, such as tetraethylammonium (TEA), apamin (APA), and glibenclamide (Glib), had no effect. However, $N^G$-nitro-L-arginine methyl ester (L-NAME) and 1H-(1,2,4)oxadiazolo (4,3-A) quinoxalin-1-one (ODQ), an inhibitor of soluble guanylate cyclase (sGC), did inhibit histamine-induced tonic relaxation. In particular, histamine-induced tonic relaxation was converted to tonic contraction by pretreatment with L-NAME. Ranitidine, the $H_2$ receptor blocker, inhibited histamine-induced tonic relaxation. These findings suggest that histamine produced relaxation in circular smooth muscle of human gastric smooth muscle through $H_2$ receptor and NO/sGC pathways.

Plasma Gastrin Concentration after a Carbohydrate Meal and a Protein Meal in Normal Human Subjects (식후 정상 한국인의 혈장 gastrin 농도)

  • This study was conducted to investigate the effect of the ingestion of rice and hamburger meals upon the plasma gastrin concentration in normal human subjects in Korea. Eight normal human subjects including male and female with the mean age of 28 years (range: 20-40 years) were studied. After an overnight(about 15 hrs) fast each subject ingested a rice meal and a hamburger meal on different days. The rice meal consisted of 250 g boiled rice, 50 g vegetables and 200 ml barley tea, corresponding to 6.8 g protein, 0.5 g fat and 81g carbohydrate and the hamburger meal consisted of 200 g hamburger, 50 g vegetables ana 200 ml milk, corresponding to 43 g protein, 43 g fat and 9 g carbohydrate. The venous blood samples were drawn before and after the ingestion of the test meal at the following times: -30, 0, 15, 30, 45, 60, 90, and 120 min, for measurement of gastrin by radioimmunoassay. The following results were obtained : 1) Plasma gastrin concentration in response to the ingestion of the rice or the hamburger meal increased significantly compared with the concentration in fasting state. 2) The increase of the plasma gastrin concentration after the hamburger meal was significantly higher than that after the rice meal. 3) There was a significant linear correlation between the postprandial peak plasma gastrin concentration after the rice meal and the concentration after the hamburger meal in each subject. It is inferred from the above results that a carbohydrate meal as well as a protein meal has a stimulatory effect on gastrin release in normal human subjects.

Taxifolin Glycoside Blocks Human ether-a-go-go Related Gene $K^+$ Channels

  • Taxifolin glycoside is a new drug candidate for the treatment of atopic dermatitis (AD). Many drugs cause side effects such as long QT syndrome by blocking the human ether-a-go-go related gene (hERG) $K^+$ channels. To determine whether taxifolin glycoside would block hERG $K^+$ channels, we recorded hERG $K^+$ currents using a whole-cell patch clamp technique. We found that taxifolin glycoside directly blocked hERG $K^+$ current in a concentration-dependent manner ($EC_{50}=9.6{\pm}0.7{\mu}M$). The activation curve of hERG $K^+$ channels was negatively shifted by taxifolin glycoside. In addition, taxifolin glycoside accelerated the activation time constant and reduced the onset of the inactivation time constant. These results suggest that taxifolin glycoside blocks hERG $K^+$ channels that function by facilitating activation and inactivation process.

The anti-microbial peptide SR-0379 stimulates human endothelial progenitor cell-mediated repair of peripheral artery diseases

  • Ischemia is a serious disease, characterized by an inadequate blood supply to an organ or part of the body. In the present study, we evaluated the effects of the anti-microbial peptide SR-0379 on the stem cell-mediated therapy of ischemic diseases. The migratory and tube-forming abilities of human endothelial progenitor cells (EPCs) were enhanced by treatment with SR-0379 in vitro. Intramuscular administration of SR-0379 into a murine ischemic hindlimb significantly enhanced blood perfusion, decreased tissue necrosis, and increased the number of blood vessels in the ischemic muscle. Moreover, co-administration of SR-0379 with EPCs stimulated blood perfusion in an ischemic hindlimb more than intramuscular injection with either SR-0379 or EPCs alone. This enhanced blood perfusion was accompanied by a significant increase in the number of CD31- and ${\alpha}$-SMA-positive blood vessels in ischemic hindlimb. These results suggest that SR-0379 is a potential drug candidate for potentiating EPC-mediated therapy of ischemic diseases.

Plasma Gastrin Concentraion after Ingestion of Sesame Gruel in Normal Human Subjects (깨죽이 정상 성인의 혈장 Gastrin 농도에 미치는 영향)

  • This study was conducted to investigate the effect of ingestion of sesame (Sesamum indicum) gruel as a nourishing meal upon the plasma gastrin concentration in normal Korean. Sixteen normal persons with no history of gastrointestinal diseases, including male and female were studied. After an overnight(about 15 hrs) fast, eight persons(mean age: 26.6, range: $20{\sim}40$ years) of them ingested a 350 ml sesame gruel corresponding to 12 g protein, 13 g fat and 99 g carbohydrate, and the remaining 8 subjects(mean age: 21.3, range: $20{\sim}24$ years) ingested a 350 ml glutinous rice gruel(control meal) corresponding to 8 g protein, 1 g fat and 115 g carbohydrate. The venous blood samples were drawn before and after the ingestion of the test meal for the measurement of gastrin by means of radioimmunoassay. 1) Plasma gastrin concentration in response to the ingestion of sesame gruel or glutinous rice gruel increased significantly compared with the concentration in fasting state. 2) Mean increment or percent increment in postprandial plasma gastrin concentration after the ingestion of sesame gruel was not significantly different from that after the control meal, i.e. the glutinous rice gruel. It is inferred from the above results that the ingestion of sesame contained in sesame gruel may have no significant influence on gastrin release in normal human subjects.
