A Study on the Antimicrobial Effect of Glyceryl Caprylate in Cosmetics (Glyceryl Caprylate의 화장품에서의 항균력에 관한 연구)

  • The antimicrobial properties of medium-chain ($C_{8-12}$) free fatty acids and their 1-monoglyceride derivatives against a wide range of microorganisms we well known. However, previous studies have been mainly focused on the antimicrobial activity against pathogenic bacteria and viruses causing diseases in human or domestic animals' infection. But, there have been few reports describing comprehensive surveys of antimicrobial effects against microorganisms in cosmetics. For a start of this study, we evaluated and compared the preservative activities of $C_8$ (glyceryl caprylate) and $C_{12}$(glyceryl laurate) 1-monoglyceride in cosmetic formulations. From the result, we found that both of them have very excellent preservative activity against bacteria, but less against fungi. And $C_8$ 1-monoglyceride was a little bit more effective than $C_{12}$ 1-monoglyceride. According to the test results to evaluate each antimicrobial activity of glyceryl caprylate towards 5 kinds of microorganisms used in preservation efficacy test in cosmetics, gram-positive bacteria S. aureus and yeast C. albicans were sensitive and mold A. niger was most tolerant to glyceryl caprylate. Therefore, we tried to improve the antimicrobial activity of glyceryl caprylate agianst mold such as A. niger so that we could make it used as a preservative for cosmetic products. As a result, we confirmed that the antimicrobial activity of glyceryl caprylate is much improved under acidic conditions in formulation. In addition, we found optimal combinations of glyceryl caprylate with other antimicrobial agents. Among tested 7 antimicrobial agent, methyparaben showed the highest preservative activity in combination with gglyceryl caprylate.

Impact of Microbiota on Gastrointestinal Cancer and Anticancer Therapy (미생물 균총이 위장관암과 항암제에 미치는 영향)

  • Human microbiota is a community of microorganisms, including bacteria, fungi, and viruses, that inhabit various locations of the body, such as the gut, oral, and skin. Along with the development of metabolomic analysis and next-generation sequencing techniques for 16S ribosomal RNA, it has become possible to analyze the population for subtypes of microbiota, and with these techniques, it has been demonstrated that bacterial microbiota are involved in the metabolic and immunological processes of the hosts. While specific bacteria of microbiota, called commensal bacteria, positively affect hosts by producing essential nutrients and protecting hosts against other pathogenic microorganisms, dysbiosis, an abnormal microbiota composition, disrupts homeostasis and thereby has a detrimental effect on the development and progression of various types of diseases. Recently, several studies have reported that oral and gut bacteria of microbiota are involved in the carcinogenesis of gastrointestinal tumors and the therapeutic effects of anticancer therapy, such as radiation, chemotherapy, targeted therapy, and immunotherapy. Studying the complex relationships (bacterial microbiota-cancer-immunity) and microbiota-related carcinogenic mechanisms can provide important clues for understanding cancer and developing new cancer treatments. This review provides a summary of current studies focused on how bacterial microbiota affect gastrointestinal cancer and anticancer therapy and discusses compelling possibilities for using microbiota as a combinatorial therapy to improve the therapeutic effects of existing anticancer treatments.

Establishment of Food Processing Methods for Larvae of Allomyrina dichotoma, Korean Horn Beetle (식약용으로의 장수풍뎅이 유충 전처리 방법에 대한 연구)

  • With the objective of developing a functional food source, we established optimal processing conditions for the larvae of Allomyrina dichotoma, which have been used in traditional medicine to treat hepatic disorders in Korea. Without suitable processing, the larvae are difficult to consume as a food because of their disgusting taste and smell; moreover, in this form they might be a potential microbial hazard. In this study, we investigated the effect of feeding material, sterilization, and powdering after freeze-drying on the food quality of the larvae of A. dichotoma and on cytotoxicity against Raw 264.7 cells. Three to five days feeding with the sawdust from discarded oak-trees is sufficient for the breeding process. The sawdust was sterilized by vapor for five minutes. Sterilization of the larvae at a high temperature ($115^{\circ}C$ for 5 min, 0.9 $kgf/cm^2$) is necessary to eliminate pathogenic bacteria and fungi. The results of the cytotoxicity assay showed no toxicity in the prepared extract from larvae of A. dichotoma. In addition, to prepare the larvae for human consumption, various feeds were used and the smell, color, and taste were evaluated. Our results suggested that larvae of A. dichotoma could be developed as food source when a suitable processing method is established.

Antimicrobial and Hemolytic Activity of Oriental Medicinal Herbs (한약재의 항균 활성 및 인간 적혈구 용혈 활성)

  • To develop the safe and natural antimicrobial agents, the 68 ethanol extracts from the 61 different kinds of oriental herbal medicine were prepared and their antimicrobial activities were evaluated. The herbal medicine used were from China (46 kinds), South Korea (14 kinds), North Korea (5 kinds) and Vietnam (3 kinds), respectively, and the root (27 species) was popular part in this study. The average water content and extraction ratio for ethanol were 7.10% and 6.75%, respectively. Determination of antimicrobial activity by disc-diffusion assay at 0.5 mg/disc concentration showed that the extract of Angelica tenuissima Nakai (china), Illicium verum, Junci medulla, Rhus javanica L., Salvia miltiorrhiza Bunge and Syzygium aromaticum has strong antimicrobial activities against different food spoilage and pathogenic bacteria and fungi. Determination of MIC and MBC/MFC further showed that the extract of Syzygium aromaticum has MIC of 1.25 mg/mL and MBC/MFC of 1.25~5.00 mg/mL against Listeria monocytogenes, Bacillus subtilis, Staphylococcus epidermidis, Staphylococcus aureus, Salmonella typhimurium, Proteus vulgaris, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Saccharomyces cerevisiae. And, the extract of Junci medulla, Rhus javanica L. and Salvia miltiorrhiza Bunge showed strong antibacterial activities with MIC of 0.08~0.63 mg/mL and MBC/MFC of 0.08~2.50 mg/mL against the tested bacteria except E. coli and P. aeruginosa. In a while, the results of hemolytic activity of 68 different herbal extracts against human red blood cells showed that the extract of Angelica tenuissima Nakai has hemolytic activity at 0.5 mg/mL concentration. Therefore, Illicium verum, Junci medulla, Rhus javanica L., Salvia miltiorrhiza Bunge and Syzygium aromaticum were finally selected for natural antimicrobial resources. Further research on active substances and the mode of action of the selected herbal medicine is necessary.
