Bacteriophage Usage for Bacterial Disease Management and Diagnosis in Plants

  • In nature, plants are always under the threat of pests and diseases. Pathogenic bacteria are one of the major pathogen types to cause diseases in diverse plants, resulting in negative effects on plant growth and crop yield. Chemical bactericides and antibiotics have been used as major approaches for controlling bacterial plant diseases in the field or greenhouse. However, the appearance of resistant bacteria to common antibiotics and bactericides as well as their potential negative effects on environment and human health demands bacteriologists to develop alternative control agents. Bacteriophages, the viruses that can infect and kill only target bacteria very specifically, have been demonstrated as potential agents, which may have no negative effects on environment and human health. Many bacteriophages have been isolated against diverse plant-pathogenic bacteria, and many studies have shown to efficiently manage the disease development in both controlled and open conditions such as greenhouse and field. Moreover, the specificity of bacteriophages to certain bacterial species has been applied to develop detection tools for the diagnosis of plant-pathogenic bacteria. In this paper, we summarize the promising results from greenhouse or field experiments with bacteriophages to manage diseases caused by plant-pathogenic bacteria. In addition, we summarize the usage of bacteriophages for the specific detection of plant-pathogenic bacteria.

Antibacterial potential of Clerodendrum inerme, crude extracts against some human pathogenic bacteria

  • This communication emphasized upon the sensitivity of the crude extracts of clerodendrum inerme against some of the human pathogenic bacteria. Five plant extracts (Petrol, Benzene, Methanol, Ethly acetate and Aqueous) under six different concentrations(500 ${\mu}g/ml$, 1 mg/ml, 2 mg/ml, 5 mg/ml, 10 mg/ml and 15 mg/ml) were tested by disk diffusion method. Methanol, Ethyl acetate and Aqueous extracts of the plant showed significant inhibition against fifteen of the eighteen bacteria tested. No earlier report on antibacterial activity of this taxon could be found in literature.

  • Rapid and accurate detection of pathogenic bacteria is crucial for various applications, including public health and food safety. However, existing bacteria detection techniques have several drawbacks as they are inconvenient and require time-consuming procedures and complex machinery. Recently, the precision and versatility of CRISPR/Cas system has been leveraged to design biosensors that offer a more efficient and accurate approach to bacterial detection compared to the existing techniques. Significant research has been focused on developing biosensors based on the CRISPR/Cas system which has shown promise in efficiently detecting pathogenic bacteria or virus. In this review, we present a biosensor based on the CRISPR/Cas system that has been specifically developed to overcome these limitations and detect different pathogenic bacteria effectively including Vibrio parahaemolyticus, Salmonella, E. coli O157:H7, and Listeria monocytogenes. This biosensor takes advantage of the CRISPR/Cas system's precision and versatility for more efficiently accurately detecting bacteria compared to the previous techniques. The biosensor has potential to enhance public health and ensure food safety as the biosensor's design can revolutionize method of detecting pathogenic bacteria. It provides a rapid and reliable method for identifying harmful bacteria and it can aid in early intervention and preventive measures, mitigating the risk of bacterial outbreaks and their associated consequences. Further research and development in this area will lead to development of even more advanced biosensors capable of detecting an even broader range of bacterial pathogens, thereby significantly benefiting various industries and helping in safeguard human health

Evaluation of Microbiological Contamination of Water Purifiers at Two Universities in Chungcheong Region

  • The purpose of this study is to investigate microbial contamination in water purifiers from two universities (A and B) in Chungcheong region and to evaluate about the harmfulness of the isolated bacteria to the human. The degree of microbiological contamination of six water purifiers at university A was investigated three times from July 2018 to September 2019, and nine water purifiers at university B were investigated in 2023. The isolated bacteria were biochemically identified using an API kit and Vitek-2 system, and then the bacteria were identified to the species level using MALDI-TOF MS. In addition, the possibility of human infection of the isolated bacteria was evaluated through a literature search. In July 2018 and September 2019, the number of bacteria isolated inside the faucet was below the acceptable standard for hot water, but exceed for cold water in all water purifiers. In January and September 2019, bacteria exceeding the acceptable standards were isolated nine times from the cold water of six water purifies (a total of 12 water purifiers). Bacteria identified by MALDI-TOF MS included anaerobic bacteria (Clostridium novyi, Clostridium themopalmarium etc.), Gram-positive bacilli (Microbacterium testaceum, Arthrobacter woluwensis etc.), and Gramnegative bacilli (Acinetobacter nosocomialis, Comamonas kerstersii etc.), which are difficult identify by biochemical methods. In conclusion, bacteria exceeding the acceptable standard were isolated from the cold water of most of the water purifiers. Most of the isolated bacteria were low-pathogenic bacteria from natural environment, but opportunistic bacteria that can cause infection in humans were also isolated from some water purifiers.

프로바이오틱스, 프리바이오틱스 및 신바이오틱스 연구동향 (Trends in studies on probiotics, prebiotics, and synbiotics)

  • Probiotics are very closely related to gut microbiome and recognized as beneficial microorganisms for our health. They have various biological effects such as inhibition of pathogenic bacteria, activation of beneficial bacteria, prevention of diarrhea and constipation, enhanced immune activity etc. Prebiotics, non-digestible carbohydrates such as galactooligosaccharide and fructooligosaccharide, are utilized by beneficial gut bacteria such as bifidobacteria and lactobacilli, resulting in production of short chain fatty acids which inhibit pathogenic bacteria in the gut and function for human health. Synbiotics are introduced for synergistic effects when probiotics are combined with prebiotics and now commercially available. At the moment many functional ingredients are developed and commercialized. Probiotics, prebiotics, and synbiotics might be hot items in the functional food market and the values will increase according to the results of human gut microbiome researches. To meet the situation, systematic and scientific studies as well as marketing effects should be accompanied.

인진쑥 추출물의 병원성 세균에 대한 생체내 증식 억제 효과 (Inhibitory Effects of Artemisia capillaris Extract on the Pathogenic Bacteria in Mice)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate the inhibitory effects of orally administrated Artemisia capillaris extracts on the pathogenic bacteria in 200 ICR mice. The experimental groups showed inhibitory effects on the bacteria in $1{\sim}3$ days after inoculation. After 21 days of inoculation, no viable bacterial cells appeared in the feces of both experimental groups while they did appear in the control group. The results of these studies indicate Artemisia capillaris extract exhibited excellent antimicrobial and inhibitory effects on the food poisoning pathogenic bacteria; S. enteritidis, E. coli O157:H7, L. monocytogenes and S. aureus.

Synergistic Antibacterial Activity of an Active Compound Derived from Sedum takesimense against Methicillin-Resistant Staphylococcus aureus and Its Clinical Isolates

  • There are a growing number of reports of hospital-acquired infections caused by pathogenic bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA). Many plant products are now being used as a natural means of exploring antimicrobial agents against different types of human pathogenic bacteria. In this research, we sought to isolate and identify an active molecule from Sedum takesimense that has possible antibacterial activity against various clinical isolates of MRSA. NMR analysis revealed that the structure of the HPLC-purified compound was 1,2,4,6-tetra-O-galloyl-glucose. The minimum inhibitory concentration (MIC) of different extract fractions against numerous pathogenic bacteria was determined, and the actively purified compound has potent antibacterial activity against multidrug-resistant pathogenic bacteria, i.e., MRSA and its clinical isolates. In addition, the combination of the active compound and β-lactam antibiotics (e.g., oxacillin) demonstrated synergistic action against MRSA, with a fractional inhibitory concentration (FIC) index of 0.281. The current research revealed an alternative approach to combating pathogenesis caused by multi-drug resistant bacteria using plant materials. Furthermore, using a combination approach in which the active plant-derived compound is combined with antibiotics has proved to be a successful way of destroying pathogens synergistically.

Antimicrobial Effect of Furaneol Against Human Pathogenic Bacteria and Fungi

  • Furaneol, a key aroma compound found in strawberry, pineapple, and processed foodstuffs, has been known to possess various biological activities on animal models. In this study, the antimicrobial effects of furaneol against human pathogenic microorganisms were investigated. The results indicated that furaneol displayed a broad spectrum of antimicrobial activities against Gram-positive and Gram-negative bacteria and fungi without hemolytic activity on human erythrocyte cells. To confirm the antifungal activity of furaneol, we examined the accumulation of intracellular trehalose as a stress response marker on toxic agents and its effect on dimorphic transition of Candida albicans. The results demonstrated that furaneol induced significant accumulation of intracellular trehalose and exerted its antifungal effect by disrupting serum-induced mycelial forms. These results suggest that furaneol could be a therapeutic agent having a broad spectrum of antimicrobial activity on human pathogenic microorganisms.

인간 피부 상재균과 병원성 세균에 대한 Paenibacillus elgii DS381과 그 항균물질의 항균활성 (Antimicrobial activity by Paenibacillus elgii DS381 and its antimicrobial substances against microbial residents on human skin and pathogenic bacteria)

  • 이 연구는 분리된 토양 세균에 의해 생성된 항균물질의 효과를 평가하기 위해 수행되었다. 2000여개의 세균 분리주 중 Paenibacillus elgii DS381이 여러 인간 피부 상재균과 병원성 세균에 대해 높은 항균활성을 나타내었다. DS381 균주는 agar well diffusion test에서 모든 대상 세균과 효모에 대해 15.3~26.0 mm 직경의 저해대를 형성하였다. DS381이 생성한 항균 펩티드는 모든 대상 미생물에 낮은 최소저해농도 (0.039-5.000 mg/ml)를 나타내었다. DS381 균주는 lipopeptide 같은 생물계면활성제 생산을 나타내었는데, 배양 상등액의 표면장력을 60.0에서 40.3 mN/m으로 낮추었다. DS381은 또한 $1.56{\pm}0.13U/ml$의 chitinase 활성도 나타내었다. 이 결과들은 P. elgii DS381이 일부 중요한 인간 피부 상재균과 병원성 세균에 대한 효율적인 생물제어제로 사용될 수 있음을 가리킨다.

  • To assess the effect of chitosan complex with metal ion on various pathogenic bacteria, the antibacterial activities were investigated. Arsenic, bismuth, calcium, iodine, iron, mercury, platinum, and silver were used as a metal ion. Staphylococcus aureus. Streptococcus mutans, Helicobacter pylori, Propionibacterium acnes and human saliva were examined. It was demonstrated that metal ions associated chitosan showed aggregation activities on various pathogens.
