The Inhibitory Effect of Gooseberry on DNCB-induced Atopic Dermatitis in vivo and in vitro

  • Generally, berry fruits have various pharmacological activities such as anti-inflammation, anti-oxidation and anti-cancer effects. The effects of gooseberry, a berry fruits, on atopic dermatitis (AD) have not been widely examined. The aim of this present study is to investigate whether gooseberry modulates AD. We examined the pharmacological effects of gooseberry on 2, 4-dinitrochlorobenzene (DNCB)-induced AD symptoms in mice. To determine the anti-atopic mechanism of gooseberry, we investigated its effects on the production of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in PMA + ionophore -stimulated human mast cells (HMC-1). The results demonstrated that gooseberry attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, gooseberry suppressed the expression of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in stimulated HMC-1. These findings demonstrate that gooseberry is potential agent for treating AD and allergic inflammation.

  • Ginseng (the root of Panax ginseng C.A $M_{EYER}$, family Araliaceae) is frequently used as a crude substance in Asian countries as a traditional medicine. The major components of ginseng are ginsenosides, which have been reported to show various biological activities including antiinflammatory activity and antitumor effect. In addition, Sugiyama et al. reported that ginsenoside Rg3 suppresses histamine release from mast cells due to stimulation with compound 48/80 in vitro. However, the antiallergic effects of ginsenoside Rh2, which is metabolized by human intestinal bacteria to ginsenoside Rg3, have not been studied. (omitted)

TSLP 유전자의 다형성은 한국인 류마티스관절염 발생에 영향을 미치치 않는다 (Thymic Stromal Lymphopoietin (TSLP) Gene Polymorphisms are not Associated with Rheumatoid Arthritis in a Korean Population)

  • TSLP 유전자는 IL-7와 유사한 새로운 조혈성 사이토카인이다. 인간의 TSLP는 상피세포, 기질세포 및 비만세포에서 만들어진다. TSLP는 류마티스관절염 환자의 윤활성 활액에서 높은 발현을 나타낸다. 이전 연구에서 우리들은 사람의 TSLP유전자에서 4개의 유전자다형성 및 한 개의 변이를 발굴하였다. 이 연구에서는, 우리들이 발굴한 TSLP유전자의 유전자다형성의 유전자형 및 대립형질의 비율을 건강한 정상인과 류마티스관절염 환자에서 비교분석하였으며, 류마티스관절염 환자에 있어서 유전자형에 따른 RF 및 anti-CCP의 정도를 비교 분석하였다. 또한, 양쪽 그룹에서 이들 유전자다형성에 의한 일배체형 비율을 비교 분석하였다. 그 결과, 류마티스관절염 환자군과 건강한 정상인 군 사이에 있어서 유전자형, 대립형질 비율뿐만 아니라 일배체형 비율에 큰 차이를 보이지 않았다. 이 결과는 TSLP유전자의 유전자다형성은 류마티스관절염 감수성에 영향을 미치지 않음을 암시한다.

쏘가리의 위장관 내분비세포에 관한 면역조직화학적 연구 (An Immunohistochemical Study on the Endocrine Cells in the Gastrointestinal Tract of the Mandarin Fish (Siniperca scherzeri))

  • 쏘가리 (Siniperca scherzeri Steindachner)의 위장관에 존재하는 내분비세포의 부위별 분포 및 출현빈도를 포유류의 peptide에 대한 7종류의 항혈청을 사용하여 면역조직화학적 방법으로 관찰하였다. 쏘가리의 위장관은 근위부에서부터 원위부까지 위, 소장 및 대상으로 3 등분 하였으며, 다양한 종류의 항혈청에 면역반응성을 나타내는 면역반응세포들이 상피세포 사이와 위샘 또는 장샘에서 관찰되었다. 상피세포 사이에서는 대부분의 면역반응세포들은 장 내강까지 신장되어 있는 긴 세포질 돌기를 함유한 방추형의 개방형 세포 (open type cell)로 관찰되었으며, 세포질 돌기 없이 원형 또는 타원형의 형태를 나타내는 폐쇄형 세포 (close type cell)들이 위 부위에서 소수 관찰되었다. 본 실험에서는 serotonin, somatostatin, pastrin, cholecystokinin (CCK)-8 및 human pancreatic polypeptide (HPP) 면역반응세포들이 관찰되었으나, insulin 및 glucagon 면역반응세포들은 관찰되지 않았다. Serotonin 및 somatostatin 면역반응세포들은 위 부위에 국한되어 각각 중등도 및 다수의 출현빈도로 관찰되었다. 또한 gastrin 면역반응세포들은 위와 소장에서 출현하였으며, 각각 소수 및 중등도의 출현빈도를 나타내었고, CCK-8 면역반응세포들은 소장에 국한되어 중등도의 출현빈도를 나타내었다. 한편 HPP 면역반응세포들은 위와 소장에서 다수 관찰되었다. 이상에서 쏘가리 위장관 내분비세포들의 부위별 분포 및 출현 빈도는 다른 경골어류에 비해 특이한 양상을 나타내는 것으로 관찰되었다.

  • The last two decades witnessed significant advances in the efforts of immune-parasitologists to elucidate the nature and role of the host mucosal defence mechanisms against intestinal nematode parasites. Aided by recent advances in basic immunology and biotechnology with the concomitant development of well defined laboratory models of infection, immunoparasitologists have more precisely analyzed and defined the different immune effector mechanisms during the infection; resulting in great improvement in our current knowledge and understanding of protective immunity against gastrointestinal (GI) nematode parasites. Much of this current understanding comes from experimental studies in laboratory rodents, which have been used as models of livestock and human GI nematode infections. These rodent studies, which have concentrated on Heligmosomoides polygyrus, Nippostrongylus brasiliensis, Strongyloides ratti/5. venezuelensis. Trichinella spiralis and trichuris muris infections in mice and rats, have helped in defining the types of T cell responses that regulate effector mechanisms and the effector mechanisms responsible for worm expulsion. In addition, these studies bear indications that traditionally accepted mechanisms of resistance such as eosinophilia and IgE responses may not play as important roles in protection as were previously conceived. In this review, we shall, from these rodent studies, attempt an overview of the mucosal and other effector responses against intestinal nematode parasites beginning with the indices of immune protection as a model of the protective immune responses that may occur in animals and man.

호장근추출물이 피부질환 및 피부미용에 미치는 영향 (The Effects of Polygonum Cuspidatum on the Skin Functions)

  • Objectives : This study was carried out to investigate the effects of Polygonum cuspidatum extract on several skin functions including inflammation and wrinkle formation. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe $TNF-{\alpha}$, IL-8 by ELISA Results : Polygonum Cuspidatum extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Polygonum Cuspidatum extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Polygonum Cuspidatum extract could be applicable for improvement of several skin functions.

Anti-allergic Effect of Aqueous Extract of Stachys riederi var.japonica Miq.in vivo and in vitro

  • The effect of aqueous extract of Stachys riederi var. japonica Miq. (Labiatae) (SRAE) on the immediate-type allergic reactions was investigated. SRAE was found to exhibit a inhibitory activity on the compound 48/80-induced systemic anaphylaxis in mice. SRAE inhibited the plasma histamine release induced by compound 48/80 in mice. In addition, SRAE also inhibited the passive cutaneous anaphylaxis reaction induced by IgE/anti-IgE in mice. The effect of SRAE on the histamine release from rat peritoneal mast cells (RPMC) was studied. SRAE inhibited the histamine release induced by compound 48/80 in RPMC. To clarify the mechanism of these inhibiting reactions, we investigated the effects of SRAE on cyclic AMP (cAMP). The level of cAMP in human leukemia cell line, HMC-1, when SRAE (1 mg/ml) was added, significantly increased compared with that of basal cells. These results indicate that SRAE may be beneficial in the treatment of immediate-type allergic reaction.

국내 실외공기의 곰팡이 분포 및 노출에 따른 염증영향평가 (Evaluation of Distribution and Inflammatory Effects of Airborne Fungus in Korea)

  • Objectives: For the risk management of airborne fungal diseases, our aim was to evaluate airborne fungi and study the toxicity associated with fungal allergic diseases using fungal species native to Korea. Methods: Fungi were isolated from outdoor air samples collected from Seoul, Incheon, Cheonan, Gwangju, Ulsan, Busan, and Jeju and tested for their cytotoxicity potential and their ability to induce proliferation and secretion of macrophage-derived chemokine (MDC) in human mast cells (HMC-1). Results: More than 18 species of fungi were collected from outdoor air in Korea over one year, and the strains were identified and systematically analyzed. The results showed that the Cladosporium (59%) and Alternaria (22%) strains are the most common in outdoor air. Three of the collected strains (Fusarium, Trichoderma, and Penicillium) showed mild toxicity in cells involved in allergic inflammation, and twelve induced cell proliferation in HMC-1 cells. More importantly, many strains (Edgeworthia, Trametes, Emmia, Irpex, Talaromyces, Penicillium, Periconia, Epicocum, Bipolaris) induced the MDC protein in activated HMC-1 cells. Conclusion: Nineteen percent of the tested strains caused cytotoxicity in mast cell lines, whereas, most of the non-toxic strains contributed to cell activity. Among the tested strains, more than 80% increased the expression of MDC protein, which contributes to the severity of atopic dermatitis, asthma, and rhinitis. It is, in fact, one of the markers for these conditions. Therefore, airborne fungus could be considered as an important marker for environmental risk management for allergic diseases in Korea.

Effect of Oral Probiotics (Bifidobacterium lactis AD011 and Lactobacillus acidophilus AD031) Administration on Ovalbumin-Induced Food Allergy Mouse Model

  • Recent study has demonstrated an increasing prevalence of food allergy in Korean children. Specific probiotic bacteria may promote potentially anti-allergenic processes through induction of Th1-type immunity and enhance the regulatory lymphocyte. This study investigated whether orally administrated probiotics could suppress allergic responses in an ovalbumin (OVA)-induced allergy mouse model. Thus, female C3H/HeJ mice were orally sensitized with OVA and cholera toxin for 4 weeks. Lactobacillus acidophilus AD031, Bifidobacterium lactis AD011, and L. acidophilus AD031 plus B. lactis AD011 were fed to mice from 2 weeks before the sensitization. The OVA-induced mice that were not treated with probiotics had significantly increased serum levels of OVA-specific IgE and IgG1, and OVA-specific IgA in feces. However, the mice treated with probiotics suppressed production of the OVA-specific IgE, IgG1, and IgA. The level of IL-4 was significantly lower, and the levels of INF-$\gamma$ and IL-10 were significantly higher in the mice treated with probiotics than that in the non-treated mice. The groups treated with probiotics had decreased levels of degranulated mast cells, eosinophil granules, and tail scabs. These results indicate that L. acidophilus AD031 and B. lactis AD011 might be useful for the prevention of allergy.
