Purpose: This study was conducted to examine the factors associated with HPV (Human Papillomavirus) related stigma, shame and intent to have HPV test among adult women. Methods: Data were collected from December 1, 2009 to January 31, 2010, and participants were 324 women who visited an obstetric gynecologic clinic. They anticipated testing positive for HPV. Then HPV related stigma, shame, intent to have HPV testing and HPV knowledge were measured. Descriptive statistics, Mann-Whitney U test, Kruskal Wallis test and multivariate adjusted logistic regression were used for data analysis. Results: The levels of stigma and shame were higher than average. Intent to have HPV test was high and HPV knowledge was low. Women who answered that HPV is not sexually transmitted had lower HPV stigma than did women who answered they didn't know (OR=0.20, 95%CI 0.06-0.68). Women with lower stigma showed lower intent to have HPV test than women with higher stigma (OR=0.46, 95%CI 0.26-0.82). Conclusion: Basic HPV information should be fully understood for women especially prior to HPV test. Normalizing HPV stigma is necessary for women who perceive HPV as sexually transmitted and women intending to have HPV test.
Purpose: The goal of this study was to examine knowledge level of human papillomavirus (HPV), cervical cancer, and vaccination status among Korean mothers' with a daughter in high school. Methods: A cross-sectional descriptive study was conducted with a convenience sample of 234 Korean mothers. Results: The mean score of HPV knowledge level was 4.21 out of 20, and cervical cancer knowledge was 3.88 out of 7. There were low levels of daughters' who received HPV vaccination (3.8%) and high levels of mothers' intention to get their daughters' vaccinated with HPV (85.0%). The major barrier to HPV vaccination was the concern for side effect from the vaccination. The most effective recommendation for HPV vaccination came from health care providers. Conclusion: HPV educational programs targeting mothers of daughters in high school is needed. Future studies need to determine actual HPV vaccination rates and factors related to high uptake using a larger sample from various socioeconomic background.
Purpose: Human papillomavirus (HPV) vaccination schedule is recommended by Strategic Advisory Group of Experts on Immunization of World Health Organization to achieve high efficacy and immunogenicity. However, the patients often cannot keep up their schedule for several reasons. Methods: Monthly numbers of visitors for HPV vaccination between 2019 and 2021 were analyzed to measure the impact of coronavirus disease-19 (COVID-19) outbreak which was the main cause of this delay. Results: In February 2020, the number of foreign patients vaccinated with HPV was dropped suddenly due to COVID-19. Prior to this impact, the average patients per month for HPV vaccination was 160 (95% confidence interval [CI]: 143-176). It was then 30 (95% CI: 20-40). However recent trends show a gradual recovery. Conclusion: If a woman is pregnant after starting the HPV vaccine series, the reminder doses should be delayed until she is no longer pregnant. If this series is interrupted for any length of time, it can be resumed without restarting the series. HPV vaccine series need to be administered with a minimum interval of 14 days before or after administration of COVID-19 vaccines.
Vulva cancer is rare among all gynecological cancer worldwide, including Thailand, and mainly affects older women. Persistent high risk type infection of human papillomavirus (HPV) is the one important factor for developing cancer. In this study, we focused on HPV DNA investigation and type-specific distribution of HPV in 25 formalin-fixed paraffin-embedded (FFPE) samples collected from Thai women with vulva cancer histologically confirmed by the National Cancer Institute, Thailand, during 2003-2011. HPV DNA detection and genotyping were undertaken with polymerase-chain reaction and enzyme-immunoassay using GP5+/bio6+ consensus specific primers and digoxigenin-labeled specific oligoprobes, respectively. Human ${\beta}$-globin genes was used as the internal control. Our results showed that 44% (11/25) of all vulva cancer samples were HPV-positive. All of them are high risk HPV type infection, detected as single (63.64%, 7/11) and/or double infections (4/11, 36.36%). HPV 16 was the most common type identified in vulva cancer, followed by HPV 35, 33, 18 and 58. In conclusion, this study presented that HPV-16 is observed at the highest frequency in this cancer, similar to cervical cancer, with HPV 18 being less frequent. Although the sample size was small and could not represent overall incidence and prevalence in Thai women, these preliminary data for vulva cancer are of interest since they reinforce the necessity for HPV screening or vaccination in Thailand.
Human papillomavirus (HPV) is the primary etiologic agent of cervical cancer. Consideration of safety and non human leukocyte antigen restriction, protein vaccine has become the most likely form of HPV therapeutic vaccine, although none have so far been reported as effective. Since tumor cells consistently express the two proteins E6 and E7, most therapeutic vaccines target one or both of them. In this study, we fabricated DC vaccines by transducing replication-defective recombinant adenoviruses expressing E6/E7 fusion gene of HPV-16, to investigate the lethal effects of specific cytotoxic T lymphocytes (CTL) against CaSki cells in vitro. Mouse immature dendritic cells (DC) were generated from bone marrow, and transfected with pAd-E6/E7 to prepare a DC vaccine and to induce specific CTL. The surface expression of CD40, CD68, MHC II and CD11c was assessed by flow cytometry (FCM), and the lethal effects of CTL against CaSki cells were determined by DAPI, FCM and CCK-8 methods. Immature mouse DC was successfully transfected by pAd-E6/E7 in vitro, and the transfecting efficiency was 40%-50%. A DC vaccine was successfully prepared and was used to induce specific CTL. Experimental results showed that the percentage of apoptosis and killing rate of CaSki cells were significantly increased by coculturing with the specific CTL (p <0.05). These results illustrated that a DC vaccine modified by HPV-16 E6/E7 gene can induce apoptosis of CaSki cells by inducing CTL, which may be used as a new strategy for biological treatment of cervical cancer.
Cervical cancer is one of the most prevalent cancers developed in women worldwide, and human papillomavirus (HPV) type 16 is the most common agent linked to human cerivical carcinoma. Viral oncogenes E6 and E7 are selectively ratined and expressed in carcinoma cells infected with human papillomavirus type 16 and cooperated with each other in immortalization and transformation of primary keratinocytes. Because of HPV oncogenesis mechanism was not completely solved, the more studies be required thoroughly. In the present study, to investigate the telomere independent role of telomerase in HPV oncogenesis, we constructed the E6 mutant, E7, E6/E7 and hTERT over-expressed stable cells with a telomerase negative cell line, SW13. Expressions of Inserted genes were measured by RT-PCR. E6, E7 and hTERT genes were well expressed in each cell lines comparing with the control groups. By analyzing the cell morphology under the microscope, hTERT clone size was a more smaller than the mock control but oncogene expressed clones were slightly lengthened the marginal region. In addition, hTERT cells has also, a tendency of brief dividing time compared to the mock control. To determine whether telomerase activity associated with a HPV oncogenesis by oncoprotein expression, we performed the PCR based TRAP assay and Northern blot analysis. In TRAP assay data, telomerase activities in hTERT and oncogene clones were more increased than the mock control. In addition, SW13/ E6/E7 cells appeared a extremely increased activity than any other clones. Induced TERT mRNA by E6/E7 wasn't, however, detected in Northern blotting. In conclusion, these findings suggest that telomerase activity closely associated the HPV oncogenesis and E6/E7 co-expression is a most important factor of telomerase activity.
Background: Persistent human papillomavirus (HPV) infection, especially with high-risk types such as HPV16 and HPV18, has been identified as the primary cause of cervical cancer. E6 and E7 are the major onco-proteins of high-risk HPVs, which are consistently expressed in HPV infected tissues but absent in normal tissues and represent ideal therapeutic targets for immunotherapy of cervical cancer. Materials and Methods: In this study, the optimized fusion gene HPV18 E6E7 (HPV18 ofE6E7) was constructed according to genetic codon usage for human genes. At the same time, for safety future clinical application, a mutant of HPV18 ofE6E7 fusion gene was generated by site-directed mutagenesis at L52G for the E6 protein and C98G for the E7 protein. Results: HPV18-E6E7 mutant (HPV18 ofmE6E7) constructed in this work not only lost the transformation capability for NIH 3T3 cells and tumorigenicity in BALB/c nude mice, but also maintained very good stability and antigenicity. Conclusion: These results suggest that the mutant should undergo further study for application as a safe antigenspecific therapeutic vaccine for HPV18-associated tumors.

High risk-human papillomavirus (HR-HPV) is known to be a major cause of cervical cancer, and coinfection of sexually transmitted pathogen (STP) has been reported to cause persistent HPV infection. However, the relationship between HPV and STP coinfection remains unclear. The purpose of this study was to analyze the coinfection rate with STP in high-risk human papillomavirus infected women in Busan and to collect basic data for the prevention of cervical lesions. This study was carried out in 355 women who had concurrent HPV and STP screening at Busan local hospital between January 2016 and December 2017. HPV and STP coinfection was found in 187 (52.7%) out of 355 cases. HR-HPV and STP coinfection was 82.9% higher than LR-HPV and STP coinfections 17.1%. In HR-HPV infection, Ureaplasma species was the most common pathogen (47.1%), followed by C. trachomatis (21.9%) and Mycoplasma species (12.3%). In the analysis of HR-HPV genotype according to STP, HPV 16 (12.0%) was the most frequent, followed by HPV 58 (11.6%), HPV 39 (11.1%) and HPV 52 (10.2%), but HPV 18 showed a low coinfection rate of 1.3%. According to the results of age, HR-HPV and STP coinfection rate was the highest at 41.9% among women aged 18 to 29. HR-HPV and Ureaplasma species showed the highest coinfection rates at all ages, followed by C. trachomatis and Mycoplasma species. Further studies with more samples will be needed to determine if the coinfection of HR-HPV and STPs is involved in the development of cervical tumors through histologic changes.
본 연구에서는 임신진단등의 간편 현장진단(point of care test, POCT)에 주로 사용되고 있는 멤브레인 측면흐름 분석기법을 사용하여 인유두종 바이러스(Human papillomavirus, HPV)의 특정 서열을 검출할 수 있는 DNA array를 개발하였다. HPV type 6, 11, 16, 18, 31, 45에 특이적인 DNA 탐침들을 측면흐름 분석용 멤브레인 표면에 고정하고, biotin이 label된 MY09/11 primer를 사용하여 얻어진 HPV PCR 반응 결과물과 탐침 사이에 hybridization 반응을 유도하였다. 이후 streptavidin이 label된 colloidal gold가 교잡물의 biotin과 반응함으로써 DNA hybridization 결과를 육안으로 확인할 수 있었다. 본 연구를 통해 개발된 HPV DNA lateral flow membrane array는 기존의 HPV DNA chip 기법과 비교하여 경제적이고 편리하게 주요 HPV type을 확인할 수 있음을 보여주었다.
