KCl Mediates $K^+$ Channel-Activated Mitogen-Activated Protein Kinases Signaling in Wound Healing

  • Background Wound healing is an interaction of a complex signaling cascade of cellular events, including inflammation, proliferation, and maturation. $K^+$ channels modulate the mitogen-activated protein kinase (MAPK) signaling pathway. Here, we investigated whether $K^+$ channel-activated MAPK signaling directs collagen synthesis and angiogenesis in wound healing. Methods The human skin fibroblast HS27 cell line was used to examine cell viability and collagen synthesis after potassium chloride (KCl) treatment by Cell Counting Kit-8 (CCK-8) and western blotting. To investigate whether $K^+$ ion channels function upstream of MAPK signaling, thus affecting collagen synthesis and angiogenesis, we examined alteration of MAPK expression after treatment with KCl (channel inhibitor), NS1619 (channel activator), or kinase inhibitors. To research the effect of KCl on angiogenesis, angiogenesis-related proteins such as thrombospondin 1 (TSP1), anti-angiogenic factor, basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF), pro-angiogenic factor were assayed by western blot. Results The viability of HS27 cells was not affected by 25 mM KCl. Collagen synthesis increased dependent on time and concentration of KCl exposure. The phosphorylations of MAPK proteins such as extracellular-signal-regulated kinase (ERK) and p38 increased about 2.5-3 fold in the KCl treatment cells and were inhibited by treatment of NS1619. TSP1 expression increased by 100%, bFGF expression decreased by 40%, and there is no significant differences in the VEGF level by KCl treatment, TSP1 was inhibited by NS1619 or kinase inhibitors. Conclusions Our results suggest that KCl may function as a therapeutic agent for wound healing in the skin through MAPK signaling mediated by the $K^+$ ion channel.

Characterization for calmodulin binding activity of IQ motifs on the IQGAP3 (IQGAP3에 존재하는 IQ 부위의 칼모듈린 결합 특성)

  • IQ motif-containing GTPase-activating proteins (IQGAPs), which are well-known $Ca^{2+}$-independent calmodulin (CaM) binding proteins, are involved in various cellular functions such as cell proliferation, carcinogenesis and cell migration. The IQGAP3 similar to IQGAP1 has four repeated IQ motifs, which are crucial for CaM binding. It has been recently shown that all four IQ motifs of the IQGAP1 could bind to CaM, while not clear the binding of four IQ motifs of the IQGAP3. In this study, we examined the binding between CaM and each IQ motif of IQGAP3. As a result, we found that IQ2 and IQ3, but not IQ1 and IQ4, have a $Ca^{2+}$-independent CaM binding activity. We also found that IQ(3.5-4.4) on the IQGAP3 has $Ca^{2+}$-dependent CaM binding activity as similar with that of IQGAP1. This finding indicates that IQ motifs of the IQGAP3 plays a dynamic role via different interaction of IQ motifs with $Ca^{2+}$/CaM or apoCaM.

Towards Musical User Interface : The Emotional Effects of Music on Home Appliances Usability (음악적 사용자 인터페이스: 음악이 가전제품에 미치는 정서적 효과)

  • Previous studies of music, user interface, and human-computer interaction have used sounds which include musical structure rather than real music. This study investigated whether real music affects objective and perceived usability. Silence, sound, and music conditions were compared in experiment 1 (kimchi refrigerator) and 2 (remote controller for air conditioner). Participants' performances of reaction time and accuracy, and the degree of subjective satisfaction were analyzed. The results showed that main effects on task performances were not different significantly; however, perceived usability of music condition was better than sound condition, which was better than silence condition. It means that musical user interface improves perceived usability while not interfering task performance. This study provides a basis of emotional and aesthetic effects of music in home appliances design, and can be applied to studies for the blind. More specific guideline for the musical user interface can be drafted if further studies consider more various tasks, context, musical structure and types for the appliances.

  • Machine to Machine (M2M) communications is communications between a business application and devices via a communication network without any direct human interaction. The aim of our research is to connect any M2M device with an M2M AS (Application server) through an IMS (IP Multimedia Subsystem) Network Core using a M2M Gateway, in order to develop an M2M Horizontal Services Platform over IMS. An IP Multimedia Subsystem (IMS) is an architectural framework defined by the wireless standards body of the 3rd Generation Partnership Project (3GPP) for delivering IP multimedia services to mobile users. This paper shows the design and implementation of a Horizontal M2M Network Services Platform over an IP Multimedia Subsystem (IMS) using the Message Session Relay Protocol (MSRP). We summarize the protocols and architectures that formed the basis for the creation of our architecture. We provide a detailed description of our architecture design, describing the call flow of the proposed architecture and the entities operating in each process. We also describe the design and implementation process detailing the different tools used, explaining the selection of each component and its importance; also how we designed and implemented the M2M gateway, M2M Application Server, Open IMS Core, business application and M2M devices.

Cardiovascular response to surprise stimulus (놀람 자극에 대한 심혈관 반응)

  • Basic emotions such as happiness, sadness, anger, fear, and disgust have been widely used to investigate emotion-specific autonomic nervous system activity in many studies. On the contrary, surprise emotion, Suggested also as one of the basic emotions suggested by Ekman et al. (1983), has been least investigated. The purpose of this study was to provide a description of cardiovascular responses on surprise stimulus using electrocardiograph (ECG) and photoplethysmograph (PPG). ECG and PPG were recorded from 76 undergraduate students, as they were exposed to a visuo-acoustic surprise stimulus. Heart rate (HR), standard deviation of R-R interval (SD-RR), root mean square of successive R-R interval difference (RMSSD-RR), respiratory sinus arrhythmia (RSA), finger blood volume pulse amplitude (FBVPA), and finger pulse transit time (FPTT) were calculated before and after the stimulus presentation. Results show significant increase in HR, SD-RR, and RMSSD-RR, decreased FBVPA, and shortened FPTT. Evidence suggests that surprise emotion can be characterized by vasoconstriction and accelerated heart rate, sympathetic activation, and increased heart rate variability, parasympathetic activation. These results can be useful in developing an emotion theory, or profiling surprise-specific physiological responses, as well as establishing the basis for emotion recognition system in human-computer interaction.

  • 정보검색 시스템을 대하는 대부분의 이용자의 대답은 '이용하기에 어렵다'라는 것이다. 기계적인 정보검색을 기본 철학으로 하는 기존의 matching paradigm은 정보 곡체를 여기 저기 내용을 옮길 수 있는 물건으로 간주한다. 그리고 기존의 정보시스템은 이용자가 시스템을 구성한 사람의 의도 (즉, indexing, cataloguing rule)를 완전히 이해한다면, 즉 완전하게 질문식(query)을 작성한다면, 효과적인 검색을 할 수 있는 그런 시스템이다. 그러나 어느 이용자가 그 복잡한 시스템을 이해하고 정보검색을 할 수 있겠는가? 한마디로 시스템을 설계한 사람의 의도로 이용자가 적응해서 검색을 한다는 것은 아주 힘든 일이다. 그러나 우리가 이용자에 대한 인식을 다시 한다면 보다 나은 시스템을 만들 수 있다고 본다. 우리 인간은 아주 창조적이어서 자기가 처한 상황에서 이치에 맞게끔 자기 나름대로의 행동을 할 수 있다(sense-making approach). 이 사실을 인식한다면, 왜 이용자들의 행동양식에 시스템 설계자가 적응을 못하는 것인가? 하고 의문을 던질 수 있다. 앞으로의 시스템이 이용자들의 자연스러운 행동 패턴에 맞게 끔 설계된다면 기존의 시스템과 함께 쉽게 이용할 수 있는 편리한 시스템이 설계될 수 있을 것이다. 그러므로 도서관 및 정보학 연구에 있어서 기존의 분류. 목록에 대한 연구와 이용자체에 대한연구(예를 들면, 몇 시에 이용자가 많은가? 어떤 종류의 책을 어떤 계충에서 많이 보는가? 도서 및 잡지가 어떻게 양적으로 성장해 왔는가? 등등의 use study)와 함께 여기서 제시한 제3의 요소인 이용자의 인식(cognition)을 시스템설계에 반드시 도입을 해야만 한다고 본다(user-centric approach). 즉 이용자를 중간 중간에서 도울 수 있는 facilitator가 많이 제공되어야 한다. 이용자의 다양한 패턴의 정보요구(information needs)에 부응할 수 있고, 질문식(query)을 잘 만들 수 없는 이용자를 도울 수 있고(ASK hypothesis: Anomolous State of Knowledge), 어떤 질문식 없이도 자유스럽게 Browsing할 수 있는(예를 들면 hypertext) 시스템을 설계하기 위해서는 눈에 보이는 이용자의 행동패턴(external behavior)도 중요하지만 우리 눈에는 보이지 않는 이용자의 심리상태를 이해한다면 훨씬 나은 시스템을 만들 수 있다. 이용자가 '왜?' '어떤 상황에서,' '어떤 목적으로,' '어떻게,' 정보를 검색하는지에 대해서 새로운 관심을 들려서 이용자들이 얼마나 우리 시스템 설계자들의 의도에 미치지 못한다는 사실을 인식 해야한다. 이 분야의 연구를 위해서는 새로운 paradigm이 필수적으로 필요하다고 본다. 단지 'user-study'만으로는 부족하며 새로운 시각으로 이용자를 연구해야 한다. 가령 새롭게 설치된 computer-assisted system에서 이용자들이 어떻게, 그리핀 어떤 분야에서 왜 그렇게 오류 (error)를 범하는지 분석한다면 앞으로의 computer 시스템 선계에 큰 도움을 줄 수 있을 것으로 믿는다. 실제로 많은 방법이 개발되고 있다. 그러면 시스템 설계자가 가졌던 이용자들이 이러 이러한 방식으로 정보검색을 할 것이라는 예측과(즉, conceptual model) 실제 이용자들이 정보검색을 할 때 일어나는 행동패턴 사이에는(즉, mental model) 상당한 차이점이 있다는 것을 알게 될 것이다. 이 차이점을 줄이는 것이 시스템 설계자의 의무라고 생각한다. 결론적으로, Computer에 대한 새로운 지식과 함께 이용자들의 인식을 연구할 수 있는, 철학적이고 방법론적인 연구를 계속하나가면서, 이용자들의 행동패턴을 어떻게 시스템 설계에 적용할 수 있는 지를 연구해야 한다. 중요하게 인식해야할 사실은 구 Paradigm을 완전히 무시하라는 것은 아니고 단지 이용자에 대한 새로운 인식을 추가하자는 것이다. 그것이 진정한 User Study가 될 수 있는 길이라고 생각하며, 컴퓨터와 이용자 사이의 '원활한 의사교환'이 필수불가결 한 지금 우리 학문이 가야 할 한 연구분야이다. Effective brain-wave DB building system using the five senses stimulation (오감자극을 활용한 효율적인 뇌파 DB구축 시스템)

  • Ubiquitous systems have grown explosively over the few years. Nowadays users' needs for high qualify service lead a various type of user terminals. One of various type of user interface, various types of effective human computer interface methods have been developed. In many researches, researchers have focused on using brain-wave interface, that is to say, BCI. Nowadays, researches which are related to BCI are under way to find out effective methods. But, most researches which are related to BCI are not centralized and not systematic. These problems brought about ineffective results of researches. In most researches related in HCI, that is to say - pattern recognition, the most important foundation of the research is to build correct and sufficient DB. But there is no effective and reliable standard research conditions when researchers are gathering brain-wave in BCI. Subjects as well as researchers do not know effective methods for gathering DB. Researchers do not know how to instruct subjects and subjects also do not know how to follow researchers' instruction. To solve these kinds of problems, we propose effective brain-wave DB building system using the five senses stimulation. Researcher instructs the subject to use the five senses. Subjects imagine the instructed senses. It is also possible for researchers to distinguish whether brain-wave is right or not. In real time, researches verify gathered brain-wane data using spectrogram. To verify effectiveness of our proposed system, we analyze the spectrogram of gathered brain-wave DB and pattern. On the basis of spectrogram and pattern analysis, we propose an effective brain-wave DB building method using the five senses stimulation.

The antimicrobial activity of Hwangheuk-san and synergy effect with oxacillin against methicillin-resistant Staphylococcus aureus (황흑산(黃黑散)의 메티실린 내성 황색포도상구균에 대한 항균 활성과 옥사실린과의 시너지 효과)

  • Objectives : Hwangheuk-san is a complex prescription composed of oriental traditional medicine and has been reported for antioxidant, antimicrobial and anticancer effects in the recent study. Methicillin-resistant Staphylococcus aureus (MRSA) is one of important causes of fatal infectious diseases such as septicemia, endocarditis, toxic shock syndrome, pneumonia, skin and soft tissue infections (SSTIs). S. aureus is reported as being for a variety of human diseases and its epidemiological relevance is mainly due to their ability of becoming highly resistant to common antimicrobials such as tetracycline, penicillin, cphalosporin and aminoglycoside. The objective of this study is to determine the antimicrobial effect of Hwangheuk-san ethanol extracts (HHS) and synergistic effects with antibiotics oxacillin against MRSA.Methods : The antimicrobial activity of HHS was measured by the disc diffusion method, broth microdilution method and the checkerboard dilution test, time-kill curve assay was performed to investigate synergistic effects with antibiotics oxacillin against MRSA.Results : HHS showed antimicrobial activity against MRSA with a MIC value of 125 ㎍/㎖. In the checkerboard test, the interaction of HHS with antibiotics oxacillin produced almost synergy or partial synergy against MRSA. This study showed that HHS reduced the MICs of oxacillin tested, and a remarkable antibacterial effect of HHS, with membrane permeability enhancers.Conclusions : These results suggest that HHS has the antimicrobial effect and synergistic effects with antibiotics oxacillin against MRSA. This study thus can be a valuable source for the development of a new drug with low MRSA resistance.

Expression Analysis of the Ligand to Ly-6E.1 Mouse Hematopoietic Stem Cell Antigen

  • Ly-6E.1 antigen was proposed as a regulatory molecule of T lymphocyte activation, a hematopoietic stem cell marker, a memory cell marker, and an adhesion molecule. Though there were several reports suggesting the presence of Ly-6 ligand, the characterization of the ligand was not yet performed, As an attempt to screen the expression of Ly-6E.1 ligand, we prepared a probe for detecting Ly-6E.1 ligand by producing a fusion protein between Ly-6E.1 and $hlgC_{r1}$, A mammalian cell expression vector with Ly-6E.$1/hlgC_{r1}$ chimeric cDNA was transfected in SP2/0-Ag14 myeloma cells, and stable transfectants were selected. The fusion protein was produced as a dimer and maintained the epitopes for monoclonal antibodies specific for Ly-6E.1 and for anti-human lgG antibody. The purified fusion protein through Gammabind G column was used for FACS analyses for the expression of Ly-6E.1 ligand. The fusion protein interacted with several cell lines originating from B cells, T cells, or monocytes. The fusion Protein also strongly stained bone marrow, lymph node, and spleen cells, but thymic cells weakly, if any. The staining was more obvious in C57BL/6 $(Ly-6^b)$ than Balb/c $(Ly-6^a)$ mice. These results suggest that the interaction of Ly-6E.1 with Ly-6E.1 ligand may function both in the stem cell environment and in the activation of mature lymphocytes. The fusion protein may be a valuable tool in characterization of biochemical properties of the Ly-6E.1 ligand and, further, in isolating its cDNA.

A Study on the Necessity of Mission Command in Navy Through the Study of Naval Battle Comparison - Focus on Battle of St. Vincent and Battle of Jutland - (해전 비교연구를 통한 해군의 임무형 지휘 필요성에 관한 연구 - 세인트 빈센트 곶 해전과 유틀란트 해전을 중심으로 -)

  • The origin of mission command can be traced to the era of the Prussian military reforms led by General Gerhard von Scharnhorst after defeats in battle of Jena and Auerstadt in 1806 against Napoleon I. Mission Command is the conduct of military operations through decentralized execution based upon mission-type orders. Commanders issue mission-type orders focused on the purpose of the operation rather than details of how to perform assigned tasks. The mission command has become the command philosophy of the German military and recently many countries in the west accept it as a command philosophy. This study compare and analyze the Battle of St. Vincent and the Battle of Jutland to make sure if the army-initiated mission commands were also useful for the navy. From the late 18th century, represented by the era of Nelson, Royal navy changed from the inherited rigid command culture to guaranteeing the disciplined initiative of its subordinate commanders. In the Battle of St. Vincent in 1797, Nelson acted contrary to the commander's orders at the crucial moment, which gave Britain a decisive victory. On the contrary more than 100 years later, the command culture of the Royal navy changed into a centralized command culture. In the Battle of Jutland in 1916, Royal Navy couldn't win because the rigid command culture did not guarantee initiative of subordinate commanders and subordinate commander's passive attitude of waiting for the commander's instructions even at critical moments. Therefore, a mission command that guarantees the initiative of subordinate commanders is a useful concept even in the navy because it makes subordinate commanders to take full advantage of a sudden change in battle. Today's advanced information and communication technologies have raised questions about mission command. But even advanced technology can't completely eliminate the fundamental nature of the war-the fog of war. War is chaotic and unpredictable. In the flood of lots of informations, senior commander's judgement is not always right because he(she) is also human, he(she) can make mistakes. In the age of informatization, mission command is still effective because it involves increasing interaction and synergy between senior and subordinate commanders by ensuring their independence. Therefore ROK navy also needs to activate mission command. ROK navy must dismantle the zero-defect mentality and apply from educations as Prussian did to establish a mission command culture.
