Objectives: Formalin is used as an anthelmintic in farms where flounder are raised. In this study, we aim to identify formaldehyde exposure levels for aquaculture industry workers and provide basic data for managing formaldehyde exposure. Methods: Exposure levels of formaldehyde in the air, including formalin spraying operations, were assessed separately for personal and area samples. In addition, considering the formalin administration method, dermal exposure to the hands was estimated when administering the chemical, and dermal exposure to the legs during water tank work was estimated by collecting water in the water tank and evaluating the amount of formaldehyde remaining. Finally, the respiratory exposure level and the estimated dermal exposure level were added to derive the total exposure level and compared with the maximum allowable human dose. Results: As a result of the airborne evaluation, the formaldehyde concentration of the worker (1 person) who performed the formalin spraying and flounder sorting was 33.61 ppb, and the arithmetic mean of formaldehyde concentrations of the workers (3 people) who only performed the flounder sorting was 3.28 ppb (range: 2.25-4.89 ppb). In the case of dermal exposure, when spraying formalin once, the amount was estimated to be 0.33-2.62 mg when wearing protective gear and 3.27-26.12 mg when not wearing it. Conclusions: There was a difference in the formaldehyde exposure level of workers depending on their operation of handling formalin and whether or not protective gear was worn. In particular, because the level of formaldehyde exposure due to dermal exposure can be significant, there is a need to improve formalin administration methods in a way that avoids skin contact as much as possible.
Noise may cause damage of the auditory system, hypertension, and cardiovascular disease. However, we haven't the data enough to be available for understanding various effects of noise on the human body. The current study was prospectively designed to investigate the changes of the cardiac factors and cerebral hemodynamics following a transient exposure to noise in young people. 80 subjects (mean aged $23.45\pm2.40$ years) participated in this experiment and were exposed to excavator-noise with 90 decibels for 15 minutes using ear-phone. Cardiac factors such as heart rate (HR), blood pressure (BP) and heart rate-systolic pressure product (RPP), and cerebral hemodynamics such as mean blood flow velocities (Vm), pulsatility indexes (PI), resistance indexes (RI) and mean blood flow velocities at breathing-hold (Vh) in the middle (MCA), anterior (ACA) and posterior cerebral arteries (PCA) were measured before (baseline) and during the noise-exposure. Although there were individual differences in above mentioned parameters, HR, systolic and diastolic BP, RPP, MCA-Vm, MCA-PI, MCA-RI, ACA-Vm, ACA-PI, ACA-RI, PCA-Vm, PCA-PI, and PCA-RI during the noise-exposure decreased compared with the baselines (P<0.05 or P<0.01), The findings of the present study suggest that a transient exposure to excavator-noise at rest causes changes in the cardiac factors and cerebral hemodynamics with individual differences. Further studies need to be carried out for clarifying the effects of longer exposure and combined mental activity with noise exposure.

Skin is continuously exposed to external stimuli including ultraviolet radiation, which is a major cause of skin photoaging. According to recent discoveries, UVA with a lower energy but deep-penetrating properties, compared to UVB, is likely to play a major part in causing skin photoaging. The clinical and histochemical changes of photoaging are well characterized, but the biochemical mechanisms are poorly understood partly due to the lack of suitable experimental systems. In this work, three-dimensional, reconstituted skin culture models were prepared. After certain period of maturation, the equivalent models were shown to be similar in structure and biochemical characteristics to normal skin. Mature dermal and skin equivalent models were exposed to sub-lethal doses of UVA, and the effects of UVA relevant to dermal photoaging were monitored, including the production of elastin, collagen, collagenase(MMP-1), and tissue inhibitor of metalloproteinases-1 (TIMP-1). Interestingly, dermal and skin equivalents reacted differently to acute and chronic exposure to UVA. Elastin production was increased as soon as one week after commencing UVA irradiation by chronic exposure, although a single exposure failed to do so. This early response could be an important advantage of equivalent models in studying elastosis in photoaged skin. Collagenase activity was increased by acute UVA irradiation, but returned to control levels after repeated exposure. On the other hand, collagen biosynthesis, which was increased by a single exposure, decreased slightly during 5 weeks of prolonged UVA exposure. Collagenase has been thought to be responsible for collagen degeneration in dermal photoaging. However, according to the results obtained in this study, elevated collagenase activity is not likely to be responsible for the degeneration of collagen in dermal photoagig, while reduced production of collagen may be the main reason. It can be concluded that reconstituted skin culture models can serve as useful experimental tools for the study of skin photoaging. These culture models are relatively simple to construct, easy to handle, and are reproducible Moreover the changes of dermal photoaging can be observed within 1-4 weeks of exposure to ultraviolet light compared to 4 months to 2 years for human or animal studies. These models will be useful for biochemical and mechanistic studies in a large number of fields including dermatology, toxicology, and pharmacology.

Objectives: The unsafe use of pesticides in agriculture represents a major hazard to human health. This study was conducted to investigate the association between pesticide exposure and health symptoms among chili farmers in northeast Thailand. Methods: This cross-sectional analytic study included 141 chili farmers in Sakon Nakhon Province, in northeast Thailand. Data regarding health symptoms were gathered using a self-report questionnaire. A medical technician tested blood cholinesterase activity using an erythrocyte method, and an occupational medicine specialist at Ramathibodi Hospital in Bangkok, Thailand reviewed the results. Associations between personal characteristics, pesticide exposure factors, and health symptoms were analyzed using multiple logistic regression. Results: Of the 141 chili farmers studied, 66.7% experienced pesticide poisoning, as indicated by below-normal cholinesterase levels. Fatigue was the most frequently reported symptom associated with pesticide exposure, affecting 37.6% of participants. This was followed by nausea and vomiting (31.9%), dizziness (14.9%), and dry throat (14.9%). Multivariate logistic regression analysis revealed that several factors were significantly associated with adverse symptoms: amount of work experience, volume of pesticides used, use of chemical pesticides, use of leaking containers during spraying, direct pesticide exposure while working, contact with pesticide-soaked clothing, consumption of food and drinks in the fields, and blood cholinesterase level indicating risk. Conclusions: This study suggests potential health risks for chili farmers stemming from exposure to and contamination by pesticides used in agricultural practices. To mitigate these risks, it is essential to supply personal protective equipment and to implement educational programs aimed at improving protective behaviors among farmers.

Naphthalene is bicyclic aromatic compound that is widely used in various domestic and commercial applications including lavatory scent disks, soil fumigants and moth balls. Exposure to naphthalene results in the development of bronchiolar damage, cataracts and hemolytic anemia in humans and laboratory animals. However, little information is available regarding the mechanism of naphthalene toxicity. We investigated gene expression profiles and potential signature genes in human hepatocellular carcinoma HepG2 cells and human promyelocytic leukemia HL-60 cells after 3 h and 48 h incubation with the IC$_{20}$ and IC$_{50}$ of naphthalene by using 44 k agilent whole human genome oligomicroarray and operon human whole 35 k oligomicroarray, respectively. We identified 616 up-regulated genes and 2,088 down-regulated genes changed by more than 2-fold by naphthalene in HepG2 cells. And in HL-60, we identified 138 up-regulated genes and 182 down-regulated genes changed by more than 2-fold. This study identified several interesting targets and functions in relation to naphthalene-induced toxicity through a gene ontology analysis method. Apoptosis and cell cycle related genes are more commonly expressed than other functional genes in both cell lines. In summary, the use of in vitro models with global expression profiling emerges as a relevant approach toward the identification of biomarkers associated with toxicity after exposure to a variety of environmental toxicants.

Temperature change is one of the major environmental factors that influence the human skin. However, the relationship between temperature and melanogenesis has received little attention. In the present study, we investigated the effects of temperature change on melanogenesis in a mouse melanocyte cell line (Mel-Ab), and primary cultured human melanocytes. We found that Mel-Ab cells cultured at low temperatures (31 and 34$^{\circ}C$) produce less melanin than cells at 37$^{\circ}C$. These results were confirmed by experiments upon human melanocytes, demonstrating that the hypopigmenting effect of low temperatures is not cell type dependent. The observed melanin production was found to be accompanied by tyrosinase activity at each temperature, indicating that tyrosinase activity is regulated by temperature. We further examined whether the incubation period at low temperatures plays an important role in the regulation of melanogenesis. Short exposures to 27$^{\circ}C$ for 1 h or 3 h did not affect tyrosinase activity or melanin synthesis, whereas long exposures to 31$^{\circ}C$ for 2 days or 6 days significantly reduced tyrosinase activity and melanin synthesis in a duration-dependent manner. Our results suggest that exposure to low temperature and the duration of this exposure are important regulators of melanogenesis.
Objective: The aim of this study is to review the literatures on the regulation, standard and guideline for the human vibration in Korea and other countries. Background: This review can be used to prevent various diseases caused by the human vibration as a basis for the development of the policy. Results: In Korea, the general employers' duties related to human vibration are set forth the Health Measures(Article 24) in the Occupational Safety and Health Act. And then an employer shall take measures to protect the health of the workers concerned by improving other working conditions relating to working hours for the vibration prevention measures referred to in Article 24 of the Act. The European Union adopted a Directive in 2002 on minimum requirements for the health and safety of workers exposed to vibration. New Regulations on Vibration at Work will be introduced in Great Britain on 2005 to implement the Directive. In the U.S., both ANSI and ACGIH adopted the ISO standard for measurement and suggested exposure action and limit values. In Japan, the Ministry of Labor decided that the vibration syndrome among operators of rock drills and riveters etc. could be included in an occupational disease(1947). In addition, ISO standard was based on proposals and draft documents of many countries such as U.K, Japan and European, etc. Conclusion: In Korea, Occupational Safety and Health Act prevent vibration to health, but do not include exposure limits. It is therefore important to consider the new duties regarding to vibration risks added to the general duties.
The eye irritation potential of drug candidates or pharmaceutical ingredients should be evaluated if there is a possibility of ocular exposure. Traditionally, the ocular irritation has been evaluated by the rabbit Draize test. However, rabbit eyes are more sensitive to irritants than human eyes, therefore substantial level of false positives are unavoidable. To resolve this species difference, several three-dimensional human corneal epithelial (HCE) models have been developed as alternative eye irritation test methods. Recently, we introduced a new HCE model, MCTT HCE$^{TM}$ which is reconstructed with non-transformed human corneal cells from limbal tissues. Here, we examined if MCTT HCE$^{TM}$ can be employed to evaluate eye irritation potential of solid substances. Through optimization of washing method and exposure time, treatment time was established as 10 min and washing procedure was set up as 4 times of washing with 10 mL of PBS and shaking in 30 mL of PBS in a beaker. With the established eye irritation test protocol, 11 solid substances (5 non-irritants, 6 irritants) were evaluated which demonstrated an excellent predictive capacity (100% accuracy, 100% specificity and 100% sensitivity). We also compared the performance of our test method with rabbit Draize test results and in vitro cytotoxicity test with 2D human corneal epithelial cell lines.

Cumulus-oocyte complexes (COCs), which contain immature oocytes, are matured in vitro for in vitro embryo production. Oocyte and cumulus cells are then separated using hyaluronidase. To date, there have only been a few reported cases of the toxic effects of hyaluronidase on porcine oocytes. The aim of this study was to compare the effects of bovine testis-derived hyaluronidase and recombinant human hyaluronidase on oocyte denudation and quality. Porcine COCs were matured for 44 h and denuded using different hyaluronidase concentrations and exposure times. Then, oocytes were activated by electrical parthenogenesis. In experiment 1, COCs were denuded using bovine-derived, ovine-derived (Hirax), and human recombinant (ALT-BC4) hyaluronidases for 10 and 20 min. In experiment 2, bovine-derived and human recombinant (ALT-BC4 and ICSI Cumulase®) hyaluronidases were used to denude the COCs for 2 and 20 min. In both experiments the oocytes were all completely denuded, and there was no degeneration. Rate of embryo development was significantly increased in group treated ALT-BC4 for 2 min and not significantly different in other treatment groups. In general it slightly decreased with longer exposure times. These results have confirmed that different sources of hyaluronidase do not have detrimental effects on the quality of porcine oocytes and suggest that the human recombinant hyaluronidase ALT-BC4 is suitable for oocyte denudation with an increased blastocyst rate.

Background: Exposure levels for heavy metals such as lead (Pb), mercury (Hg), and cadmium (Cd) have increased due to human activities. They are known to be a public health concern. Objectives: This study aimed to determine the exposure levels to heavy metals in the blood and urine of South Korean adults and to present the contribution rate of exposure pathways using an exposure algorithm for men aged 19~64, women aged 19~64, and all seniors aged 65 or older. Methods: We analyzed data from the Korean National Environmental Health Survey (KoNEHS) Cycle 4 (2018~2020). A total of 2,646 participants aged ≥19 years were included. Multiple regression analysis was performed to determine the factors affecting heavy metal concentrations. The contribution rate was calculated by applying three exposure algorithms for ingestion, inhalation, and dermal exposure. Results: Factors that commonly affect heavy metal concentrations in blood and urine were gender and age. The main influencing factors for Pb and Cd were education level and smoking status, while frequency of fish consumption and of alcohol consumption were indicated to be the main influencing factors for mercury. The contribution rates of lead and cadmium from food ingestion were 78.03~79.62% and 88.39~92.89%, respectively. Additionally, the highest contribution for mercury was accounted for by food at 81.69~85.77%. As a result of the risk assessment, cadmium was found to pose a potential health risk a with total cancer risk (TCR) of more than 1×10-6. Conclusions: The KoNEHS could be an important study for determining the level of exposure to heavy metals and their influencing factors. Integrated exposure to heavy metals could assess the main exposure pathways, and this methodology could be applied to exposure management of heavy metals.
