미세먼지는 유해한 다양한 작은 입자를 가진 대기오염 물질로서 산화적 및 염증성 반응을 촉진하여 다양한 질환의 발병과 진행에 관여하는 것으로 알려졌다. 본 연구에서는 외부 오염물질에 직접적으로 노출되는 1차 노출 기관인 안구를 미세먼지로부터 보호할 수 있는 약재를 선정하기 위해, 인간 각막상피세포에서 후보 약물들의 방어 효능을 평가했습니다. 그 결과, 12종의 후보 약재 중 PM2.5에 의한 세포 독성 억제효능을 보인 후보 약재 5가지(천문동, 석창포, 황련, 감국 및 금잔화)를 선별하였다. 이들 후보 물질들의 항산화 활성을 평가하기 위하여 ROS 소거능을 조사한 결과, 석창포, 천문동 및 황련 추출물이 유의한 효과를 나타내었으며, 이는 미토콘드리아 활성 보존 효능과 다소 연관성이 있었다. 또한, 이들은 PM2.5에 의한 DNA 손상을 차단할 수 있었음을 8-OHdG 생성 및 γ-H2AX 발현 분석을 통하여 확인하였다. 본 연구의 결과는 PM2.5에 대한 각막상피세포의 보호 신규 천연물의 탐색을 위한 자료로 활용될 것이다.
점안액 보존제 성분으로 이용되는 Benzalkonium Chloride(BAC)가 배양 각막 상피세포에 미치는 영향을 조사하기 위해 본 연구를 시행하였다. BAC를 세포에 0.0001%~0.01%로 15분 동안 처리하여 24시간 후 회복 효과를 조사하였다. MTT assay를 이용하여 세포 생존율을 산정하고 Hoechst 33342로 염색질 응축을 조사하였다. Fas 발현 여부를 조사하기 위해 western blot과 immunocytochemistry를 이용하였으며 DNA fragmentation은 agarose gel을 이용한 전기영동을 시행하였다. BAC를 0.005% 이상 처리하면 세포괴사가 일어나는 반면에 저농도의 BAC는 세포고사를 유도하는 것으로 나타났으며 BAC로 Fas 발현이 유도될 수 있다는 것을 보여주고 있다. 본 연구를 통해 점안액의 대표적인 보존제인 BAC의 부작용에 대한 각막세포 차원의 연구가 필요하며 세포독성이 없는 새로운 보존제의 개발이 필요한 것으로 나타났다.
Genetic diversity of 18 Acanthamoeba isolates from ocean sediments was evaluated by comparing mitochondrial (mt) DNA RFLP, 18S rDNA sequences and by examining their cytopathic effects on human corneal epithelial cells versus reference strains. All isolates belonged to morphologic group II. Total of 16 restriction phenotypes of mtDNA from 18 isolates demonstrated the genetic diversity of Acanthamoeba in ocean sediments. Phylogenetic analysis using 18s rDNA sequences revealed that the 18 isolates were distinct from morphological groups I and III. Fifteen isolates showed close relatedness with 17 clinical isolates and A. castellanii Castellani and formed a lineage equivalent to T4 genotype of Byers' group. Two reference strains from ocean sediment, A. hatchetti BH-2 and A. griffini S-7 clustered unequivocally with these 15 isolates. Diversity among isolates was also evident from their cytopathic effects on human corneal cells. This is the first time describing Acanthamoeba diversity in ocean sediments in Korea.
본 연구는 mycoplasma가 오염된 배양 각막 상피세포에 anti-FAS and anti-FAS ligand antibody를 노출시킨 후 세포고사 메커니즘을 조사하기 위해 시행하였다. 배양각막 상피세포에 anti-FAS antibody 와 anti-FAS ligand antibody를 2일과 4일 동안 처리하여 주어진 기간 동안 배양하였다. 배양 중인 각막상피세포가 mycoplasma sp.에 의해 오염된 것이 밝혀졌다. mycoplasma removal agent(MRA)가 세포주로부터 세균을 제거하기 위해 이용되었다. MRA를 세포주에 첨가하여 1주일 동안 배양하였다. 그 후 각막상피 세포주는 MRA가 포함되지 않는 배양액에서 수세대 동안 배양되어 커버글라스에 계대배양하여 50-80% 채워졌을 때 MRA 제거여부를 확인하기 위한 검사 키트에 제공된 Hoechst staining을 이용하여 염색하였다. 세포고사 실험은 MRA 제거 전과 제거 후에 시행되었다. mycoplasma가 오염된 배양 각막상피세포에 대한 anti-FAS and anti-FAS ligand antibody의 영향을 알아보기 위해 Hoechst 33342 staining과 Annexin V-FITC and Propidium Iodide Staining을 이용하여 세포고사 유도를 확인하였다. 본 연구는 anti-FAS antibody가 배양각막 상피세포에서 시간과 농도에 비례하는 메커니즘으로 세포고사를 유도한다. mycoplasma가 오염된 세포주는 FAS 유도 세포고사의 민감성을 증가시키는 것으로 나타났다.
목적: 시판되고 있는 소프트콘택트렌즈용 다목적용액이 사람의 각막상피세포에 미치는 세포독성을 이미지 분석법을 이용하여 평가하고자 하였다. 방법: 각막상피세포를 6종류의 다목적용액(A~F)이 0.05~50% 포함된 배양액에서 각각 2시간, 12시간, 24시간, 48시간 동안 배양하였다. 배양 후 각막상피세포를 고정한 다음 Draq 5로 염색하고 공초점현미경과 ImageXpress $Ultra^{TM}$를 이용하여 세포형태를 관찰하고 세포생존율과 세포자살(apoptosis)을 비교하였다. 결과: 각막상피세포 생존율과 세포자살은 다목적용액을 2시간 처리한 경우에는 모든 제품에서 대조군과 차이가 없었으나, 12시간 이상 처리한 경우에는 B~F 제품에서 대조군의 52~75% 수준으로 감소하였고, 세포자살은 모든 제품에서 차이가 없었다. 24~48시간 처리한 경우에는 생존율이 29~73% 수준으로 감소하였고(p<0.05), 세포자살은 199~526% 증가하였다. 제품별로는 다목적용액 D, E, F가 A보다 각막상피세포 생존율을 감소시켰고 세포자살을 증가시켰다(p<0.05). 결론: 저농도의 다목적용액은 각막상피세포 독성에 영향을 미치지 않지만 고농도의 다목적용액은 각막상피세포의 자살을 유도하고 세포생존율을 저하시킬 수 있으므로 다목적용액의 성분은 소독기능이 우수하고 독성이 없는 성분으로 개발되어야 할 것으로 사료된다.

FK506 binding protein 12 (FK506BP) belongs to a family of immunophilins, and is involved in multiple biological processes. However, the function of FK506BP in corneal disease remains unclear. In this study, we examined the protective effects on dry eye disease in a Botulinum toxin A (BTX-A) induced mouse model, using a cell-permeable PEP-1-FK506BP protein. PEP-1-FK506BP efficiently transduced into human corneal epithelial cells in a time- and dose-dependent manner, and remained stable in the cells for 48 h. In addition, we demonstrated that topical application of PEP-1-FK506BP was transduced into mouse cornea and conjunctiva by immunohistochemistry. Furthermore, topical application of PEP-1-FK506BP to BTX-A-induced mouse model markedly inhibited expression levels of pro-inflammatory cytokines such as interleukin-$1{\beta}$ (IL-$1{\beta}$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and macrophage inhibitory factor (MIF) in corneal and conjunctival epithelium. These results suggest PEP-1-FK506BP as a potential therapeutic agent for dry eye diseases.
To evaluate the toxicity of ophthalmic drug, the Draize test and Bovine Corneal Opacity and Permeability (BCOP) test commonly used. In Draize test, experimental animals were under stress and pain due to long-term exposure of drug. In addition, regarding physiological functions, animal model is not perfectly reflected a human eye condition. Although some models such as $EpiOcular^{TM}$, HCE model, LabCyte Cornea-Model, and MCTT $HCE^{TM}$ were already presented advanced cornea ex-vivo model to replace animal test. In this sense, cornea tissue structure mimicked ex-vivo toxicity model was fabricated in this study. The corneal epithelial cells (CECs) and keratocytes (CKs) isolated from rabbit eyeball were seeded on non-patterned silk film (n-pSF) and patterned silk film (pSF) at $32,500cells/cm^2$ and $6,500cells/cm^2$. Sequentially, n-pSF and pSF were stacked to mimic a multi-layered stroma structure. The thickness of films was about $15.63{\mu}m$ and the distance of patterns was about $3{\mu}m$. H&E stain was performed to confirm the cell proliferation on silk film. F-actin of CKs was also stained with Phalloidin to observe the cytoskeletal alignment along with patterns of the pSF. In the results, CECs and CKs were shown the good cell attachment on the n-pSF and pSFs. Proliferated cells expressed the specific phenotype of cornea epithelium and stroma. In conclusion, we successfully established the ex-vivo cornea toxicity model to replace the eye irritation tests. In further study, we will set up the human ex-vivo cornea toxicity model and then will evaluate the drug screening efficacy.

Acanthamoeba keratitis (AK) is a rare infectious disease and accurate diagnosis has remained arduous as clinical manifestations of AK were similar to keratitis of viral, bacterial, or fungal origins. In this study, we described the production of a polyclonal peptide antibody against the adenylyl cyclase-associated protein (ACAP) of A. castellanii, and evaluated its differential diagnostic potential. Enzyme-linked immunosorbent assay revealed high titers of A. castellanii-specific IgG and IgA antibodies being present in low dilutions of immunized rabbit serum. Western blot analysis revealed that the ACAP antibody specifically interacted with A. castellanii, while not interacting with human corneal epithelial (HCE) cells and other causes of keratitis such as Fusarium solani, Pseudomonas aeruginosa, and Staphylococcus aureus. Immunocytochemistry (ICC) results confirmed the specific detection of trophozoites and cysts of A. castellanii co-cultured with HCE cells. The ACAP antibody also specifically interacted with the trophozoites and cysts of 5 other Acanthamoeba species. These results indicate that the ACAP antibody of A. castellanii can specifically detect multiple AK-causing members belonging to the genus Acanthamoeba and may be useful for differentially diagnosing Acanthamoeba infections.
Multipurpose contact lens disinfecting solutions (MPDS) are widely used to cleanse and disinfect microorganisms. However, disinfection efficacy of these MPDS against Acanthamoeba cyst remain insufficient. 2, 6-dichlorobenzonitrile (DCB), a cellulose synthesis inhibitor, is capable of increasing the amoebical effect against Acanthamoeba by inhibiting its encystation. In this study, we investigated the possibility of DCB as a disinfecting agent to improve the amoebicidal activity of MPDS against Acanthamoeba cyst. Eight commercial MPDS (from a to h) were assessed, all of which displayed insufficient amoebicidal activity against the mature cysts. Solution e, f, and h showed strong amoebicidal effect on the immature cysts. Amoebicidal efficacy against mature cysts remained inadequate even when the 8 MPDS were combined with $100{\mu}M$ DCB. However, 4 kinds of MPDS (solution d, e, f, and h) including $100{\mu}M$ DCB demonstrated strong amoebicidal activity against the immature cysts. The amoebicidal activity of solution d was increased by addition of DCB. Cytotoxicity was absent in human corneal epithelial cells treated with either DCB or mixture of DCB with MPDS. These results suggested that DCB can enhance the amoebicical activity of MPDS against Acanthamoeba immature cyst in vitro.
소프트 콘택트렌즈는 HEMA(Hydroxyethyl Methacrylate)를 재질로 하여 사용하기 시작하여 현재는 실리콘을 이용한 재질까지 발달하고 있다. 본 연구는 국내에서 유통되는 7개 회사의 콘택트렌즈 재질에 대한 세포독성과 보존 용액의 세포독성을 평가하기 위해 시행하였다. 수집된 콘택트렌즈를 $121^{\circ}C$에서 1시간 동안 용출한 용액과 렌즈 보존용액을 배양 생쥐 섬유모세포와 각막 상피세포에 25% 농도로 48시간 동안 처리하여 세포증식 저해 정도를 조사하였다. 렌즈의 용출액은 세포독성이 없었으나 보존용액 중 3개회사 제품에서 세포증식 저해가 나타났다. 본 연구는 렌즈의 재질이나 보존용액에 대한 독성평가의 중요한 지침으로 이용될 수 있을 것으로 사료된다.
