A comparative study of pretreatment and extraction methods was conducted to investigate their effects on nutritional and bioactive components during hot-water extraction of Acanthopanax senticosus. The herb samples, ground and sifted $(8\~20\;mesh)$ were pretreated with ultrasound (20 kHz), microwave (2,450 MHz) or roasting $(180^{\circ}C)$ for 10 min, and then extracted by a boiling $(180^{\circ}C)$ under atmospheric pressure or a pressured boiling $(121^{\circ}C)$ for 2 hr. In general, the pretreatments improved the extraction efficiency and the DPPH radical scavenging activity of extract. Particularly, the extraction yields of soluble solids, total sugar and phenolic compounds were increased in microwaving methods, and the protein yield and the radical scavenging activity were higher in roosting pretreatment. Yields of soluble solids, total sugar, protein and phenolic compouds in pressured extraction were higher than those in nonpressured extraction. These results suggested that pretreatment including microwave radiation, sonication and roasting and pressured extraction can be utilized for improving the extraction efficiency of A. senticosus.

Recently, silk sericin has attracted the attention of researchers owing to its useful properties as a biomaterial including 1) good wound healing and cell activities, 2) fast gelation character, and 3) high water retention property. In the present study, silk sericin was prepared using different extraction times in hot water and the effect of extraction time on the rheological properties of sericin solutions and gels was examined. It was found that the production yield of sericin increased with extraction time. The shear viscosity of sericin solutions and gels decreased with increasing extraction time due to a decrease in sericin molecular weight. When the sericin solution transformed to a gel, the viscosity increased and the shear thinning behavior was more evident. In addition, the shear stress measurements indicated that the slip between the sericin samples and the measuring plate of the rheometer was increased by the gelation of sericin. The compression strength of sericin gel could be increased remarkably (by more than 100 fold) by preparation using the freezing and thawing method.

Energy performance of electric heater and geothermal source heat pump type hot air dryers are compared in this study. For set temperatures of $30^{\circ}C$, $35^{\circ}C$ and $40^{\circ}C$, radish is dried from initial mass 60 kg until it gets 5 kg, where the difference equals the amount of water removed. As set temperature is increased, drying time is shortened for both electric heater and heat pump types, however energy efficiency is decreased due to increasing electricity consumption. Moisture extraction rate(MER) of electric heater is 2.58~2.84 kg/h, and for heat pump type 2.56~2.71 kg/h, showing little difference between the two types. Specific moisture extraction rate (SMER) of electric heater is 0.94~0.96 kg/kWh, and for heat pump type 1.72~2.21 kg/kWh. SMER of heat pump type is greater by 0.78~1.25 kg/kWh than the electric heater hot air dryer, which is 1.8~2.3 times better in terms of energy efficiency.

Objectives : This study was designed to investigate the analgesic effects of Taxus cuspidata pharmacopuncture(TCP) at KI03 on formalin-induced pain in rats and to evaluate the efficiency of different extraction methods of Taxus cuspidata(TC). Methods : 30 rats were divided into 5 groups, each consisting of 6 rats. Each of the groups was treated two times. The first treatment for its right KI03 was as follows: rats were treated with normal saline(NOR), normal saline(CON), hot-water extraction of TC(W), ethanol extraction of TC(E), and ultrasonification extraction of TC(U). The second treatment was given 35 mins after the first one. Rats in NOR were treated with normal saline at their hind-paw. All groups, except NOR, were treated with formalin(5 %, $40{\mu}{\ell}$) at their hind-paw. To evaluate pain behavior, Ultrasonic vocalization(USV) was examined to be around 18~30 kHz. After analysis of USV, blood samples were taken from the rats for analysis of Substance P, aspartate aminotransferase(AST) and alanine aminotransferase(ALT). Results : In the experimental groups, USV was significantly decreased compared with CON and similar to that of NOR in the early phase. But there were no significant differences among the three extraction methods of TC. In addition, Substance P of W was significantly decreased. AST and ALT activation showed no significant differences among the 5 groups. Conclusion : These results show that regardless of extraction methods of TC, TCP at KI03 has analgesic effects in the early phase on formalin-induced pain in rats. In particular, hot-water extraction of TC significantly reduces Substance P activation.

In order to optimize the extract condition and improve physiological activity of the extract form Tricholoma matsutake, experiments related to extraction methods, totale yield, content of total soluble polysaccharide, SOD-like activity, total polyphenol amount, and volatile flavor compound and the others were carried out, results were obtained as following: Compare with traditional hydrothermal extraction method (Hot water extraction : HWEW), it illustrates that the low temperature extraction method which combines a supersonic waves and microwave (Supersonic microwave extraction : SMEW) causes of increasing the total yield, total soluble polysaccharide. As to the anti-oxident effect, SMEW method leds to increasing of the SOD-like activity, total polyphenol amount as well. Also, cytotoxic effect and growth inhibitory effect against cancer cell line are much higher in SMEW method than HWEW method, especially SMEW5 extracts treated by supersonic 15 min. and microwave 120W, 3 min. and 2 times. The main volatile flavor compound and infinitesimal volatile flavor compound both increase significantly by SMEW method. It is concluded the main components of the volatile flavor compounds extracted from Tricholoma matsutake are 1-octen-3-0l, Methyl cinnamate, 2-octeno1 et al. alcohol typies. Consequently, SMEW5 method is considered as the most effective one for anti-oxidant and is prior to any other methods. And the optimun conditions of this method are : supersonic waves (supersonic, 25KHz, 50W) 15 minutes, microwave spectroscopy (microwave, 2,450MHz, 120W) 3 minutes, and every treatment is performed once followed twice repeats.
In preparing sodium alginates from sea tangle (Laminaria japonica) powder using the Mexican process, alkaline extraction, conversion to alginic acid and reversion to sodium alginates were used to increase purity. The effect of hot water treatment and dialysis on measuring the average molecular weight of sodium alginates were investigated. Intrinsic viscosity and average molecular weight of sodium alginates after dialysis were higher than those before dialysis. Average molecular weight of sodium alginates treated with hot water was higher than that without. Hot treatment has little effect on the ash content of sodium alginates. Ash content of sodium alginates before dialysis were $27-30\%$ those after dialysis were $10\%.$ After dialysis, Na content was highest $(89-91\%),$ K was $11-12\%,$ Ca was $1.9\%,$ and Mg was $0.05\%.$ Ash content of alginates had little effect on average molecular weight. SAV (slope of apparent viscosity) of alginates solution after dialysis showed higher values than before. SAV of the alginates with hot water treatment were higher than without treatment.
This study was carried out to develop a traditional functional beverage by using hot-water extraction of dried peels of Korean tangerine. The pH of the hot-water extracts of dried tangerine peels decreased as the extraction time increased. The acidity and viscosity of the extracts increased as the extraction time increased. The antioxidative activity of the extracts during the extraction was monitored by measuring the electron donating ability. The electron donating abilities of the extracts were in the range of 80.93-83.27%. Extraction time did not affect the antioxidative activity of the extracts. The fibrinolytic activity of the extracts increased as the extraction time increased. The pH of the beverage made with the peel extract was not affected by the extraction time and the kind of sweetener added. The viscosity of the beverage increased as the extraction time increased. In sensory evaluation the highest score was obtained in the beverage samples made with the ones extracted for 180 min and added with sugar. The aboveresults indicate that the dried peel of Korean tangerine can be used as a functional material in beverage industry.

The purpose of this study was to investigate the antioxidant activity and β-glucan content of extracts extracted by varying the temperature at 30, 55 and 80℃ after hot air drying or freeze drying of Pleurotus ostreatus and Pleurotus eryngii. For the analysis antioxidant activity of each mushroom, β-glucan, total phenol, flavonoid contents, and DPPH·ABTS+·Nitrite assay were measured. Also, the β-glucan content, total flavonoid content and ABTS+ scavenging activity increased with freeze drying rather than hot air drying, and increased with increasing extraction temperature in both mushrooms. However, the total phenol and nitrite scavenging activity increased with hot air drying rather than freeze drying, and decreased with increasing temperature in both mushrooms. DPPH scavenging activity was not significant in both mushrooms, but decreased with increasing extraction temperature. Pearson's correlations between total flavonoid content and antioxidant activities were r=0.719~0.753 (p<0.01). As a result, the β-glucan content, total flavonoid content, and ABTS radical cation scavenging activity were highest during freeze drying and extraction at 80℃. And the highest total phenol content, DPPH radical scavenging activity and nitrite scavenging activity were obtained during hot air drying and extraction at 30℃.
Vascular dementia is characterized by white matter lesions involving the demyelination and activation of astrocytes and microglia. In a previous study, we showed that the supernatant of a laboratory-scale, hot water extract of ground whole wheat (TALE) attenuated white matter injury and astrocytic activation in a rat model of bilateral common carotid artery occlusion (BCCAO). In the present study, we made several modifications to the hot water extraction process to remove starch and enable large-scale production. We used wheat bran (WB), which contains less starch, instead of ground whole wheat. In addition, we removed starch granules with a decanter before hot water extraction. The final product, wheat bran extract (WBE), contained 2.42% arabinose, a surrogate marker of arabinoxylan, which is an active constituent of WBE. Supplementation of the rat model of BCCAO with WBE (400 mg/kg/day) for 33 days attenuated white matter injury, which was assessed by Luxol Fast Blue staining, in the corpus callosum (cc) and optic tract (opt) regions. Attenuation of white matter injury in the opt region was accompanied by improvement of the pupillary light reflex. Immunochemical staining revealed that supplementation with WBE reduced astrocytic activation in the cc and opt regions and reduced microglial activation in the opt region. These findings indicate that supplementation with WBE is effective at attenuating white matter injury accompanied by the inhibition of astrocytic and microglial activation. Therefore, extracts from WB, a cheap by-product of wheat milling, can be developed as a nutraceutical to prevent vascular dementia, a disease for which there is no approved pharmaceutical treatment.
The purpose of this study was to investigate the dyeing properties and anti-microbial ability of silk and wool fabrics dyed with Terminalia chebula Retzius(TCR) extract using two extraction solvent, hot water and methanol. Dyeing properties of fabrics were studied by investigating the characteristics of colorant, changes in dye uptake under different dyeing conditions, and by investigating color change when mordants were applied. Also, color fastness, and antimicrobial activity of dyed fabrics were estimated. Regardless of extraction solvent type, colorant showed maximum absorption wavelength at 280 nm and 578 nm, which implied that tannin was the major pigment component of TCR. Also, through FT-IR spectrum result, it was confirmed that tannin of TCR methanol extract was hydrolysable tannin. But for the hot water extract, it was only assumed that its tannin was condenced tannin. Fabric dyed with hot water solvent extract showed higher dye uptake than fabric dyed with methanol solvent extract, dye uptake increasing by higher concentration of the dye, longer dyeing time and higher dyeing temperature. And the absorption curve between TCR extract and protein fiber was shaped in the form of Langmuir adsorption isotherm. Fabric dyed without mordant was yellow in color, and when dyed with mordant, fabric showed various colors depending on mordant types except Sn. Color fastness to washing was generally fine and color fastness to light was moderate. But color fastness to rubbing and dry cleaning was outstanding. Lastly, dyed fabrics showed very good antimicrobial activity of 99.9% against Staphylococcus aureus and Kiebsiella pneumoniae.
