Development of New Potato Cultivars for the Utilization of Healthy Food with High Biological Function

  • Potatoes have been recognized for a long time as one of the major food crops as well as horticultural crops. Potato production as a table food has been decreased in developed countries, while it has been steadily increased in the third world countries for it importance as food source. It is a new trend to look for the food, not only as a feeding crop but also healthy food. It is also time for the potato producers to look for the potato having high economic value as found in medicinal plants. There are great diversities in potato species, indicating that valuable compounds can be found in different amounts, depending on potato species. We screened the cultivars, breeding clones, and germplasms based on the vitamin C, Vitamin E, antioxidant compounds, diverse sugar types, important amino acids, and other valuable compounds. We could select the breeding clones KC003, 98Wl17, 99j717, and Vally 8 (A group) due to their high levels of antioxidant compounds, and it can be said that most of the red and purple colored potato clones belong to the A group. In the contents of essential amino acids, ‘Taebook Valley’,‘Summer Valley’ and other breeding clones were found to be high in amount. We also made crosses between breeding clones with high biological function and low agronomic traits and low biological function with high quality in agronomic characteristics . The patterns of genetic trends of these offsprings in comparison with their parents will be reported as well. And the potential of using potato as antibody production of anti-cancer will be discussed.

  • Recently, agriculture and the environment has changed dramatically due to global warming and abnormal weather. In particular, it is necessary to develop new agricultural techniques according to transforming the growing environment of agricultural crops. Therefore, "Smart Farm" building technology for controlling agricultural environment and improving efficiency for ICT technology development has recently been introduced. However, in reality, systematic and objective evaluation items are absent at various levels and management levels that affect the management environment of the smart farm. In this research, it derived the importance index among the factors associated with Smart Farm technology by AHP method. As a result, in order to evaluate comprehensive operation and management of the smart farm, the two evaluation fields(sensor device and control/information management system) were selected as the top evaluation items. These results mean that system that can detect the growth environment information of agricultural crops and control the growing environment is more important than anything, when smart farm is applied. It is judged that the results of this research can be used as basic data for making evaluation indicators associated with the introduction of smart palm technology in the future.

Analysis of Pathogenic Microorganism's Contamination and Heavy Metals on Kimchi Cabbage by Cultivation Methods in Korea (재배농법에 따른 국내산 배추의 위해미생물 및 중금속 오염평가)

  • Kimchi cabbage is one of the four major vegetable crops in Korea. The total annual production of kimchi cabbage, the main material of kimchi, was 20,559 tons in 2015. Kimchi cabbage is one of the majer crops produced by farmers which accounts for about 80% of the total leaf vegetable production in Korea. As the consumption of environmental-friendly agricultural products increases, food safety is one of the major public health concerns. We analyzed the biological hazards of kimchi cabbage produced by two types of cultivation methos such as organic farming and conventional farming using various culture media and microscopy. A total of 432 samples were analysed for presence of sanitary indicator microorganisms (aerobic plate count, coliform count, yeast & mold) and food-borne pathogens (Staphylococcus aureus, environmental Listeria, Bacillus cereus). The population of sanitary indicating microorganisms and food borne pathogens was under 5 Log CFU/g in all tested samples. The results of total microorganism numbers of leaf surface showed a positive correlation to those of soil samples. Additionally, we examined chemical factors such as pesticide residues and heavy metals in soil samples. All tested samples did not shown contamination levels higher than the standard limit.

  • Over the past decade, double-stranded RNA (dsRNA)-mediated gene silencing technology has progressed significantly for pest management in agriculture and for protecting beneficial insects from pathogens. Recently, breakthroughs in RNA interference (RNAi) applications for insect pest management by academia and commercial entities have provided RNAi products as commercial biopesticides. Although RNAi technology has vast potential and advantages for pest control, challenges, and limitations remain in practical applications. This review explores current challenges in the development of dsRNAs as a pest management tool and considers new approaches to overcome biological and environmental obstacles, such as poor stability and resistance.

Growth Response of Lettuce after Application of Mixed Organic Fertilizer Containing Bacillus amyloliquefaciens HSOB-7 and Saccharomyces cerevisiae HSOB-8 (Bacillus amyloliquefaciens HSOB-7와 Saccharomyces cerevisiae HSOB-8 함유 유기질비료 처리 시 상추의 생육)

  • Bacillus spp. and Saccharomyces spp. are effective microorganisms that are applied as microbial fertilizers for crops in Korea. Mixed organic fertilizer (MOF) has been used in the Korean organic agriculture to supply nutrients to crops. This study was conducted to investigate the effects of MOF containing Bacillus amyloliquefaciens HSOB-7 and Saccharomyces cerevisiae HSOB-8 (MOFBS) on the soil chemical properties and growth of lettuce (Lactuca sativa). Growth factors of lettuce were higher after applying different formulation types of MOFBS, such as powder and pellet, than those of NF, and not significantly different than those of control (microbe-free MOF). Compared with the control, application of recommended amount of MOFBS (MOFBS 250 kg/10a, MOFBS treatment) and double the recommended amount (MOFBS 500 kg/10a, 2MOFBS treatment) did not significantly change the growth factors of lettuce. After the application of two formulation types or two different amounts of MOFBS, soil chemical properties such as electrical conductivity, organic matter (OM), exchangeable Ca, and cation exchangeable cation (CEC) increased. These results indicated that although the application of MOFBS improved soil fertility including OM and CEC, it did not significantly affect lettuce growth.

FISH Karyotype Analysis of Four Wild Cucurbitaceae Species Using 5S and 45S rDNA Probes and the Emergence of New Polyploids in Trichosanthes kirilowii Maxim

  • Wild relative species of domesticated crops are useful genetic resources for improving agronomic traits. Cytogenetic investigations based on chromosome composition provide insight into basic genetic and genomic characteristics of a species that can be exploited in a breeding program. Here, we used FISH analysis to characterize the ploidy level, chromosome constitution, and genomic distribution o f 5S and 4 5S r ibosomal DNA (rDNA) in four wild Cucurbitaceae species, namely, Citrullus lanatus (Thunb.) Mansf. var. citroides L. H. Bailey (2n = 22), Melothria japonica Maxim. (2n = 22), Sicyos angulatus L. (2n = 24), and Trichosanthes kirilowii Maxim. (2n = 66, 88, 110 cytotypes), collected in different areas of Korea. All species were diploids, except for T. kirilowii, which included hexa-, octa-, and decaploid cytotypes (2n = 6x = 66, 8x = 88, and 10x = 110). All species have small metaphase chromosomes in the range of $2-5{\mu}m$. The 45S rDNA signals were localized distally compared to the 5S rDNA. C. lanatus var. citroides and M. japonica showed one and two loci of 45S and 5S rDNA, respectively, with co-localization of rDNA signals in one M. japonica chromosome. S. angulatus showed two co-localized signals of 5S and 45S rDNA loci. The hexaploid T. kirilowii cytotype showed five signals each for 45S and 5S rDNA, with three being co-localized. This is the first report of hexaploid and decaploid cytotypes in T. kirilowii. These results will be useful in future Cucurbitaceae breeding programs.

Application of UAV-based RGB Images for the Growth Estimation of Vegetable Crops

  • On-site monitoring of vegetable growth parameters, such as leaf length, leaf area, and fresh weight, in an agricultural field can provide useful information for farmers to establish farm management strategies suitable for optimum production of vegetables. Unmanned Aerial Vehicles (UAVs) are currently gaining a growing interest for agricultural applications. This study reports on validation testing of previously developed vegetable growth estimation models based on UAV-based RGB images for white radish and Chinese cabbage. Specific objective was to investigate the potential of the UAV-based RGB camera system for effectively quantifying temporal and spatial variability in the growth status of white radish and Chinese cabbage in a field. RGB images were acquired based on an automated flight mission with a multi-rotor UAV equipped with a low-cost RGB camera while automatically tracking on a predefined path. The acquired images were initially geo-located based on the log data of flight information saved into the UAV, and then mosaicked using a commerical image processing software. Otsu threshold-based crop coverage and DSM-based crop height were used as two predictor variables of the previously developed multiple linear regression models to estimate growth parameters of vegetables. The predictive capabilities of the UAV sensing system for estimating the growth parameters of the two vegetables were evaluated quantitatively by comparing to ground truth data. There were highly linear relationships between the actual and estimated leaf lengths, widths, and fresh weights, showing coefficients of determination up to 0.7. However, there were differences in slope between the ground truth and estimated values lower than 0.5, thereby requiring the use of a site-specific normalization method.

Estimation of Transpiration Rate with a Metering Pump and its Application in Soilless Culture System (정량펌프를 이용한 무토양재배시스템의 증산량 추정 및 그 응용)

  • Nutrient supplying by a metering pump often produces the inaccuracy in the rate of inflow than expected. In this study, we developed the transpiration estimation system using the metering pump to measure the continuous supplying rate as well as to estimate the accurate transpiration rate. The system showed the stable characteristics by eliminating the fluctuations in the head loss of nutrient tank. The direct proportionality between the supplying time and the supplied nutrient solution was obtained. The exact correlation between the integrated solar radiation and the transpiration rate using the system was calculated, and correlation coefficients between the two factors were 0.98 in the NFT system and 0.92 in the aggregate system. This results suggest that the integrated solar radiation was an important factor to directly decide the supplying volume of nutrient solution in soilless culture system. The deveolped system using the metering pump in the study was able to control the supply of the nutrient solution to the crops adjusting to the variation of solar radiation.

Current status and prospects of genomics and bioinformatics in grapes (포도 유전체 연구현황 및 전망)

  • Grape is one of the important fruit crops around the world, and exposed to disease and pests, and internal or environmental stresses in the vineyards. Breeding and cultivation of new varieties of high quality-grapes resistant to diseases and pests and tolerant to stresses are the most important steps in the grape production. However, conventional breeding has laborious and time-consuming procedures in maintaining and selecting seedlings in the fields. Development of molecular breeding technology through understanding of molecular mechanism of useful traits can be used as an alternative strategy to improve the efficiency of grape breeding program by cross hybridization in grape development programs. The completion of the grape genome sequencing project provided the way to discover the novel genes and to analyze their functions. Comparative genomics, transcriptomic analysis, and the genome-wide identification and analysis of useful genes as well as development of molecular marker for valuable traits could provide novel insights into fruit quality and the responses to diseases and stresses, and can be used as important information in molecular breeding programs for grape development.

Stable Production Technique of Paprika (Capsicum annuum L.) by Hydrogen Peroxide Treatment at Summer (여름철 과산화수소를 이용한 파프리카(Capsicum annuum L.) 안정생산기술)

  • Hydrogen peroxide, which is used in various crops as an oxidizer to improve high temperature adaptation, was evaluated on the effects on productivity and disease incidence in paprika (Capsicum annuum L.) by periodic leaf spray at summer. Hydrogen peroxide treatment not only increased the leaf thickness and SPAD (chlorophyll content) but also the fruit set numbers per plant by 2. Hydrogen peroxide content increase in leaf resulted in increase of catalase and peroxidase activities, and the powdery mildew disease (Leveillula taurica) was also suppressed by the treatment. Transpiration was improved by the reduced leaf stomata resistance in the hydrogen peroxide treatment. Therefore, hydrogen peroxide leaf spray is recommended for improvement of summer productivity in paprika.
