Differential Inhibitory Activities of Four Plant Essential Oils on In Vitro Growth of Fusarium oxysporum f. sp. fragariae Causing Fusarium Wilt in Strawberry Plants

  • The objective of this study was to determine inhibitory activities of four volatile plant essential oils (cinnamon oil, fennel oil, origanum oil and thyme oil) on in vitro growth of Fusarium oxysporum f. sp. fragariae causing Fusarium wilt of strawberry plants. Results showed that these essential oils inhibited in vitro conidial germination and mycelial growth of F. oxysporum f. sp. fragariae in a dose-dependent manner. Cinnamon oil was found to be most effective one in suppressing conidial germination while fennel oil, origanum oil and thyme oil showed moderate inhibition of conidial germination at similar levels. Cinnamon oil, origanum oil and thyme oil showed moderate antifungal activities against mycelial growth at similar levels while fennel oil had relatively lower antifungal activity against mycelial growth. Antifungal effects of these four plant essential oils in different combinations on in vitro fungal growth were also evaluated. These essential oils demonstrated synergistic antifungal activities against conidial germination and mycelial growth of F. oxysporum f. sp. fragariae in vitro. Simultaneous application of origanum oil and thyme oil enhanced their antimicrobial activities against conidial germination and fungal mycelial growth. These results underpin that volatile plant essential oils could be used in eco-friendly integrated disease management of Fusarium wilt in strawberry fields.

Theoretical Design for the Production of Quinoa (Chenopodium quinoa Willd.) in a Closed Plant Factory

  • Quinoa (Chenopodium quinoa Willd.) is a grain crop with high nutritional value. The leaves and sprouts of quinoa can also be consumed either raw or cooked, providing considerably nutritional value as well as high antioxidant and anticancer activities. This study was carried out to obtain basic data to assist in the practical design of a plant factory with artificial lighting for the cultivation of quinoa as a leafy vegetable. We estimated the energy content of the quinoa and the electrical energy required to produce this crop. The yield was 1,000 plants per day, with a planting density and light intensity of $0.015m^2$ ($15{\times}10cm$) and $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. The total number of plants, cultivation area, and electricity consumption were estimated to be 25,000, $375m^2$, and $93,750{\mu}mol{\cdot}s^{-1}$, respectively. White fluorescent lamps were used at a power of 20.4 kW from 1,857 fluorescent lamps (FL, 55 W), and the cost for electricity was approximately 1,820 dollars (exchange rate of $1 = 1,200 won) per month. For a daily harvest of 1,000 plants per day in a closed plant factory, the estimated light installation cost, total installation cost, and total production cost would be 15,473, 46,421, and 55,704 dollars, respectively. The calculated production cost per plant, including labor costs, would be 27 cents for the 25-day cultivation period, with a marketable ratio of 80%. Considering the annual total expenses, income, and depreciation costs, the selling price per plant was estimated to be approximately 56 cents.

Rapid Detection of SdhBP225F and SdhBH272R Mutations in Boscalid Resistant Botrytis cinerea Strains by ARMS-PCR

  • $SdhB^{P225F}$ and $SdhB^{H272R}$ mutations have been found associated with boscalid resistance in Botrytis cinerea from strawberry in Shanghai, China. For rapid detection of two mutations, tetra-primers were designed and optimized to gain the relatively high accuracy and specificity based on the ARMS-PCR technique, by which resistance can be identified with different lengths of products on agarose gels. The tetra-primer ARMS-PCR systems for $SdhB^{P225F}$ and $SdhB^{H272R}$ were validated by 9 SdhB-squenced strains repeatedly. Then, sensitivity of 30 more strains were also tested by the methods, which were accordant with genotypes by sequencing and the sensitivity of conidial germination to boscalid by 100%. Thus, the methods developed in this study are proved to be rapid, inexpensive, accurate and practical for resistance detection of Botrytis cinerea caused by $SdhB^{P225F}$ and $SdhB^{H272R}$ mutations.

Application of Volatile Antifungal Plant Essential Oils for Controlling Pepper Fruit Anthracnose by Colletotrichum gloeosporioides

  • Anthracnose caused by Colletotrichum gloeosporioides has been destructive during pepper fruit production in outdoor fields in Korea. In vitro antifungal activities of 15 different plant essential oils or its components were evaluated during conidial germination and mycelial growth of C. gloeosporioides. In vitro conidial germination was most drastically inhibited by vapour treatments with carvacrol, cinnamon oil, trans-cinnamaldehyde, citral, p-cymene and linalool. Inhibition of the mycelial growth by indirect vapour treatment with essential oils was also demonstrated compared with untreated control. Carvacrol, cinnamon oil, trans-cinnamaldehyde, citral and eugenol were among the most inhibitory plant essential oils by the indirect antifungal efficacies. Plant protection efficacies of the plant essential oils were demonstrated by reduced lesion diameter on the C. gloeosporioides-inoculated immature green pepper fruits compared to the inoculated control fruits without any plant essential oil treatment. In planta test showed that all plant essential oils tested in this study demonstrated plant protection efficacies against pepper fruit anthracnose with similar levels. Thus, application of different plant essential oils can be used for ecofriendly disease management of anthracnose during pepper fruit production.

Genetic diversity analysis of Glycyrrhiza uralensis using 8 novel polymorphic microsatellite markers

  • Licorice plant (Glycyrrhiza spp.) is an important herb, but the major portion of the national demand is imported to Korea because the domestic production base is vulnerable. We performed basic molecular breeding research for domestic cultivation and production. All publicly available G. uralensis EST sequences, which totaled 56,089, were assembled into 4,821 unigenes and examined for microsatellites. Eight polymorphic microsatellite loci were identified and 16 G. uralensis and 6 G. glabra accessions, which were collected from different locations, were genotyped using the microsatellites. Genetic diversity within the accessions was estimated by construction of a dendrogram. The dendrogram was clustered into two groups. The results showed that there is a correlative genetic relationship between species. The microsatellite markers were found to be useful for diversity analysis as they are able to successfully distinguish the Glycyrrhiza accessions.

2007년 우리나라 주요 작물 바이러스병 발생 상황 (Survey of Viral Diseases Occurrence on Major Crops in 2007)

  • The severe damage induced by the important viruses of Rice stripe virus (RSV), Cucumber green mottle mosaic virus (CGMMV), Melon necrotic spot virus (MNSV), Tomato spotted wilt virus (TSWV) and Tomato bushy stunt virus (TBSV) was described on major crops in Korea. In 2007, the plot incidence rate of RSV was 100% on the precocious rice cultivars at the Western coastal provinces of Gyeonggido, Chungcheongnamdo, Jellabugdo and Jellanamdo, and Jejudo. RSV occurred in 2,441 ha with incidence rate of 70% over at 5 areas of Seocheon, Seosan, Boryung, Hongsung and Buyou in Chungcheongnamdo. At 4 areas of Buan, Gimje, Gunsan and Gochang in Jellabukdo, RSV occurred in 2.016 ha. CGMMV occurred on watermelon in 4.6 ha at Cheongyang area, and its outbreak was also 890 ha on oriental melon for 120 farmers with the incidence area of 23% against total cultivation areas of Seongju. MNSV was recorded firstly on watermelon in 2006 at Andong and it spread to 3 areas of Hapcheon, Gochang and Yanggu. TSWV occurred firstly at Danggin in Chungcheongnamdo in 2005. TSWV in 2006 spread to 6 areas; Taian, Hongsung and Seosan in Chungcheongnando, Namwon in Jellabukdo, and Sunchon and Kwangju in Jellanamdo. In 2007, TSWV covered 17 areas of western and southern parts; the 5 area including Taian in Chungcheongnamdo, Kwangju in Jellanamdo, Bucheon in Gyunggido, and so forth. TBSV was described firstly on table tomato at Sacheon in Kyungsangnamdo in 2004. TBSV occurred on cherry tomato at Chungju in 2006 and on table tomato at Busan area.

장미 줄기에 발생하는 식물병의 종류 및 증상 (Plant Diseases Occurring on Rose Stem)

  • 장미는 세계적으로 가장 많이 재배되고 있는 화훼작물로서 최근에 양액재배 및 토양재배 되는 장미에서 발생하는 줄기마름증상에 대한 연구를 수행하였다. 장미의 가지나 줄기가 말라죽는 증상은 크게 4 가지 병에 의한 것으로 조사되었는데, Botrytis cinerea에 의한 잿빛곰팡이병, Coniothyrium fuckelii에 의한 가지 마름병, Colletotrichum gloeosporioides에 의한 탄저병, 그리고 Pythium sp. 에 의한 뿌리썩음병이었다. 이들 병의 진단과 방제에 도움이 되고자 병 증상과 병원균의 특성에 대하여 기술하였다.
