THE ANTIMICROBIAL EFFECT OF HORSERADISH(ARMORACIA RUSTICANA) ROOT EXTRACTS AGAINST STREPTOCOCCUS MUTANS ISOLATED FROM HUMAN DENTAL PLAQUE (치태에서 분리된 Streptococcus mutans에 대한 서양산 고추냉이(Armoracia rusticana) 뿌리 추출물의 항균효과)

  • Recently interesting in development of antimicrobial agent from natural origin has been increased in these days. Many studies have been reported antimicrobial effect of Horseradish(Armoracia rusticana) root extracts against various microorganisms such as Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa, Vibrio parahaemolyticu, Staphylococcus aureus, Bacillus subtilis, Aspergillus parasiticus, Helicobacter pylori. The main component related to antimicrobial activity in horseradish is well known as allyl isothiocyanate(AIT). In this study, we investigated the antimicrobial effects of Horseradish(Armoracia rusticana) root extracts against Streptococcus mutans isolated from human dental plaque, Streptococcus mutans reference strain and compared with that of chlorhexidine. Horseradish root extracts and chlorhexidine were tested to determine their minimum inhibitory concentration( MIC) and minimum bactericidal concentration(MBC). The result of this study can be summerized as follows; 1. Horseradish root extracts showed antimicrobial effect against both S. mutans isolated strain and reference strain, their MIC were respectively $0.083{\sim}0.25%$ ($833.33{\sim}2500 ppm$), 0.25% (2500 ppm). Horseradish root extracts showed antimicrobial effect against S. mutans isolated strain at same or slightly lower concentration compared with MIC of reference strain. 2. $0.083{\sim}0.25%$ horseradish root extracts showed similar antimicrobial effect with chlorhexidine ($0.0021{\sim}0.0041%$).

  • In this study, the antimicrobial effects of Horseradish (Armoracia rusticana) root extracts against oral pathogens were investigated, and also compared with that of chlorhexidine. The following 7 microorganisms were used in this study, Streptococcus mutans ATCC 25175, Streptococcus sobrinus(d) ATCC 27607, Lactobacillus casei ATCC 393, Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212, Actinobacillus actinomycetemcomitans ATCC 29522. Candida albicans ATCC 10261. Horseradish root extracts and chlorhexidine were tested to determine their minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC). The results of this study can be summarized as follows : 1. Horseradish root extracts showed antimicrobial effect against the tested oral pathogens. MIC and MBC of this extracts were 30-125, 125-500ppm, respectively. Especially, it was the most effective against C. albicans of other tested microorganisms. 2. Chlorhexidine also showed antimicrobial effect against the tested oral pathogens. MIC of chlorhexidine range between 0.15 and 2.5%, MBC are 0.4-2.5%. In conclusion, it was suggested that AIT had similar antimicrobial effects in the lower concentration, compared with that of chlorhexidine.

  • Microorganisms are the main causative factors of pulpal and periapical diseases, therefore successful endodontic treatment is depend on the effective elimination of intracanal bacterial populations. Many studies have been reported antimicrobial effect of Allyl isothiocyanate (AIT) which the principle ingredient of Horseradish (Armoracia rusticana) root extracts. The purposes of this study are to evaluate the antimicrobial effectiveness of Horseradish root extracts against Enterococcus faecalis in root canals of extracted human teeth and compare to sodium hypochlorite (NaOCl). Extracted human mandibular premolar root canals were infected with E. faecalis for 21 days, and then irrigated with Horseradish root extracts, NaOCl solution and saline. After canal irrigation, first samples (S1) were taken. After first sampling, the canals were additionally incubated 7 days, and then second samples (S2) were taken. The samples were inoculated on EHI agar plate to determine the colony forming units (CFU). 1. Mean values of CFU in S1 were $5.815{\times}10^3$ CFU/ml at Horseradish groups, and $3.465{\times}10^3$ CFU/ml at NaOCI groups. There was no statistically significant differences (p=0.086). 2. Mean values of CFU in S2 were $3.100{\times}10^3$ CFU/ml at Horseradish groups, and $5.252{\times}10^5$ CFU/ml at NaOCI groups. There was statistically significant difference (p<.05). 3. There was no statistically significant differences (p=0.076) between S1 and S2 at Horseradish groups in the mean values of CFU. However, there was statistically significant differences (p<.05) between S1 and S2 at NaOCI groups in the mean values of CFU.

STUDY ON COMPARISON OF ANTIMICROBIAL ACTIVITY BETWEEN HORSERADISH(ARMORACIA RUSTICANA) ROOT EXTRACTS AND SYNTHETIC ALLYL ISOTHIOCYANATE AGAINST ORAL PATHOGENIC MICROORGANISMS (구강 내 미생물에 대한 서양산 고추냉이 (Horseradish, Armoracia rusticana) 뿌리 천연추출물과 합성 Allyl isothiocyanate의 항균활성 비교)

  • Allyl isothiocyanate (AIT), the principle ingredient of antimicrobial ingredients from horseradish root, can be prepared from extracts of horseradish root or synthetic method. It is reported that the horseradish root extract has the antimicrobial effect against various oral microorganisms, while there is no further study about the antimicrobial effect against the oral microorganisms of synthetic AIT derived from synthetic method. The aim of the study is to compare the difference of the antimicrobial effect between horseradish root extracts and synthetic AIT. To evaluate the antimicrobial effect, we measured the minimum inhibitory concentration(MIC) and minimum bactericidal concentration (MBC), and the results are like following. 1. The MIC of horseradish root extract against 7 kinds of oral pathogenic microorganisms is about 117$\sim$1,750 ppm(0.0117$\sim$0.175%), and the MIC of the synthetic AIT is about 344$\sim$3,000 ppm(0.0344$\sim$0.3%), which have the antimicrobial effects against all kinds of microorganisms. 2. The MBC of the horseradish root extracts against the 7 kinds of oral microorganisms is about 625.2$\sim$6,000 ppm(0.06252$\sim$0.6%), and the MBC of the synthetic AIT is about 1,750$\sim$7,000 ppm(0.175$\sim$0.7%), which have the antimicrobial effects against all kinds of microorganisms.

  • Objective : The aim of this study was to determine the antimicrobical effect of horseradish root extracts against Enterococcus faecalis and Fusobacterium nucleatum isolated from oral cavity compared with reference strain, and compared with that of chlorhexidine. Method : Horseradish root extracts and chlorhexidine were sequentially diluted and tested against anaerobes(E. faecalis, F. nucleatum) isolated from children's oral cavity. The microbes were anaerobically incubated and the minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were detected. Results :1. Horseradish root extracts showed antimicrobial effect against E. faecalis isolated strain at same or slightly higher concentration compared with MIC of reference strain. 2. $625.0\sim1,250.0{\mu}g/ml$ horseradish root extracts showed similar antimicrobial effect with chlorhexidine($7.8\sim15.6{\mu}g/ml$). 3. Horseradish root extracts showed antimicrobial effect against F. nucleatum isolated strain at same or slightly higher concentration compared with MIC of reference strain. 4. $78.1\sim312.5{\mu}g/ml$ horseradish root extracts showed similar antimicrobial effect with chlorhexidine($7.8\sim15.6{\mu}g/ml$). Conclusions : The results of this study confirm that horseradish root extracts has antimicrobial effect against anaerobes isolated from oral cavity as well as reference strain. And we found the potential of horseradish root extracts as a canal irrigant or disinfectant.

A CONTROL OF PLAQUE, GINGIVITIS AND ANTI-CARIES EFFECT OF DENTIFRICE CONTAINING HORSERADISH EXTRACTS (서양산 고추냉이 추출물이 함유된 세치제의 효능에 대한 임상적 실험연구)

  • The purpose of this study was to find out the effect of dentifrice containing Horseradish (Armoracia rustica) root extracts on improvement of oral hygiene. The clinical evaluation was done participated by 80 adults volunteers who provided a informed consent for their participation. The participants were divided into two groups ; control group (using toothpaste without Horseradish extracts) and experimental group (using toothpaste containing Horseradish extracts). The checklist of the experiment includes the plaque index, gingival index and anti-caries activity. The anti-caries activity was measured by cultivating Streptococcus mutans for 48 hours then estimating the number of increased bacteria using Dentocult SM. The double-blind method was used in this study. The results of this study can be summarized as follows; 1. The plaque formation decreased 28% in control group, 58% in experimental group using dentifrice with Horseradish extracts compared to baseline data. 2. For gingival index, control group and experimental group showed 26% and 40% decrease compared to baseline data, respectively. 3. The score of caries activity test decreased by 4% for control group and 69% for experimental group compared to baseline data. According to the results, a dentifrice containing Horseradish extracts can improve oral hygiene.

THE ANTIMICROBIAL EFFECT OF HORSERADISH (ARMORACIA RUSTICANA) ROOT EXTRACTS AGAINST OBLIGATE ANAEROBES IN ROOT CANAL (치근관 내 편성 혐기성 세균에 대한 서양산 고추냉이 뿌리 추출물의 항균효과)

  • When the symptom of periapical infection is not released by mechanical instrumentation. anti-microbial agents including antibiosis become necessary in order to remove microorganisms from the root canal. Since anti-microbial agents of natural origins are currently popular, more natural remedies are being sought out. As it turns out, it is well known isothiocyanates (ITCs) in horseradish root extract have anti-microbial activity from many studies. In this research, anti-microbial effects of horseradish root extract and chlorhexidine, a typical anti-microbial agent, were investigated and compared against two kinds of obligate anaerobes. Fusobacterium nucleatum and Prevotella nigrescens, that are often discovered in infected root canal, and Clostridium perfringens, which is resistant to antibiotics and frequently used as a control strain for antibacterial studies 1. The MIC and MBC of horseradish root extract were ranged from 87 to 470 ppm and from 156 to 625 ppm against three kinds of obligate anaerobes, respectively. Horseradish root extract showed the strongest anti-bacterial activity (MBC, 156 ppm) against F. nucleatum and also showed anti-bacterial activity against antibiotic resistant obligate anaerobes. C. perfringens. 2. The MIC and MBC of chlorhexidine were ranged from 3.12 to 6.25 ppm and 10.94 ppm against three kinds of obligate anaerobes, respectively. 3. The MIC with 87-470 ppm of horseradish root exact has the same growth inhibiting effect as the one of 3.12-6.25 ppm of chlorhexidine. Likewise, the MBC with 156-625 ppm of horseradish has the similar bactericidal effect as 10.94 ppm of chlorhexidine.
