Effect of KiFAY on Performance, Insulin-like Growth Factor-1, and Thyroid Hormones in Broilers

  • A comparative study was performed to investigate the efficacy of KiFAY as a feed additive on performance parameters, thyroid, and pancreatic hormone levels in broilers. Ninety birds (Vencobb 400) were randomly divided into three groups viz., Control (no DL-methionine supplementation), Treatment1 (containing added DL-methionine) and Treatment 2 (containing KiFAY and without DL-methionine supplementation). The performance parameters (weekly body weight, body weight gain, feed intake, and feed consumption ratio) were recorded and calculated during the whole study of 4 weeks. Analyses of insulin and insulin-like growth factor (IGF 1), triiodothyronine (T3), thyroxine (T4) and thyroid stimulating hormone (TSH) were performed at the end of the study. The results show that birds on supplementation of KiFAY performed significantly (p<0.001) better than other treatments. The weekly body weight, body weight gain, feed in-take and feed consumption ratio improved in KiFAY treated birds. The study found an increase in insulin and IGF1 levels (p<0.001) in KiFAY compared with the other treatments. Serum T3, T4, and TSH levels in the Treatment 2 were higher than other treatments (p<0.001). The KiFAY supplementation was able to improve performance with associated responses at a hormonal level in broilers.

Effect of Recombinant Bovine Somatotropin on Growth of Olive Flounder, Paralichthys of olivaceus

  • The recombinant bovine somatotropin (rBST) was administered to the oliver flounder, Paralichthys olivaceus, to know the effects and optimal administration frequency and dosage of the hormone. The experiment was conducted with three different treatment groups (A, B and C) designated based on the duration and administration frequency of rBST and one control (D) from April 14, 1996 to March 16, 1997. The fish of hormone treated groups grew 7.86 to 10.07% (47.45 to 60.75 g in weight) better than the control at the end of the experiment (P<0.05). No significant differences in their growth were detected among treatment groups. The distinct growth improvement was recognized four weeks after completion of the first four hormone administration. When considering water temperatures measured from the experimental tanks, the effect of rBST on the flounder was greater during the period showing relatively lower temperature. The survival rates were higher in treatment groups than in the control, revealing 98.3% (A), 98.4% (B), 97.7% (C) and 93.1% (D) during the first stage of culture; 92.7% (A), 91.3% (B), 86.7% (C) and 80.0% (D) during the second stage of culture.

Effect of rGH on Body Growth and Udder Development on Korean Native Heifers (외인성 성장호르몬이 한우의 성장 및 유방의 발달에 미치는 영향)

  • This experiment was carried out with 12 Korean native heifers(8~12month old, body weight, 160~240kg) raised at a farm of Chang-Soo Livestock Cooperatives to evaluate the effects of rGH(recombinant growth hormone) on serum concentrations of growth hormone, estrogen, and IGF-I, weight gain, teat volume gain and processing enzyme activity of IGF-I, binding protein III at 28 day intervals. Animals used were injected with 250mg rGH at 14 day intervals from December to Ferbruary in 1994. The significant difference was found in the group of treatment on the 4th week in the endogenous GH(p<.01) and 8th week in estrogen and IGF-I(p<.05) after injectin of rGH in Korean native heifers. There were significant differences between control group and treatment group in weight and teat volume on 8th week after treatment(p<.05). Processing enzyme activity before injection of rGH were low. However, heifers injected with 250mg of rGH showed that processing enzyme activity of IGF binding protein was highly increased throughout the experiment. Present results suggest that injection of exogenous rGH to heifers can increase the growth performance and udder development of Korean native heifers by the endogenous hormonal changes.

  • Prader-Willi syndrome (PWS) is a genetic disorder that is considered, especially on child, to cause poor feeding, hypotonia, failure to thrive, developmental delay and hypogonadism which is known to affect between 1 in 10,000 and 30,000 people. The children with PWS are viewed as affected by growth hormone (GH) insufficiency, although the exact mechanisms of GH deficiency are not fully understood. However, the benefits of GH treatment in children with PWS are well established. Myers, et al. (2006), Grugni, et al. (2016) indicated its positive effects on linear growth, body composition, motor function, respiratory function and psychomotor development. Despite of its effectiveness and advantages had been well known and proven in many other studies, there is only one recombinant GH product that is approved for PWS in Korea, $Genotropin^{(R)}$, till now. A phase III clinical study of GH treatment with $Eutropin^{TM}$, in 34 Korean PWS children is in progress, which is expected to have comparable effects and safety profile with the active control by assessing auxological changes such as height standard deviation score, body composition changes such as lean body mass and percent body fat, motor and cognitive development using Bayley scale, and safety profiles.

Efficacy of corifollitropin alfa followed by recombinant follicle-stimulating hormone in a gonadotropin-releasing hormone antagonist protocol for Korean women undergoing assisted reproduction

  • Objective: To evaluate the effect of a gonadotropin-releasing hormone (GnRH) antagonist protocol using corifollitropin alfa in women undergoing assisted reproduction. Methods: Six hundred and eighty-six in vitro fertilization-embryo transfer (IVF)/ intracytoplasmic sperm injection (ICSI) cycles were analyzed. In 113 cycles, folliculogenesis was induced with corifollitropin alfa and recombinant follicle stimulating hormone (rFSH), and premature luteinizing hormone (LH) surges were prevented with a GnRH antagonist. In the control group (573 cycles), premature LH surges were prevented with GnRH agonist injection from the midluteal phase of the preceding cycle, and ovarian stimulation was started with rFSH. The treatment duration, quality of oocytes and embryos, number of embryo transfer (ET) cancelled cycles, risk of ovarian hyperstimulation syndrome (OHSS), and the chemical pregnancy rate were evaluated in the two ovarian stimulation protocols. Results: There were no significant differences in age and infertility factors between treatment groups. The treatment duration was shorter in the corifollitropin alfa group than in the control group. Although not statistically significant, the mean numbers of matured (86.8% vs. 85.1%) and fertilized oocytes (84.2% vs. 83.1%), good embryos (62.4% vs. 60.3%), and chemical pregnancy rates (47.2% vs. 46.8%) were slightly higher in the corifollitropin alfa group than in the control group. In contrast, rates of ET cancelled cycles and the OHSS risk were slightly lower in the corifollitropin alfa group (6.2% and 2.7%) than in the control group (8.2% and 3.5%), although these differences were also not statistically significant. Conclusion: Although no significant differences were observed, the use of corifollitropin alfa seems to offer some advantages to patients because of its short treatment duration, safety, lower ET cancellation rate and reduced risk of OHSS.

Development of Two-Component Nanorod Complex for Dual-Fluorescence Imaging and siRNA Delivery

  • Recently, multifunctional nanomaterials have been developed as nanotherapeutic agents for cellular imaging and targeted cancer treatment because of their ease of synthesis and low cytotoxicity. In this study, we developed a multifunctional, two-component nanorod consisting of gold (Au) and nickel (Ni) blocks that enables dual-fluorescence imaging and the targeted delivery of small interfering RNA (siRNA) to improve cancer treatment. Fluorescein isothiocyanate-labeled luteinizing hormone-releasing hormone (LHRH) peptides were attached to the surface of a Ni block via a histidine-tagged LHRH interaction to specifically bind to a breast cancer cell line, MCF-7. The Au block was modified with TAMRA-labeled thiolated siRNA in order to knock down the vascular endothelial growth factor protein to inhibit cancer growth. These two-component nanorods actively targeted and internalized into MCF-7 cells to induce apoptosis through RNA interference. This study demonstrates the feasibility of using two-component nanorods as a potential theranostic in breast cancer treatment, with capabilities in dual imaging and targeted gene delivery.

Treatment of Differentiated Thyroid Cancer (분화된 갑상선암의 치료)

  • During the last several decades, prognosis of differentiated thvroid cancers improved markedly, mainly due to refinement of surgical techniques and routine use of radioactive iodine. Total or neat-total thyroidectomy is to be performed as the first line of treatment. Routine remnant ablation just after surgery decreases recurrence and increases survival. After then patients are maintained on suppressive dose of thyroid hormone to keep endogenous TSH below normal. Regular follow-up of the patients with serum thyroglobulin measurement and iodine whole body scan after thyroid hormone withdrawal or under recombinant human thyrotropin stimulation select high risk patients with recurrence or metastatic disease and treatment with therapeutic dose of radioactive iodine prolongs survival and sometimes leads cure. Currently recommended diagnostic and therapeutic strategies, present controversies and future directions are discussed.

Effects of Taxilli Ramulus Extract on Bone Metabolism of Ethanol Treated Rats (상기생이 ethanol을 장기 투여한 흰쥐의 골 대사에 미치는 영향)

  • Objectives : To investigate the effect of Taxilli Ramulus (TR) extract on bone metabolism of ethanol-treated animal model. Methods : The changes of serum calcium, calcitonin, estrogen level, a1ka1ine phosphatase activity, osteocalcin, parathyroid hormone content and urine calcium level were observed with ethanol treatment for 60 days. The results were compared with an ethanol- TR extract double treatment group. Results : We observed increment of serum osteocalcin, parathyroid hormone content, alkaline phosphatase activity and urine calcium level by chronic ethanol feed and they were recovered to near normal level with Taxilli Ramulus extract treatment. Weight gain, serum calcium level, calcitonin and estrogen content were remarkably reduced with ethanol treatment and their levels were normalized by Taxilli Ramulus extract. Conclusions : These results showed that Taxilli Ramulus extract have the ability to recover to normal in the body an abnormal calcium metabolism process due to external factors. These results suggested that Taxilli Ramulus extract have preventive effects on calcium concentration loss and osteoporosis.

The influence of gonadotropin-releasing hormone agonists on anthropometric change in girls with central precocious puberty

  • Purpose: The potential effect of gonadotropin-releasing hormone agonist (GnRHa) treatment on the weight of girls with central precocious puberty (CPP) remains a controversy. We investigated anthropometric changes during and after GnRHa treatment among girls with CPP. Methods: This retrospective study evaluated data from 127 girls with CPP who received GnRHa treatment for ${\geq}2years$. Height, weight, and body mass index (BMI) values were compared at the baseline (visit 1), after 1 year of GnRHa treatment (visit 2), the end of GnRHa treatment (visit 3), and 6-12 months after GnRHa discontinuation (visit 4). Results: The height z score for chronological age (CA) increased continuously between visit 1 and visit 4. No significant differences were observed in BMI z score for CA between visits 1 and 4. However, an increasing trend in the BMI z score for bone age (BA) was observed between visits 1 and 4. The numbers of participants who were of normal weight, overweight, and obese were 97, 22, and 8, respectively, at visit 1, compared to 100, 16, and 11, respectively, at visit 4 (P=0.48). Conclusion: Among girls with CPP, the overall BMI z score for CA did not change significantly during or after GnRHa treatment discontinuation, regardless of their BMI status at visit 1. However, the BMI z score for BA showed an increasing trend during GnRHa treatment and a decreasing trend after discontinuation. Therefore, long-term follow-up of BMI changes among girls with CPP is required until they attain adult height.

A Case Report for One Case of Thyrotoxic Periodic Paralysis Patient

  • Objective : The thyrotoxic periodic paralysis mainly appears in case of hyperthyroidism and the amyotonia is the characteristic main symptom. Through the deteriorated case and treatment by oriental medicine, the purpose of this research is to study the change of symptoms and the treatment effect for one case of a thyrotoxic periodic paralysis patient, Methods and Results : The treatment was carried out with oriental medicine as a main treatment and by the oriental medicinally deteriorated case, acupuncture treatment, moxibustion treatment and herbal medicine treatment were used as the main treatment methods. Also based on the symptoms which could appear with hyperthyroidism, the changes of symptoms were evaluated once a day with 4 levels of subjective scales that the patient was feeling, by dividing with 4 aspects of totalis symptoms, cardiovascular symptoms, musculoskeletal symptoms and other symptoms. In addition, it was shown that through the continuous hematologic test, the subjective and objective symptoms were mostly decreased during the treatment periods by comparing with the objective thyroid hormone (T3, T4, TSH) levels and the electrolyte levels. Conclusion : With this result, it was considered that oriental medicinal treatment was very effective for the thyrotoxic periodic paralysis through the oriental medicinally deteriorated case.
