A Report on the Effect of Jowisengchung-tang in 2 Cases of Precocious Puberty (조위승청탕(調胃升淸湯)을 투여한 성조숙 증상을 주소로 한 여아의 한방치료 2례)

  • Objectives: These cases are that of three girls who are diagnosed as precocious puberty. It seems that the incidence of precocious puberty is rapidly increasing these days. In addition to the psychosocial disturbances associated with precocious puberty, the premature pubertal growth spurt and the accelerated bone maturation result in reduced adult height. The aim of this study was to report the effect of Jowisengchung-tang in 3 cases of precocious puberty. Methods: Retrospective and comparative analysis of 3 children who had been diagnosed with preco cious puberty. Anthropometric measurements including height, weight, body fat, body fat percent, AHP and body mass index were measured. Endocrine investigations including estradiol, luteinizing hormone(LH), FSH were conducted. Pubertal stages were determined with a questionnaire using Tanner stages. Jowisengchung-tang was given to 3 precocious puberty girls. Results: After treatment, the hormone(estradiol index) and accompanying symptoms (breast bud with elevation of breast and papilla; enlargement of areola) were reduced compared with first visit day. Conclusions: We may conclude that Korean traditional treatment of Jowisengchung-tang is effective in patients with precocious puberty and menopausal symptoms.

Transmucosal Delivery of Luteinizing Hormone-Releasing Hormone: Effect of Medium Chain Fatty Acid Salts on Stabilization of LHRH in Mucosal Homogenates in vitro. (황체호르몬 유리호르몬의 경점막 수송: 가토 점막균질액 중에서 중쇄지방산염의 LHRH에 대한 안정화 효과)

  • In order to investigate the feasibility of transmucosal delivery of the model peptide, LHRH, metabolism of LHRH and inhibition effect of medium chain fatty acid salts were studied in rabbit mucosal homogenate. LHRH incubated in homogenates of rectal(RE), nasal(NA) and vaginal(VA) mucosa were assayed by HPLC. Five to six degradation products of LHRH were deterted and the degradation of LHRH$(500\;{\mu}g/ml)$ followed the first order kinetics. The main degradation products were found as $LHRH^{1-5}(M-I)$, $LHRH^{1-3}(M-II)$ and $LHRH^{1-6}(M-III)$ by the method of amino acid analysis. The half-lives of LHRH in the mucosal homogenates were found to be less than 20 min at protein concentration of 2.5 mg/ml with the order of VA>NA>RE mucosal homogenate. Medium chain fatty acid salts such as sodium caprylate $(C_8)$, sodium caprate $(C_{10})$ and sodium laurate $(C_{12})$ at the concentration of $0.5%{\sim}1.0%$ inhibit the proteolysis of LHRH significantly. The addition of sodium laurate(0.5%) into the NA and VA mucosal homogenates protected LHRH completely from the degradation.

  • A computer simulation of biochemical process was carried out using Macintosh-based BioDesignerTM developed at Bioprocess and Engineering Center(BPEC) of MIT. Based on the assumptions and flask culture experiments, a porcine growth hormone (PGH, Porcine Somatotropin) production process was simulated by a two-stage continuous culture. The economical and sensitivity analyses were evaluated for the scale-up production of PGH. A high return on investment (ROI, 104%/year) suggested that the process be profitable. However, sensitivity analysis indicated that ROI was dependent on the yield of PGH, selling price, dose and the achievement of projected market penetration.

  • An immunoassay techniques using monoclonal antibodies have been developed of the quantitative analysis of endogenous hormones in plants. In order to be useful for immunoassay, both a high degree of specificity and a high affinity are required. A system is described for production of hybridomas which secrete antibodies against the plant hormone. Using the system we were able to produce hybridmas with the desired antibody specificity by cell fusion and culture method. For a number of obvious reasons, monoclonal antibodies(mAb) were superior to polyclonal antibodies.

  • Transcript profiling is a particularly valuable tool in the field of steroid receptor biology, as these receptors are ligand-activated transcription factors and therefore exert their initial effects through altering gene expression in responsive cells. Also, an awareness of endocrine disrupting chemicals (EDCs) and their potential screening methods to identify endocrine activity have been increased. Here we developed an in-house cDNA microarray, named KISTCHIP-400 ver. 1.0, with 416 clones, based on public database and research papers. These clones contained estrogen, androgen, thyroid hormone & receptors, sex hormone signal transduction & regulation, c-fos, c-myc, ps2 gene, metabolism related genes etc. Also, to validate the KISTCHIP-400 ver. 1.0, we investigated gene expression profiles with reference hormones, $10^{8}\;M\;17{\beta}-estradiol,\;10^{-7}\;M\;testosterone\;and\;10^{-7}\;M$ progesterone in MCF-7 cell line. As the results, gene expression profiles of three reference hormones were distinguished from each other with significant and identified 33 $17{\beta}-estradiol$ responsive genes. This study is in first step of validation for KISTCHIP-400 ver. 1.0, as following step transcriptional profile analysis on not only low concentrations of EDCs but suspected EDCs using KISTCHIP-400 ver. 1.0 is processing. Our results indicate that the developed microarray may be a useful laboratory tool for screening EDCs and elucidating endocrine disrupting mechanism.

  • Objectives: After the discontinuance of the Women's Health Initiative (WHI)clinical trial in the U.S. in July 2002, I thought that the domestic medical field would come to a crisis in hormonal treatment for perimenopausal and postmenopausal women. However, the domestic condition of HRT has barely changed. This study was designed to investigate world research trends in HRT and bring forward the response of the Traditional Korean Medicine (TKM) Research Group. Methods: I investigated recent domestic ;md world research trends about HRT and climacteric syndrome. Based on this analysis, I predicted western medical research trends and direction for HRT and medical care of perimenopausal and postmenopausal women. Results and Conclusions: I propose that we (TKM Research Group) have to explain the side effects of HRT for postmenopausal women and try to suggest complementary therapy for it. For that aim, the Association of Korean Medicine's effort is very important, as well as personal practitioner's actions.

  • Prader-Willi syndrome (PWS) is a complex multisystem genetic disorder that is caused by the lack of expression of paternally inherited imprinted genes on chromosome 15q11-q13. This syndrome has a characteristic phenotype including severe neonatal hypotonia, early-onset hyperphagia, development of morbid obesity, short stature, hypogonadism, learning disabilities, behavioral problems, and psychiatric problems. PWS is an example of a genetic condition caused by genomic imprinting. It can occur via 3 main mechanisms that lead to the absence of expression of paternally inherited genes in the 15q11.2-q13 region: paternal microdeletion, maternal uniparental disomy, and an imprinting defect. Over 99% of PWS cases can be diagnosed using DNA methylation analysis. Early diagnosis of PWS is important for effective long-term management. Growth hormone (GH) treatment improves the growth, physical phenotype, and body composition of patients with PWS. In recent years, GH treatment in infants has been shown to have beneficial effects on the growth and neurological development of patients diagnosed during infancy. There is a clear need for an integrated multidisciplinary approach to facilitate early diagnosis and optimize management to improve quality of life, prevent complications, and prolong life expectancy in patients with PWS.

A Study on Vitamin D Status and Factors Affecting It in Young Adults (정상 성인의 혈청 Vitamin D 수준과 이에 영향을 주는 변인에 관한 연구)

  • In this study, the serum level of 25-hydroxyvitamin D(25-(OH)D) was measured by high pressure liquid chromatography(HPLC), and factors affecting it were investigated in 72 young adults age ranging from 21 years to 39 years with normal bone density. The mean level of serum 25-(OH)D was 20.0$\pm$6.8ng/ml in males and 26.1$\pm$12.3ng/ml in females, which was significantly higher in females (p<0.01). The serum level of parathyroid hormone(PTH) showed a negative correlation with that of 25-(OH)D(p<0.05). Time spent outdoors in a day correlated positively with the serum level of 25-(OH)D(p<0.01). During the day, a specific time between 12:00 a.m. and 2:00 p.m. showed the most significant correlation with the level of 25-(OH)D(p<0.005). Among the nutrients studied, fat and vitamin D intake were positively correlated with the serum 25-(OH)D level. Stepwise multiple regression analysis showed that the serum level of 25-(OH)D could be fit by vitamin D intake(34.7% explained), serum PTH level (27.3% explained) and the time spent outdoors during the specific time(28.4% explained).1996)

Biochemical and Structural Analysis of Hormone-sensitive Lipase Homolog EstE7: Insight into the Stabilized Dimerization of HSL-Homolog Proteins

  • Hormone sensitive lipase (HSL) plays a major role in energy homeostasis and lipid metabolism. Several crystal structures of HSL-homolog proteins have been identified, which has led to a better understanding of its molecular function. HSL-homolog proteins exit as both monomer and dimer, but the biochemical and structural basis for such oligomeric states has not been successfully elucidated. Therefore, we determined the crystal structure of HSL-homolog protein EstE7 from a metagenome library at $2.2\;{\AA}$ resolution and characterized the oligomeric states of EstE7 both structurally and biochemically. EstE7 protein prefers the dimeric state in solution, which is supported by its higher enzymatic activity in the dimeric state. In the crystal form, EstE7 protein shows two-types of dimeric interface. Specifically, dimerization via the external ${beta}8$-strand occurred through tight association between two pseudosymmetric folds via salt bridges, hydrogen bonds and van der Waals interactions. This dimer formation was similar to that of other HSL-homolog protein structures such as AFEST, BEFA, and EstE1. We anticipate that our results will provide insight into the oligomeric state of HSL-homolog proteins.

Stress Responses of Eurasian Otter (Lutra lutra) in Relation to Habitats and Season (계절과 서식지 유형에 따른 국내 Eurasian otter (Lutra lutra)의 스트레스 반응)

  • In order to understand the stress responses, we measured the concentration of stress hormones by using the fresh feces of wild otters in the Gwangju stream and Seomjin river during summer and winter in 2018. We compared seasonal and regional number of spraints (=otters' feces), temperature, altitude, water width, land cover, and water quality with stress hormones. The stress hormone levels were higher in summer than in winter, and significantly different to season and region. In addition, the number of feces were different according to the season and region, and showed a negative correlation with stress hormone. This study suggested assessing stress hormones as another promising method for the analysis of status of wild otters and it will contribute to establish the management and conservation policy, especially in urban area.
