A cDNA encoding the defensin homologue of a novel family member was isolated from the cDNA library of the firefly,Pyrocoelia rufa. Sequence analysis of the cDNA encoding the defensin homologue of P. rufa resulted that the 165 bp cDHA has an open reading frame of 55 amino acid residues. The deduced amino acid sequences of the defensin homologue gene from P. rufa showed identity to known mammalian defensins. Also 6 cystein residues in the P. rufa defensin homologue gene were conserved in the same position as those of known mammalian defensins. The result suggested that P. rufa defensin homologue is a novel member of the insect defensin family. Southern blot analysis suggests that there may be a single copy number of the P.rufa defensin homologue gene and their fat body-specific expression pattern at the transcriptional level was confirmed by Northern blot analysis.

This study investigated on concentrations of PCDD/Fs and the homologue profiles deposited on pine needles and soil collected from different areas. As, in an area, the concentration of ${\sum}PCDD/Fs$ of pine needles decreased, the percentages of the higher chlorinated homologues of pine needles decreased, and those of lower chlorinated ones increased. However on the contrary the percentages of the higher chlorinated homologues of soil increased and those of lower chlorinated ones decreased. The concentrations of PCDD/Fs deposited on pine needles only depended upon those of PCDD/Fs in air, while the concentration of PCDD/Fs deposited on soil depended upon those of organic matter contents as well as those of PCDD/Fs in air. The results showed that pine needles rather than soil were adequate as a passive sampler.

A cDNA encoding a cathepsin D homologue was cloned from a cDNA library of the mulberry longicorn beetle, Apriona germari. Sequence analysis of the cDNA encoding the cathepsin D homologue of A. germari revealed that the 1,158 bp cDNA has an open reading frame of 386 amino acid residues. The deduced protein sequence of the A. germari cathepsin D homologue shows high homology with cathepsin D in insects, Aedes aegypti (68.2% amino acid similarity) and Drosophila melanogaster (67.2% amino acid similarity). Two aspartic residues and six cystein residues in the A. germari cathepsin D homologue are present at identical locations in all of the other catepsins D. Unlike cathepsins D in two insect species, A. gemari cathepsin D homologue appears to have two putative glycosylation sites, rather than one. Phylogenetic analysis revealed the A. germari cathepsin D homologue is more closely related to insect cathepsins D than to the other animal cathepsins D. Northern blot analysis suggests that A. germari cathepsin D homologue gene is expressed in most if not all, body tissues.
Probing Streptomyces albus ATCC 21838 chromosomal DNA with a proline tRNA sequence resulted in an isolation of a putative integrating element in the 6.4-kb EcoRI fragment. It was found that Streptomyces lividans TK-24 transformed with a cloned DNA fragment on a multicopy plasmid, produced a higher level of spore pigment and mycelial red pigment on a regeneration agar. Furthermore, the transformant S. lividans TK-24 produced a markedly increased level of undecylprodigiosin in a broth culture. A nucleotide sequence analysis of the cloned region revealed several open reading frames homologous to the integrases of integrating plasmids or temperate bacteriophages, signal-transducing regulatory proteins with a conserved ATP-binding domain, oxidoreductases ($\beta$-ketoacyl reductase), and an AraC-like transcriptional regulator. To examine the effect on antibiotic production, each coding region was overexpressed separately from the other genes in the region in S. lividans TK-24 with; pJHS3044 for the expression of the signal-transducing regulatory protein homologue, pJHS3045 for the homologue of oxidoreductase, and pJHS3051 for the homologue of the AraC-like transcriptional regulator. Phenotypic studies of S. lividans TK-24 strains harboring plasmids for the overexpression of individual genes suggested the following effects of the genes on antibiotic production: The oxidoreductase homologue stimulated the production of actinorhodin and undecylprodigiosin, which was influenced by the culture conditions; the homologue of the AraC-like transcriptional regulator was the most effective factor in antibiotic production within all the culture conditions tested; the signal-transducing regulatory protein homologue repressed the effect due to the homologue of the AraC-like transcriptional regulator, however, the antibiotic production was derepressed upon entering the stationary phase.
A study on the bioconversion of soluble starch to cyclodextrin(CD) homologue by CGTase was performed in the membrane-enzyme reactor equipped with a dead-end type membrane module. in the batch reactor, the total conversion of soluble starch to CD homologue was decreased rapidly from a maximum value of 45 % with increasing reaction time due to the product inhibition and breakdown of CD homologue to the reducing sugars. However, in the membrane-enzyme reactor, the total conversion of soluble starch was maintained at a constant value of 35 % throughout the reaction, since the membrane(MWCO = 10,000) promptly separated CD homologue from the reaction mixture. After the macdon for 24 hr in the membrane-enzyme reactor using a 10 % soluble starch solution, the cumulative production amount of CD homologue was about 3.7 kg/m$^2$ at the operating pressure of 2 atm.

This study was performed to investigate the behavior of chlorobenzenes (CIBZS) and chlorophenols (CIPhs) in a thermally treated MSWI fly ash. The experiment was carried out in a fixed bed reactor at the temperature range of 300~$600^{\circ}C$. Reaction time range was between 30 and 120 minutes, and NB and 02 gases were used as carrier gas. The decomposition rate of CIBZS was more affected by reaction time than by the reaction temperature. The decomposition rate of CIPhs was affected by both parameters. Decomposition rate of CIBZS and CIPhs reached 80.4% and 96.6% at $600^{\circ}C$, 120 min, respectively. Considering the effect of O2 content, decomposition rate of CIBZS and CIPhs was the highest at 10% of O2 content. Declorination and decomposition reactions Pere investigated by analyzing homologue distribution. Higher chlorinated CIBZS and CIPhs homologue decreased but lower chlorinated compounds increased with the increase of temperature. Effect of O2 on the homologue distribution of these compounds was not clear in the range of our experiment conditions.
The aim of this study was to investigate deposition characteristics of atmospheric PCDD/Fs on pine needles and soil. Concentration of PCDD/Fs on soil were higher than those in pine needles, being 7.4 times for ${\Sigma}PCDDs$, 2.3 times for ${\Sigma}PCDFs$, and 5.4 times for ${\Sigma}PCDD/Fs$ respectively. The ratios of the concentration of each homologue in soil to that in pine needles increased as the number of chlorine atoms increased. The concentration (%) of each congener to ${\Sigma}PCDD/Fs$ was higher in the one with a small number of chlorine atoms substituted, such as tetra- to hexa-homologues in pine needle samples and in one with a large number of chlorine atoms, such as hepta- to octa-homologues in soil samples. Lower-chlorinated homologues of PCDD/Fs were mainly accumulative deposited in pine needles, whereas higher-chlorinated ones were on soil.

Homologue and isomer patterns of polychlorinated dibenzo-p-dioxins(PCDDs) and polychlorinated dibenzofurans(PCDFs) congeners formed from phenols in the gas-phase at $600{\sim}700^{\circ}C$ and via particle-mediated reactions at $400^{\circ}C$ were studied in an isothermal flow reactor. A mixture of 20 phenols in relative concentrations found in a municipal waste incinerator(MWI) stack gas was used for this study. PCDDs and PCDFs homologue and isomer patterns obtained from the phenol. From the phenol experiments, gas-phase formation at $600{\sim}700^{\circ}C$ favors PCDFs formation whereas particle-mediated formation at $400^{\circ}C$ favors PCDDs formation. DD and DF were most abundant homologue groups, PCDDs and PCDFs homologue fraction decreased with increasing number of chlorine substituents. PCDDs and PCDFs homologue and isomer fractions were almost constant from gas-phase formation and particle-mediated formation. Unsubstituted phenol, which was present in high concentration, played a significant role in the formation of PCDD/Fs congeners under both sets of experimental conditions.

The Sec61 trimeric complex ($\alpha$,$\beta$, and ${\gamma}$ subunits) is one of the Sec-complex responsible for post-translational protein translocation across the endoplasmic reticulum membrane in diverse organisms. In this study, a cDNA encoding the Sec61p ${\gamma}$ subunit homologue was isolated from the cDNA library of the mole cricket, Gryllotalpa orientalis. Sequence analysis of a 442-bp cDNA clone showed it to contain an open reading frame of 68 amino acid residues consisted of 204-bp. The homologues of the gene were found in the GenBank database in a diverse organism including insect, mammals, fungi, and plants. The deduced amino acid sequence of Sec61p ${\gamma}$ subunit homologue of the mole cricket showed the highest homology to the gene of the singly known insect, Drosophila melanogester (93% identity), and the least homology to that of the baker's yeast, Saccharomyces cerevisiae (37.2%). Phylogenetic analysis also confirmed a close relationship between the insect Sec61p ${\gamma}$ subunit homologues of G. orientalis and D. melanogester. Hydropathy analysis of the cricket mole and published other data suggested that the hydrophobic segment close to C-terminus is predicted to be the putative membrane anchor, Multiple alignment of the Sec61p ${\gamma}$ subunit homologue among several organisms showed the presence of several conserved domains including the conserved proline at position 28.
Bacillus sp. secretes high levels of extracellular enzymes into the culture medium. The signal recognition particle (SRP) and the SRP receptor play a central role in targeting pre secretory proteins to the translocase. By the analysis of the DNA microarray of B. lentimorbus WJ5, it was detected that WJ5m12, antifungal activity deficient mutant induced by gamma radiation, had a down-regulated expression of the SRP receptor gene (B. subtitis srb homologue, srbL). To determine the relationship of SRP receptor to antifungal activity, srbL of B. lentimorbus WJ5 was amplified by PCR and ligated into pQE30 vector, and then transferred into WJ5m12. The transformant, WJ5m12::srbL, recovered the antifungal activity. From the 2-DE analysis, the several presecretory proteins accumulated in the mutant cell and decreased to a level of the wild type in WJ5m12::srbL. It seems that the srbL could play an important role in the secretion of the antifungal activity related proteins of B. lentimorbus WJ5.
