This research was performed to investigate the protective effect of Chungpesagan-tang (CPS) against ischemic damage in PC12 cells. To elucidate the mechanism of the protective effect of CPS on ischemic insult, cell viability and changes in activities of Superoxide dismutase, Glutathione Peroxidase, Catalase, Caspase 3 and the production of Malondialdehyde were observed after treating PC12 cells with CPS which was metabolized by rat liver homogenate. Pretreatment of CPS with liver homogenate increased its protective effect against ischemic insult by reducing the harmful effect of CPS itself. (omitted)

This study was designed to investigate the antioxidant effects of dietary xanthophylls supplementation in broiler breast and thigh meat homogenates during incubation at 37$^{\circ}C$ for 0, 2, 4, 8, 16 hours respectively. Experimental treatments were divided into control, lutein, canthaxanthin, astaxanthin and capxanthin fed meats. The supplementation levels of to chicks were adjusted to 30 ppm in feeds. The 30 ${\mu}$M FeCl$_3$ and 100 ${\mu}$M ascorbic acid were added to meat homogenates in order to catalyze lipid oxidation. In breast meat homogenates, the TBARS(O.D) of all treatments at 2 hour was significantly(p〈0.05) increase. In thigh meat homogenates, the highest TBARS(O.D) value of all treatments appeared at 16 hour incubation and TBARS(O.D) value of all treatments was significantly(p〈0.05) lower than that control during incubation time. The TBARS(O.D) of lutein treatment in breast meat homogenate at 8 hour and 16 hour were significantly(p〈0.05) lower than those of treatments. Also, astaxanthin treated in thigh meat homogenate of the 2 hour and 4 hour and lutein treatment in thigh meat homogenate at 8 hour and 16 hour were significantly(p〈0.05) lower than other treatments. In conclusion, dietary xanthophyll treatments in breast and thigh meat homogenates showed more antioxidant effect to lipid oxidation than control. Especially, lipid oxidation inhibited significantly in lutein fed breast meat, and lutein and astaxanthin fed thigh meat homogenates.

This experiment was carried out to investigate variation of plasma lipid level and liver peroxide level(Thiobarbituric acid, T.B.A) caused by tube feeding of Pteridium sp. extracts for seven weeks in rats. A seven week rat's plasma triacylglyceride (T.G.) level showed remarkably higher than that of the control group and that of the three week rats. On the other hand, no remarkable differences of plasma phospholipid, free fatty acid and thiobarbituric acid level have been recognized. Liver homogenate lipid peroxides value of seven week feeding rats showed remarkably higher level compared with that of the control group and the three weeks rats. So it could not be agreed that short time feeding of Pteridium sp. plant or even it's concentrated extracts influenced to the levels of liver homogenate lipid peroxides, plasma lipids and its peroxides.

In order to elucidate the correlation between the lipid peroxidation and hepatotoxicity, the formation of malondialdehyde (MDA) in liver homogenate and serum, and the transaminase activities were determined in intoxicated by ascorbic acid-Fe$\^$2+/-ADP in rat. In a model of ascorbic acid-Fe$\^$2+/-ADP hepatotoxicity, G009, which was obtained from Ganoderma lucidum IY009, exhibited anti-lipid peroxidative effect in rat liver homogenate, and that MDA values of the liver homogenate decreased from 48.1% to 74.8% in comparision to controls (p<0.01) Also, the MDA formation in serum inhibited 66.5% at 100 mg/kg of G009. Serum levels of glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) in peroxidizer-induced rats treated with G009 was decreased compared with control. Especially, The formation of lipid peroxides in serum was related to GPT levels. These results that G009 has a protective effect on ascorbic acid-Fe$\^$2+/-ADP-induced hepatic injury through an inhibition of lipid peroxidation in liver.
A cepfialosporin with an aminothiazoiylmethoxyimino-type side chain at the 7 position and bicyclic quinolone dithicarbamate at the 3' position was synthesized. It has broad and potent antivacterial activity in vitro. The antibacterial spectrum reflects contributions of both the cephalosporin moiety and the quinolone moiety. Thus, this compound was named DACD implying a dualaction cephalosporin derivative. In this paper, the physicochemical proper-ties (lipid-water partition, pKa), stability and pharmacokinetics of DACD were determined and compared with cefotaxime 3'-norfloxacin dithiocarbamate (CENO). Stability tests were studied in pH 1.20, 6.80 and 8.00 buffers and in the presence of AB type human plasma, rat liver homogenate and its .betha.-lactamase. The pharmacokinetic parameters of DACD were evaluated in mice after a single intravenous dose of 40 mg/kg. The results are as follows. The lipid-water partition coefficient of DACD was higher than that of CENO. The calculated pKa values of CENO and DACD, were 6.82$\pm$0.03, 7.53$\pm$0.21, respectively. In the hydrolysis test, half-lives (t$^{1/2}$) of CENO and DACD was 66.0 hr and 80.0 hr in pH 6.80 buffer, 190 hr and 91.4 hr in pH 8.00 buffer. CENO and DACD were rapidly hydrolyzed in human plasma and in rat liver hornogenate. Half-lives (t$_{1/2}$ of CENO and DACD were 1.29 hr and 1.15 hr in hyman plasma, 0.62 hr and 0.71 hr rat liver homogenate. In $\beta$-lactamase stability test, CENO and DACD were very stable to the .betha.-lactamases obtained from three different strains. Half-life (t$_{1/2}$) and areas under the curve (AUC) in mice were 2.33 hr and 15.97 (mg.h/1), respectively.
To avoid the intrinsic problem of aggregation associated with the traditional solution-phase refolding process, we propose a solid-phase refolding method integrated with expanded bed adsorption chromatography. The model protein used was a fusion protein of recombinant human growth hormone and a glutathione S transferase fragment. It was demonstrated that the EBA-mediated refolding technique could simultaneously remove cellular debris and directly renature the fusion protein inclusion bodies in the cell homogenate with much higher yields and less agregation. To demonstrate the applicability of the method, we successfully tested the three representative types of starting materials, i. e., rhGH monomer, washed inclusion bodies, and the E. coli homogenate. This direct and simplified refolding process could also reduce the number of renaturation steps required and allow refolding at a higher concentration, at approximately 2 mg fusion protein per ml of resin. To the best of our knowledge, it is the first approach that has combined the solid-phase refolding method with expanded bed chromatography.
The change of cell counts of Vibrio parahaemolyticus in fish muscle by the storage time and temperature was examined to get basic informations for precautionary steps against food poisoning of slices of raw fish (sashimi). There fore, we inoculated fish homogenate of oceanic bonito (Katsuwonus pelamis), yellow tail (Seriola quinqueradiata) with Kanagawa positive Vibrio parahaemolyticus and stored it at $30^{\circ}C,\;18^{\circ}C,\;4^{\circ}C\;and\;-20^{\circ}C$ for 24 hours. The number of the Vibrio parahaemolyticus upon fish homogenate stored at $30^{\circ}C\;and\;18^{\circ}C$ decreased for the first two hours and increased thereafter. When the fish homogenates inoculated with Vibrio parahaemolyticus at about $10^3$ per gram were stored at $18^{\circ}C\;and\;30^{\circ}C$ for 10 hours, the cell numbers increased about 10 times and 1,000 times initial cell numbers, respectively. The survival rate of Vibrio parahaemolyticus was about $20\%$, when the inoculated fish homogenates were stored at $-20^{\circ}C$ for 24 hours. Vibrio parahaemolyticus inoculated in fish homogenates was decreased by about $10\%$ of initial cell numbers by the storage at $4^{\circ}C$ for 4 hours and it was decreased by about $50\%$ after 24 hours storage of the samples at the same temperature. The decreasing rate of inoculated Vibrio parahaemolyticus in fresh fish muscle homogenate was higher than that in frozen fish muscle homogenate during the storage time at a refrigerator.
Cooking characteristics of coated rice with water homogenate of citrus fruit peels (1% for rice) were investigated. The color of the coated rice both before and after cooking was dark yellow. The total content of carotenoids, hesperidin and naringin were 10.74, 2173.12 and 1468.40 mg% for citrus fruit peels, 0.46, 108.65 and 73.35 % for its water homogenate, 0.12, 21.73 and 14.62 mg% for coated rin, and 0.05, 8.67 and 5.87 mg% fur cooked coated rice, respectively. Citrus fruits peel contained 94.22 mg% of asparagine, 24.88 mg% of methionine, 19.64 mg% of alanine, and 15.37 mg% of ${\gamma}$-aminoisobutyric acid as the majority free amino acids, accounting for 70% of the total free amino acids present. Total free amino acid content of the cooked coated rice increased by 15% compared to those of cooked uncoated rice. The majority of minerals in the citrus fruit peels were K and Ca, accounting for 56% of total minerals present. The mineral content of cooked coated rice was generally higher than that of the cooked uncoated rice. The cooked coated rice showed comparable hardness, gumminess and brittleness, but higher cohesiveness and springiness than the cooked uncoated rice. There were no differences in sweet and bitter taste between the cooked uncoated and coated rice. However, the cooked coated rice showed higher sensory scores fur color acceptability, savory taste and overall acceptability than the cooked uncoated rice.
Abilities of various edible plants and natural antioxidants to protect brain against oxidative damages were evaluated using brain homogenate of perfused Sprague-Dawley rat. Oxidative damage, expressed as lipid peroxidation (LPO), indicating total quantity of malondialdehyde and 4-hydroxyalkenal, increased from 4.1 to 6.9nmol/mg protein by treatment of $2.5{\mu}M$ ferrous sulfate and 7.5mM hydrogen peroxide as source of reactive oxygen species (ROS) on brain homogenate for 10min at $37^{\circ}C$ Mallow(88%) in leafy vegetables, small potato (93%) in root vegetables, green red pepper (76%) in fruit vegetables, and avocado (96%) in fruits showed highest LPO inhibition capacities. Ability of mushrooms decreased in order of nameko, shiitake, pine mushroom, oyster mushroom, and new type pine mushroom. Among natural antioxidants tested, (+)catechin (91%), (-)epigallocatechin gallate (85%), (-)epicatechin gallate (83%), and kaempferol(83%) showed high LPO inhibition capacities.
This study was carried out to investigate the effects of Salicornia herbacea L. (SH) on carbon tetrachloride $(CCl_4)-induced$ hepatotoxicity. Sprague-Dawley rats were intraperitoneally administered the SH at 100 mg/kg per day for two weeks. Then single dose of $CCl_4$ (3.3 ml/kg) was injected into rats. Twelve hours later, they were anesthesized with ether and dissected. $SH-CCl_4-administered$ group showed $65.56\%\;and\;59.04\%$ of inhibitory effects in aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities, compared to $CCl_4-treated$ group (p<0.05). Malonedialdehyde (MDA) levels of $SH-CCl_4-administered$ group in liver homogenate and mitochondria were significantly inhibited by $53.74\%,\;89.86\%$, and respectively, compared to $CCl_4-treated$ group (p<0.05). Superoxide dismutase (SOD) activities of $SH-CCl_4-administered$ group in liver homogenate and mitochondria were significantly inhibited by $42.51\%,\;and\;38.42\%$, respectively, compared to $CCl_4-treated$ group (p<0.05). The histological examinations showed that the liver cell necrosis and centrilobular congestive aggregation induced by $CCl_4$ were clearly eliminated by the administration of SH. These results suggest that SH could have the protective effects against hepatotoxicity.
