염산 가수분해 추출법으로 조제된 자하거 약침액의 생리활성 효과 (Biological Activities of Hominis Placenta Herbal Acupuncture prepared by Hydrochloric Acid Hydrolysis)

  • 본 연구의 목적은 자하거약침액의 항산화기능에 관련한 생리활성을 보기 위하여 대한약침학회에서 제공받은 자하거 약침액을 시료로 사용하였다. 총 폴리페놀 함량은 자하거 약침액 원액에서 24.641 mg/L로 이는 산수유 등과 같은 약용식물과 비슷한 수준의 함량이다. 자하거약침액원액에서 DPPH radical 소거능과 Superoxide dismutase(SOD) 유사활성 측정은 각각 49.4%, 50.87%였다. 또한 ABTS radical 소거능과 Nitrite radical 소거능(pH1.5)에서는 각각 50.01%, 52.8%로 ABTS radical과 Nitrite radical 소거능이 우수하였다. 본 실험결과로볼 때, 자하거 약침액은 항산화능력은 뛰어난 것으로 사료된다. 하지만 현재 자하거 약침액과 관련된 실험이 부족한 실정이기 때문에 앞으로 이에 관련된 더 많은 연구가 필요하다고 생각된다.

A Pilot Study on Single-dose Toxicity Testing of Hominis placenta Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: This study was performed to analyze the toxicity and to find the lethal dose of the test substance Hominis placenta pharmacopuncture when used as a single-dose in 6 week old, male and female Sprague-Dawley (SD) rats. Methods: All experiments were conducted at Biotoxtech (Chungwon, Korea), an institution authorized to perform non clinical studies, under the regulations of Good Laboratory Practice (GLP). SD rats were chosen for the pilot study. Doses of Hominis placenta pharmacopuncture extracts, 0.125, 0.25 and 0.5 mL, were administered to the experimental group, and 0.5 mL doses of normal saline solution were administered to the control group. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results: No deaths or abnormalities occurred in any of the groups. Also, no significant changes in body weights were observed among the groups, and no significant differences in hematology/biochemistry, necropsy, and histopathology results were noted. Hematologically, some changes in the male rats in two experimental groups were observed, but those changes had no clinical or toxicological meaning because they were not dose dependent. Histopathological tests on the injected parts showed cell infiltration in the male rats in one of the experimental groups; however, that result was due to spontaneous generation and had no toxicological meaning. Therefore, this study showed that Hominis placenta pharmacopuncture had no effect on the injected parts in terms of clinical signs, body weight, hematology, clinical chemistry, and necropsy. Conclusion: As a result of single-dose tests of the test substance Hominis placenta pharmacopuncture in 4 groups of rats, the lethal dose for both males and females exceeded 0.5 mL/animal. Therefore, the above findings suggest that treatment with Hominis placenta pharmacopuncture is relatively safe. Further studies on this subject are needed.

합곡, 수삼리 자하거약침요법을 병행한 복합 한방치료의 수부냉증 치험례 (A Case Report of Cold Sensitivity of Hand by Korean Medical Treatment with Hominis Placenta Pharmacopunture at LI4 and LI10)

  • Objectives : The purpose of this study is to report the clinical effects of Korean medical treatment with Hominis Placenta Pharmacopunture(HPP) on cold hypersensitivity of hands. Methods : A 46-year-old female patient whose chief complaint was cold hypersensitivity of hands was treated with Placenta Pharmacopunture at LI4 and LI10, ten times for a month. Acupuncture and herbal medicine were added depending on the symptoms. Digital Infrared Thermal Imaging(D.I.T.I) was taken and Visual Analog Scale(VAS) was examined before and after each treatment. Results : After the treatments, the symptoms were improved. Conclusions : DITI and VAS showed that the patient's cold hypersensitivity of hands was improved after Korean medical treatment with Hominis Placenta Pharmacopuncture.

자하거약침의 월경통에 대한 치료효과 (Effects of Hominis Placenta Herbal Acupuncture(HPA) on Menstrual Cramps)

  • Purpose : The purpose of this study was to identify the effects of Hominis Placenta Herbal Acupuncture(HPA) combined with Acupuncture compared with Acupuncture therapy. Methods : Data was collected from October 1st 2004 to February 28th 2005. A total of 8 women participated in this study. Of all subjects, 4 and 4 subjects were assigned to the experimental and the control group respectively. Subjects were treated three times a week. We used Visual Analogue Scale(VAS) for evaluating the effects of HPA. Results : There showed statistically boardline significance between the experimental group and control group(p=0.057). Conclusion : The results showed HPA has significant effects on menstrual cramps.

  • Hominis Placenta has a broad array of clinical applications in Korean medicine, including treatment of inflammatory conditions such as rheumatoid arthritis. The purpose of this study is to explore the global gene expression profiles in human RAW 264.7 cell lines treated with Hominis Placenta herbal-acupuncture solution (HPHAS) using microarray analysis. The RAW 264.7 cells were treated with lipopolysaccharide (LPS), HPHAS, or both. Of the 8,170 genes profiled in this study, with a cut-off level of two-fold change in the expression, 72 genes (CTD1, regulating synaptic membrane exocytosis 2, etc.) were upregulated and 135 genes(splicing factor, arginine/serine-rich 1, actinin, alpha 1, etc.) downregulated following LPS treatment. One gene (acrosin) was upregulated and 12 genes (phospholipase A2, group IB, neurofilament, heavy polypeptide 200kDa, etc.) were downregulated following HPHAS treatment. Eleven genes (RAB27A, member RAS oncogene family, eosinophil peroxidase, etc.) were upregulated and 16 genes (V-maf musculoaponeurotic fibrosarcoma oncogene homolog G (avian), RW1 protein, etc.) were downregulated following co-stimulation of HPHAS and LPS. It is thought that microarrays will play an ever-growing role in the advance of our understanding of the pharmacological actions of HPHAS in the treatment of arthritis. Further studies, however, are required to concretely prove the effectiveness of HPHAS.

Immunomodulatory Effects of Hominis Placenta Extract Injection into an Acupuncture Point on the Experimental Subcutaneous Tumor Model of Mice

  • Hominis placenta (HP) has been used as an agent for promoting physiological function in traditional asian medicine. The present study was peformed to investigate whether HP acupuncture treatment in an experimental tumor mice model inhibit tumor growth through immunomodulatory effects. Mice were inoculated subcutaneously with colon26-L5 cells on the back. Three days after tumor inoculation, HP herbal acupuncture treatment was conducted on BL18 acupoint every other day for three weeks. HP Herbal acupuncture treatment significantly suppressed the primary tumor growth and prolonged survival rate. To evaluate immunomodulatory effect of HP acupuncture, splenocytes proliferation assay, fluorescence-activated cell sorting (FACS) and ELISA for IFN- ${\gamma}$, and IL-4 cytokine level. HP herbal acupuncture enhanced the mitogenic activity of Balb/c whole splenocytes induced by various mitogenic stimuli and increased immune cell population such as T cell, B cell, Th cell, Tc cell and Macrophages. HP herbal acupuncture caused a marked increase of production of Th1 cytokine (IFN- ${\gamma}$ ,) and decrease of production of Th2 cytokine (IL-4). These results indicated that HP herbal acupuncture suppresses tumor growth through a mechanism leading to a Th1 dominant immune state.

신경뿌리 병증, 경추부의 증례보고 1례 (A Case Report of Cervical Radiculopathy)

  • Objectives: The purpose of this study is to report the clinical effects of Korean medical treatment on the patient with cervical radiculopathy. Methods: A 16-year-old female patient, who was suffering from the cervical radiculopathy on her Lt. shoulder: ROM limit, swelling and arthralgia, was treated by herbal medicine, physical therapy, acupuncture and pharmacupuncture. Results: As a result, the patient's left upper limb disability was improved by Korean herbal medicine (ssanghwatang hap gamiseokyungtang), hominis placenta (Ja-ha-geou) pharmacupuncture and physical therapy. Conclusions This study showed that Korean medical treatments can be an effective choice for cervical radiculopathy.

  • Objective : This study has been designed and performed to identify the effect on insomnia patients according to the injection of herbal medicine induced from the Hominis placenta. Methods : Injection on G20(風池), G20(池) in insomnia patients who were admitted in Dong-Seo Oriental Medical Hospital every day for 5days, without western medicine. Results: Total duration of sleeping time showed 0.68 hours of increased. (p<0.005 respectively)

말초성 안면신경마비에 대한 황련해독탕약침과 자하거약침의 효능 : 후향적 비교 연구 (Efficacy between Hwangryunhaedok-tang Pharmacopuncture Therapy and Hominis Placenta Pharmacopuncture Therapy on Peripheral Facial Paralysis : Retrospective Comparision Study)

  • Objectives : The purpose of this study is to compare the effects of Hwangryunhaedok-tang pharmacopuncture therapy with Hominis Placenta pharmacopncture therapy in hospitalized patients with peripheral facial paralysis. Methods : We investigated 34 cases of patients with peripheral facial paralysis who were admitted into the Dept. of Acupuncture & Moxibustion of Dongshin University Suncheon Oriental Hospital from February 1, 2014 to June 31, 2015. Subjects were divided into two groups, Hwangryunhaedok-tang pharmacopuncture group(HR group), and Hominis Placenta Pharmacopuncture group(JH group). HR group was treated by Hwangryunhaedok-tang pharmacopuncture five times a week and JH group was treated by Hominis Placenta Pharmacopuncture five times a week. And both groups were treated by acupuncture, electroacupuncture, herbal medicine therapy, western drug therapy equally. To investigate the effectiveness of treatment, we used House-Brackmann Grading System, Yanagihara's unweighted grading system and Sunnybrook facial grading scale at before admission and after admission. Results : Each scores by 3 evaluation methods improved both in two groups. However, there were no significant differences in improvement between two groups. Conclusions : These results suggest that the Hwangryunhaedok-tang pharmacopuncture therapy is as effective as Hominis Placenta pharmacopuncture therapy to improve symptoms of peripheral facial paralysis.

SAM p6 mouse에서 자하거 주사제의 골다공증 개선에 대한 연구 (Effects on osteoporosis of Senescence Accelerated Mice (SAM) P6 by Injection of Placenta extracts ($Beuronel^{(R)}$))

  • Objectives : The purpose of this study was to examine the effects of the Hominis placenta extracts on improvement of senile osteoporosis (Type 2) in SAM P6. In traditional Oriental medicine, Placenta (Jahageo) is indicated for insufficiency of blood and tidal fever due to steaming bone disorder. Methods : At 10 weeks after birth, Placenta ex. was given 100 mL, 200 ml/kg/day, i.p. 3 times a week for 4 weeks in SAM P6 mice. And then we measured complete blood cells, blood urea nitrogen, creatinine, inorganic phosphate and total iron. In addition, we tested bone mineral density (BMD) using the soft X-ray. Results : As a result, in SAM P6, injection of Placenta ex. (200 mL) increased in RBC, HB and PLT, in comparison with control group. It was also found that the inorganic phosphate levels increased significantly in the injection groups of the Placenta ex. from that of the control group, but blood urea nitrogen was no significant. Moreover, Placenta ex. were showed a trend of increase in bone mineral density (BMD) of the vertebrae lumbales and tibia in P6 mice. Conclusions : These findings suggest that Placenta ex. are effective in preventing bone loss in SAMP6.
