Identification of Pedigree by Bovine Blood Typing in Holstein (젖소에 있어서 혈액형분석에 의한 혈통확인에 관한 연구)

  • This experiment was carried out to clarify the pedigree identification from blood typing of 301 Hoisteins in National Animal Breeding Institute(N.A.B.I.). Twenty kinds of standard reagent standardized by Insternational Society for Animal Blood Group Research provied from KNC improvement center, N, L, C, F. were used as the reference reagents in this study. The highest frequency of antigenic facfors was obtained from X$_2$in blood typing of 301 Holsteins. The frequency of X$_2$ was 0.714.In A blood system, four kinds of phenogroups were observed. The gene frequencies of Al and Z' phenogroups were equally 0.027.This frequency was greatly lower than those of breeds of Southern European and Zebu cattle. In B blood Systern, nineteen kinds of blood type were appeared. The appearance frequency of Gx blood type was 0.259, whish was higher than the others. In C blood system, thirty kinds of blood type were observed. The appearance frequency of X$_2$ blood type was the highest(0.189). In F blood system, three kinds of alleles were detected. The gene frequency of F allele was higher than that of V(0.105). However, the frequency of F allele(0.327) was greatly lower than that of "- /- " allele. In S blood system, twelve kinds of blood type were appeared and showed sirnilar appearance frequencies except " - / - " allele. From the results of the pedigree identification from 8 sires and 28 progenies of them, the accuracy of pedigree identification was 92.9%.ification was 92.9%.

Identification of Genomic Differences between Hanwoo and Holstein Breeds Using the Illumina Bovine SNP50 BeadChip

  • The use of genomic information in genomic selection programs for dairy and beef cattle breeds has become a reality in recent years. In this investigation, we analyzed single-nucleotide polymorphisms (SNPs) for Hanwoo (n=50) and Holstein (n=50) breeds using the Illumina Bovine SNP50 BeadChip to facilitate genomic selection and utilization of the Hanwoo breed in Korea. Analysis of the entire genomes showed different spectra of SNP frequencies for Hanwoo and Holstein cattle. The study revealed a highly significant (p<0.001) difference between Hanwoo and Holstein cattle in minor allele frequency (MAF). The average MAFs were $0.19{\pm}0.16$ and $0.22{\pm}0.16$ for Hanwoo and Holstein, respectively. From the total of 52,337 SNPs that were successfully identified, about 72% and 79% were polymorphic in Hanwoos and Holsteins, respectively. Polymorphic and fixed SNPs were not distributed uniformly across the chromosomes within breeds or between the two breeds. The number of fixed SNPs on all chromosomes was higher in Hanwoo cattle, reflecting the genetic uniqueness of the Hanwoo breed. In general, the rate of polymorphisms detected in these two breeds suggests that the SNPs can be used for different applications, such as whole-genome association and comparative genetic studies, and are a helpful tool in developing breed identification genetic markers.

Analysis of total mixed ration (TMR) nutrition and metabolic diseases in Korean dairy farm (국내 고능력우 Holsteins 농가의 TMR 영양성분 및 대사성 질병 분석)

  • A variety of livestock feed resources were used in Korean dairy farm due to a lack of the endemic feed. However, there is inadequate real farm data to support farmers' decisions on the choice of options. The main objective of this study was to evaluate the nutritional value of total mixed ration (TMR) as well as the metabolic diseases status in Korean dairy farms. TMR samples were collected from nine feed companies and eight selected self-formulated by the dairy farms. The nutrient contents were examined by AOAC methods. The frequency of metabolic diseases such as ketosis and hypocalcemia were surveyed. The average moisture content was 36.2% although the min. and max. value were varied from 21.7% and 50.6% among farms. The mean${\pm}$standard deviation of crude fiber (CF), crude ash (CA), ether extract (EE), and crude protein (CP) were $21.4{\pm}2.5$, $4.6{\pm}0.4$, $3.2{\pm}0.5$ and $9.8{\pm}1.7$, respectively. However, the average ADF and NDF was $17.3{\pm}3.7$ and $31.0{\pm}5.7$, respectively. The compositions of TMR were varied significantly among the dairy farms. The frequency of clinical Ketosis (CK), subclinical ketosis (SCK) and hypocalcemia were higher in early lactation period with 4.5%, 11.0% and 3.0%, respectively. Also, the frequency of SCK was higher than CK and hypocalcemia throughout the lactation. Periodic TMR nutrient analysis based on herd production or physiology change would maximize the effects of TMR feeding. Furthermore, the study results would be useful to the farm practitioner and producer for their farm management.

Determination of in situ degradation parameters and feeding level of pineapple (Ananas comosus L.) cannery by-product to Hanwoo steers

  • Objective: The objectives of this study were to determine the in situ degradation parameters and appropriate feeding level of pineapple cannery by-products (PCB) based on the growth performance and blood parameters of growing Hanwoo (Bos taurus coreanae) steers fed various levels of PCB. Methods: Two ruminally cannulated Holstein cows were used for in situ disappearance rate measurements. Nylon bags (5×10 cm, 45 ㎛ pore size) filled with 5 g of PCB in triplicate were inserted into the ventral sac of the two cannulated Holsteins cows and incubated for 0, 2, 4, 8, 16, 24, and 48 h. A total of 16 castrated growing Hanwoo steers (12.5±0.5 months old, 302.9±25.7 kg of initial body weight [BW]) were used for the experiment. Animals were stratified by initial BW and then randomly assigned to one of four experimental diets (0%, 1.5%, 3.0%, or 6.0% of PCB, on the dry matter [DM] basis) fed for 91-d, including 30-d of adaptation. Results: Soluble fraction a of DM and crude protein (CP) was 61.9% and 86.0%, fraction b of DM and CP was 32.7% and 11.2%, and indigestible fraction c of DM and CP was 5.4% and 2.8%. The 6.0% PCB feeding group showed lower productivity compared to animals in the other treatments. Increasing the dietary level of PCB did not alter DM intake, but it was numerically lowest in the 6.0% feeding group. The gain to feed ratio was linearly decreased by increasing of PCB. The quadratic broken-line test estimated that 2.5% (DM basis) was the maximum feeding level of PCB in growing Hanwoo steers (y = 0.103 - 0.001×[1.245-x]2, R2 = 0.18). Conclusion: Diets containing up to 2.5% PCB can be fed to growing Hanwoo steers without adverse effects on growth performance.

Characterization of the DGAT1 Gene in the Korean Holstein Dairy Cattle Population (한국 Holstein종 유우집단의 DGAT1 유전자의 특성분석)

  • This study was conducted to characterize the DGAT1 gene in Korean Holstein dairy cattle population and examine the relationship of DGAT1 polymorphisms with milk yield and milk fat yield for the genetic improvement of Korean Holstein dairy cattle. Results indicated that the 411 bp PCR products were successfully amplified by DGAT1 specific primers. Sequence analysis indicated that the DGTA1 Q allele had AUG (Lysine, K) nucleotide sequences in 216-218 bp and q allele had GCG (Alanine, A) sequences in the same position. Nucleotide sequence homology between the DGAT1 sequences generated in this study showed 100% homology with bovine DGAT1 sequences in the NCBI database. The genotype frequencies of DGAT1 QQ, Qq, and qq were 16.43, 36.43, and 47.14%, respectively, in Korean Holstein dairy cattle population. The observed Q and q allele frequencies were 0.35 and 0.65, respectively. Statistically significant (P<0.05) results were identified for milk yield and milk fat yield for the DGAT1 genotypes. The Qq genotype Holsteins have significantly higher milk yield and milk fat yield than those of the QQ and qq genotype Holsteins(P<0.05).

Effects of Higher Dietary Cation with or without Protected Fat and Niacin on the Milk Yield and Thermoregulatory Ability in Holsteins During Summer Heat Stress (여름철 고온스트레스 기간에 고 양이온 사료와 반추위 보호지방과 나이아신의 추가공급시 착유우의 유생산 및 체온조절에 미치는 영향)

  • The objective of this study was to evaluate the effects of dietary cation-anion difference (DCAD) with or without ruminally protected fat and niacin on the thermoregulatory ability, milk yield and milk composition of lactating dairy cows during summer in Korea. Thirty mid-lactating Holstein cows (134±12.4 DIM and 23.4±2.3kg/d of milk yield) were divided into three groups (10 animals/group). Cows were housed in a free-stall barn and were provided with forced- air ventilation (wind velocity = 4 m/s) using 41 cm diameter fans. Diet one was formulated to contain low DCAD (+15 DCAD) while the remaining two diets were higher in DCAD (+30 DCAD). One higher DCAD diet was formulated to contain by-pass fat and the second higher DCAD diet contained the niacin along with by-pass fat. The maximum ambient temperature during July was 28.5℃ which could be seen as a period of mild heat stress. As summer progressed, August was characterized as a severe heat stress condition with maximum ambient temperature (32.4℃) and THI (74.0). Dry matter, crude protein and total digestible nutrients intake was not affected by the DCAD level and supplementation of ruminally protected fat or niacin. Milk production was higher in cows fed diets supplemented with fat and niacin than those fed un-supplemented diet. No difference in milk yield was observed in cows fed diets supplemented with fat or niacin plus fat. Milk fat and rectal temperature were not affected by the DCAD level and supplementation of ruminally protected fat or niacin. However, respiration rate was decreased in cows fed diets supplemented with either fat or fat and niacin compared to those fed. The results of the present study indicated that higher DCAD (+30) and supplementation of fat along with niacin can somehow mitigate the negative effects of heat stress on milk yield and physiology of lactating Holsteins during July and August in Korea. In present study reduced respiration rate and increased milk yield in lactating cows may be attributed to the cooling effect of supplemented fat along with vasodilatory functions of niacin.   • This study was carried out to investigate effect of claw trimming on milk yield and its composition in Holstein at different lactation stages. 1 . There was no difference in daily milk yield between control and claw trimming in early, mid and late lactating Holsteins. 2. Somatic cell count (SCC) was lower in early lactation and it was higher in late lactation when claws were trimmed in Holstein. However, claw trimming did not affect SCC during mid lactation in Holstein. 3. Milk fat, protein and total solids were decreased during late lactation in Holstein after claw trimming. However, milk composition was not affected by claw trimming in early and mid lactating Holsteins.

Evaluation of Microbially Ensiled Spent Mushroom (Pleurotus osteratus) Substrates (Bed-Type Cultivation) as a Roughage for Ruminants (균상재배 느타리버섯부산물 생균발효사료의 반추동물 조사료원으로서의 가치 평가)

  • An in situ ruminal disappearance trial and an in vivo sheep metabolism trial were conducted to evaluate the nutritional value of spent mushroom substrate (SMS, originated from Pleurotus osteratus bed-type cultivation). The raw SMS was ensiled (ESMS) for 30 days with 5% (w/w, DM basis) molasses, 0.5% (v/w) yeast (Saccharomyces cerevisiae) and 0.5% (v/w) lactic acid bacteria (Lactobacillus plantarum). Two ruminally cannulated Holsteins (average BW 620 kg) were used to evaluate in situ disappearance. Six sheeps (average BW 48 kg) were fed, in $3{\times}3$ Latin square design, rice straw alone (Control), 25% (ESMS-25) and 50% (ESMS-50) of rice straw were replaced with ensiled SMS. For an in situ trial, ruminal DM and neutral detergent fiber (NDF) disappearance of SMS were increased after ensiling (P<0.05). For a sheep trial, protein and fiber (NDF, acid detergent fiber, crude fiber) digestibilities were decreased (P<0.05), crude ash digestibility was increased (P<0.001), and nitrogen retention was not affected (P>0.05) as rice straw was replaced with ensiled SMS. Ruminating time was decreased by an average of 28% by feeding ensiled SMS (P<0.05). Ensiled SMS (Bed-type cultivation) had 76% of energy value of rice straw. Consequently, ensiled SMS (Bed-type cultivation, 100% cotton waste) could be used as a roughage source appropriate for maintenance type rations for ruminants.

Results of Embryo Transfer with Hanwoo Embryos Produced In-Vivo or In-Vitro to Holstein Cows as Recipients (체내 또는 체외에서 생산된 한우 수정란을 젖소 수란우에 이식한 결과)

  • This study was performed to investigate the result that in-vivo or in-vitro embryos of Hanwoo cows were transferred to Holstein cows. Seventeen Hanwoo cows were used as donors for production of in-vivo embryos and fresh hanwoo in-vivo embryos were transferred to 1,150 Holsteins. And 2 embryos were transferred to 188 Holstein recipients to produce twin calves. Diagnosis on pregnancy was performed by rectal palpation at $60\sim90$ days after transfer. The pregnancy rate of Holstein recipients was 55.8% after transfer with Hanwoo in-vivo embryos and 38.2% after transfer with Hanwoo in-vitro embryos. The delivery rate of pregnant Holstein recipients was 88.4% after transfer with Hanwoo in-vivo embryos and 75.6% after transfer with Hanwoo in-vitro embryos. The rate of delivery of Holstein recipients transferred with two Hanwoo embryos was 36.2% and the rate of twin production was 25.9%. The rate of twin production by embryo transfer with in-vivo embryos was 30.4%, whereas the fate with in-vitro embryos was 15.6%. The pregnancy rate according to the grade of corpus luteum of Holstein recipients transferred with Hanwoo in-vitro embryos was 41.5 and 36.0% for A and B grade, respectively. The pregnancy rate according to the transfer in site in the uterine lumen of recipients was 40.9 and 32.7% for anterior and middle site, respectively. The pregnancy rate according to day of embryo transfer after estrus of recipients was 45.5, 38.8 and 39.7% for day 6, day 7 and day 8, respectively. There was difference of pregnancy rate according embryo transfer technician ($30.5\sim45.8%$) individual dairy farm ($21.1\sim51.0%$). These results are supposed to indicate that the rate of pregnancy after transfer with Hanwoo embryos to Holstein recipients was similar to that within the same breed, and consequently that this method would be beneficial to enhance the productivity in Hanwoo reproduction.

(Co)heritability of acetone and β-hydroxybutyrate concentrations in raw milk related to ketosis in Holsteins (홀스타인 젖소의 케톤증과 관련된 원유속 아세톤과 β-히드록시부틸산 함량에 대한 (공)유전력)

  • This experiment was conducted to estimate the heritability and coheritablity of daily milk yield, acetone and ${\beta}$-hydroxybutyrate (BHBA) concentrations in raw milk. The average concentrations of acetone and BHBA were $135.54{\pm}96.29{\mu}mol$ and $61.08{\pm}66.76{\mu}mol$, respectively, and the differences between high group and low group cows were highly significant (p <0.01). The estimates of heritability of daily milk yield, acetone and BHBA concentrations were in the range of 0.18~0.21, 0.11~0.13 and 0.01~0.02, respectively. The estimate of heritability of $Log_e$acetone did not change much, while that of $Log_eBHBA$ increased to 0.03~0.04. The estimates of phenotypic and genetic correlation coefficients between acetone and BHBA were 0.44 and 0.48, respectively. In low milk yield group, the coheritability estimates of BHBA and $Log_eBHBA$ when selection was for daily milk yield were 0.26 and 0.32, respectively. These were higher than the coheritability estimate of acetone when selection was for daily milk yield. The same trend was noted in the coherihability estimates from the whole records using both high and low milk yield groups together. BHBA concentration seemed to be more effectively responding than acetone concentration when selection was for daily milk yield.
