This research was investigated the relationship between the number of the transferable embryos and estrus expression rate, BCS (Body Condition Score), which affect the nutritional state of the cow, in Holstein donor cows. CIDRs were inserted into the vaginas of twenty two head of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. When BCS was $$\leq_-$$2.5, the total number of collected ova was 7.3 + 1.9, which is significantly lower (p<0.05) than the numbers 15.4 + 2.8 and 15.4 + 2.1 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively. Whereas the numbers of transferable embryos were 5.2 + 1.4 when BCS was $$\leq_-$$2.5, which was smaller than the numbers 6.0 + 2.1 and 8.5 + 1.8 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively; however, the differences were not significant. As for estrus induction rate, the cow groups whose BCSs were 2.75 and $$\geq_-$$3.0 showed 100.0% and 95.0%, respectively. Whereas the cow group whose BCS was $$\leq_-$$2.5 showed 57.1%, and the differences were significant (p< 0.05). As for estrous expression rate, the cow groups whose BCSs were $$\leq_-$$2.5, 2.75 and $$\geq_-$$3.0 showed 100.0%, 100.0% and 85.7%, respectively; however, the differences were not significant. According to the result of this research, it is considered that the total number of collected ova and the number of transferable embryos will be affected by the nutritional state before and after in vivo embryo production and superovulation treatment, and that although the mechanism is not clear, poor stockbreeding management and nutritional level would cause the decrease of ovum recovery rate and the number of transferable embryos in high-producing cows. On the other hand, diverse researches on the superovulation treatment method that is suitable for high-producing Holstein donor cows would contribute to preventing ovarian cyclicity disorder, as well as to the early multiplication of cows with superior genes by increasing the utilization value of donor cows.
Prolactin (PRL) plays an important role in promoting mammalian mammary gland development, and milk production during lactation. Therefore the PRL gene was chosen as a candidate gene for milk traits in Holstein dairy cows. PCR-SSCP and PCR-RFLP were used to analyze genetic variations in the 5' regulation region of the PRL gene. In this part of the gene, two new polymorphic sites were detected in the Chinese Holstein dairy cows. One was a XbaI-RFLP locus, and the other was an SSCP locus. Statistical analysis showed that the XbaI-RFLP locus and the SSCP locus had a significant positive effect on milk traits.
The objectives of this study were to evaluate the effects of different supplemental levels of Saccharomyces cerevisiae fermentation product (SCFP; Original XP; Diamond V) on lactation performance in Holstein dairy cows under heat stress. Eighty-one multiparous Holstein dairy cows were divided into 27 blocks of 3 cows each based on milk yield ($23.6{\pm}0.20kg/d$), parity ($2.88{\pm}0.91$) and day in milk ($204{\pm}46d$). The cows were randomly assigned within blocks to one of three treatments: 0 (control), 120, or 240 g/d of SCFP mixed with 240, 120, or 0 g of corn meal, respectively. The experiment was carried out during the summer season of 2014, starting from 14 July 2014 and lasting for 9 weeks with the first week as adaption period. During the experimental period, average daily temperature-humidity index (measured at 08:00, 14:00, and 20:00) was above 68, indicating that cows were exposed to heat stress throughout the study. Rectal temperatures tended to decrease linearly (p = 0.07) for cows supplemented with SCFP compared to the control cows at 14:30, but were not different at 06:30 (p>0.10). Dry matter intake was not affected by SCFP supplementation (p>0.10). Milk yield increased linearly (p<0.05) with increasing levels of SCFP. Feed efficiency (milk yield/ dry matter intake) was highest (p<0.05) for cows fed 240 g/d SCFP. Cows supplemented with SCFP gained (p<0.01) body weight, while cows in the control lost body weight. Net energy balance also increased linearly (p<0.01) with increasing levels of SCFP. Concentrations of milk urea nitrogen (p<0.01) decreased linearly with increasing levels of SCFP, while no difference (p>0.10) was observed among the treatments in conversion of dietary crude protein to milk protein yield. In summary, supplementation of SCFP alleviated the negative effect of heat stress in lactating Holstein dairy cows and allowed cows to maintain higher milk production, feed efficiency and net energy balance. Effects of SCFP were dose-dependent and greater effects were observed from higher doses.

Background: The milk yield can be affected by the frequency of milking per day, in dairy cows. Previous studies have shown that the milk yield is increased by 6.25 % per lactation when the milking frequency is increased from 2 to 3 times per day while the somatic cell count is decreased. To investigate the effect of milking frequency (3X vs. 4X) on milk yield and it's genetic parameters in the first and second lactations of the Iranian Holstein dairy cows, a total of 142,604 test day (TD) records of milk yield were measured on 20,762 cows. Results: Heritability estimates of milk yield were 0.25 and 0.19 for 3X milking frequency and 0.34 and 0.26 for 4X milking frequency throughout the first and second lactations, respectively. Repeatability estimates of milk yield were 0.70 and 0.71 for 3X milking frequency and 0.76 and 0.77 for 4X milking frequency, respectively. In comparison with 3X milking frequency, the milk yield of the first and second lactations was increased by 11.6 and 12.2 %, respectively when 4X was used (p < 0.01). Conclusions: Results of this research demonstrated that increasing milking frequency led to an increase in heritability and repeatability of milk yield. The current investigation provided clear evidences for the benefits of using 4X milking frequency instead of 3X in Iranian Holstein dairy cows.

The fertility of dairy cows has been declining worldwide. The number of services per conception has increased, and repeat breeder (RB) cows are considered important in the dairy industry. However, there has been little research on RB cows in Korea. The objective of this study was to investigate the relationship between serum biochemical profiles and RB Holstein cattle in Korea and compare the results with those of studies conducted abroad. In addition, we investigated hidden factors that are needed for RB cattle to become pregnant. Overall, 34 Korean Holstein Friesian cows were divided into three groups: pregnant with normal cycling (PNC), pregnant with repeat breeder (PRB), and non-pregnant with repeat breeder (NRB). Blood samples were collected from the jugular vein, and serum parameters (ALT, AST, ALP, GGT, LD, CK, TP, ALB, GLOB, TBIL, BUN, CRE, GLU, TC, TG, NEFA, CA, MG, and IP) were analyzed. GGT levels were significantly different among groups (P=0.01). The concentration of GGT was the highest in the NRB and the lowest in the PNC. In contrast to the findings of other studies, there were no differences in GLU, TP, TC, and BUN levels. This study is the first to investigate serum biochemistry in RB cattle and provides results that differ from those of previous studies; these findings would help establish a novel approach to improve fertility of RB cattle.

Background: Research on loci influencing milk production traits of dairy cattle is one of the main topics of investigation in livestock. Many genomic regions and polymorphisms associated with dairy production have been reported worldwide. In this context, the purpose of this study was to identify candidate loci associated with milk yield in Argentinean dairy cattle. A database of candidate genes and single nucleotide polymorphisms (SNPs) for milk production and composition was developed. Thirty-nine SNPs belonging to 22 candidate genes were genotyped on 1643 animals (Holstein and Holstein x Jersey). The genotypes obtained were subjected to association studies considering the whole population and discriminating the population by Holstein breed percentage. Phenotypic data consisted of milk production values recorded during the first lactation of 1156 Holstein and 462 Holstein ${\times}$ Jersey cows from 18 dairy farms located in the central dairy area of Argentina. From these records, 305-day cumulative milk production values were predicted. Results: Eight SNPs (rs43375517, rs29004488, rs132812135, rs137651874, rs109191047, rs135164815, rs43706485, and rs41255693), located on six Bos taurus autosomes (BTA4, BTA6, BTA19, BTA20, BTA22, and BTA26), showed suggestive associations with 305-day cumulative milk production (under Benjamini-Hochberg procedure with a false discovery rate of 0.1). Two of those SNPs (rs43375517 and rs135164815) were significantly associated with milk production (Bonferroni adjusted p-values < 0.05) when considering the Holstein population. Conclusions: The results obtained are consistent with previously reported associations in other Holstein populations. Furthermore, the SNPs found to influence bovine milk production in this study may be used as possible candidate SNPs for marker-assisted selection programs in Argentinean dairy cattle.
This study aimed to improve the reproductive efficiency of dairy herds by comparison and analyzing estrous appearance rate, conception and non-conception rate according to the stage of lactation using the lactation and reproductive records of average (less than 10,000 liters milk in 305 days) and high yielding (more than 10,000 liters milk in 305 days) Holstein cows (n=102). Milk production and reproduction data were collected between January 2010 and December 2012 from Holstein cows kept in the commercial dairy farms. Average (n=32) and high yielding (n=24) Holstein cows used to analyze the relationship between milk yield and reproductive performance. Our results showed that estrous appearance rate according to the stage of lactation was 25.0% (30~59d), 40.6% (60~89d), 25% (90~110d) and 9.4% (>111d) in average yielding cows and 16.7% (30~59d), 20.8% (60~89d), 12.5% (90~110d) and 50.0% (>111d) in high yielding cows, respectively. Conception rate according to the stage of lactation was 87.5% (30~59d), 61.5% (60~89d), 75.0% (90~110d) and 66.7% (>111d) in average yielding cows and 25.0% (30~59d), 0% (60~89d), 33.3% (90~110d) and 50.0% (>111d) in high yielding cows, respectively. Days between parturition and conception was 23.7% (<149d), 0% (150~209d) and 0% (>210 d) in average yielding cows and 69.0% (<149 d), 77.8% (150~209d) and 38.9% (>210d) in high yielding cows, respectively. Conception rate from 110 days postpartum in high yielding cows was 41.7% (110~150d), 50.0% (151~180d) and 50.0% (>181d). Body condition score (BCS) in 120 days postpartum was $2.64{\pm}0.1$ in average yielding cows and $2.28{\pm}0.1$ in high yielding cows, respectively.
This research was investigated the relationship, in high-producing Holstein donor cows, between the number of the transferable embryos and the blood serum concentrations of Blood Urea Nitrogen (BUN), glucose and cholesterol, which affect the nutritional state of cows. CIDRs were inserted into the vaginas of twenty two heads of Holstein cows, regardless of estrous cycle. The total numbers of ova collected from 3 experimental groups whose blood BUN concentrations were <10 mg/dl, 11~18 mg/dl and ${\geq}19$ mg/dl were 8.9, 12.5 and 19.0, respectively; whereas the numbers of transferable embryos were 5.8 + 1.9, 7.9 + 2.8 and 5.2 + 1.4, respectively. When glucose concentration was <60 mg/dl, the total number of collected ova was 9.9, which was smaller than when the concentration was 60~70 mg/dl or ${\geq}70$ mg/dl. When glucose concentration was 60~70 mg/dl, the number of transferable embryos was 7.1 + 2.4, which was slightly larger than the numbers 6.4 + 2.1 and 6.1 + 1.7 that were obtained when the concentrations were <60 mg/dl and ${\geq}70$ mg/dl, respectively ; however, the differences were not significant (p>0.05). When cholesterol concentrations were <150 mg/dl, 150~200 mg/dl and ${\geq}200$ mg/dl, the total numbers of collected ova were 11.2, 11.3 and 8.6, respectively. Whereas the numbers of transferable embryos were 7.1 + 2.1, 7.3 + 1.9 and 5.6 + 1.3, respectively ; however, the differences were again not significant (p>0.05). The result of this research showed no significant difference in ovum recovery rate and the number of transferable embryos according to major metabolite concentrations in high-producing Holstein donor cows. However, it is considered that the failure of maintaining proper nutritional status would cause the fall in in vivo embryo productivity.
This study was conducted to investigate the effects of supplementing additional sucrose, in the form of black sugar (BS), into the diet of Holstein dairy cows on dry matter intake (DMI), milk yield, and milk composition. Eight Holstein dairy cows ($741{\pm}65.8kg$ body weight) were divided into two groups, including the control and BS groups. Animals in the control group were offered a total mixed ration (TMR) ad libitum, and the BS group was offered TMR with 300 g of BS/head/d. After two weeks of adaptation period, the animal performance, including DMI, milk yield and milk composition, was measured. Cows supplemented with BS appeared to consume more feed than that by the controls (i.e., 17.08 and 18.28 kg/d for the control and BS groups, respectively). However, there were no significant differences between treatments. Milk yield or milk composition, such as milk fat, milk protein, lactose, solids-non-fat, total solids and pH, did not differ between treatments. However, there was a significant difference (p<0.05) in the concentration of milk urea nitrogen (MUN). The MUN concentration of the BS group was approximately 15% lower than that of the control group (i.e., 18.75 vs. 16.05 mg/dL for the control and BS groups, respectively), which suggests improved nitrogen metabolism in the animals. The somatic cell count was numerically lower in the cows of the BS group compared to those in the control group. However, a significant difference was not noted due to the substantial amount of variation among cows. In terms of the trace mineral composition for milk, the concentration of Cu from BS animals was higher (p<0.05) than that of the control animals. In summary, supplementing the diets of dairy cows with BS marginally affected animal performance and improved nitrogen metabolism. The level of supplementation and other factors, such as animal variation were discussed.
