Hypoxic condition enhances chondrogenesis in synovium-derived mesenchymal stem cells

  • Background: The chondrogenic differentiation of mesenchymal stem cells (MSCs) is regulated by many factors, including oxygen tensions, growth factors, and cytokines. Evidences have suggested that low oxygen tension seems to be an important regulatory factor in the proliferation and chondrogenic differentiation in various MSCs. Recent studies report that synovium-derived mesenchymal stem cells (SDSCs) are a potential source of stem cells for the repair of articular cartilage defects. But, the effect of low oxygen tension on the proliferation and chondrogenic differentiation in SDSCs has not characterized. In this study, we investigated the effects of hypoxia on proliferation and chondrogenesis in SDSCs. Method: SDSCs were isolated from patients with osteoarthritis at total knee replacement. To determine the effect of oxygen tension on proliferation and colony-forming characteristics of SDSCs, A colony-forming unit (CFU) assay and cell counting-based proliferation assay were performed under normoxic (21% oxygen) or hypoxic (5% oxygen). For in vitro chondrogenic differentiation, SDSCs were concentrated to form pellets and subjected to conditions appropriate for chondrogenic differentiation under normoxia and hypoxia, followed by the analysis for the expression of genes and proteins of chondrogenesis. qRT-PCR, histological assay, and glycosoaminoglycan assays were determined to assess chondrogenesis. Results: Low oxygen condition significantly increased proliferation and colony-forming characteristics of SDSCs compared to that of SDSCs under normoxic culture. Similar pellet size and weight were found for chondrogensis period under hypoxia and normoxia condition. The mRNA expression of types II collagen, aggrecan, and the transcription factor SOX9 was increased under hypoxia condition. Histological sections stained with Safranin-O demonstrated that hypoxic conditions had increased proteoglycan synthesis. Immunohistochemistry for types II collagen demonstrated that hypoxic culture of SDSCs increased type II collagen expression. In addition, GAG deposition was significantly higher in hypoxia compared with normoxia at 21 days of differentiation. Conclusion: These findings show that hypoxia condition has an important role in regulating the synthesis ECM matrix by SDSCs as they undergo chondrogenesis. This has important implications for cartilage tissue engineering applications of SDSCs.

Coactivity of Mast Cells and Stem Cells on Angiogenesis and Antioxidants' Potentials at Inflammation, Proliferation, and Tissue Remodeling Phases of Wound

  • Background Reactive oxygen species cause serious damage to the physiological function of tissues. Determination of total antioxidant capacity of skin tissue is one of the determinants of damaged tissue function. Mast cells (MCs) are one of the groups of cells that are invited to the site of injury. The healing process begins with the rapid release of various types of MCs' intermediate factors at the site of injury. Bone marrow mesenchymal stem cell (BMMSC) production and secretion have been shown to regenerate the skin. The aim of this research was to evaluate the wound-healing and antioxidant effects of BMMSCs per MCs. Methods Fifty-four albino Wistar male rats were divided into three groups: (1) nonsurgery, (2) surgery, and (3) surgery + BMMSCs. Groups 2 and 3 were operated with a 3 × 8 cm flap and in group 3, cell injections (7 × 109 cell injection at the time of surgery) were performed. After days 4, 7, and 15, percentage of the surviving tissue, histological characteristics, superoxide dismutase (SOD) activity, and amount of malondialdehyde (MDA) were measured in the groups. For results, Graph Pad Prism 8 software was used, and data were analyzed and compared by analysis of variance and Tukey test. Results BMMSCs' application decreased the amount of MDA, increased SOD activity and survival rate of the flaps, and improved the histological characteristics. Conclusion This study revealed the protective effects BMMSCs alongside MCs against oxidative stress on the survival of the flaps. However, for clinical use, more research is needed to determine its benefits.

A Chronic-Low-Dose Exposing of DEHP with OECD TG 443 Altered the Histological Characteristics and Steroidogeic Gene Expression of Adrenal Gland in Female Mice

  • Phthalates and their metabolites are well-known endocrine disrupting chemicals. Di-(2-ethylhexyl) phthalate (DEHP) has been widely used in industry and the exposing possibility to adult is high. In this study, DEHP was treated (133 ㎍/L and 1,330 ㎍/L in drinking water) according to the OECD test guideline 443 to mature female mice and their adrenal gland were examined for histological characteristics and steroidogenic gene expression. The wet weight of the adrenal gland was increased in all administrated groups compared to control. The diameter of zona fasciculata (ZF) was increased by DEHP in both outer ZF and inner ZF but there was no difference in morphology of the cells and arrangements into zona between groups. In addition, the arrangement of extracellular matrix was not different between control and DEHP groups. CYP11B1 was mainly localized at ZF and the intensity was not different between groups. DAX1 was localized in zona glomerulosa (ZG) and ZF, and its expression levels were decreased by DEHP administration. Its level was lower in DEHP133 group than DEHP1330 group. On the other hand, CYP17A1 was localized in ZG of DEHP1330 group. These results suggest that chronic low-dose DEHP exposing may modify the microstructure and function of the adrenal cortical cortex.

광주기에 따른 비단털쥐(Mammalia: Rodentia: Cricetidae) 암컷의 발정주기 변화 (Estrus Cycles of the Female Tscherskia triton (Mammalia: Rodentia: Cricetidae) according to the Photoperiod)

  • 비단털쥐 (Tscherskia triton)는 중국 북부와 한국, 극동러시아 지역 등 국한된 지역에 분포하며, 번식, 성장과 발달 및 분포에 관한 정보는 알려져 있으나 발정주기와 생식기관에 관한 번식생물학적 연구는 거의 연구된 바 없다. 이에 본 연구는 제주도에서 포획된 개체를 사육하면서 광주기에 따른 암컷 비단털쥐의 생식주기 및 생식기관의 특징을 밝혀 비단털쥐의 번식생물학적 정보를 제공하고자 진행하였다. 연구결과, 평소 사육환경(16L : 8D)에서의 비단털쥐 발정주기는 4~5일이었고, 발정 단계는 발정전기, 발정기, 발정후기, 발정휴지기의 4단계로 구분되었다. 광주기 8L : 16D 환경에서 비단털쥐의 발정주기는 6~12일째부터 발정휴지기 상태가 계속 유지되었다. 광주기에 따른 비단털쥐 생식기관의 조직도 차이를 보였다. 16L : 8D 경우에는 발달된 난소 내에 다수의 성숙 난포와 황체가 관찰되었고, 자궁내막도 두껍고 분비샘도 잘 발달되어 있었으나 8L : 16D 조건에서는 원시난포와 제2차, 3차 난모세포가 관찰되고 얇은 자궁내막과 발달되지 않는 자궁분비샘을 확인하였다. 이에 따라 광주기에 따른 비단털쥐의 생식주기 변화와 난소와 자궁의 조직학적 차이를 확인하였다. 이 결과는 비단털쥐의 번식생물학적 측면에서 매우 중요한 단서가 되는 자료이기에 생물종 다양성 유지를 위한 자료로 널리 활용될 것이라 판단된다.

Tensile Characteristics and Behavior of Blood Vessels from Human Brain in Uniaxial Tensile Test

  • The rupture of blood vessels in the human brain results in serious pathological and medical problems. In particular, brain hemorrhage and hematomas resulting from impact to the head are a major cause of death. As such, investigating the tensile behavior and rupture of blood vessels in the brain is very important from a medical point of view. In the present study, the tensile characteristics of the blood vessels in the human brain were analyzed using a quasi-static uniaxial tensile test, and the properties of the arteries and veins compared. In addition, to compare the tensile behavior and demonstrate the validity of the experimental results, blood vessels from the legs of pigs were also tested and analyzed. The overall results were in accordance with the histological structures and previous medical reports.

Characteristics of Hodgkin Lymphoma in a Defined Group of Iranian Pediatric Patients

  • This study was conducted to describe the characteristics of Hodgkin lymphoma in Iranian children. In a referral center for pediatric oncology (Mofid Hospital) in Tehran, patient data over a 10-year period were retrieved and recorded accordingly. Among 82 cases, 73.2% were male, 26.8% were female, and 70.7% were 5-9 years old. About 40% of patients were in stage III and 42.7% had systemic signs. Cervical nodes were commonly involved (91.5%). The most frequent histological subtype was mixed cellularity. The main hematological features were anemia (47.6%), lymphopenia (20.7%), and eosinophilia (8.7%). Survival rate was 72%, and 8.4% of patients were deceased. A 3% recurrence rate was observed in our patients. A significant relationship was found between the stage of disease and systemic signs (P<0.0005, ${\chi}^2$). Despite diagnosis of Hodgkin lymphoma in many children in Iran being made in higher stages, the mortality rate is relatively low.

Agrobacterium rhizogenes에 의하여 형질전환된 인삼 모상근의 세포 유전학적 및 조직학적 특성 (Cytogenetic and Histological Characteristics of Ginseng Hairy Root Transformed by Agrobacterium rhizogenes)

  • Ginseng (Panax ginseng C.A. Meyer) hairy root transformed with Agrobacterium rhizogenes (strain A4) was examined cytogenetically and histologically to assess its characteristics. The optimum growth of hairy root obtained in hormone-free MS medium (sucrose 30 g/L, pH5-6) for long period cultures. All hairy root strains (HB1, HB2, HB3) had the 2n diploid number (2n=48) of chromosomes in root tip cells. There were no alteration in chromosome structure except in one cell of HB3 strain. Results of SDS-PAGE showed a few difference in pattern and number of bands between normal and hairy root of ginseng. The root anatomy of normal root and hairy root differed each other. The hairy root had a clearly defined vascular strand, and the morphology of cortical cells were disorganised with large intercellular spaces.

소아 Helicobacter pylori 위염에서 균 박멸과 관련된 위염의 내시경 및 조직학적 변화에 대한 연구 (Evaluation of Endoscopic and Histological Change of Gastritis Related to Helicobacter pylori Eradication in Children)

  • 목 적: 소아 H. pylori 위염에서 균 박멸 후의 조직학적 변화에 대한 연구는 거의 없는 실정이며 치료전의 조직소견과 균 박멸과의 관련성 또한 잘 밝혀져 있지 않다. 이에 저자들은 H. pylori 위염 소아들을 대상으로 치료 전후의 내시경 및 조직소견을 분석하여 균 박멸로 초래되는 위염의 내시경적, 조직학적 변화 및 균 박멸의 성공에 영향을 주는 치료 전 조직소견들을 알아보고자 하였다. 방 법: 95년 6월부터 99년 5월까지 만성 복통으로 내원하여 치료 전후 내시경 생검을 시행받은 소아들 중 조직 검사상 균이 관찰되면서 urease 검사가 양성인 만성위염 소아 42명을 대상으로 하였다. 박멸요법 후 4주 이후에 추적 내시경 검사를 시행하여 위 두 가지 검사 및 배양 검사가 모두 음성인 경우를 균 박멸로 판정하였고, 조직 소견상 위전정부의 균밀도 및 염증 소견들을 최근의 시드니 위염분류법에 따라 0, 1, 2, 3으로 등급화하여 분석하였다. 또한 OAC로 치료받은 32명의 소아들을 박멸에 성공한 군과 실패한 군으로 나누어 각각의 경우 시드니 위염분류법에 따른 치료 전 조직 소견에 차이가 있는지를 분석하였다. 결 과: 1) 균이 박멸된 소아들(27례)의 경우 결절성 위염은 치료 전 89%에서 치료 후 63%로 유의하게 감소하였다(p<0.05). 조직 소견상 호중구 침윤등급은 평균 2.06에서 0.24로 현저하게 감소하였고(p<0.01), 만성염증세포 침윤등급은 평균 2.61에서 1.89로 유의하게 감소하였으나(p<0.05) 완전히 호전된 예는 없었다. 림프여포는 48%에서 15%로 유의하게 감소하였고(p<0.05), 점막 상피손상은 치료 후 모두 소실되었다. 균박멸에 실패한 소아들(15례)의 경우는 치료 전후 결절성 위염의 빈도, 조직 소견상 호중구 및 만성염증세포 침윤등급, 림프여포의 빈도에 유의한 변화가 없었다. 2) OAC 세 약제 치료 후 성공적으로 균이 박멸된 소아들(18례)과 균 박멸에 실패한 소아들(14례)의 치료전 내시경 소견과 조직소견을 비교하였을 때 결절성 위염의 빈도, 호중구 및 만성 염증세포침윤등급, 림프 여포 및 상피손상의 빈도에 모두 유의한 차이가 없었으나, 균밀도는 균박멸에 실패한 소아들에서 더 높은 경향을 보였다(p=0.072). 결 론: 소아 H. pylori 위염에서 균박멸 치료 4주 이후 호중구 침윤은 현저한 감소를 보이며, 만성염증세포 침윤정도, 림프여포와 내시경상 결절성 위염도 의미 있는 호전을 보인다. 시드니 위염분류법에 따른 치료 전의 조직학적인 등급 중 균밀도는 높을수록 균박멸이 낮은 경향이 있어서 치료의 성공여부를 예측하는 데 이용될 수 있는 가능성이 있다.

  • 기아에 따른 버들치 Rhynchocypris oxycephalus의 생화학적, 조직학적 변화를 파악하기 위해, 간의 간중량지수, 단백질 함량, RNA 량 및 DNA 량 조사와 더불어 간세포 및 중장 상피 세포의 조직학적 조사를 실시하였다. 기아군은 포식군에 비해 간의 단백질 함량, RNA 량 및 DNA 량에서 높은 값을 보였지만, 먹이 공급 중단은 간 중량지수, 간 세포 핵 크기 및 중장 상피 세포 핵 높이에 있어서 감소를 나타내었다. 본 실험에서 조사된 RNA-DNA 비는 버들치의 영양 상태 파악 지수로 유용하였으며 또한, 버들치 표본 시기에서의 높은 성장 단계인지 혹은 낮은 성장 단계인지의 성장 활성 파악을 가능케하였다. 본 연구 결과의 자료를 구체적으로 해석하였으며, 아울러 다수의 생물학적 중요 연관 사항들을 고찰하였다.

Right-Sided and Left-Sided Colon Cancers are Two Distinct Disease Entities: an Analysis of 200 Cases in Pakistan

  • Background: There is growing evidence that there are differences in histological and genetic characteristics along with clinical behavior between right- and left-sided colon carcinomas. We have compared various parameters of the two types and assessed associations of the results with prognosis in patients in Pakistan. Materials and Methods: We reviewed 200 cases from our institutional database; 100 cases of right-sided and 100 cases of left-sided colon cancer. Parameters including age, gender, TNM stage, histological features and clinical outcome were analyzed. Results: The patients with right-sided colon cancer were significantly older as compared to their counterparts with left-sided cancer. They presented with a lumbar mass rather than symptoms of obstruction and perforation as seen in left-sided colon cancers, and the histology showed higher percentage of poorly differentiated tumors with advanced pT stage. Moreover, Crohn's-like reactions, intra tumoral lymphocyte responses and other poor prognostic factors like lymph vascular invasion and perineural invasion were more common in right-sided cancers. Conclusions: We found that right- and left-sided colon cancers are different from each other in terms of clinical presentation, histology and clinical behavior. Right-sided colon cancers are more aggressive and are associated with poorer clinical outcome as compared to left sided colon cancers in our population.
