Objectives: Bufonis venonum (BV) is toad venom and is the dried, white secretions of the auricular and the skin glands of toads. This study was performed to evaluate the toxicity of intravenous injection of Bufonis venonum pharmacopuncture (BVP) through a single-dose test with sprague-dawley (SD) rats. Methods: Twenty male and 20 female 6-week-old SD rats were injected intravenously in the caudal vein with BVP or normal saline. The animals were divided into four groups with five female and five male rats per group: the control group injected with normal saline, the low-dosage group injected with 0.1 mL/animal of BVP, the medium-dosage group injected with 0.5 mL/animal of BVP and the high-dosage group injected with 1.0 mL/animal of BVP. We performed clinical observations every day and body weight measurements on days 3, 7 and 14 after the injection. We also conducted hematology, serum biochemistry, and histological observations immediately after the observation period. Results: No mortalities were observed in any experimental group. Paleness occurred in the medium- and the high-dosage groups, and congestion on tails was observed in females in the medium- and the high-dosage groups. No significant changes in weight, hematology, serum biochemistry, and histological observations that could be attributed to the intravenous injection of BVP were observed in any experimental group. Conclusion: The lethal dose of intravenously-administered BVP in SD rats is over 1.0 mL/animal.
In the present study, the anti-inflammatory effect of "Jungcheonhwadamgangki-tang ga Antler" water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on body weight and ear weights and histopathological observation of induced ear were conducted with ear histomorphometry. The obtained results were as follows. The increases of absolute and relative ear weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, thickness and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler. In addition, the increases of ear thickness half and thickness full detected in the vehicle control were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang ga Antler-dosing groups. Base on these results, it is concluded that Jungcheonhwadamgangki-tang ga Antler extracts has clear anti-inflammatory effect on the acute inflammation such as bronchial asthma.
Primordial germ cells (PGCs) of White Leghorn chicken embryos as a donor were transferred to Rhode Island Red chicken embryos as a recipient. At 48-50 h (stage 13-15) of incubation of fertilized eggs, donor PGCs, which were taken out from blood vessels of donor embryos, were injected into blood vessels of recipient embryos. Sex of the treated embryos was determined after the transfer of PGCs using remaining blood samples. In the present experiments, survival rate of the treated embryos was 33.3% for homo-sexual and 35.4% for hetero-sexual transfers of PGCs, respectively, when determined at 17 days of incubation. In this study, most of the treated embryos could not survive more than 18 days of incubation, though the reason for that was not clarified in the present work. The gonalds removed from embryos that died after 18 days of incubation and the organs from newly hatched chicks were examined for morphological and histological features. The gonads removed from the embryos with homo-sexual transfer of PGCs showed normal development in appearance. On the contrary, some (35.3%) of the embryos with hetero-sexual transfer of PGCs possessed abnormal gonads similar to ovotestis by histological observation. In cases where the gonads developed to be normal organs (64.7%) the sex of embryos was the same as recipient ones. The present results suggest that hetero-sexual transfer of the PGCs may bring about the possibility of development of the embryos bearing sexually different gametes, spermatogonia or oogonia.
The androgenic gland secretes a hormone, androgenic gland hormone, which is believed to act on the differentiation of the primary, secondary, and behavioral sex characteristics in most malacostracan crustaceans. Based on the changes in gonado-somatic indexe and histological observation of testis and androgenic gland cell, testicular maturation and spermatogenesis of M. nipponense occurred early in summer (May to July), and generally spermatogenesis was absent in August. It could be, concluded that May to July is the period when adult males of this species are sexually active, and androgenic gland showed signs of increased secretory activity.
Background: In the early stages of development, teeth and lingual papillae are induced and developed through special and complex epithelial-mesenchymal interactions. Tooth completion indicates the beginning of the weaning phase, and accordingly, many oral tissues and organs are completed, and it is thought that their developmental completion times are related to each other. The purpose of this study was to clarify the embryonic and neonatal development of the filiform papillae and mandibular molar tooth, and discuss the developmental relationship between these organs by comparing the developmental completion times. Methods: Embryos at embryonic day 15 (EM15), 17 (EM17), and 21 (EM21) and mice at neonatal day 1 (NE1), 5 (NE5), 10 (NE10), and 21 (NE21) were used for experimentation. Tissues dissected from embryos and mice were fixed, and processed for histological analysis. Sections from the tissues were stained with hematoxylin and eosin for observation under a light microscope. Results: Based on the histological analysis results, the developmental process of the lingual epithelium covering the dorsal surface of the tongue was classified into three stages: initiation, morphogenesis, and functional. The development of the filiform papillae begins at EM17; undergoes rapid morphological changes in epithelial cells at EM21, PN1 and PN5, and reaches the functional stage at PN10, which is the sucking phase. Tooth development begins at EM13 or 15 and is completed at NE21 through prenatal and postnatal development. Conclusion: The development of the filiform papillae was initiated late and completed quickly through embryonic and neonatal development in comparison with the mandibular molar tooth. The filiform papillae are considered to play an important role in sucking rather than mastication as it is completed in the sucking phase.
Histological changes occurred by freezing storage and subsequent thawing, and changes of muscle fiber after heating and drying of the eel (Anguilla japonica) were observed under microscope . The results are as follows : (1) After one month of freezing storage the muscular tissue produced considerable number of extracellular and intracellular ice crystals. The sample stored at the temperature of $-40^{\circ}C$ produced ice crystals inside the muscle cells while sample stored at $-20^{\circ}C$, outside. (2) No changes were observed in the hypodermic fat after thawing regardless of storage temperature, while insufficient recovering of muscle cells were detected in the muscular tissue. Muscular tissues which have been stored $-20^{\circ}C$ showed severe change in shape due to dehydration. (3) Microscopic observation on muscle homogenate showed loss of transparency due to free-zing, disfiguration and contraction by drying and water seperation, and elasticity by heating.

The aim of this study was to investigate the effects of caponization on the bone characteristics, biomechanical property and histology in Taiwan country chickens fed to market age of 26 wks. Male Taiwan country chickens $D{\times}L_2$ were caponized or sham-operated at 8 wks of age, and selected healthy sham-operated and completely caponized chickens (prominent degenerated comb) were selected at 16 wks old and fed to 26 wks old for the trials. Fifteen intact male chickens (Intact), sham-operated chickens (Sham) and caponized chickens (Capon) were assigned for trial 1, and sixteen Intact and Capon were assigned for trial 2. Results in trial 1 showed that the abdominal fat and relative abdominal fat weights of Capon were significantly heavier than Intact and Sham (p<0.05), while the tibia weight and relative weight were the lightest (p<0.05). The tibia breaking strength, bending moment and stress of Capon were the poorest among groups (p<0.05). The trial 2 produced the similar observation that Capon were significantly lighter than Intact (p<0.05) in the tibia weight, relative tibia weight and their biomechanical properties. On histological determinations, Capon showed a thinner cartilage end and fewer chondrocytes (about 50%) and trabecular, and bigger marrow cavity; while decreased hemopoietic cells number with increased adipocytes than Intact observed by H&E stain and at low magnification. At high magnification, Capon showed a decrease in the chondrocyte size by 33 to 50%, with smaller nucleus located near the cell membrane, and exhibited monocellular form chondrocytes. Capon also showed a less strongly acidic sulfated mucosubstance with weaker dyeing property within cartilage zone, and smaller chondrocytes size by Alcian blue stain.
The objective of this study was to investigate the effects of implanted chitosan applied to surgically created wound in Japanese Macaque monkeys. 4 healthy Japanese Macaque monkeys were used. A 4 cm straight skin incision was made and undermined skin ($4{\times}4cm$) over on the 2 monkeys both sides of the dorsal midline, and a 4 cm circular skin incision was made on 2 monkeys both sides of the dorsal midline. One wound (left side) was implanted 1 mg (straight incision) and daily 0.2 mg (circular incision) of cotton type chitosan and the other wounds were treated with normal saline (3 ml) in monkeys. Each straight wound was closed with two interrupted sutures of 2-0 sutures. The monkey's circular skin incision is opened. At 14 days after initial wounding, each wound was taken for histological observations in monkeys. The inflammatory cells in the chitosan group are observed less than the control group, the collagen and the fibrin in the chitosan are observed more than the control group in monkeys. So the wound healing is moderately enhanced for chitosan treatment. The fibroblasts and the capillaries increased for chitosan treatment. The treatment of chitosan in wound is to promote healing.
Vitamin D is one of important factors involved in the regulation of bone metabolism. In osteoporosis, the therapeutic effect of vitamin D on the healing process of fracture has still been controversial. These studies were designed to understand the healing process of normal fibular fracture and the therapeutic effects of $1\alpha$, 25 dihydroxycholecalciferol on the osteoporotic fracture in rats. The simple transverse fractures of rat fibulae were produced with a rotating diamond saw. The histological and ultrastructural changes of rats were observed. The histological and ultrastructural studies revealed the healing of the fibular fracture in the 5th week after simple transverse fracture. The osteoporosis impaired more the healing of osteoporotic fibular fracture than normal non-osteoporotic fibular fracture. The healing process of osteoporotic fracture was facilitated by the treatment with $1\alpha$, 25 dihydroxycholecalciferol, however, was delayed more than the healing process of normal fracture. These results suggest that $1\alpha$, 25 dihydroxycholecalciferol was effective for reducing the deleterious effects of osteoporosis in fracture healing.

Background: Progressed tissue culture techniques have allowed us to easily obtain mass products of tissue-cultured mountain ginseng over 100 yr old (TCMG-100). We investigated the effects of TCMG-100 extract on erectile function using in vitro and in vivo studies. Methods: To examine the relaxation effects and mechanisms of action of TCMG-100 on rabbit cavernosal strips evaluated in an organ bath. To investigate the long-term treatment effect of TCMG-100, 8-wk administration was performed. After administration of TCMG-100, intracavernosal pressure, cyclic guanosine monophosphate and nitric oxide (NO) levels of cavernosal tissue, serum testosterone level, histological observation of collagen fiber, endothelium, smooth muscle cell, and transforming growth factor-${\beta}1$ were investigated. Results: TCMG-100 extract displayed dose-dependent relaxation effects on precontracted rabbit corporal smooth muscle. The TCMG-100-induced relaxation was significantly reduced by removing the endothelium, and treatment with an NO synthase inhibitor or NO scavenger. Eight weeks of TCMG-100 administration increased intracavernosal pressure in a rat model. The levels of cyclic guanosine monophosphate and NO in the corpus callosum and serum testosterone level were also increased by TCMG-100 treatment. Furthermore, histological evaluation of collagen, smooth muscle, and endothelium showed increases in endothelium and smooth muscle, and a decrease in transforming growth factor-${\beta}1$ expression. Conclusion: These relaxation effects on corporal smooth muscle and increased erectile function suggest that TCMG-100 might be used as an alternative herbal medicine to improve erectile function.
