The Effects of Elbow Joint Angle on the Mechanical Properties of the Common Extensor Tendon of the Humeral Epicondyle

  • The purpose of this study was to determine the effects of elbow joint angle on mechanical properties, as represented by ultimate load, failure strain and elastic modulus, of bone-tendon specimens of common extensor tendon of the humeral epicondyle. Eight pairs of specimens were equally divided into two groups of 8 each, which selected arbitrarily from left or right side of each pair, positioned at 45$^{\circ}$ and 90$^{\circ}$ of elbow flexion and subjected to tension to failure in the physiological direction of the common extensor tendon. For comparison of the differences in the failure and elastic modulus between tendon and the bone-junction, data for both were evaluated individually. Significant reduction in ultimate load of bone-tendon specimens was shown to occur at 45$^{\circ}$. The values obtained from the bone-tendon junctions with regard to the failure strain were significant higher than those from tendon in both loading directions, but the largest failure strain at the bone-tendon junction was found at 45$^{\circ}$. The elastic modulus was found to decrease significantly at the bone-tendon junction when the loading direction switched from 90$^{\circ}$ to 45$^{\circ}$. Histological observation, after mechanical tensile tests, in both loading directions showed that failure occurred at the interface between tendon and uncalcified fibrocartilage in the thinnest fibrocartilage zone of the bone-tendon junction. We concluded that differences in measured mechanical properties are a consequence of varying the loading direction of the tendon across the bone-tendon specimen.

  • 1. The weights of ovary and uternus in the all treated groups were lighter than those in control group showing significantly differences from 4 weeks after treatment. The significant was not recognized between PTU and Thx. groups, and Thyro. and control groups. 2. In the histological changes of ovary, follicles were disa, pp.ared and alignment of membrana granulosa disnitergration in cell change were serious in Thx. and PTU groups and slight in Thyro. group. 3. In the histological changes of uterus, endometrial epithelium and lamina propria were atrophied from 3 weeks after treatment in Thx. group, from 4 weeks in PTU groups and from 5 weeks in Thyro. group. Muscular layers were atrophied with time elapse in the all treated groups. 4. The changes of the concentrations of serum progesterone at all observation times in Thx. and PTU groups were significantly decreased in comparison with those in control group. While those in Thyro. group were significantly increased in comparison with those in control group. 5. The concentration of serum estradiol-17$\beta$ at all observation times in all experimental groups were below 27.2pg/ml. Therefore, we did not detect any changes of the concentrations.

The Cytotoxicity of Rat Hepatocytes on Viscum album Lectin (겨우살이 lectin의 흰쥐 간세포 독성에 미치는 영향)

  • In the present study, the author investigated to the cytotoxocity in cultured rat hepatocytes of Viscum album lectin. The cytotoxcity effect in Viscum album lectin on the activity of LDH was also investigated. Viscum album lectin significantly increased LDH leakage into medium of hepatocytes treated or untreated with $CCl_4$ (p<0.001). However, Viscum album lectin significantly increased LDH leakage from $CCl_4$-induced hepatocyte (p<0.001). There was a significant increase in LDH levels relative to the control group. Histological observation basically supported the result obtained from LDH assay. The livers of rats challenged with $CCl_4$ produced a marked increased cytoplasmic vacuoles and inflammatory cells in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered DMSO alone did not alter the normal hepatic architecture. Histological observation of liver section in rat treated 72 hrs with either Viscum album lectin $CCl_4$-induced liver damage showed number of cytoplasmic vaculoe and necrotic cell. The number of inflammatory cell increased markedly. This results suggest to the conclusion that Viscum album lectin has a effect of hepatotoxicity activator.

Effect of Salvia miltiorrhiza Extract in Urethane-induced Lung Tumorigenesis in A/J Mice (단삼(丹蔘) 추출액이 Urethan으로 유발된 생쥐의 폐암에 미치는 영향)

  • Objectives: The aim of this study was to determine the effects of Salvia miltiorrhiza (SM) extract in urethane-induced lung tumorigenesis in A/J mice. Methods : We examined change of body weight, histological, apoptosis, immunohistochemical and gene expression of cyclooxygenase (COX-2) in lung tumors. Mice were divided into 3 groups: normal, saline, and experimental group administered SM extract after injection with urethane. Results : Histological observation showed shrunken alveoli in the control group, but recovered from damage in the SM extract administered group. The COX-2 positive materials were observed in the smooth muscle of terminal bronchiole and alveoli from the control group, but these positive materials decreased in the SM extract treatment group. The results of electron microscopical observation, dilated capillary and degenerated endothelia were observed in the control group. The apoptotic nuclei increased more in the control group compared with the normal and SM extract administered groups. Serial sections of the whole lung showed solid and papillary tumors. The size and number of tumors decreased in the SM groups compared with the control groups. Conclusions : These results suggest the possibility that SM may exert an anti-tumor effect on urethane-induced lung tumorigenesis.

Effect of Lipid Metabolism in Viscum album Lectin on Rats (겨우살이 Lectin이 지질대사에 미치는 영향)

  • This study was carried out to investigate effect of lipid metabolism in Viscum album lectin on rats. The lectin was purified by sepharose 4B affinity chromatography and gel filtration using sephadex G-150 with plant material from Viscum album collected in Mt. Duk Yui. After 72 h of $CCl_4$ injection (in olive oil, 1:1, 2 mg/kg) there was a significant increase in serum total cholesterol and triglycerige levels relative to the control group. However, treatment of both Viscum album and purified lectin were significantly decreased lipid parameters against the $CCl_4$-induced. Histological observation basically supported the result obtained from serum lipid assay. The livers of rats challenged with $CCl_4$ produced a marked increase of cytoplasmic vacuoles in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered olive oil alone did not alter the normal hepatic architecture. Histological observation of the liver section in rats treated 72 h with either Viscum album purified lectin or $CCl_4$-induced liver lipogenesis showed decreased numbers of cytoplasmic vaculoes and necrotic cells. The normal hepatic architectural pattern was observed in Hematoxylin-eosin stain. These results suggest that Viscum album lectin has a possible protective effect of lipid metabolim in rats.

  • Flowergenic callus was induced from the ovary surface of Allium fistulosum L. cultured on MS medium containing 0.5mg/L NAA and 0.5mg/L BA or 0.5mg/L kinetin. After 3-4 weeks of culture, the flower buds were developed from flowergenic callus. The continuous production of flowergenic callus was proliferated, when subcultured on the medium containing 0.5mg/L NAA and 0.5mg/L kinetin. However, frequency of flower bud formation from flowergenic callus was decreased as the subculture was repeated. Histological observation reveals that the developmental pattern of flower bud from flowergenic callus was closely similar to that of natural flowers.

  • Objective : To investigate effects of herbal preparations formulated(Gihyeolgwanjeolbang-A, CWS-A) on complete Freund's adjuvant (CFA)-induced arthritis in rats. Method : Arthritis was induced by injecting CFA subcutaneously into the left knee joint and paw, and the herbal preparations formulated(CWS-A I, 82.3 mg/kg ; CWS-A II, 164.6 mg/kg) was administered orally (i.g.) for 10 consecutive days beginning on day 10 after the injection. External shape, paw edema, inflammatory cytokines tumor necrosis factor alpha (TNF-alpha) and interleukin-1 beta (IL-1beta), and histological observation were assessed. Result : In swelling of the paw, CWS-A I and CWS-A II group in 15 days and 20 days were significantly reduced compared to controls. In serum ALT, CWS-A I and CWS-A II group were significantly reduced compared to controls. In TNF-${\alpha}$, CWS-A I and CWS-A II group were a tendency reduced compared to controls. In HGB, HCT, MCHC of erythrocytes, CWS-A II group was increased compared to controls. In histological observations, CWS-A II group was observed synoviocytes more than control group and was observed proteoglycans in the deep layers. Conclusion : The data suggest that CWS-A significant anti-arthritic effects that may be mediated by suppressing inflammatory parameters.

Histological Observation on the Symbiotic Relationships Between Gastrodia elata and Rhizomorph of Armillaria mellea (뽕나무버섯균 균사속과 천마의 공생관계에 대한 조직학적 관찰)

  • The histological studies were conducted to investigate the symbiotic relationships between Gastrodia elata and Armillaria mellea by using light and electron microscopes. The fungus, A. mellea, penetrated into the cortex of G. elata, in which endomycorrhizal mycelia in the cortical cells appeared to be dissolved and digested, and seemed to be consequently used as nutritional sources for G. elata growth. Staining of infected tissues revealed that protein- and fat-like substances were localized in the cells. The nuclei of cells infected by the fungal mycelia were hypertrophied 1.5 to 2 times as those without the fungal infection.

The Histological Study about the Burn Caused by Moxibustion Using the Commercial Indirect Moxibustion (상용 간접구로 인한 화상에 대한 조직학적 연구)

  • Objectives : Burn is the important adverse effect of the moxibustion treatment. This research does with the purpose to utilize as basic data which the results of histological observation of the burn tissue which generates when operating various commercial indirect moxa (CIM) on the skin, evaluate the burn because of the surgical operation. Methods : It operated 6 kinds of CIM on the depilated abdomen of the rat. By using the H-E staining and TUNEL assay, the image was observed and analyzed. Results : It could be the moxibustion medical treatment using the CIM generated in the first degree burn until the third degree. And in some case, burn has not generated. By using the H-E stain, the second-degree burn and third degree was observed, and it was observed the first degree burn by using TUNEL assay. In the first degree burn, TUNEL reaction in the epidermal layer was confirmed. The damage to the dermal layer was observed in more than 2 degree burn. In the third degree, the tissue degeneration to the subcutaneous fat was observed but the thickness of the skin tissue was not observed. Conclusions : Basic data classifying the burn generated by the CIM treatment through the histological observation of the burn tissue caused by the CIM treatment were built according to this research result.

The Study of Behavior and Histological Change on Treadmill Exercise Intensity after Spinal Cord Injury in Rats (트레드밀 강도에 따른 운동이 척수손상 흰쥐의 행동학적 변화와 조직학적 변화에 관한 연구)

  • This study was designed to investigate the effect of treadmill exercise of low-intensity and high-intensity on the functional recovery and histological change in spinal cord injury rats. Sprague-Dawley rats were experimented(n=15) for this research. Spinal cord injury was induced by the NYU drop impacter device after laminectomy. After operation, rats were test at modified Tarlov scale at 3 days, and divided into the control group(n=5), experimental group I(n=5, low-intensity treadmill) and experimental group II(n=5, high-intensity treadmill). The rats were disciplined from 7 day through 21 day. Functional recovery was evaluated by the BBB scales and the Grid Walk test for the progressive locomotor recovery at 3, 7, 14, 21 days. Histopathological studies for the muscle in order to observation the change of damage and size of the organized surface which is visible visually it executed hematoxylin & eosin stain. According to the result of 4 weeks of treadmill exercise, group II showed improvement than group I of motor behavior after spinal cord injury.
