형질전환 감자 소괴경의 발달단계에 따른 Patatin Promoter-GUS 유전자의 발현 분석 (Distinct Spatio-temporal Expression Patterns of Patatin Promoter-GUS Gene Fusion in Transgenic Potato Microtubers)

  • 본 실험은 감자괴경에 특이적으로 나타나는 patatin promoter에 의한 외부 도입 유전자의 발현 양상을 파악하고자 수행되었다. Patatin promoter에 의하여 GUS 유전자의 발현이 조절되도록 pATGUS 벡터를 제작한 후 Agrobacterium tumefaciens LBA4404를 이용하여 감자의 잎 절편에 형질전환하였다. 대조구로 GUS 유전자의 상시발현 벡터인 pBI121을 사용하였으며, 항생제를 포함한 재분화 배지에서 개체를 유도한 결과 8주 후부터 신초를 관찰할 수 있었다. NPTII 유전자의 삽입여부를 PCR로 검정한 후, 선별된 형질전환체의 소괴경 형성을 위해 sucrose 농도를 높인 배지에서 1주일 간격으로 줄기의 하단 부분을 배양하였다. 주별로 시료를 채취한 후, RNA gel blot 분석을 해 본 결과 CaMV35S promoter에 의한 GUS 발현은 소괴경의 전단계에서 고르게 발현되는 반면, 괴경-특이적인 patatin promoter의 경우 감자 줄기에서는 관찰이 어려웠고, 주별 발현율은 1주부터 5주까지는 점점 증가하다가 그 이후부터는 점차 감소함을 알 수 있었다. 또한, GUS의 효소활성 역시 mRNA의 발현율과 비례함을 알 수 있었다. 제시된 실험결과들로 보아 감자 괴경에서의 patatin promoter에 의한 GUS 유전자의 발현은 5주경에 가장 높게 나타났으며, 이러한 결과들로 보아 patatin promoter에 의해 감자 소괴경내로 도입된 외래 유전자의 발현을 확인하기 가장 좋은 시기는 소괴경 형성 후 5주째임을 알 수 있었다.

어패류 및 종에 내재되어 있는 ${\beta}-Galactosidase$의 활성 비교 (Comparative Study on Endogeneous Activities of ${\beta}-Galactosidase$-like Enzyme in Several Finfishes and Shellfishes)

  • 어류 및 폐류내에 내재하는 내재성 ${\beta}-galactosidase$의 활성도를 분석함으로서 외래유전자의 이식시 기초자료로 활용하고자 어류 6종 및 패류 4종을 대상으로 본 실험을 행하였다. 어류에 있어서의 X-gal 염색 결과 혈청 및 근육을 제외한 모든 조직에서 모두 positive ( + ) 염색 양상을 보였다. 패류에 있어서도 어류와 같은 양상을 보여 근육을 제외한 모든 조직에서 모두 positive 염색 양상을 보였으며 더욱이 참굴에 있어서는 폐각근 및 근육에 있어 매우 진한 염색 반응을 보였다. 근육으로 부터 DNA를 추출한 후 PCR을 수행한 결과 모든 종에서 positive (+) 결과를 나타내었으며, 어패류간 그리고 각 개체간 차이는 없었다. 각 장기별 ${\beta}-galactosidase$의 활성도 측정결과 모든 종에 있어서 혈청의 활성도는 무시할 만한 수준이었고, 근육에서 가장 낮게 나타났다. 미꾸라지와 잉어에서는 신장에서 가장 높은 활성을 보였고, 틸라피아는 소화관에서 가장 높은 값을 보였다. 해산어인 넙치의 경우 돌가지미, 문치가자미의 경우에 비해 간에서 활성이 높았다. 그러나 돌가자미 및 문치가자미인 경우 신장에서 가장 높은 값을 보였다. 패류에 있어서는 소화맹낭에서 가장 높은 활성값을 나타내었고 폐각근 및 외투막에서 매우 낮은 활성값을 나타내었다. 그러나, 참굴에 있어서는 참전복, 피조개 및 진주조개 보다는 상대적으로 폐각근 및 외투근에서 높은 활성을 나타내었다.

A Role for Arabidopsis miR399f in Salt, Drought, and ABA Signaling

  • MiR399f plays a crucial role in maintaining phosphate homeostasis in Arabidopsis thaliana. Under phosphate starvation conditions, AtMYB2, which plays a role in plant salt and drought stress responses, directly regulates the expression of miR399f. In this study, we found that miR399f also participates in plant responses to abscisic acid (ABA), and to abiotic stresses including salt and drought. Salt and ABA treatment induced the expression of miR399f, as confirmed by histochemical analysis of promoter-GUS fusions. Transgenic Arabidopsis plants overexpressing miR399f (miR399f-OE) exhibited enhanced tolerance to salt stress and exogenous ABA, but hypersensitivity to drought. Our in silico analysis identified ABF3 and CSP41b as putative target genes of miR399f, and expression analysis revealed that mRNA levels of ABF3 and CSP41b decreased remarkably in miR399f-OE plants under salt stress and in response to treatment with ABA. Moreover, we showed that activation of stress-responsive gene expression in response to salt stress and ABA treatment was impaired in miR399f-OE plants. Thus, these results suggested that in addition to phosphate starvation signaling, miR399f might also modulates plant responses to salt, ABA, and drought, by regulating the expression of newly discovered target genes such as ABF3 and CSP41b.

소라(Batillus cornutus) 외투막의 미세구조 (Mantle Ultrastructure of the Spiny Top Shell, Batillus cornutus (Gastropoda: Turbinidae))

  • 소라 (Batillus cornutus) 외투막의 조직화학적 특성 및 미세구조를 광학현미경과 투과전자현미경을 이용하여 기재하였다. 소라의 외투막은 결합조직층을 단층상 피층이 위아래로 둘러싸고 있는 구조였으며, 상피층은 내장 기관계 쪽을 향하고 있는 내부상피층과 패각 쪽의 외부상피층으로 구분된다. 상피층에 의해 둘러싸인 결합조직층은 주로 교원섬유와 근섬유다발로 구성되며, 혈림프동이 발달하고 있었다. 외투막의 각정부 상피층에 분포하는 점액세포들은 산성 및 중성 점액다당류를 함유하고 있었으며, 중간부와 가장자리의 상피층에 분포하는 점액세포들은 비황화 강산성의 당단백질 점액성분을 함유하는 것으로 나타났다. 외투막의 두께, 상피층의 두께, 혈림프동의 면적은 각정부에서 가장자리로 갈수록 낮아지는 경향을 보였다. 투과전자현미경으로 관찰하였을 때 외투막의 상피층은 단층으로 원주형 상피세포, 섬모세포, 흡수세포, 분비세포로 구성되어 있었다. 외투막 상피세포들은 원주형으로 장방형의 핵을 가지며, 자유면에는 미세융모들이 발달되어 있었다. 상피세포 사이에는 폐쇄띠, 폐쇄대와 수지상막구조로 연결되어 있었다. 섬모세포는 자유면에 섬모와 미세융모를 가지며, 세포질의 상부에는 다수의 미토콘드리아를 가지고 있었다. 상피층에서는 흡수기능을 가지는 두 종류의 세포가 관찰되었다. 이들은 원주형으로 미세융모, 음소포, 미토콘드리아 그리고 전자밀도가 다양한 용해소체들을 함유하고 있었다. 소라 외투막에서는 네 종류 (A, B, C, D)의 분비세포를 구분할 수 있었는데, 이들은 모두 단세포선으로 확인되어 대부분의 복족류와 이매패류의 외투막에서 보고된 선세포들과 유사한 특징을 보였다.

Agrobacterium을 이용한 Phosphinothricin Acetyl Transferase의 도라지로의 형질전환 (Agrobacterium-mediated Transformation of PAT into Platycodon grandiflorum A. De. candolle)

  • 본 연구는 Agrobacterium을 매개로 도라지에 PAT 유전자를 도입하여 ‘바스타’에 저항성을 가지는 형질전환 도라지를 개발하는 기술을 확립하기 위하여 수행되었다. 종자를 무균적으로 발아시킨 후 10일 된 미성숙 자엽과 성숙엽에 Agrobac-terium을 접종하고 1/10 MS 배지에서 48시간 동안 공동 배양하였다. 공동배양 후 부정아 유도를 위해 MS 선발배지 (0.2 $mg/{\ell}$ NAA, 1.0 $mg/{\ell}$ BA, 3% 설탕, pH 5.8; 3 $g/{\ell}$ gelrite, 100 $mg/{\ell}$ kanamycin, 500 $mg/{\ell}$ carbenicillin)에 치상하여 배양한 결과 미성숙 자엽의 절편에서 형질전환체로 추정되는 부정아가 형성되었고, 선발배지에 2회 계대배양하여 형질전환 추정체를 선발하였다. 이러한 형질전환 추정체는 GUS, PCR 분석 및 RT-PCR 분석에 의하여 형질전환체로 확인되었다. 또한 10 $mg/{\ell}$ 의 phosphinothricin이 함유된 배지에서 배양하여 형질전환 여부를 확인하였고, 순화재배 후 0.3% ‘바스타’를 살포한 결과 형질전환 도라지는 제초제에 저항성을 보였다. ‘바스타’에 저항성을 보인 도라지 식물체는 정상적인 생육을 계속하여 개화하였다.

  • Purpose : These studies were undertaken to evaluate the effects of Allii tuberosi Semen (ATS) on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Materials and Methods : We used the 8-week-old mice and administered the 0.2 ml extract solution of ATS in the different concentration (0.1 mg/ml, 1 mg/ml, 10 mg/ml and 100 mg/ml) once a day for 60 days. The control group was administered the distilled water in the same way. After the administration of each extract solution, we examined the number of total, motile and normal sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed the histological changes of isolated testis. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The concentration of total sperm, the motility and normality of spermatozoa were significantly increased in ATS groups, especially in 1 and 10 mg/ml groups, compared to control group. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the ATS groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the ATS groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the ATS groups compared to the control group, especially in 1 mg/ ml group. The activity of testicular catalase was increased in ATS groups. Conclusion : This study shows that ATS has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase and testicular peroxidase. We can suggest that ATS extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Expression of the Floral Repressor miRNA156 is Positively Regulated by the AGAMOUS-like Proteins AGL15 and AGL18

  • The regulation of flowering time has crucial implications for plant fitness. MicroRNA156 (miR156) represses the floral transition in Arabidopsis thaliana, but the mechanisms regulating its transcription remain unclear. Here, we show that two AGAMOUS-like proteins, AGL15 and AGL18, act as positive regulators of the expression of MIR156. Small RNA northern blot analysis revealed a significant decrease in the levels of mature miR156 in agl15 agl18 double mutants, but not in the single mutants, suggesting that AGL15 and AGL18 co-regulate miR156 expression. Histochemical analysis further indicated that the double mutants showed a reduction in MIR156 promoter strength. The double mutants also showed reduced abundance of pri-miR156a and pri-miR156c, two of the primary transcripts from MIR156 genes. Electrophoretic mobility shift assays demonstrated that AGL15 directly associated with the CArG motifs in the MIR156a/c promoters. AGL18 did not show binding affinity to the CArG motifs, but pull-down and yeast two-hybrid assays showed that AGL18 forms a heterodimer with AGL15. GFP reporter assays and bimolecular fluorescence complementation (BiFC) showed that AGL15 and AGL18 co-localize in the nucleus and confirmed their in vivo interaction. Overexpression of miR156 did not affect the levels of AGL15 and AGL18 transcripts. Taking these data together, we present a model for the transcriptional regulation of MIR156. In this model, AGL15 and AGL18 may form a complex along with other proteins, and bind to the CArG motifs of the promoters of MIR156 to activate the MIR156 expression.

쇄양(鎖陽)의 농도별(濃度別) 투약(投藥)이 수컷 생쥐 생식능력(生殖能力)에 미치는 영향(影響) (Effects of Cynomorii Herba Extract Solution on Reproductive Capacities in Mice)

Materials and Methods : We used the 2-month-old mice and administered 0.2ml extract solution of CH in the 0.1mg/ml, 1mg/ml, 10mg/ml and 100mg/ml once a day for 60days. The control group was administered the distilled water in the same way. After the administration of extract solution, we examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We observed the histological changes of isolated testis and compared to the testicular tissue especially seminiferous tubules between control and CH groups by histochemical method. Results : The concentration of total sperm and the motility of spermatozoa were significantly increased in the 1mg/ml, 10mg/ml and 100mg/ml CH groups, especially in 10mg/ml group, compared to the control group. The significant differences were observed in the normality of spermatozoa of the CH groups compared to the control group. In the histolocal analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the CH groups compared to the control group. Also, the activity of hyaluronidase was significantly increased in the CH groups compared to the control group. In the antioxidant activity analysis, the activities of testicular peroxidase and testicular catalase were significantly increased in the CH groups compared to the control group, respectively. Conclusion : This study shows that CH has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We can suggest that CH extract solution be useful for the treatment of male sexual dysfunctions and infertility.

농도별(濃度別) 파극(巴戟)투약(投藥)이 숫컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響) (Dose Dependent Effect of Morindae officinalis Radix Extract Solution on the Reproductive Capacities in the Mice)

  • Purpose : These studies were undertaken to evaluate the effects of the administration of different concentrated Morindae officinalis Radix extract solution on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Methods : We used the 2-month-old mice and administered the extract solution of Morindae officinalis Radix in the different concentration once a day for 60 days. The control group was administered the normal saline in the same way and duration. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed changes of isolated testis before and after administration of Morindae officinalis Radix extract solutions the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The significant dose-dependent differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. Conclusion : This study shows that Morindae officinalis Radix has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase and testicular peroxidase. We can suggest that Morindae officinalis Radix extract solution be useful for the treatment of male sexual dysfunctions and infertility.

The Calmodulin-Binding Transcription Factor OsCBT Suppresses Defense Responses to Pathogens in Rice

  • We previously isolated the OsCBT gene, which encodes a calmodulin (CaM)-binding protein, from a rice expression library constructed from fungal elicitor-treated rice suspension cells. In order to understand the function of OsCBT in rice, we isolated and characterized a T-DNA insertion mutant allele named oscbt-1. The oscbt-1 mutant exhibits reduced levels of OsCBT transcripts and no significant morphological changes compared to wild-type plant although the growth of the mutant is stunted. However, oscbt-1 mutants showed significant resistance to two major rice pathogens. The growth of the rice blast fungus Magnaporthe grisea, as well as the bacterial pathogen Xanthomonas oryzae pv. oryzae was significantly suppressed in oscbt-1 plants. Histochemical analysis indicated that the hypersensitive-response was induced in the oscbt-1 mutant in response to compatible strains of fungal pathogens. OsCBT expression was induced upon challenge with fungal elicitor. We also observed significant increase in the level of pathogenesis-related genes in the oscbt-1 mutant even under pathogen-free condition. Taken together, the results support an idea that OsCBT might act as a negative regulator on plant defense.
