Mast cells play an important role in early and late phase allergic reactions through allergen and IgE-dependent release of histamine, proteases, prostaglandins, and several multifunctional cytokines. In this study, we investigated whether Cudrania tricuspidata extract (CTE) suppresses IgE-mediated allergic responses in mast cells, an allergic animal model, and its mechanism of action in mast cells. We found that CTE inhibited IgE-mediated degranulation and cytokine production in rat basophilic leukemia (RBL)-2H3 mast cells and bone marrow-derived mast cells (BMMC), as well as passive cutaneous anaphylaxis (PCA) in mice. With regard to its mechanism of action, CTE suppressed the activating phosphorylation of spleen tyrosine kinase (Syk), a key enzyme in mast cell signaling processes and that of LAT, a downstream adaptor molecule of Syk in $Fc{\varepsilon}RI$-mediated signal pathways. CTE also suppressed the activating phosphorylation of mitogen-activated protein (MAP) kinases and Akt. The present results strongly suggest that the anti-allergic activity of CTE is mediated through inhibiting degranulation and allergic cytokine secretion by inhibition of Syk kinase in mast cells. Therefore, CTE may be useful for the treatment of allergic diseases.
Mahwang-Shin-Gung-San has been found inhibiting the mast cell-mediated anaphylactic reaction. This report describes an inhibitory effect of Mahwang-Shin-Gung-San (MSGS) on the immediate-type cutaneous allergic reactions. MSGS has concentration -dependently inhibited the ear swelling response induced by compound 48/80 in mouse by intradermal injection. The mast cells in mouse ear tissue undergone ear-swelling response by compound 48/80 were stained by alcian blue/nuclear fast red. MSGS significantly inhibited the compound 48/80-induced degranulation from mast cells in ear tissue. MSGS concentration-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. We also studied the effect of MSGS on mast cell-dependent passive cutaneous anaphylaxis (PCA) activated by dinitrophenyl IgE antibody. MSGS showed potent inhibition of PCA by oral administration. These results indicate that MSGS inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.
In this report, we investigated the effect of aqueous extract of vinegar treated small black soybean (Glycine max Merr.) (Leguminosae) (VSBS) on mast cell-mediated allergic reaction and pro-inflammatory cytokine secretion. VSBS inhibited compound 48/80-induced systemic reactions. VSBS attenuated immunoglobulin (Ig) E-mediated passive cutaneous anaphylaxis. In addition, VSBS decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated secretion of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6 and interleukin (IL)-8 in human mast cells. Our findings provide evidence that VSBS inhibits mast cell-derived allergic reactions.
Organic substances are released from phytoplankton cells during all phases of growth. The type and amounts of organic substance excreted and the effects of nutrient limitation are often highly species-specific. Dinoflagellate, Cochlodinium polykrikoides grown in batch culture produced an exopolysaccharide. Exopolysaccharide and intracellular polysaccharide concentrations increased as C. polykrikoides cultures progressed from exponential phase, through stationary phase, to declining phase. In the exponential phase, the concentration of exopolysaccharide was relatively low, but in the stationary phase, it showed a rapid increase which seemed to coincide with the depletion of nitrate from the medium. Of the 20 amino acids analyzed, proline dominated in the organic matter of all cultures ranging from 48.2 to 79.9 nmol L–1, and constituting the 20-90% of total amino acids, and followed by histamine varying from 0.7 to 47.5 nmol L–1. Leucine and cysteine were also abundant in the stationary phase. The release rates of exopolysaccharide and intracellualr polysaccharide were higher the end of stationary phase than in the exponential phase. Exopolysaccharide concentration per cell was more than two times higher during the end of stationary phase than that in exponential phase. C. polykrikoides produced extracellular polysaccharide at a rate of 47.04 pg cell–1 day–1.
Flos magnoliae (FM), the dry flower buds of Magnolia officinalis or its related species, is a traditional herbal medicine commonly used in Asia for symptomatic relief of and treating allergic rhinitis, headache, and sinusitis. Although several studies have reported the effects of FM on store-operated calcium entry (SOCE) via the ORAI1 channel, which is essential during intracellular calcium signaling cascade generation for T cell activation and mast cell degranulation, the effects of its isolated constituents on SOCE remain unidentified. Therefore, we investigated which of the five major constituents of 30% ethanoic FM (vanillic acid, tiliroside, eudesmin, magnolin, and fargesin) inhibit SOCE and their physiological effects on immune cells. The conventional whole-cell patch clamp results showed that fargesin, magnolin, and eudesmin significantly inhibited SOCE and thus human primary CD4+ T lymphocyte proliferation, as well as allergen-induced histamine release in mast cells. Among them, fargesin demonstrated the most potent inhibitory effects not only on ORAI1 (IC50 = 12.46 ± 1.300 μM) but also on T-cell proliferation (by 87.74% ± 1.835%) and mast cell degranulation (by 20.11% ± 5.366%) at 100 μM. Our findings suggest that fargesin can be a promising candidate for the development of therapeutic drugs to treat allergic diseases.
Background: Ginsenoside Rg3 (G-Rg3) is the major bioactive ingredient of Panax ginseng and has many pharmacological effects, including antiadipogenic, antiviral, and anticancer effects. However, the effect of G-Rg3 on mast cell-mediated allergic inflammation has not been investigated. Method: The antiallergic effects of G-Rg3 on allergic inflammation were evaluated using the human and rat mast cell lines HMC-1 and RBL-2H3. Antiallergic effects of G-Rg3 were detected by measuring cyclic adenosine monophosphate (cAMP), detecting calcium influx, and using real-time reverse transcription polymerase chain reaction, enzyme-linked immunosorbent assay, Western blotting, and in vivo experiments. Results: G-Rg3 decreased histamine release from activated mast cells by enhancing cAMP levels and calcium influx. Proinflammatory cytokine production was suppressed by G-Rg3 treatment via regulation of the mitogen-activated protein kinases/nuclear factor-kappa B and receptor-interacting protein kinase 2 (RIP2)/caspase-1 signaling pathway in mast cells. Moreover, G-Rg3 protected mice against the IgE-mediated passive cutaneous anaphylaxis reaction and compound 48/80-induced anaphylactic shock. Conclusion: G-Rg3 may serve as an alternative therapeutic agent for improving allergic inflammatory disorders.
본 연구는 쌀겨의 사람유래 암세포 증식 억제효과(in vitro)와 사람유래 비만세포주(HMC-1)의 활성 억제효과 및 흰쥐 복강 비만세포로부터 히스타민 분비에 미치는 영향을 관찰하였다. Hep3B와 HeLa 세포에 대한 형태변화는 쌀겨의 물 추출물과 60% 에탄을 추출물 처리군 모두 농도에 비례하여 암세포의 밀도가 감소하는 경향을 보였고, 시료를 고농도로 처리할수록 사멸세포 형태인 원형의 세포 수가 증가하였다. Hep3B 세포에 대한 증식 억제능은 쌀겨 물 추출물이 60% 에탄을 추출물보다 높았고, $100{\mu}g/mL$와 $1,000{\mu}g/mL$ 농도에서 물 추출물이 60% 에탄을 추출물보다 각각 20%, 55% 더 높은 효과가 있었다. HeLa세포에 대한 증식 억제능은 쌀겨 물 추출물과 60% 에탄을 추출물에서 비슷한 경향을 보였으나 $1,000{\mu}g/mL$와 $3,000{\mu}g/mL$ 농도에서 물 추출물이 60% 에탄을 추출물보다 각각 5%, 27% 더 높았다. 사람의 비만세포주(HMC-1)에서 $Fc{\varepsilon}RI$, tryptase, c-kit mRNA 발현양에 미치는 효과를 관찰한 결과 GAPDH의 발현양은 대조군과 시료를 처리한 실험군에서 모두 비슷하게 나타났다. 비만세포의 $Fc{\varepsilon}RI$ mRNA와 c-kit의 mRNA 발현양은 쌀겨 물 추출물군보다 60% 에탄올 추출물에서 더 많이 억제되는 것으로 나타났으나 tryptase mRNA 발현양은 억제하지 못한 것으로 관찰되었다. 비만세포의 히스타민 분비에 미치는 영향은 무처리 대조군에 비해 $5{\mu}g/mL$ compound 48/80을 처리하였을 때 히스타민 분비량이 유의하게 증가하였으며, 쌀겨 물 추출물 $0.01{\mu}g/mL$에서 약 87%의 억제율을 나타내었고, 에탄을 추출물 $100{\mu}g/mL$에서 약 86%의 억제율을 보였다.
상백피의 기능성식품 소재로서의 이용 가능성을 조사하기 위하여 상백피 물 추출물의 항산화 활성 및 in vitro에서의 항천식 효과를 확인하였다. 항산화 측정 결과는 총 페놀함량은 35.2 mg/g로 나타났으며, DPPH radical 소거능은 35.3~59.8%, hydroxyl radical 소거능은 62.1~78.8% 범위로 높은 항산화 효능을 나타내었다. DNP-IgE와 HSA로 활성화된 RBL-2H3 세포에서의 항천식 효과는 농도가 증가할수록 유의적으로 $\beta$-hexosaminidase를 감소시켰으며, 염증성 cytokine인 IL-4, TNF-$\alpha$ 사이토카인 분비량 또한 유의적(p<0.05)으로 감소시켰다. 결과적으로 상백피는 높은 항산화력을 가지며, 항천식과 관련된 지표들을 유의적으로 감소시킴으로써 천식 예방 및 개선을 위한 기능성 소재로서 이용가치가 높을 것으로 사료된다.
갈조류인 감태(Ecklonia cava)의 에틸아세테이트 분획으로부터 phloroglucinol, eckol, eckstolonol, triphlorethol-A, phlorofucofuroeckol A, dieckol의 6종의 phlorotannin이 분리되었으며, NMR 및 MS분석으로 구조가 규명되었다. 감태의 주요 활성물질 분석을 위하여, 분리된 6종 물질에 대한 항산화, 항염증 및 항알러지 효과를 평가하였다. 그 중 phlorofucofuroeckol A와 triphlorethol-A가 라디컬(DPPH, ABTS) 소거 활성이 높았으며, 염증 반응에 대한 활성을 분석하기 위하여 LPS를 처리한 대식세포주인 Raw264.7에서 산화질소(NO) 생성 억제활성을 측정한 결과, phlorofucofuroeckol A의 NO 생성 억제 효과가 가장 큰 것으로 나타났다. 또한, 알러지 반응에 대한 억제활성을 분석하기 위하여 IgE-항체로 활성화시킨 비만세포주 RBL-2H3에서 분비되는 ${\beta}-Hexosaminidase$를 측정한 결과, dieckol이 농도의존적으로 가장 높은 억제 효과를 나타냈다. 따라서, 본 연구에서 항염증 및 항알러지 활성을 중심으로 분리된 6종의 플로로탄닌 중 phlorofucofuroeckol A와 dieckol은 감태의 알러지 염증 억제활성을 나타내는 주요 물질로 사료된다.
In vitro 및 in vivo에서 모두 효과를 나타내는 새로운 여드름 개선 성분을 개발하였다. 특히, in vitro에서 효과가 검증된 많은 원료들이 실제 여드름환자에게서 실질적인 효능을 나타내지 못하는 경우가 많아, 본 연구에서는 임상에서 실질적인 효과를 보일 수 있는 원료의 개발에 초점을 맞추었다. 우선적으로 여드름에 효과가 높은 것으로 알려져 있거나 여드름개선 효과가 기대되는 천연물질의 추출물을 대상으로 소규모의 예비임상시험을 통해 효능을 확인하였다. 여러 가지 후보물질 중에서 발아콩과 목련박피 2종의 추출물이 여드름 및 여드름에 의한 홍조현상과 흉터에 뚜렷한 효과를 나타내어 이 두 천연물의 혼합추출물을 SeleMix AN이라 명명하였다. 200여 명의 예비임상시험을 통해서 SeleMix AN의 임상적 효과를 확인한 후 상기 물질의 여드름 개선 작용기작을 규명하기 위한 in vitro 효능테스트를 실시하여 P. Acne 성장억제 효과(시료농도 0.0125%), 16.9%의 히스타민 분비저해효과와 함께, 인간유래 섬유아세포의 활성을 대조군 대비 57% 높여 주는 실험결과를 확인하였다. 최종적으로 분당 서울대병원과의 공동연구를 통해 23명의 여성여드름환자를 대상으로 한 임상평가를 실시하여 새로운 여드름 개선성분의 효능을 검증하였다. 여름에 실시된 최종임상에서 SeleMix AN이 함유된 시료는 피지분비량이 증가하고 여드름 발병율이 높아지는 계절적인 영향에도 불구하고 4주만에 특히, 염증성 병변을 대조군 대비 통계적으로 유의차 있게 감소시키는 뛰어난 여드름 개선 결과를 얻을 수 있었다.
