히스티딘으로 표지된 람다 박테리오파아지 꼬리 단백질 J와 대장균 K-12와의 결합 (Binding of the His-tagged Tail Protein J of Bacteriophage Lambda with Escherichia coli K-12)

  • 본 연구에서는 미생물 검출용 바이오센서 제작을 위해 재조합 람다 파아지 꼬리 단백질 J을 센싱 요소로 활용가능한지를 조사하였다. 융합 단백질의 입체 장애를 최소화하기 위해 J 단백질 절편의 N-말단을 6X His-tag로 융합하고 HisTALONTM 컬럼으로 정제하였다. 정제 단백질은 약 38 kDa 크기를 SDS-PAGE에서 나타내며 anti-His 단클론 항체와 반응하였다. Anti-His 단클론 항체는 6HN-J를 처리한 E. coli K-12와 특이적으로 결합하나 BSA 처리하거나 6HN-J 처리한 다른 미생물들(Salmonella typhimurium, Bacillus subtilis, Pseudomonas aeruginosa)과는 결합되지 않음을 보여주었다. 또한 정제 단백질과 숙주세포의 결합은 온전한 람다 파아지의 in vivo 숙주 표면 흡착을 약 $1{\mu}g/ml$ 단백질 농도에서 50%, $25{\mu}g/ml$ 단백질 농도에서 거의 완전히 방해하였다. 결론적으로 재조합 6HN-J 단백질은 탁월한 선택성과 선별성으로 인해 바이오센서의 제작에서 센싱 요소로 활용 가능함을 시사한다.

Expression and Purification of Intact and Functional Soybean (Glycine max) Seed Ferritin Complex in Escherichia coli

  • Soybean seed ferritin is essential for human iron supplementation and iron deficiency anemia prevention because it contains abundant bioavailable iron and is frequently consumed in the human diet. However, it is poorly understood in regards its several properties, such as iron mineralization, subunit assembly, and protein folding. To address these issues, we decided to prepare the soybean seed ferritin complex via a recombinant DNA approach. In this paper, we report a rapid and simple Escherichia coli expression system to produce the soybean seed ferritin complex. In this system, two subunits of soybean seed ferritin, H-2 and H-1, were encoded in a single plasmid, and optimal expression was achieved by additionally coexpressing a team of molecular chaperones, trigger factor and GroEL-GroES. The His-tagged ferritin complex was purified by $Ni^{2+}$ affinity chromatography, and an intact ferritin complex was obtained following His-tagged enterokinase (His-EK) digestion. The purified ferritin complex synthesized in E. coli demonstrated some reported features of its native counterpart from soybean seed, including an apparent molecular weight, multimeric assembly, and iron uptake activity. We believe that the strategy described in this paper may be of general utility in producing other recombinant plant ferritins built up from two types of subunits.

Specific bovine antibody response against a new recombinant Cryptosporidium parvum antigen containing 4 zinc-finger motifs

  • A Cryptosporidium parvum sporozoite and oocyst λgt11 cDNA library was screened with a hyperimmune rabbit serum that was developed against insoluble fragments of ultrasonicated oocysts. A clone named Cp22.4.1 encoding a protein of 231 amino acids with 4 zinc-finger domains characterized by a Cys-X2-Cys-X4-His-X4-Cys motif was isolated and characterized. There was a complete match between the sequencing data of the coding region of Cp22.4.1 and the corresponding gene at chromosomal level. Cloning in a pBAD-TOPO-TA expression vector permitted to evaluate the antigenicty of the recombinant His-tagged antigen. This antigen was recognized by 2 out of 5 sera from Cruptosporidium immune calves and not by sera from parasite naive animals.

Production and Evaluation of Monoclonal Antibodies Against Recombinant Coat Protein of Lily mottle virus for Western Blotting and Immono-blot Analysis

  • Lily mottle virus (LMoV) causes flower quality reduction in Lilium spp. The coat protein gene was RT-PCR-amplified from total RNA extracted from infected lily leaves and the amplified fragment was cloned into the pRSET expression vector tagged with a His-MBP. The plasmid of recombinant coat protein was used to transform an Escherichia coli strain pLysS and was expressed. The coat protein was purified by affinity chromatography using a Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE. The in vitro-expressed protein was used for immunization of mice. The polyclonal and monoclonal antibodies reacted specifically for the detection of LMoV in lily extracts in Western blot. Moreover the monoclonal antibodies reacted with lily extracts in DAS-ELISA with no unspecific or heterologous reactions against other non-serologically related viruses, but the polyclonal antibodies revealed a weak reaction against both infected lily and healthy control.

  • 전염성 췌장괴저바이러스 (Infectious pancreatic necrosis virus) DRT 주의 VP1유전자를 대 장균 발현운반체와 Baculovirus에 삽입하여 대장균과 진핵세로에서 VP1단백질의 발현을 연구하였다. 재조합체 pMal-pol 클론 [7]에서 2.7 Kb 단편인 VP1 유전자를 제한효소 XbaI으로 절단하여 Baculovirus 운반체인 pBacPAK9에 클로닝하여 pBacVP1이라 명명하였다. 이 pBacVP1에 클로닝된 VP1유전자를 제한효소 SacI과 PstI으로 절단하여 대장균 발현 운반체인 pQE-30에 클로닝하여 pQEVP1이라 명명하였다. 또한 VP1 단백질의 C-말단에 6개의 히스티딘 $6{\times}His$이 붙어 있는 단백질을 만들기 위하여, pQEVP1 클론의 His부위를 EcoRI으로 절단하고, 또한 pBacVP1을 EcoRI으로 절단하여 생긴 부위에His-EcoRI DNA 단편을 교체시켜 재클로닝하여 pBacHis-VP1을 만들었다. pBacHis-VP1 DNA와 Bsu36I로 처리된 LacZ-Hyphantria cunea nuclear polyhedrosis virus (LacZ-HcNPV)를 함께 lipofectin을 이용하여 곤충세포 (Spodoptera frugiperda cell)에 동시 감염을 시켜서 재조합 바이러스를 선발하여, VP1-HcNPV-1이라 명명하였다. pQEVP1 클론은 6개의 히스티딘 단편이 부착된 VP1단백질을 Ni-NTA resin 크로마토그래피법으로 정제하여 SDS-PAGE와 Western blot으로 확인하였고, 단백질의 활성과 구조에 영향을 주지 않는 6개의 히스티딘 단편 ($6\;{\times}\;His$)이 부착된 94 kDa의 VP1단백질을 정제할 수 있었다. 또한 재조합 바이러스에 감염된 곤충세포에서 VP1 단백질이 발현된 것을 전기영동과 Western blot으로 검색을 한 결과 95 kDa VP1 단백질이 발현이 되었음을 확인하였다.

TK-PTP, Protein Tyrosine Phosphatase from Hyperthermophilic Archaeon Thermococcus kadakaraensis KODI : Enzymatic Characteristics and Isolation of its Substrate Proteins

  • The Tk-ptp gene encoding a protein tyrosine phosphatase (PTPase) from the hyperthermophilic archaeon Thermococcus kodakaraensis KODI was cloned and sequenced. Sequence analysis indicated that Tk-ptp encoded a protein consisting 147 amino acid residues (16,953 Da). The wild type and the mutants were expressed in Escherichia coli cells as His-tagged fusion proteins and examined for enzyme characteristics. Tk-PTP possessed two unique features that were not found in eucaryal and bacterial counterparts. First, the recombinant Tk-PTP showed the phosphatase activity not only for the phosphotyrosine but also phosphoserine. Second, the conserved Asp (Asp-63), which was considered to be a critical residue, was not involved in catalysis. In order to know a specific substrate for Tk-PTP, C93S mutant was used to trap substrate protein. Proteins of 120, 60 and 53 kDa were isolated specifically from KODI cell lysates by affinity chromatography with Tk-PTP-C93S. It is suggested that these proteins are tyrosine-phosphorylated substrates of Tk-PTP.

Cloning and Characterization of Cycloinulooligosaccharide Fructanotransferase (CFTase) from Bacillus polymyxa MGL21

  • Microorganism producing extracellular CFTase was isolated from soil and designated as Bacillus polymyxa MGL21. The gene encoding the CFTase (cft) from B. polymyxa MGL21 was cloned and sequenced. The ORF of the cf gene was composed of 3,999 nucleotides, encoding a protein (1,333 amino acids) with a predicted molecular mass of 149,375 Da. Sequence analysis indicated that CFTase was divided into five distinct regions. CFTase contained three regions of repeat sequences at the N-terminus and C-terminus. The endo-inulinase region of homology (ERH) of CFTase was similar to that of Pseudomonas mucidolens endo-inulinase ($50\%$ identity, 259 amino acids). Furthermore, CFTase possessed a highly conserved core region, which is considered to be functional for the hydrolysis reaction of inulin. The cft gene was expressed in a His-tagged form in Escherichia coli cells, and the His-tagged CFTase was purified to homogeneity. The optimal temperature and pH for CFTase activity were found to be $50^{\circ}C$ and 9.0, respectively. The enzyme activity was completely inhibited by 10 mM $Ag^+\;and\;Cu^2+$. Thin-layer chromatography analyses indicated that CFTase catalyzed not only the cyclization reaction ut also disproportionation and hydrolysis reactions as well.

Purification and Characterization of Hrp1, a Homolog of Mouse CHD1 from the Fission Yeast Schizosaccharomyces pombe

  • Hrp1, of Schizosaccharomyces pombe, is a new member of the SW12/SNF2 protein family that contains a chromodomain and a DNA binding domain as well as ATPase/7 helicase domains. This configuration suggests that Hrp1 could be a homolog of mouse CHD1, which is thought to function in altering the chromatin structure to facilitate gene expression. To understand the enzymatic nature of Hrp1 we purified the 6-Histidine-tagged Hrp1 protein (6$\times$His-Hrp1) to homogeneity from a S. pombe Hrp1-overexpressing strain and hen examined its biochemical properties. We demonstrate that the purified 6$\times$His-Hrp1 protein exhibited a DNA-binding activity with a moderate preference to the (A+T)-rich tract in double-stranded NA via a minor groove interaction. However, we failed to detect any intrinsic DNA helicase activity from the purified Hrp1 like other SW12/SNF2 proteins. These observations suggest that the DNA binding activities of Hrp1 may be involved in the remodeling of the chromatin structure with DNA-dependent ATPase. We propose that Hrp1 may function in heterochromatins as other proteins with a chromo- or ATPase/helicase domain and play an important role in the determination of chromatin architecture.

Expression and Purification of a Functional Recombinant Aspartate Aminotransferase (AST) from Escherichia coli

  • Aspartate aminotransferase (AST; E.C. 2.6.1.1), a vitamin B6-dependent enzyme, preferentially promotes the mutual transformation of aspartate and ${\alpha}$-ketoglutarate to oxaloacetate and glutamate. It plays a key role in amino acid metabolism and has been widely recommended as a biomarker of liver and heart damage. Our study aimed to evaluate the extensive preparation of AST and its application in quality control in clinical laboratories. We describe a scheme to express and purify the 6His-AST fusion protein. An optimized sequence coding AST was synthesized and transformed into Escherichia coli BL21 (DE3) strain for protein expression. Ideally, the fusion protein has a volumetric productivity achieving 900 mg/l cultures. After affinity chromatography, the enzyme activity of purified AST reached 150,000 U/L. Commutability assessment between the engineered AST and standard AST from Roche suggested that the engineered AST was the better candidate for the reference material. Moreover, the AST showed high stability during long-term storage at $-20^{\circ}C$. In conclusion, the highly soluble 6His-tagged AST can become a convenient tool for supplying a much better and cheaper standard or reference material for the clinical laboratory.
