Anti-inflammatory, anti-arthritic and analgesic activity of each herbal extract, which is extracted from Bacopa monnieriis, Cassia fistula and Phyllanthus polyphyllus, respectively. The treatment of herbal extract exhibited anti-inflammatory effect as a dose-dependent manner, from 1.25 mg/kg to 12.5 mg/kg, in acute inflammatory models (carrageen and egg-albumin induced rat hind paw edema). It also elicited significant anti-inflammatory activity in chronic inflammatory models (cotton pellet granuloma and Freund's adjuvant induced polyarthritis in rat). In cotton pellet granuloma test, the extract exhibited the inhibitory effect of 23 and 57% at the dose of 6.25 and 12.5 mg/kg, respectively. In Freund's adjuvant induced model, the treatment of the extract of 1.25, 6.25 and 12.5 mg/kg showed the inhibitory effect of 23, 56 and 66% at 8 days, respectively. In the acetic acid-induced model, the extract significantly reduced abdominal writhing in mice when compared to the control group, reducing the mean number of writhing from $41{\pm}2$ in the control group to $17{\pm}3$ and $15{\pm}2$ at the dose of 6.25 and 12.5 mg/kg. From these experiments, the extract, which was extracted from the combination of Bacopa monnieriis, Cassia fistula and Phyllanthus polyphyllus, (w/w/w = 1/2/1) is surprisingly found a significant analgesic and anti-inflammatory activity.
In order to evaluate the improvement effects of head of Panax ginseng on chronic arthritis, we have investigated the activity of butanol fraction (BuOH fraction) in vitro and in vivo system. BuOH fraction showed significant inhibition on the elastase activity. Anti-arthritic activity of BuOH fraction was also examined on type II collagen-induced arthritis in DBA/1J mice. Mice were immunized with injection of type II collagen emulsified in Freund's complete adjuvant, followed by a booster injection 21 days later. BuOH fraction(BHPG) was administered at an oral dose of 500mg/kg for 2 weeks from the 1st day boost. The hind paw edema was significantly decreased in the group of treatment with BuOH fraction compared to control. In collagen-induced DBA/1J mice, BuOH fraction did not affected the collagen antibody titer but significantly inhibited the tumor necrosis factoralpha(TNF-${\alpha}$) activity. These results were confirmed with histological evaluation of joint tissues. This study may raise the possibility that the usage of BuOH fraction of head of Panax ginseng as alternative medicine for the relief and prevention of rheumatoid arthritis symptoms.
Some pharmacological activities of Ulmus davidiana var. japonica were evaluated using its methanol extract (UDE). An acute anti-inflammatory activity of UDE was assessed using carrageenan-induced hind paw edema in rats. UDE exhibited an antioxidant activity when assayed by a stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH). Dose-dependent anti-nociceptive activity of UDE was assessed using the acetic acid-induced writhing test in mice. UDE was able to diminish the reactive oxygen species (ROS) level in the lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. UDE also suppressed production of nitric oxide and induction of inducible nitric oxide synthase and cyclooxygenase-2 in the stimulated macrophages cells. collectively, the results imply that U. davidiana var. japonica has antioxidant and anti-nociceptive activities in addition to anti-inflammatory activity.
To develop a clinically available saponin- or sapogenin complex from Oriental medicines, the EtOH extract (KPRG-A) was obtained by extracting from the four crude drugs, Kalopanacis Cortex, Platycodi Radix, Rubi Fructus and Glycyrrhizae Radis. The BuOH fraction (KPRG-B), a crude saponin complex, was prepared by fractionating KPRG-A, which were further completely hydrolyzed to afford the sapogenin complex (KPRG-D). In an attempt to find the antinoicpetive effects of the saponin complex and sapogenin complex, KPRG-C, and -D, were assayed by writhing-, hot plate-, and tail-flick tests using mice or rats. The three samples were also subjected to antiiflammatory tests using serotonin-induced and carrageenan-induced hind paw edema mice and rats, respectively. The three samples significantly reduced inflammations and pains of the experimental animal. The potency were found in the order of KPRG-D> KPRG-C> KPRG-B. The most active sample, KPRG-D, caused no death, no body increase or no anatomical pathlogic change even at 2,000 mg/kg dose. These results suggest that a sapogenin complex, KPRG-D, which was found to contain mainly hederagenin, platycodigenin, polygalacic acid, 23-hydroxytormentic acid, glycyrrhetic acid together with minor triterpene acids, could be a potential candidate for antiinflammatory therapeutics.

The purpose of this research was to investigate the effect of Kamidangkwieumja(KDEJ) water extract on the allergy reaction in mice. The results were obtained as follows: 1. The passive cutaneous anaphylaxis induced by egg albumin in fat was not affected. 2. The lethal anaphylaxis induced by platelet activating factor in mice. was inhibited. 3. The degranulation of peritoneal mast cells induced by compound 48/80 was not affected. 4. The acute hind paw edema was inhibited after 2hours later when it was induced by histamine. 5. The permeability of evans blue into peritoneal cavity induced by acetic acid was not affected. 6. Arthus reaction in mice was not affected. 7. The delayed type hypersensitivity induced by SRBC was inhibited. 8. The contact dermatitis induced by DNFB was not affected. 9. The hemagglutination titer induced by SRBC was inhibited. 10. The writhing syndrome induced by acetic acid was inhibited. 11. The population of heper T cells in mice thymus was enhanced. 12. The phagocytic activity of peritoneal macrophages was enhanced. 13. The production of nitric oxide from peritoneal macrophages was not affected. These results suggest that the anti-allergy effect of KDEJ is caused by steroidlike and enhanced immune action. The steroidlike action of KDEJ correspond with steroid-applying-method that frequently used in clinic, so it is used io treatment of psoriasis. The research on anti-allergy of KDEJ might has to be continued.
In order to investigate the effect of 'Whangryonhaedok-Tang' on the anti-inflammatory, ileum of mice, blood pressure, blood glucose, total cholesterol and antibacterial activity, those pharmacological actions were conducted. The results of this studies were summarized as follows; Rat hind paw edema induced by acetic acid was inhibited. In mice, tone of intestinal smooth muscle was suppressed with the treatment of sample and intestinal contractions induced by $BaCl_2$ was inhibited. Hypertensive effect was not observed in rabbits but was observed by the water extract of 'Whangryonhaedok-Tang' excluded Phellodendri Cortex in rabbits. The increased blood glucose levels in alloxandiabetic rabbits were significantly decreased by the oral administration of the sample I after 6 days. The total cholesterol levels in alloxan-induced rabbits were significantly decreased by the oral administration of the sample I after 9 days. Antibacterial activity was observed. In addition, we thought the pharmacological actions were significantly noted by the sample III (Methanol soluble fraction) because of the purification of effective components.

In order to examine the anti-arthritic properties of ShinEumHur(SEH)-herbal acupuncture, an adjuvant-induced arthritic rat was generated by the intra-articular injection of dried cells of Mycobacterium tuberculosis emulsified in squalene into the right knee joint. Fifty microliter of SEH extract was injected into Zusanli(ST36) acupoint on the ipsilateral hind paw every other day for 2 weeks. The body weight, knee circumference, squeak threshold, and weight distribution ratio were analyzed as the assessment methods addressing arthritic symptoms such as arthritic pain, edema, and tenderness. The weight distribution ratio was measured by a digital-type analgesia instrument using the dual channel scale that separately measures the weight the arthritic rat distributes to each hind paw, and thus quantifies both of swelling and pain severities at once. The therapeutic effects of SEH-herbal acupuncture, assessed by squeaking threshold and weight distribution ratio, were observed on 8th day after the arthritis induction as compared to saline group and control group. On 10th day, SEH-herbal acupuncture therapy significantly started to alleviate the growing pattern of knee circumference of an arthritic rat in the range of 0.2㎝. However, the loss of body weight was not significantly recovered. Taken together, the SEH-herbal acupuncture exhibited the significant therapeutic efficiency to treat adjuvant-induced monoarthritis in rat.

Objectives : Illicium verum Hook. f. has been known to possess antimicrobial, antioxidant, antifungal, anti-inflammatory, insecticidal, analgesic, sedative, convulsive activities, it has been rarely conducted to evaluate the immuno-biological activity. The present study was examined to evaluate the anti-inflammatory effects of the Illicium verum Hook. f. water extracts (IVE) in vivo and in vitro. Methods : Cell viability was measured by MTT assay. The relative levels of NO were measured with Griess reagent. iNOS, COX-2, $NF-{\kappa}B$ and target proteins were detected by immunoblot analysis, and levels of cytokines were analyzed by ELISA kit. Anti-edema effect was determined in the carrageenan (CA)-induced paw edema model in rats. Results : All dosages of IVE used in MTT assay had no significant cytotoxicity. The increases of NO production and iNOS expression were detected in LPS-treated cells compared with control. However, these increases were attenuated by treatment with IVE. Also, IVE reduced the elevated production of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 by LPS. IVE inhibited the $p-I{\kappa}B$ and translocation of $NF-{\kappa}B$ to nuclear. Furthermore, IVE significantly inhibited the increases of hind paw swelling, skin thicknesses and inflammatory cell infiltrations induced by CA injection. Therefore, IVE will be favorably inhibited the acute edematous inflammations. Conclusion : These results provide evidences that anti-inflammatory effect of IVE is partly due to the reduction of some inflammatory mediators by suppression of $NF-{\kappa}B$ pathway.

The purpose of this study was to observe the effects of Achyranthes Radix(AR) and electroacupuncture(EA) in rats with rheumatoid arthritis induced by type II collagen for 28 days. Control group was daily administered 0.9% NaCl 0.5 $m{\ell}$, Group I was daily administered 0.9% NaCl 0.5 $m{\ell}$ to arthritic rats, Group II was orally administered with Achyranthes Radix 500 mg/kg 0.5 $m{\ell}$ to arthritic rats. Group III was given 2 Hz EA of chok samni acupoint(ST36) in the test group for 30 min/days to arthritic rats. Group IV was daily orally administered with Achyranthes Radix 500 mg/kg 0.5 $m{\ell}$ and 2 Hz EA of chok samni acupoint(ST36) in the test group for 30 min/days to arthritic rats. This studies have been designed to evaluate the hind paw edema, assessment of arthritis indices, analgetic effects by analysis of blood chemistry(WBC, CRP, ALP, AST). In each group, histologic observations, Safranin O-fast green stain were observed and analyzed. The following results were obtained. Group II, III, IV were significantly decreased arthritis indices and the rate of paw edema compared with Group I . Especially group IV was the most significantly decreased. The WBC, CRP, AST, ALT was that Group II, III, IV were significantly decreased compared with Group I . In conclusion, Achyranthes Radix and Ea contribute to the improvement of blood chemistry and change in safranin O-fast green by knee joint of arthritic rats.

This study was performed to investigate the effects in inflammatory and pain status on rheumatoid arthritis(RA) induced rats by swimming exercise and Achyranthes Radix(AR) extracts ointment according to the application methods with the change of motor-behavioral and histochemistry study through the change of safranin o-fast green stain in the knee joint and prostaglandin $E_2(PGE_2)$ concentration production in serum for 28 days. They were randomly divided into four groups; Group I: RA induction, Group II: application of only swimming exercise after RA induction, Group III: application of only AR extracts ointment after RA induction, Group IV: application of both AR extracts ointment and swimming exercise after RA induction. The following results were obtained. Volume change of hind paw edema and arthritis indices test and arthritic dorsal flexion & plantar flexion pain test, group II, III, IV were showed that significantly decrease to each scores compared with group I (p<.01). Safranin o-fast green stain were showed histological indices, group II, III, IV were showed that significantly decrease the scores of cellular infiltration and synovial hyperplasia(p<.05), pannus formation and cartilage destruction(p<.01) compared with group I. Group IV were the most decreased compared with group II, III. The $PGE_2$ concentration of the group II, III, IV were decreased compared to the group I (p<.01) In conclusion, swimming exercise and AR extracts ointment acts were the most therapeutic intervention in inflammatory and pain control of RA induced rats.
